| 1 | Temozolomide resistance in glioblastoma multiforme显示文摘Temozolomide(TMZ)is an oral alkylating agent used to treat glioblastoma multiforme(GBM)and astrocytomas.However,at least 50%of TMZ treated patients do not respond to TMZ.This is due primarily to the over-expression of O6-methylguanine methyltransferase(MGMT)and/or lack of a DNA repair pathway in GBM cells.Multiple GBM cell lines are known to contain TMZ resistant cells and several acquired TMZ resistant GBM cell lines have been developed for use in experiments designed to define the mechanism of TMZ resistance and the testing of potential therapeutics.However,the characteristics of intrinsic and adaptive TMZ resistant GBM cells have not been systemically compared.This article reviews the characteristics and mechanisms of TMZ resistance in natural and adapted TMZ resistant GBM cell lines.It also summarizes potential treatment options for TMZ resistant GBMs. | Sang Y.Lee | 2016 | Genes & Diseases2016,3,3: | 26 |

| 4 | Real-time forecasts of the COVID-19 epidemic in China from February 5th to February 24th,2020显示文摘The initial cluster of severe pneumonia cases that triggered the COVID-19 epidemic was identified inWuhan,China in December 2019.While early cases of the disease were linked to a wet market,human-to-human transmission has driven the rapid spread of the virus throughout China.The Chinese government has implemented containment strategies of city-wide lockdowns,screening at airports and train stations,and isolation of suspected patients;however,the cumulative case count keeps growing every day.The ongoing outbreak presents a challenge for modelers,as limited data are available on the early growth trajectory,and the epidemiological characteristics of the novel coronavirus are yet to be fully elucidated.We use phenomenological models that have been validated during previous outbreaks to generate and assess short-term forecasts of the cumulative number of confirmed reported cases in Hubei province,the epicenter of the epidemic,and for the overall trajectory in China,excluding the province of Hubei.We collect daily reported cumulative confirmed cases for the 2019-nCoV outbreak for each Chinese province from the National Health Commission of China.Here,we provide 5,10,and 15 day forecasts for five consecutive days,February 5th through February 9th,with quantified uncertainty based on a generalized logistic growth model,the Richards growth model,and a sub-epidemic wave model.Our most recent forecasts reported here,based on data up until February 9,2020,largely agree across the three models presented and suggest an average range of 7409e7496 additional confirmed cases in Hubei and 1128e1929 additional cases in other provinces within the next five days.Models also predict an average total cumulative case count between 37,415 and 38,028 in Hubei and 11,588e13,499 in other provinces by February 24,2020.Mean estimates and uncertainty bounds for both Hubei and other provinces have remained relatively stable in the last three reporting dates(February 7th e 9th).We also observe that each of the models predicts that the epidemic has reached saturation in both Hubei and other provinces.Our findings suggest that the containment strategies implemented in China are successfully reducing transmission and that the epidemic growth has slowed in recent days. | K.Roosa Y.Lee R.Luo A.Kirpich R.Rothenberg J.M.Hyman P.Yan G.Chowell | 2020 | Infectious Disease Modelling2020,5,1: | 12 |

| 7 | Tissue distribution and tumor uptake of folate receptor–targeted epothilone folate conjugate,BMS-753493,in CD2F1 mice after systemic administration显示文摘To assess targeting of an epothilone folate conjugate(BMS-753493) to the folate receptor(FR)-overexpressed tumor in mice bearing both FRt and FR– tumors,a series of experiments were conducted by quantitative whole-body autoradiography(QWBA) and LC–MS/MS following i.v.administration of BMS-753493 or its active moiety,BMS-748285 in mice bearing FRt(98M109) and FR–(M109) tumors.QWBA showed [3H]BMS-753493–derived radioactivity was extensively distributed to various tissues.The FR over-expressing 98M109 tumors showed consistently higher level of radioactivity than FR-negative tumors(i.e.,M109 tumors) up to 48 h post dose of [3H]BMS-753493,despite the magnitude of difference between the tumors is relatively small(generally 3 5-fold).The radioactivity level in 98M109 tumors was 2 12-fold of normal tissues except intestine/content at 48 h post dose.No selective radioactivity uptake into 98M109 tumors over M109 or normal tissues was observed after i.v.administration of the active epothilone,[3H]BMS-748285.LC–MS/MS measurements demonstrated that the concentrations of BMS-748285,presumably from hydrolysis of the folate conjugate,in 98M109 tumors were greater than those in M109 tumors after i.v.administration of BMS-753493(2–3-fold) whereas no differential uptake in the tumors following BMS-748285 administration.Those data were consistent with radioactivity determinations.Those results demonstrated that the folate conjugation in BMS-753493 enabled moderately preferential distribution of the active epothilone to FR overexpressing 98M109 tumors,thereby supporting targeted delivery of cytotoxics through the folate receptor. | Hong Shen Lifei Wang Weiqi Chen Krista Menard Yang Hong Yuan Tian Samuel J.Bonacorsi W.Griffith Humphreys Francis Y.Lee Jinping Gan | 2016 | Acta Pharmaceutica Sinica B2016,6,5: | 3 |
