1Exosome and Exosomal MicroRNA: Trafficking, Sorting, and Function显示文摘Exosomes are 40–100 nm nano-sized vesicles that are released from many cell types into the extracellular space. Such vesicles are widely distributed in various body fluids. Recently,m RNAs and micro RNAs(mi RNAs) have been identified in exosomes, which can be taken up by neighboring or distant cells and subsequently modulate recipient cells. This suggests an active sorting mechanism of exosomal mi RNAs, since the mi RNA profiles of exosomes may differ from those of the parent cells. Exosomal mi RNAs play an important role in disease progression, and can stimulate angiogenesis and facilitate metastasis in cancers. 4TNF-α suppresses sweat gland differentiation of MSCs by reducing FTO-mediated m^6 A-demethylation of Nanog mRNA显示文摘An effect of inhibition of tumor necrosis factor-α(TNF-α)on differentiation of mesenchymal stromal cells(MSCs)has been demonstrated,but the exact mechanisms that govern MSCs differentiation remain to be further elucidated.Here,we show that TNF-αinhibits the differentiation of MSCs to sweat glands in a specific sweat gland-inducing environment,accompanied with reduced expression of Nanog,a core pluripotency factor.We elucidated that fat mass and obesity-associated protein(FTO)-mediated m^6 A demethylation is involved in the regulation of MSCs differentiation potential.Exposure of MSCs to TNF-αreduced expression of FTO,which demethylated Nanog m RNA.Reduced expression of FTO increased Nanog m RNA methylation,decreased Nanog m RNA and protein expression,and significantly inhibited MSCs capacity for differentiation to sweat gland cells.Our finding is the first to elucidate the functional importance of m^6 A modification in MSCs,providing new insights that the microenvironment can regulate the multipotency of MSCs at the post-transcriptional level.Moreover,to maintain differentiation capacity of MSCs by regulating m^6 A modification suggested a novel potential therapeutic target for stem cellmediated regenerative medicine.Yihui Wang Rui Wang Bin Yao Tian Hu Zhao Li Yufan Liu Xiaoli Cui Liuhanghang Cheng Wei Song Sha Huang Xiaobing Fu 2020Science China(Life Sciences)2020,63,1:9
5Premature Tapetum Degeneration: a Major Cause of Abortive Pollen Development in Photoperiod Sensitive Genic Male Sterility in Rice显示文摘Photoperiod-sensitive genic male-sterile (PSGMS) rice (Oryza sativa L.), a natural mutant found in the rice cultivar Nongken 58, is very useful for the development of hybrid rice cultivars. Despite its widespread use in breeding programs, the initial stage of the abortive development of PSGMS rice and the possible cytological mechanisms of pollen abortion have not been determined. In the present study, a systematic cytological comparison of the anther development of PSGMS rice with its normal fertile counterpart is conducted. The results show that pollen abortion in PSGMS rice first occurs before the pollen mother cell (PMC) stage, and continues during the entire process of pollen development until pollen degradation. The abortive process was closely associated with the abnormal behavior of the tapetum. Although tapetum degeneration in PSGMS rice initiates already at the PMC stage, it proceeds slowly and does not complete until the breakdown of the pollen. 6Air-sea heat flux exchange over the South China Sea under different weather conditions before and after southwest monsoon onset in 2000显示文摘With the data observed from the Second SCS Air-Sea Flux Experiment on the Xisha air-sea flux research tower, the radiation budget, latent, sensible heat fluxes and net oceanic heat budgetswere caculated before and after summer monsoon onset. It is discovered that, after summer monsoononset, there are considerable changes in air-sea fluxes, especially in latent heat fluxes and net oceanicheat budget. Furthermre, the analyzed results of five synoptic stages are compared. And the character-istics of the flux transfer during different stages around onset of South China Sea monsoon are discussed.The flux change shows that there is an oceanic heat accumulating process during the pre-onset and thebreak period, as same as oceanic heat losing process during the onset period. Moreover, latent fluxes,the water vapor moving to the continent, even the rainfall appearance in Chinese Mainland also can beinfluenced by southwester. Comparing Xisha fluxes with those obtained from the Indian Ocean and thewestern Pacific Ocean, their differences may be obeerved. 