1Autophagy inhibition enhances apigenin-induced apoptosis in human breast cancer cells显示文摘Apigenin (4',5,7-trihydroxyflavone) is a member of the flavone subclass of flavonoids present in fruits and vegetables. The involvement of autophagy in the apigenin-induced apoptotic death of human breast cancer cells was investigated. Cell proliferation and viability were assessed by 3-(4,5-dimethylthiazol- 2-yl)-2,5-diphenyltetrazolium bromide (MTT) and clonogenic assays. Flow cytometry, fluorescent staining and Western blot analysis were employed to detect apoptosis and autophagy, and the role of autophagy was assessed using autophagy inhibitors. Apigenin dose- and time-dependently repressed the proliferation and clonogenic survival of the human breast cancer T47D and MDA-MB-231 cell lines. The death of T47D and MDA-MB-231 cells was due to apoptosis associated with increased levels of Caspase3, PARP cleavage and Bax/Bcl-2 ratios. The results from flow cytometry and fluorescent staining also verified the occurrence of apoptosis. In addition, the apigenin-treated cells exhibited autophagy, as characterized by the appearance of autophagosomes under fluorescence microscopy and the accumulation of acidic vesicular organelles (AVOs) by flow cytometry. Furthermore, the results of the Western blot analysis revealed that the level of LC3-II, the processed form of LC3-I, was increased. Treatment with the autophagy inhibitor, 3-methyladenine (3-MA), significantly enhanced the apoptosis induced by apigenin, which was accompanied by an increase in the level of PARP cleavage. Similar results were also confirmed by flow cytometry and fluorescence microscopy. These results indicate that apigenin has apoptosis-and autophagy-inducing effects in breast cancer cells. 2Validation of American Joint Committee on Cancer 8th edition of TNM staging in resected distal pancreatic cancer显示文摘BACKGROUND In order to improve risk stratification and clinical management of the pancreatic ductal adenocarcinoma(PDAC),the American Joint Committee on Cancer(AJCC)has published its eighth edition staging manual.Some major changes have been introduced in the new staging system for both T and N categories.Given the rarity of resectable disease,distal pancreatic cancer is likely underrepresented in the published clinical studies,and how the impact of the staging system actually reflects on to clinical outcomes remain unclear.AIM To validate the AJCC 8th edition of TNM staging in distal PDAC.METHODS A retrospective cohort study was performed in seven academic medical centers in the United States.Clinicopathological prognostic factors associated with progression-free survival(PFS)and overall survival(OS)were evaluated through univariate and multivariate analyses.RESULTS Overall,454 patients were enrolled in the study,and were divided into 2 subgroups:Invasive intraductal papillary mucinous neoplasms(IPMN)(115 cases)and non-IPMN associated adenocarcinoma(339 cases).Compared to invasive IPMN,non-IPMN associated adenocarcinomas are more common in relatively younger patients,have larger tumor size,are more likely to have positive lymph nodes,and are associated with a higher tumor(T)stage and nodal(N)stage,lymphovascular invasion,perineural invasion,tumor recurrence,and a worse PFS and OS.The cohort was predominantly categorized as stage 3 per AJCC 7th edition staging manual,and it’s more evenly distributed based on 8th edition staging manual.T and N staging of both 7th and 8th edition sufficiently stratify PFS and OS in the entire cohort,although dividing into N1 and N2 according to the 8th edition does not show additional stratification.For PDAC arising in IPMN,T staging of the 7th edition and N1/N2 staging of the 8th edition appear to further stratify PFS and OS.For PDAC without an IPMN component,T staging from both versions fails to stratify PFS and OS.CONCLUSION The AJCC 8th edition TNM staging system provides even distribution for the T staging,however,it does not provide better risk stratification than previous staging system for distal pancreatic cancer.Feng Yin Mohammed Saad Hao Xie Jingmei Lin Christopher R Jackson Bing Ren Cynthia Lawson Dipti M Karamchandani Belen Quereda Bernabeu Wei Jiang Teena Dhir Richard Zheng Christopher W Schultz Dongwei Zhang Courtney L Thomas Xuchen Zhang Jinping Lai Michael Schild Xuefeng Zhang Xiuli Liu 2020World Journal of Gastrointestinal Pharmacology and Therapeutics2020,11,2:1
