1Shock Waves Increase T-cell Proliferation or IL-2 Expression by Activating p38 MAP Kinase显示文摘Shock waves were elicited by transient pressure disturbances, which could be used to treat musculoskeletal disorders. In present studies, we investigated whether the low-density shock waves (LDSWs),which are able to damage plasma membrane without impairing the vimentin or other organelles, might augment T-cell proliferation as well as IL-2 expression, and if mitogen activated protein kinase p38 (p38 MAPK) might be an underlying mechanism through which the LDSWs enhanced T-cell function. We found that the LDSWs increased activation of p38 MAPK in Jurkat T cells. The LDSWs alone didn't result in the T-cell proliferation and IL-2 expression. However, in combination with other stimuli, LDSWs could augment the T-cell proliferation and IL-2 expression. Inhibition of p38 MAPK using SB203580 reduced the stimulatory effects of the LDSWs, which indicated that the LDSWs enhanced IL-2 expression through a mechanism that involved p38 MAPK activation. 2Carboxyfluorescein Diacetate Succinimidyl Ester Fluorescent Dye for Cell Labeling显示文摘Our objective was to study the properties of the carboxyfluorescein diacetate succinimidyl ester (CFDA-SE) and the methodology of cell labeling using CFDA-SE fluorescent dye. First, we analyzed the kinetics of CFDA-SE fluorescent dye intensity over time. Second, we determined the optimal concentration of CFDA-SE fluorescent dye for cell labeling. Third, we tested the toxicity of CFDA-SE fluorescent dye on labeled cells. Finally, we determined the optimal staining time of CFDA-SE fluorescent dye for cell labeling.The results show that the optimal concentration of CFDA-SE fluorescent dye for cell labeling varies according to different cell types. CFDA-SE fluorescent dye is non-toxic to cells as the cell death rate caused by CFDASE labeling is below 5%. The optimal cell labeling time was determined to be 8 min of incubation with CFDA-SE fluorescent dye. We concluded that the advantages of using CFDA-SE fluorescent dye for cell labeling are as follows: (1) the binding of CFDA-SE fluorescent dye to cells is stable; (2) CFDA-SE fluorescent dye is not toxic and does not modify the viability of labeled cells; and (3) CFDA-SE fluorescent dye is a suitable fluorochrome for cell labeling.Xiao-QiWANG Xiu-MeiDUAN Li-HuaLIU Yan-QiuFANG YanTAN 2005Acta Biochimica et Biophysica Sinica2005,37,6:8

4Anti-oxidative effect of zinc in human umbilical cord mesenchymal stem cells显示文摘Human umbilical cord mesenchymal stem cells(HUC-MSCs)are pluripotent and functional in many biological processes,by which releasing secretary factors to promote the self-repairing of damaged tissue or developing into functional cell at local organ.However,there is a high risk that oxidative stress would reduce the pluripotency and factor-secretion during the preparation and transplantation.Therefore,reducing oxidative stress is expected to improve the efficacy of HUC-MSCs therapy.Zinc[Zn] is an essential trace element which involves in the resistance of oxidative stress.To investigate Zn-regulated signaling pathways,we have profiled the gene expression at transcriptome level in primary HUC-MSCs treated with zinc sulfate,followed with GO and KEGG gene enrichment analysis.Zn treatment improved signal pathways for mineral absorption,cell growth,and cell death.Zn deficiency was mimicked by TPEN administration,which suppressed cell proliferation and reduced the expression of HUC-MSCs surface stem cell markers CD73,CD90 and CD105 by flow cytometry.Nuclear factor erythrocyte 2 related factor 2(Nrf2)plays an important role in antioxidant biological processes.In vitro treatment of Zn significantly increased Nrf2 and Sirt3 expression at gene level and protein level respectively.Zn supplementation inhibited TPEN-induced failure of cell survival and reversed the reduction of Nrf2 and Sirt3 expression,which further reduced the production of ROS.Zn successfully presented its antioxidation effect by activating Nrf2/Sirt3 signaling pathway in HUC-MSCs.Zn supplementation may improve the efficacy of HUC-MSCs therapy with reduced oxidative stress.Xiaodan Lu Yifan Lin Xiuying Lin Qiang Zhang Zihang Wang Xuguang Mi Ruobing Wang Xiaofang Zhang Xu Luan Yan Liu Bing Li YanTan Yanqiu Fang 2021Biophysics Reports2021,7,2:0