8Stratigraphy of the Triassic-Jurassic Boundary Successions of the Southern Margin of the Junggar Basin,Northwestern China显示文摘The Triassic-Jurassic(Tr-J) boundary marks a major extinction event,which(~200 Ma) resulted in global extinctions of fauna and flora both in the marine and terrestrial realms.There prevail great challenges in determining the exact location of the terrestrial Tr-J boundary,because of endemism of taxa and the scarcity of fossils in terrestrial settings leading to difficulties in linking marine and terrestrial sedimentary successions.Investigation based on palynology and bivalves has been carried out over a 1113 m thick section,which is subdivided into 132 beds,along the Haojiagou valley on the southern margin of the Junggar Basin of the northern Xinjiang,northwestern China.The terrestrial Lower Jurassic is conformably resting on the Upper Triassic strata.The Upper Triassic covers the Huangshanjie Formation overlaid by the Haojiagou Formation,while the Lower Jurassic comprises the Badaowan Formation followed by the Sangonghe Formation.Fifty six pollen and spore taxa and one algal taxon were identified from the sediments.Based on the key-species and abundance of spores and pollen,three zones were erected:the Late Triassic(Rhaetian) Aratrisporites-Alisporites Assemblage,the Early Jurassic(Hettangian) Perinopollenites-Pinuspollenites Assemblage,and the Sinemurian Perinopollenites-Cycadopites Assemblage.The Tr-J boundary is placed between bed 44 and 45 coincident with the boundary between the Haojiagou and Badaowan formations.Beds with Ferganoconcha(?),Unio-Ferganoconcha and Waagenoperna-Yananoconcha bivalve assemblages are recognized.The Ferganoconcha(?) bed is limited to the upper Haojiagou Formation, Unio-Ferganoconcha and Waagenoperna-Yananoconcha assemblages are present in the middle and upper members of the Badaowan Formation.The sedimentary succession is interpreted as terrestrial with two mainly lake deposit intervals within Haojiagou and Badaowan formations,yielding fresh water algae and bivalves.However,the presence of brackish water algae Tasmanites and the marine-littoral facies bivalve Waagenoperna from the Badaowan Formation indicate that the Junggar Basin was influenced by sea water caused by transgressions from the northern Tethys,during the Sinemurian.SHA Jingeng Vivi VAJDA PAN Yanhong Linda LARSSON YAO Xiaogang ZHANG Xiaolin WANG Yaqiong CHENG Xiansheng JIANG Baoyu DENG Shenghui CHEN Siwei PENG Bo 2011Acta Geologica Sinica(English Edition)2011,85,2:7

10High performance hydroxyapatite ceramics and a triply periodic minimum surface structure fabricated by digital light processing 3D printing显示文摘High performance hydroxyapatite(HA)ceramics with excellent densification and mechanical properties were successfully fabricated by digital light processing(DLP)three-dimensional(3D)printing technology.It was found that the sintering atmosphere of wet C02 can dramatically improve the densification process and thus lead to better mechanical properties.HA ceramics with a relative density of 97.12%and a three-point bending strength of 92.4 MPa can be achieved at a sintering temperature of 1300℃,which makes a solid foundation for application in bone engineering.Furthermore,a relatively high compressive strength of 4.09 MPa can be also achieved for a DLP-printed p-cell triply periodic minimum surface(TPMS)structure with a porosity of 74%,which meets the requirement of cancellous bone substitutes.A further cell proliferation test demonstrated that the sintering atmosphere of wet CO2 led to improve cell vitality after 7 days of cell culture Moreover,with the possible benefit from the bio-inspired structure,the 3D-printed TPMS structure significantly improved the cell vitality,which is crucial for early osteogenesis and osteointegration.Yongxia YAO Wei QIN Bohang XING Na SHA Ting JIAO Zhe ZHAO 2021Journal of Advanced Ceramics2021,10,1:5
11Bushen Qiangjin capsule(补肾强筋胶囊) inhibits the Wnt/β-catenin pathway to ameliorate papain-induced knee osteoarthritis in rats显示文摘OBJECTIVE:To evaluate the molecular mechanism underlying the beneficial effect of Bushen Qiangjin capsule(补肾强筋胶囊,BSQJ),a Traditional Chinese Medicine,on knee osteoarthritis(KOA).METHODS:In the present study,32 female Sprague-Dawley rats were randomly divided into four groups:control,KOA,high-dose BSQJ(H-BSQJ),and low-dose BSQJ(L-BSQJ).After successfully establishing the KOA model by intra-articular injection of papain,H-BSQJ and L-BSQJ groups were intragastrically administered 0.243 and 0.122 g/kg BSQJ,respectively,daily for 6 weeks.At the end of the experiment,knee articular cartilage