| 1 | Relationship between the imaging features and pathologic alteration in hepatoma of rats显示文摘AIM: The imaging features of MRI and DSA, using the modelsof implanted and induced hepatoma, were investigated in rats.METHODS: CBRH3 cancer cells were implanted for differentliver site of rat liver and the diethylnitrosoamine was givenorally to rats in order to induce liver cancer. Both experimentalgroups were detected by magnetic resonance imaging (MRI),digital subtraction angiography (DSA) and morphologic assay.RESULTS: Hypointensity on T1WI and homogenous highsignal intensity on T2WI in MRI, and ring-like abnormal stainon DSA were found in implanted cancer. Induced cancersappeared as homogeneous or heterogeneous hypointensityon T1WI (10 cases), and equal or slight high intensity onT2WI (8 cases), but some as hypointensity on T2WI (2 cases).CONCLUSION: The imaging features of implanted cancerswere similar to that of human liver metastases. Therefore, itcould serve as an experimental model of human liver metastatictumor. The imaging feature of induced cancers, whereas, weresimilar to that of human primary liver cancer. | 2 | Analysis of gene expression profile of pancreatic carcinoma using cDNA microarray显示文摘AIM: To identify new diagnostic markers and drug targets,the gene expression profiles of pancreatic cancer were compared with that of adjacent normal tissues utilizing cDNA microarray analysis.METHODS: cDNA probes were prepared by labeling mRNA from samples of six pancreatic carcinoma tissues with Cy5dUTP and mRNA from adjacent normal tissues with Cy3dUTP respectively through reverse transcription. The mixed probes of each sample were then hybridized with 12 800cDNA arrays (12 648 unique human cDNA sequences), and the fluorescent signals were scanned by ScanArray 3 000scanner (General Scanning, Inc.). The values of CyS-dUTP and Cy3-dUTP on each spot were analyzed and calculated by ImaGene 3.0 software (BioDiscovery, Inc.). Differentially expressed genes were screened according to the criterion that the absolute value of natural logarithm of the ratio of Cy5-dUTP to Cy3-dUTP was greater-than 0.69.RESETS: Among 6 samples investigated, 301 genes, which accounted for 2.38% of genes on the microarry slides,exhibited differentially expression at least in 5. | 3 | Effects of Changtai granules, a traditional compound Chinese medicine, on chronic trinitrobenzene sulfonic acid-induced colitis in rats显示文摘AIM: To study the effects of Changtai granules (CTG), a traditional compound Chinese medicine, on chronic trinitrobenzene sulfonic acid-induced colitis in rats. METHODS: Healthy adult Sprague-Dawley (SD) rats of both sexes, weighing 250-300 g, were employed in the present study. The rat colitis models were induced by 2, 4,6-trinitrobenzene sulfonic acid (TNBS) enemas at a concentration of 100 mg/kg in 50% ethanol. The experimental animals were randomly divided into dexamethasone (DX) treatment, CTG treatment, and model control groups, which were intracolicly treated daily with DX (0.2 mg/kg), CTG at doses of 2.9, 5.7 and 11.4 g crude drug/kg, and the equal amount of saline respectively from 6 h following induction of the colitis in rats inflicted with TNBS to the end of study. A normal control group of rats treated without TNBS but saline enema was also included in the study. After 3 wk of treatment, the animals were assessed for colonal inflammatory and ulcerative responses with respect to mortality, frequency of diarrhea, histology and myeloperoxidase activity (MPO).RESULTS: The therapeutic effect of CTG on ulcerative colitis (UC) was better than DX. CTG effectively inhibited the activity of granulocytes, macrophages and monocytes in a dosedependent manner. Also it reduced MPO and formation of inflammation in colonic mucosal tissue.
