1Atmospheric fine particles in a typical coastal port of Yangtze River Delta显示文摘In recent decades, coastal ports have experienced rapid development and become an important economic and ecological hub in China. Atmospheric particle is a research hotspot in atmospheric environmental sciences in inland regions. However, few studies on the atmospheric particle were conducted in coastal port areas in China, which indeed suffers atmospheric particle pollution. Lack of the physicochemical characteristics of fine particles serves as an obstacle toward the accurate control for air pollution in the coastal port area in China. Here, a field observation was conducted in an important coastal port city in Yangtze River Delta from March 6 to March 19, 2019. The average PM2.5 concentration was 63.7 ±27.8 μg/m^3 and NO3^-, SO4^2-, NH4^+, and organic matter accounted for ?60% of PM 2.5. Fe was the most abundant trace metal element and V as the ship emission indicator was detected. Transmission electron microscopy images showed that SK-rich, soot, Fe, SK-soot and SK-Fe were the major individual particles in the coastal port. V and soluble Fe were detected in sulfate coating of SK-Fe particles. We found that anthropogenic emissions, marine sea salt, and secondary atmosphere process were the major sources of fine particles. Backward trajectory analysis indicated that the dominant air masses were marine air mass, inland air mass from northern Zhejiang and inland-marine mixed air mass from Shandong and Shanghai during the sampling period. 2Linsitinib (OSI‑906) modulates brain energy metabolism and seizure activity in the lithium‑pilocarpine rat model显示文摘Background:Epileptic seizure is a process of energy accumulation,bursting,and depletion accompanied by the production,spread,and termination of epileptic discharges.The energy required for a seizure is mainly provided through mitochondrial production of ATP.Mitochondrial diseases often lead to epileptic seizures,and energy depletion caused by seizures can lead to mitochondrial dysfunction.The energy metabolism has become a key target for treatment of epileptic diseases.Method:The effect of OSI-906,an insulin receptor(IR)/insulin-like growth factor 1 receptor(IGF-1R)inhibitor,on behaviors and electroencephalographic activity in the lithium-pilocarpine rats were tested.18F-FDG positron emission tomography(PET)/computed tomography(CT)was performed to detect the relative whole-brain glucose uptake values.Electron microscopy was performed to observe the ultrastructure of neuronal and mitochondrial damage.The changes in blood glucose at different time points before and after the intervention were tested and the effects of OSI-906 on IR/IGF-1R and downstream Akt signaling in the context of seizures were evaluated.Results:The OSI-906 treatment applied 3 days before the pilocarpine-induced seizures significantly reduced the seizure severity,prolonged the seizure latency and decreased the EEG energy density.MicroPET/CT revealed that 50 mg/kg of OSI-906 inhibited the 18F-FDG glucose uptake after epileptic seizures,suggesting that OSI-906,through inhibiting IR/IGF-1R and the downstream AKT signaling,may regulate the excessive energy consumption of the epileptic brain.The OSI-906 treatment also reduced the mitochondrial damage caused by epileptic seizures.Conclusion:The IR/IGF-1R inhibitor OSI-906 can significantly reduce the sensitivity and severity of pilocarpineinduced seizures by inhibiting the IR/IGF-1R and the downstream Akt signaling pathway.Guohui Jiang Shenglin Wang Mingyue Chen Xiaomi Ding Weiwei He Li Wang Shunxian Wang Juming Yu Xiaoming Wang 2021Acta Epileptologica2021,3,1:0
3Potentials of miR-9-5p in promoting epileptic seizure and improving survival of glioma patients显示文摘Background Epilepsy affects over 70 million people worldwide;however,the underlying mechanisms remain unclear.MicroRNAs(miRNAs)have essential functions in epilepsy.miRNA-9,a brain-specific/enriched miRNA,plays a role in various nervous system diseases and tumors,but whether miRNA-9 is involved in epilepsy and glioma-associated epilepsy remains unknown.Therefore,we aimed to explore the potential role of miR-9-5p in seizures and its effect on the survival of glioma patients,in order to provide new targets for the treatment of epilepsy and glioma.Methods The YM500v2 database was used to validate the expression of hsa-miR-9-5p in tissues.Moreover,qRT-PCR was performed to investigate the expression of miR-9-5p in temporal lobe epilepsy patients and rats with lithium-pilocarpine-induced seizures.Recombinant adeno-associated virus containing miR-9-5p was constructed to overexpress miR-9-5p in vivo.The effects of miR-9-5p on the behavior and electroencephalographic activities of the lithium-pilocarpine rat model of epilepsy were tested.Bioinformatics analysis was used to predict the targets of miR-9-5p and explore its potential role in epilepsy and glioma-associated epilepsy.Results The expression of miR-9-5p increased at 6 h and 7 days after lithium-pilocarpine-induced seizures in rats.Overexpression of miR-9-5p significantly shortened the latency of seizures and increased seizure intensity at 10 min and 20 min after administration of pilocarpine(P<0.05).Predicted targets of miR-9-5p were abundant and enriched in the brain,and affected various pathways related to epilepsy and tumor.Survival analysis revealed that overexpression of miR-9-5p significantly improved the survival of patients from with low-grade gliomas and glioblastomas.The involvement of miR-9-5p in the glioma-associated epileptic seizures and the improvement of glioma survival may be related to multiple pathways,including the Rho GTPases and hub genes included SH3PXD2B,ARF6,and ANK2.Conclusions miR-9-5p may play a key role in promoting epileptic seizures and improving glioma survival,probably through multiple pathways,including GTPases of the Rho family and hub genes including SH3PXD2B,ARF6 and ANK2.Understanding the roles of miR-9-5p in epilepsy and glioma and the underlying mechanisms may provide a theoretical basis for the diagnosis and treatment of patients with epilepsy and glioma.Shenglin Wang Xuzhi He Nana Bao Mingyue Chen Xiaomi Ding Ming Zhang Li Zhao Shunxian Wang Guohui Jiang 2022Acta Epileptologica2022,5,4:0