tissues of rats were collected for evaluation by hematoxylin and eosin staining,Safranin O-Fast Green staining,and terminal deoxynucleotidyl transferase-mediated d UTP nick-end labeling assay.Serum interleukin-1βand tumor necrosis factor-αlevels of rats were detected with an enzyme-linked immunosorbent assay method.Gene expression of Wnt-4,β-catenin,Frizzled-2,glycogen synthase kinase-3β(GSK-3β),cysteinyl aspartate-specific proteinases 3 and 9(caspases 3 and 9),collagen typeⅡalpha 1(Col2 a1),and matrix metalloproteinases 1 and 13(MMP-1 and MMP-3)of rat knee articular cartilage was quantified by reverse transcription-quantitative polymerase chain reaction analysis.Wnt-4,β-catenin,Frizzled-2,GSK-3β,cleaved caspase-3,and cleaved caspase-9 protein expression in rat knee articular cartilage was determined by western blot analysis.RESULTS:BSQJ obviously reduced pathological damage and matrix degradation of articular cartilage in KOA rats.Compared with the KOA group,H-BSQJ rats exhibited downregulated m RNA and protein expression of Wnt-4,β-catenin,Frizzled-2,and caspase-3,as well as upregulated m RNA and protein expression of GSK-3β.In addition,H-BSQJ significantly increased m RNA expression of Col2 a1 and decreased m RNA expression of MMP-1 and MMP-13.CONCLUSION:BSQJ exerted a beneficial effect on KOA by a mechanism involving downregulation of the Wnt/β-catenin pathway,which inhibited both cartilage extracellular matrix degradation and chondrocyte apoptosis to ameliorate KOA in rats.YAO Nan CHEN Guocai LU Yanyan XU Xuemeng ZHAO Chuanxi HUANG Xuejun LIU Wengang PENG Sha WU Huai 2021Journal of Traditional Chinese Medicine2021,41,6:5
12Redirecting differentiation of mammary progenitor cells by 3D bioprinted sweat gland microenvironment显示文摘Background:Mammary progenitor cells(MPCs)maintain their reproductive potency through life,and their specific microenvironments exert a deterministic control over these cells.MPCs provides one kind of ideal tools for studying engineered microenvironmental influence because of its accessibility and continually undergoes postnatal developmental changes.The aim of our study is to explore the critical role of the engineered sweat gland(SG)microenvironment in reprogramming MPCs into functional SG cells.Methods:We have utilized a three-dimensional(3D)SG microenvironment composed of gelatin-alginate hydrogels and components from mouse SG extracellular matrix(SG-ECM)proteins to reroute the differentiation of MPCs to study the functions of this microenvironment.MPCs were encapsulated into the artificial SG microenvironment and were printed into a 3D cell-laden construct.The expression of specific markers at the protein and gene levels was detected after cultured 14 days.Results:Compared with the control group,immunofluorescence and gene expression assay demonstrated that MPCs encapsulated in the bioprinted 3D-SG microenvironment could significantly express the functional marker of mouse SG,sodium/potassium channel protein ATP1a1,and tend to express the specific marker of luminal epithelial cells,keratin-8.When the Shh pathway is inhibited,the expression of SG-associated proteins in MPCs under the same induction environment is significantly reduced.Conclusions:Our evidence proved the ability of differentiated mouse MPCs to regenerate SG cells by engineered SG microenvironment in vitro and Shh pathway was found to be correlated with the changes in the differentiation.These results provide insights into regeneration of damaged SG by MPCs and the role of the engineered microenvironment in reprogramming cell fate.Rui Wang Yihui Wang Bin Yao Tian Hu Zhao Li Yufan Liu Xiaoli Cui Liuhanghang Cheng Wei Song Sha Huang Xiaobing Fu 2019Burns & Trauma2019,7,1:5
13Consensus on the digestive endoscopic tunnel technique显示文摘With the digestive endoscopic tunnel technique(DETT), many diseases that previously would have been treated by surgery are now endoscopically curable by establishing a submucosal tunnel between the mucosa and muscularis propria(MP). Through the tunnel, endoscopic diagnosis or treatment is performed for lesions in the mucosa, in the MP, and even outside the gastrointestinal(GI) tract.At present, the tunnel technique application range covers the following:(1)Treatment of lesions originating from the mucosal layer, e.g., endoscopic submucosal tunnel dissection for oesophageal large or circular early-stage cancer or precancerosis;(2) treatment of lesions from the MP layer, per-oral endoscopic myotomy, submucosal tunnelling endoscopic resection, etc.; and(3) diagnosis and