1Photocrosslinkable Col/PCL/Mg composite membrane providing spatiotemporal maintenance and positive osteogenetic effects during guided bone regeneration显示文摘Guided bone regeneration membranes have been effectively applied in oral implantology to repair bone defects.However,typical resorbable membranes composed of collagen(Col)have insufficient mechanical properties and high degradation rate,while non-resorbable membranes need secondary surgery.Herein,we designed a photocrosslinkable collagen/polycaprolactone methacryloyl/magnesium(Col/PCLMA/Mg)composite membrane that provided spatiotemporal support effect after photocrosslinking.Magnesium particles were added to the PCLMA solution and Col/PCLMA and Col/PCLMA/Mg membranes were developed;Col membranes and PCL membranes were used as controls.After photocrosslinking,an interpenetrating polymer network was observed by scanning electron microscopy(SEM)in Col/PCL and Col/PCL/Mg membranes.The elastic modulus,swelling behavior,cytotoxicity,cell attachment,and cell proliferation of the membranes were evaluated.Degradation behavior in vivo and in vitro was monitored according to mass change and by SEM.The membranes were implanted into calvarial bone defects of rats for 8 weeks.The Col/PCL and Col/PCL/Mg membranes displayed much higher elastic modulus(p<0.05),and a lower swelling rate(p<0.05),than Col membranes,and there were no differences in cell biocompatibility among groups(p>0.05).The Col/PCL and Col/PCL/Mg membranes had lower degradation rates than the Col membranes,both in vivo and in vitro(p<0.05).The Col/PCL/Mg groups showed enhanced osteogenic capability compared with the Col groups at week 8(p<0.05).The Col/PCL/Mg composite membrane represents a new strategy to display space maintenance and enhance osteogenic potential,which meets clinical needs.Feilong Wang Dandan Xia Siyi Wang Ranli Gu Fan Yang Xiao Zhao Xuenan Liu Yuan Zhu Hao Liu Yongxiang Xu Yunsong Liu Yongsheng Zhou 2022Bioactive Materials2022,7,7:1
2A surface-eroding poly(1,3-trimethylene carbonate)coating for magnesium based cardiovascular stents with stable drug release and improved corrosion resistance显示文摘Magnesium alloys with integration of degradability and good mechanical performance are desired for vascular stent application.Drug-eluting coatings may optimize the corrosion profiles of magnesium substrate and reduce the incidence of restenosis simultaneously.In this paper,poly(trimethylene carbonate)(PTMC)with different molecular weight(50,000 g/mol named as PTMC5 and 350,000 g/mol named as PTMC35)was applied as drug-eluting coatings on magnesium alloys.A conventional antiproliferative drug,paclitaxel(PTX),was incorporated in the PTMC coating.The adhesive strength,corrosion behavior,drug release and biocompatibility were investigated.Compared with the PLGA control group,PTMC coating was uniform and gradually degraded from surface to inside,which could provide long-term protection for the magnesium substrate.PTMC35 coated samples exhibited much slower corrosion rate 0.05μA/cm^(2)in comparison with 0.11μA/cm^(2)and 0.13μA/cm^(2)for PLGA and PTMC5 coated counterparts.In addition,PTMC35 coating showed more stable and sustained drug release ability and effectively inhibited the proliferation of human umbilical vein vascular smooth muscle cells.Hemocompatibility test indicated that few platelets were adhered on PTMC5 and PTMC35 coatings.PTMC35 coating,exhibiting surface erosion behavior,stable drug release and good biocompatibility,could be a good candidate as a drug-eluting coating for magnesium-based stent.Hongyan Tang Shuangshuang Li Yuan Zhao Cunli Liu Xuenan Gu Yubo Fan 2022Bioactive Materials2022,7,1:1
3The PCK2-glycolysis axis assists three-dimensional-stiffness maintaining stem cell osteogenesis显示文摘Understanding mechanisms underlying the heterogeneity of multipotent stem cells offers invaluable insights into biogenesis and tissue development. Extracellular matrix (ECM) stiffness has been acknowledged as a crucial factor regulating stem cell fate. However, how cells sense stiffness cues and adapt their metabolism activity is still unknown. Here we report the novel role of mitochondrial phosphoenolpyruvate carboxykinase (PCK2) in enhancing osteogenesis in 3D ECM via glycolysis. We experimentally mimicked the physical characteristics of 3D trabeculae network of normal and osteoporotic bone with different microstructure and stiffness, observing that PCK2 promotes osteogenesis in 3D ECM with tunable stiffness in vitro and in vivo. Mechanistically, PCK2 enhances the