| 8 | Effect of coating material and lubricant on forming force and surface defects in wire drawing process显示文摘A pilot wire drawing machine as well as wire end-pointing roller was developed. Using these machines, a wire drawing test for four different coating materials and two different lubricants was performed as the reduction ratio increased from 10% to 30%. Materials used for a substrate in this study are plain carbon steel (AISI1045) and ultra low carbon bainite steel. To compute the friction coefficient between the coating layer of wire and the surface of die for a specific lubricant, a series of finite element analyses were carried out. SEM observations were also conducted to investigate the surface defects of wire deformed. Results show that the behavior of drawing force varies with the lubricant-type at the initial stage of drawing. The powder-typed lubricant with a large particle causes the retardation of full lubrication on the entire contact surface and the local delamination of coating layer on the wire surface. | 11 | Experimental and finite element analysis for fracture of coating layer of galvannealed steel sheet显示文摘Mechanical properties of galvannealed (GA) steel sheet used for automotive exposed panel and predicted failure phenomenon of its coating layer were evaluated using finite element method. V-bending test was performed to understand better the fracture of coating layer of GA steel sheet during plastic deformation. Yield strength of the coating layer was calculated by using a relative difference between hardness of coating layer measured from the nano-indentation test and that of substrate. To measure shearing strength at the interface between substrate and coating layer, shearing test with two specimens attached by an adhesive was carried out. Using the mechanical properties measured, a series of finite element analyses coupled with a failure model was performed. Results reveal that the fracture of coating layer occurs in an irregular manner at the region where compressive deformation is dominant. Meanwhile, a series of vertical cracks perpendicular to material surface are observed at the tensile stressed-region. It is found that 0.26-0.28 of local equivalent plastic strain exists at the coating and substrate at the beginning of failure. | 12 | Unburned Carbon Loss in Fly Ash of CFB Boilers Burning Hard Coal显示文摘The unburned carbon loss in fly ash of circulating fluidized bed (CFB) boilers, most of which are burning active fuels such as lignite or peat, is normally very low. However, most CFB boilers in China usually burn hard coals such as anthracite and bituminous coal and coal wastes, so the carbon content in the fly ash from these boilers is higher than expected. This paper investigates the source of unburned carbon in the fly ash of CFB boilers burning hard coal through a series of field tests and laboratory investigations. The char behavior during combustion, including fragmentation and deactivation, which is related to the parent coal, has an important impact on the carbon burnout in CFB boilers. The research shows that char deactivation occurs during char burnout in fluidized bed combustion, especially for large particles of low rank coal. The uneven mixing of solids and air in the core region of the furnace also causes poor burnout of carbon in CFB fly ash. An index describing the volatile content (as dry ash free basis) over the heating value is proposed to present the coal rank. The coal combustion efficiency is shown to be strongly connected with this coal index. 目的:评估开始他汀治疗与慢性心力衰竭患者发生死亡或住院危险的关系。
设计、地点和患者:于确诊心力衰竭的患者中进行的趋势校正队列研究。患者适合降脂治疗,但从1996年1月1日至2004年12月31日,在北加利福尼亚一家综合性保健救助系统治疗前未予他汀治疗。他汀的使用于药房数据库通过填写的门诊处方确定。
主要观测指标:在中位数2.4年的随访期间,心力衰竭患者之全因死亡和住院。在多变量校正潜在混淆因素后,我们检查了他汀治疗与总不良事件以及与有或无冠心病亚组不良事件的独立关系。
结果:在24598例以前未使用他汀的心力衰竭成年患者中,开始他汀治疗(n=12648;51.4%)的患者较为年轻,更可能为男性,更可能患心血管病、糖尿病和高血压。有8235例患者死亡。使用意向治疗方法,他汀的使用与心力衰竭患者发生死亡(使用他汀治疗组与未使用他汀治疗组年龄、性别校正后死亡率为14.5/100病人-年比25.3/100病人-年;校正后风险比,0.76[95%可信区间,0.72~0.80])和住院(年龄、性别校正后发生率为21.9/100病人-年比31.1/100病人-年;校正风险比,0.79[95%可信区间,0.74~0.85])的危险较低相关,即使在校正他汀使用趋势、胆固醇水平、其他心血管药物或潜在混淆因素后结果亦如此。他汀的使用与有或无已知冠心病患者发生不良结局之校正后风险较低相关。