treatment of lesions outside the GI tract, such as resection of lymph nodes and benign tumour excision in the mediastinum or abdominal cavity. With the increasing number of DETTs performed worldwide, endoscopic tunnel therapeutics, which is based on DETT, has been gradually developed and optimized. However, there is not yet an expert consensus on DETT to regulate its indications, contraindications, surgical procedure, and postoperative treatment.The International DETT Alliance signed up this consensus to standardize the procedures of DETT. 14An Arabidopsis thaliana (Ler) inbred line AFDL exhibiting abnormal flower development mainly caused by reduced AP1 expression显示文摘The genetic network controlling flowering and flower development consists of a set of floral integrator genes that play a role in light sensing,hormone signaling and developmental pathways.These integrators activate the expression of meristem identity genes LEAFY (LFY) and APETALA1 (AP1) to initiate the flowering transition.However,how the expression of key genes,such as AP1,responds to diverse signals during flower development remains largely unknown.Here,we report that an Arabidopsis abnormal flower development inbred line (AFDL) exhibits a phenotype similar to the ap1 mutant,with delayed flowering time and a high frequency of transition of flower meristems into inflorescence meristems after flowering.The flower organs with an abnormal first whorl lack the second whorl and the increased number of inflorescences at the first-and second-whorl positions most closely resembled the phenotypes of ap1/cal double mutants.Interestingly,both normal and abnormal flowers coexisted in a single individual.Microarray and quantitative real-time PCR analysis revealed that the expression of AP1 was significantly reduced,while the expression of its interacting genes TERMINAL FLOWER 1 (TFL1),SHORT VEGETATIVE PHASE (SVP),AGAMOUS-like 24 (AGL24),SEPALLATA (SEP) and CAULIFLOWER (CAL) and upstream genes FLOWERING LOCUS C (FLC) and FLM were increased in AFDL,which could serve to explain its phenotype.The expression of genes responsive to different stimuli dramatically changed in AFDL relative to the wild type,as revealed by the differential display of transcripts,indicating that this expression variation is subject to a threshold,leading to an on/off expression pattern of the master regulatory gene (such as AP1) of flower development.QI XiaoLi JIANG Yao TANG Fang WANG MinJie HU JianJun ZHAO ShuTang SHA Wei LU MengZhu 2011Chinese Science Bulletin2011,56,1:4

16A novel architecture of electro-hydrostatic actuator with digital distribution显示文摘To address the flow imbalance rapidly of the asymmetric Electro-Hydrostatic Actuator(EHA),this paper presents a novel architecture of asymmetric EHA with the Digital Distribution(EHA-DD).It also improves the flow nonlinearity and control accuracy of the pump,especially in the low-speed condition.The digital distribution with two High-Speed on–off Valves(HSVs)not only balances flow instead of the check valves,but also replaces the pump at the low-speed to control output flow by adjusting the PWM signal.The pump and HSVs are the crucial components to control flow output.Firstly,the flow calculation models of the pump and the duty ratio inversed model of the HSV are obtained through experimental tests and the identification method.To get the control input signal for the required flow,the pump speed and PWM duty ratio for the HSVs are inversed to compensate for flow output.Further,a multimode digital flow distribution control method based on pump speed,mainly including the pump-controlled mode for large flow demand and valve-controlled mode for little flow demand,is proposed to control the accurate flow output actively.The step extension experiments based on the flow calculation models are conducted on the EHA-DD prototype under elastic,opposite varying load.The results demonstrate that the EHADD realizes little position error by accurate flow control,and it is also beneficial to improve the service life of the pump.Jing YAO Pei WANG Zhaosheng DONG Dongting JIANG Tong SHA 2021Chinese Journal of Aeronautics2021,34,5:3