rate-limiting metabolic enzyme pallet isoform phosphofructokinase (PFKP) in 3D ECM, and further activates AKT/extracellular signal-regulated kinase 1/2 (ERK1/2) cascades, which directly regulates osteogenic differentiation of MSCs. 5Elaidic acid-induced intestinal barrier damage led to gut-liver axis derangement and triggered NLRP3 inflammasome in the liver of SD rats显示文摘Previous studies have shown that trans fatty acids(TFA) are associated with several chronic diseases,the gut microbiota is directly influenced by dietary components and linked to chronic diseases.Our research investigated the effects of elaidic acid(EA),a typical TFA,on the gut microbiota to understand the underlying mechanisms of TFA-related chronic diseases.16S rDNA gene sequencing on faecal samples from Sprague-Dawley rats were performed to explore the composition change of the gut microbiota by EA gavage for 4 weeks.The results showed that the intake of EA increased the abundance of well-documented harmful bacteria,such as Proteobacteria,Anaerotruncus,Oscillibacter and Desulfovibrionaceae.Plus,EA induced translocation of lipopolysaccharides(LPS) and the above pathogenic bacteria,disrupted the intestinal barrier,led to gut-liver axis derangement and TLR4 pathway activation in the liver.Overall,EA induced intestinal barrier damage and regulated TLR4-MyD88-NF-κB/MAPK pathways in the liver of SD rats,leading to the activation of NLRP3 inflammasome and inflammatory liver damage.Hui Liu Xuenan Li Lu Li Yucai Li Haiyang Yan Yong Pang Wenliang Li Yuan Yuan 2024Food Science and Human Wellness2024,13,3:0
6Hydrogen-bonded organic framework for red light-mediated photocatalysis显示文摘The development of heterogeneous molecule-based catalysts for red light-mediated photocatalysis is still challenging due to the improper light absorption for most materials and the photoactivity deactivation for solid assembly.Herein,red light photocatalysis with a hydrogen-bonded organic framework(HOF)is established.This HOF,named HOF-66,is formed from the self-assembly of guanine-decorated naphthalenediimide(NDI)molecule through hydrogen-bonded guanine-quadruplex nodes,showing square grid supramolecular layers confirmed by powder X-ray diffraction analysis.In contrast to unsubstituted NDI HOF,introduction of ethylamino groups to NDI core in HOF-66 tunes strong electronic maximum absorption peak to 619 nm,allowing red light photocatalysis of singlet oxygen evolution proved by 1,3-diphenylisobenzofuran degradation and electron spin resonance determination.Particularly,under the same conditions,the sulfide oxidation rate in the presence of HOF-66 was 28 times higher compared to its unsubstituted analogue.This work integrates the molecular design and aggregation effect towards the application of HOFs,opening a new gate for red light photocatalysts.Haofei Zhao Ziwen Zhou Xuenan Feng Chao Liu Hui Wu Wei Zhou Hailong Wang 2023Nano Research2023,16,7:0

8Elaidic acid leads to mitochondrial dysfunction via mitochondria-associated membranes triggers disruption of mitochondrial calcium fluxes显示文摘Elaidic acid(EA)stimulation can lead to endoplasmic reticulum stress(ERS),accompanied by a large release of Ca^(2+),and ultimately the activation of NLRP3 inflammasome in Kupffer cells(KCs).Mitochondrial instability or dysfunction may be the key stimulating factors to activate NLRP3 inflammasome,and sustained Ca^(2+)transfer can result in mitochondrial dysfunction.We focused on KCs to explore the damage to mitochondria by EA.After EA stimulation,cells produced an oxidative stress(OS)response with a significant increase in ROS release.Immunoprecipitation experiments and the addition of inhibitors revealed that the increase in the level of intracellular Ca^(2+)led to Ca^(2+)accumulation in the mitochondrial matrix via mitochondria-associated membranes(MAMs).This was accompanied by a significant release of m ROS,loss of MMP and ATP,and a significant increase in mitochondrial permeability transition pore opening,ultimately leading to mitochondrial instability.These findings confirmed the mechanism that EA induced mitochondrial Ca^(2+)imbalance in KCs via MAM,ultimately leading to mitochondrial dysfunction.Meanwhile,EA induced OS and the decrease of MMP and ATP in rat liver,and significant lesions were found in liver mitochondria.Swelling of the inner mitochondrial cristae and mitochondrial vacuolization occurred,with a marked increase in lipid droplets.Hui Liu Xuenan Li Ziyue Wang Lu Li Yucai Li Haiyang Yan Yuan Yuan 2024Food Science and Human Wellness2024,13,1:0
