1In vivo self-assembled small RNAs as a new generation of RNAi therapeutics显示文摘RNAi therapy has undergone two stages of development,direct injection of synthetic siRNAs and delivery with artificial vehicles or conjugated ligands;both have not solved the problem of efficient in vivo siRNA delivery.Here,we present a proof-of-principle strategy that reprogrammes host liver with genetic circuits to direct the synthesis and self-assembly of siRNAs into secretory exosomes and facilitate the in vivo delivery of siRNAs through circulating exosomes.By combination of different genetic circuit modules,in vivo assembled siRNAs are systematically distributed to multiple tissues or targeted to specific tissues(e.g.,brain),inducing potent target gene silencing in these tissues.The therapeutic value of our strategy is demonstrated by programmed silencing of critical targets associated with various diseases,including EGFR/KRAS in lung cancer,EGFR/TNC in glioblastoma and PTP1B in obesity.Overall,our strategy represents a next generation RNAi therapeutics,which makes RNAi therapy feasible.Zheng Fu Xiang Zhang Xinyan Zhou Uzair Ur-Rehman Mengchao Yu Hongwei Liang Hongyuan Guo Xu Guo Yan Kong Yuanyuan Su Yangyang Ye Xiuting Hu Wei Cheng Jinrong Wu Yanbo Wang Yayun Gu Sheng-feng Lu Dianqing Wu Ke Zen Jing Li Chao Yan Chen-Yu Zhang Xi Chen 2021Cell Research2021,31,6:10
2An experimental study on microscopic characteristics of gas-bearing sediments under different gas reservoir pressures显示文摘Gas-bearing sediments are widely distributed in five continents all over the world.Most of the gases exist in the soil skeleton in the form of discrete large bubbles.The existence of gas-phase may increase or decrease the strength of the soil skeleton.So far,bubbles’structural morphology and evolution characteristics in soil skeleton lack research,and the influence of different gas reservoir pressures on bubbles are still unclear.The micro characteristics of bubbles in the same sediment sample were studied using an industrial CT scanning test system to solve these problems.Using the image processing software,the micro variation characteristics of gas-bearing sediments in gas reservoir pressure change are obtained.The results show that the number and volume of bubbles in different equivalent radius ranges will change regularly under different gas reservoir pressure.With the increase of gas reservoir pressure,the number and volume of tiny bubbles decrease.In contrast,the number and volume of large bubbles increase,and the gas content in different positions increases and occupies a dominant position,driving the reduction of pore water and soil skeleton movement.Zhenqi Guo Tao Liu Lei Guo Xiuting Su Yan Zhang Sanpeng Li 2021Acta Oceanologica Sinica2021,40,10:2
3Effect of Penetration Rates on the Piezocone Penetration Test in the Yellow River Delta Silt显示文摘Partial drainage often occurs during piezocone penetration testing on Yellow River Delta silt because of its intermediate physical and mechanical properties between those of sand and clay.Yet,there is no accurate understanding for the range of penetra-tion rates to trigger the partial drainage of silt soils.In order to fully investigate cone penetration rate effects under partial drainage condi-tions,indoor 1 g penetration model tests and numerical simulations of cavity expansion at variable penetration rates were carried out on the Yellow River Delta silt.The boundary effect of the model tests and the variation of key parameters at the different cavity ex-pansion rates were analyzed.The 1 g penetration model test results and numerical simulations results consistently indicated that the penetration rate to trigger the partially drainage of typical silt varied at least three orders of magnitude.The numerical simulations also provide the reference values for the penetration resistance corresponding to zero dilation and zero viscosity at any given normalized penetration rate for silt in Yellow River Delta.These geotechnical properties can be used for the design of offshore platforms in Yel-low River Delta,and the understanding of cone penetration rate effects under the partially drained conditions would provide some technical support for geohazard evaluation of offshore platforms.ZHANG Jiarui MENG Qingsheng ZHANG Yan FENG Xiuli WEI Guanli SU Xiuting LIU Tao 2022Journal of Ocean University of China2022,21,2:1