17Tailoring bioinks of extrusion-based bioprinting for cutaneous wound healing显示文摘Extrusion-based bioprinting (EBB) holds potential for regenerative medicine. However, the widely-used bioinks of EBB exhibit some limitations for skin regeneration, such as unsatisfactory bio-physical (i.e., mechanical, structural, biodegradable) properties and compromised cellular compatibilities, and the EBB-based bioinks with therapeutic effects targeting cutaneous wounds still remain largely undiscussed. In this review, the printability considerations for skin bioprinting were discussed. Then, current strategies for improving the physical properties of bioinks and for reinforcing bioinks in EBB approaches were introduced, respectively. Notably, we highlighted the applications and effects of current EBB-based bioinks on wound healing, wound scar formation, vasculari-zation and the regeneration of skin appendages (i.e., sweat glands and hair follicles) and discussed the challenges and future perspectives. 183D-printable supramolecular hydrogels with shear-thinning property: fabricating strength tunable bioink via dual crosslinking显示文摘3-dimensional(3D)bioprinting technology provides promising strategy in the fabrication of artificial tissues and organs.As the fundamental element in bioprinting process,preparation of bioink with ideal mechanical properties without sacrifice of biocompatibility is a great challenge.In this study,a supramolecular hydrogel-based bioink is prepared by polyethylene glycol(PEG)grafted chitosan,α-cyclodextrin(α-CD)and gelatin.It has a primary crosslinking structure through the aggregation of the pseudo-polyrotaxane-like side chains,which are formed from the host-guest interactions betweenα-CD and PEG side chain.Apparent viscosity measurement shows the shear-shinning property of this bioink,which might be due to the reversibility of the physical crosslinking.Moreover,withβ-glycerophosphate at different concentrations as the secondary crosslinking agent,the printed constructs demonstrate different Young's modulus(p<0.001).They could also maintain the Young's modulus in cell culture condition for at least 21 days(p<0.05).By co-culturing each component with fibroblasts,CCK-8 assay demonstrate cellular viability is higher than 80%.After bioprinting and culturing,immunofluorescence staining with quantification indicate the expression of Ki-67,Paxillin,and N-cadherin is higher in day 14 than those in day 3(p<0.05).Oil red O and Nissl body specific staining reflect strength tunable bioink may have impact on the cell fate of mesenchymal stem cells(p<0.05).This work might provide new idea for advanced bioink in the application of re-establishing complicated tissues and organs.Tian Hu Xiaoliang Cui Meng Zhu Man Wu Ye Tian Bin Yao Wei Song Zhongwei Niu Sha Huang Xiaobing Fu 2020Bioactive Materials2020,5,4:3
19Clinical practice guidelines for treating headache with Traditional Chinese Medicine: quality assessment with the appraisal of guidelines for research and evaluation Ⅱ instrument显示文摘OBJECTIVE: To critically appraise the methodological quality of clinical practice guidelines for headache produced over the last two decades, including those covering specific interventions using Traditional Chinese Medicine.METHODS: The guidelines on headache disorders were obtained by searching a number of databases, including Pub Med, EMBASE, Web of Science,Chinese Biomedical Literature Database, China National Knowledge Infrastructure Database, China Science and Technology Journal Database, and Wanfang database, three guideline-related databases [Guideline-International Network, National Guideline Clearinghouse, and Medlive], and the records of organizations that develop guidelines. The publication date was limited to the period from January 1996 to June 2015. The search terms 'headache', 'headache disorders', 'cephalalgia', 'migraine', 'tension-type headache', 'practice guideline', 'consensus ', 'statement', 'regulation', and 'recommendation' were used in the 'Me SH' and 'Free-text' fields. The guidelines were independently appraised by four researchers using the Appraisal of Guidelines for Research and Evaluation Ⅱ instrument.RESULTS: A total of 23 guidelines published between 1998 and 2014 were reviewed. The overall consistency of the four appraisers was good [interclass correlation coefficient 0.84; 95% confidence interval(CI) 0.82-0.86]. The mean(standard deviation) scores for scope and purpose, stakeholder involvement, rigor of development, clarity of presentation, applicability, and editorial independence were 52.1(18.0), 39.5(17.1), 33.4(21.0), 49.8(21.9),23.8(19.3), and 24.2(23.7). Only two guidelines were recommended, 12 were recommended with modification, and nine were not recommended.CONCLUSION: Physical Traditional Chinese Medicine therapies were recommended to treat headache. The overall quality of headache guidelines was low in China, but evidence-based guidelines are gradually becoming mainstream.
