3Structure-guided protein design of fluoroacetate dehalogenase for kinetic resolution of rac-2-bromobutyric acid显示文摘R-2-Bromobutyric acid is a very important intermediate for the synthesis of agrochemicals and pharmaceuticals.Bioresolution of rac-2-bromobutyric acid(rac-2-BBA)provides a promising process for R-2-bromobutyric acid(R-2-BBA)production.The fluoroacetate dehalogenase(FAcD)has been always studied in the defluorination process.We found that FAcD RPA1163 showed detectable activity but no enantioselectivity towards rac-2-BBA.The iterative saturation mutagenesis(ISM)of FAcD RPA1163 resulted in a mutant H155V/W156R/Y219M,which catalyzed the kinetic resolution of rac-2-BBA to produce R-2-BBA with enhanced activity and enantioselectivity(99.3%ee).The high preference for S-2-bromobutyric acid(S-2-BBA)is of synthetic value.Molecular docking analysis indicated that the H155V/W156R/Y219M mutation reduced steric hindrance and broadened the halide pocket.It is not only the steric hindrance but also the electrostatic environment that has an effect on the activity and enantioselectivity.Shu Wang Zhanbing Cheng Yanbing Xu Lu Yang Jian-Bo Wang Zhenhua Tian Xudong Qu 2020Green Synthesis and Catalysis2020,1,1:0
4Haplotype variation and KASP markers for SiPSY1-A key gene controlling yellow kernel pigmentation in foxtail millet显示文摘Carotenoid biosynthesis and accumulation are important in determining nutritional and commercial value of crop products.Yellow pigmentation of mature kernels caused by carotenoids is considered a vital quality trait in foxtail millet,an ancient and widely cultivated cereal crop across the world.Genomic regions associated with yellow pigment content(YPC),lutein and zeaxanthin in foxtail millet grains were identified by genome-wide association analysis(GWAS),and SiPSY1(Phytoene synthase 1 which regulates formation of the 40-carbon backbone of carotenoids)was confirmed as the main contributor to all three components by knockout and overexpression analysis.SiPSY1 was expressed in seedlings,leaves,panicles,and mature seeds,and was subcellularly localized to chloroplasts.Transcription of SiPSY1 in 15 DAP immature grains was responsible for YPC in mature seeds.Selection of SiPSY1 combined with increased YPC in mature grains during domestication of foxtail millet was confirmed.Haplotype analysis suggested that expression level of SiPSY1 could be a selection target for future breeding programs,and a KASP marker was developed for selection of favorable SiPSY1 alleles in breeding.The results of this work will benefit nutritional and commercial improvement of foxtail millet varieties,as well as other cereal crops.Rongjun Zuo Yanyan Zhang Yanbing Yang Chunfang Wang Hui Zhi Linlin Zhang Sha Tang Yanan Guan Shunguo Li Ruhong Cheng Zhonglin Shang Guanqing Jia Xianmin Diao 2023The Crop Journal2023,11,6:0

6Self-assembled ternary hybrid nanodrugs for overcoming tumor resistance and metastasis显示文摘Traditional chemotherapy exhibits a certain therapeutic effect toward malignant cancer,but easily induce tumor multidrug resistance(MDR),thereby resulting in the progress of tumor recurrence or metastasis.In this work,we deigned ternary hybrid nanodrugs(PEI/DOX@CXB-NPs)to simultaneously combat against tumor MDR and metastasis.In vitro results demonstrate this hybrid nanodrugs could efficiently increase cellular uptake at pH 6.8 by the charge reversal,break lysosomal sequestration by the proton sponge effect and trigger drugs release by intracellular GSH,eventually leading to higher drugs accumulation and cell-killing in drug-sensitive/resistant cells.In vivo evaluation revealed that this nanodrugs could significantly inhibit MDR tumor growth and simultaneously prevent A549 tumor liver/lung metastasis owing to the specifically drugs accumulation.Mechanism studies further verified that hybrid nanodrugs were capable of down-regulating the expression of MDR or metastasis-associated proteins,lead to the enhanced anti-MDR and anti-metastasis effect.As a result,the multiple combination strategy provided an option for effective cancer treatment,which could be potentially extended to other therapeutic agents or further use in clinical test.Xu Cheng Dapeng Li Jiaxi Xu Bing Wei Qin Fang Longshun Yang Yanbing Xue Xin Wang Rupei Tang 2021Acta Pharmaceutica Sinica B2021,11,11:0