3The Coevolutionary Relationship of Technology, Market and Government Regulation in Telecommunications显示文摘The telecommunications industry has been undergoing tremendous technological changes, and owning to continuous technological advancement, it has maintained sustained prosperity and development. In this paper, the interplay between technology, market and government in telecommunications is discussed briefly, and then we introduce technology and government into the traditional SCP(Structure – Conduct – Performance) paradigm to develop an industry analysis framework called TGM(SCP)(Technology – Government – Market(Structure – Conduct – Performance)). Based on this framework, we present the spiral coevolution model which elaborates on the interaction mechanism of technological innovation with government regulation and market dynamics from the perspective of industry evolution. Our study indicates that the development of the telecommunications industry is the result of the coevolution of technology, government regulation and market forces, and among the three actors, technology is the fundamental driving force. Relative to the 'invisible hand'(market) and 'visible hand'(government), we conceptualize technology as the 'third hand', which fundamentally drives the development of telecommunications industry in coordination with the other two hands. 4Mapping HⅠ in the NGC 4636 Galaxy Group with FAST显示文摘This paper presents data from a 21 cm H I emission drift scan observation of a field partially covering the NGC 4636 galaxy group with the Five-hundred meter Aperture Radio Telescope(FAST). We construct a pipeline to reduce the data, and use So Fi A for source finding. When not contaminated by Radio Frequency Interference(RFI), the FAST observations are capable of detecting all of the galaxies previously detected by the Arecibo Legacy Fast ALFA(ALFALFA) survey in the same region. Comparing to ALFALFA for the detections in common, the FAST data show consistent integrated spectra when the H I disks are spatially unresolved, and capture more flux when the H I disks are resolved. The FAST data further reveal 10 new detections in the region mutually covered with ALFALFA, and 18 new detections beyond the footprint of ALFALFA. All of the new detections have the matching optical counterparts. For the member galaxies of the NGC 4636 group, the detection limit of FAST is deeper by 0.4 dex in H I mass than that of the ALFALFA data. After correcting for the incompleteness caused by RFI contamination, we show that the H I detection rate of galaxies rises steeply with radius out to the virial radius of the group, and flattens beyond that. We also examine four spatially resolved galaxy systems with potential tidal interaction features in detail. 7Cryptic divergences and repeated hybridizations within the endangered “living fossil” dove tree(Davidia involucrata) revealed by whole genome resequencing显示文摘The identification and understanding of cryptic intraspecific evolutionary units(lineages) are crucial for planning effective conservation strategies aimed at preserving genetic diversity in endangered species.However, the factors driving the evolution and maintenance of these intraspecific lineages in most endangered species remain poorly understood. In this study, we conducted resequencing of 77 individuals from 22 natural populations of Davidia involucrata, a “living fossil” dove tree endemic to central and southwest China. Our analysis revealed the presence of three distinct local lineages within this endangered species, which emerged approximately 3.09 and 0.32 million years ago. These divergence events align well with the geographic and climatic oscillations that occurred across the distributional range.Additionally, we observed frequent hybridization events between the three lineages, resulting in the formation of hybrid populations in their adjacent as well as disjunct regions. These hybridizations likely arose from climate-driven population expansion and/or long-distance gene flow. Furthermore, we identified numerous environment-correlated gene variants across the total and many other genes that exhibited signals of positive evolution during the maintenance of two major local lineages. Our findings shed light on the highly dynamic evolution underlying the remarkably similar phenotype of this endangered species.