4Research on Corrosion Effect of Sulfate Ions on Concrete Under Wetting-Drying Cycle显示文摘Saline soil is widely distributed in the marine sediments along the coast of the world and the arid-semi-arid areas of the Middle East and Iraq,and calcium sulfate erosion has become one of the important factors affecting the durability of concrete in this area.In order to clarify the mechanism of sulfate ion damage to concrete,this paper mainly takes saline soil with high sulfate content in coastal area as well as arid-semi-arid area as the research object,and uses indoor geotechnical test,field test and numerical simulation to study the influence of different dry-wet cycle times on the unconfined compressive strength of concrete test blocks,and puts forward the relationship between the erosion arrival depth and time of sulfate ion in concrete,so as to predict the long-term erosion depth by using the erosion depth of sulfate ion in concrete in short time.The results show that the shorter the erosion time when the erosion reaches a certain depth,and the larger the erosion reaches when the erosion time is the same,the faster the erosion reaches the depth with the increase of erosion time.Compared with rectangular section concrete,circular section concrete penetrates faster.The results of this study can provide a reference for the durability design of concrete in saline soil sites containing sulfate.WANG Xiuhai TIAN Zhuangcai ZHANG Yanan SU Xiuting LIU Hongjun LIU Tao 2022Journal of Ocean University of China2022,21,1:0
5CPT-Based estimation of undrained shear strength of fine-grained soils in the Huanghe River Delta显示文摘The Huanghe River(Yellow River)Delta has a wide distribution of fine-grained soils.Fluvial alluviation,erosion,and wave loads affect the shoal area,resulting complex physical and mechanical properties to sensitive finegrained soil located at the river-sea boundary.The cone penetration test(CPT)is a convenient and effective in situ testing method which can accurately identify various soil parameters.Studies on undrained shear strength only roughly determine the fine content(FC)without making the FC effect clear.We studied four stations formed in different the Huanghe River Delta periods.We conducted in situ CPT and corresponding laboratory tests,examined the fine content influence on undrained shear strength(S_(u)),and determined the cone coefficient(N_(k)).The conclusions are as follows.(1)The fine content in the area exceeded 90%,and the silt content was high,accounting for more than 70%of all fine particle compositions.(2)The undrained shear strength gradually increased with depth with a maximum of approximately 250 kPa.When the silt content was lower than 60%–70%,the undrained shear strength decreased.(3)The silt and clay content influenced undrained shear strength,and the fitted f_(s)h/q_(t) function model was established,which could be applied to strata with a high fine content.The cone coefficients were between 20 and 25,and the overconsolidated soil layer had a greater cone coefficient.Zhongnian Yang Xuesen Liu Lei Guo Yuxue Cui Xiuting Su Chao Jia Xianzhang Ling 2022Acta Oceanologica Sinica2022,41,5:0
6Constructing high-capacity and flexible aqueous zinc-ion batteries with air-recharging capability using organic cathodes显示文摘Flexible aqueous zinc-ion batteries(AZIBs)with air-recharging capability are a promising self-powered system applied in future wearable electronics.It is desired to develop high-capacity air-rechargeable AZIBs.Herein,we developed a flexible AZIB with air-recharging capability based on trinitrohexaazatrinaphthylene(TNHATN)cathode and a ZnSO_(4)electrolyte.The flexible Zn//TNHATN battery exhibits high volumetric energy density(21.36 mWh/cm^3)and excellent mechanical flexibility.Impressing,the discharged flexible Zn//TNHATN battery can be chemical self-charged via the redox reaction between TNHATN cathode and O_(2)from the air.After oxidation in air for 15 h,such flexible Zn//TNHATN battery can deliver a high specific capacity of 320 mAh/g at 0.5 A/g,displaying excellent air-recharging capability.Notably,this flexible Zn//TNHATN battery also works well in chemical or/and galvanostatic charging mixed modes,showing reusability.This work provides a new insight for designing flexible aqueous self-powered systems.Xiaojuan Chen Haoqi Su Baozhu Yang Xiaocen Liu Xiuting Song Lixin Su Gui Yin Qi Liu 2024Chinese Chemical Letters2024,35,3:0
