Non-coding RNAs:the new central dogma of cancer biology显示文摘The central dogma of molecular biology states that the functions of RNA revolve around protein translation.Until the last decade, most researches were geared towards characterization of RNAs as intermediaries in protein translation, namely, messenger RNAs(mRNAs)as temporary copies of genetic information, ribosomal RNAs(rRNAs)as a main component of ribosome,or translators of codon sequence(t RNAs).The statistical reality, however,is that these processes account for less than 2%of the genome, and insufficiently explain the functionality of 98%of transcribed RNAs.Recent discoveries have unveiled thousands of unique non-coding RNAs(ncRNAs)and shifted the perception of them from being'junk'transcriptional products to'yet to be elucidated'—and potentially monumentally important—RNAs.Most ncRNAs are now known as key regulators in various networks in which they could lead to specific cellular responses and fates. In major cancers,ncRNAs have been identified as both oncogenic drivers and tumor suppressors, indicating a complex regulatory network among these ncRNAs.Herein,we provide a comprehensive review of the various ncRNAs and their functional roles in cancer, and the pre-clinical and clinical development of nc RNA-based therapeutics.A deeper understanding of ncRNAs could facilitate better design of personalized therapeutics. | Phei Er Saw Xiaoding Xu Jianing Chen Er-Wei Song | 2021 | Science China(Life Sciences)2021,64,1: | 21
Spatiotemporal variations of nitrate sources and dynamics in a typical agricultural riverine system under monsoon climate显示文摘Nitrogen pollution is a serious environmental issue in the Danjiangkou Reservoir region(DRR),the water source of the South-to-North Water Diversion Project of China. In this research, seasonal surveys and a bi-weekly time series survey were conducted in the Qihe River Basin, one of the most densely populated agricultural basins in the DRR.Hydrochemical compositions(NO3^-Cl^-),dual isotopes(δD-H2O,δ^18O-H2O,δ^15N-NO3^-,andδ^18O-NO3^-),and a Markov Chain Monte Carlo isotope mixing model were jointly applied to unravel the sources, migrations, and transformations of the nitrate(NO3^-)in the basin. It was revealed that the mixing between different sources was the main process controlling the isotopic compositions of the riverine NO3the upper-middle reaches. In contrast, denitrification occurred in the lower reaches. For the first time, the sources of NO3^-quantified at a basin scale in the DRR.Overall, the river transported 484.2 tons/year of NO3^-N to the reservoir,of which 32.6%,36.4%,28.0%,and 3.0%was from soil organic nitrogen, chemical fertilizer, residential sewage and atmospheric precipitation, respectively. The NO3^-N fluxes of the different sources were regulated by the monsoon climate and anthropogenic activities. For example, high precipitation and intense fertilization resulted in severe nonpoint source pollution.Denitrification thrived in soils and reservoirs in wet seasons.Temperature could regulate the migration, nitrification and denitrification processes.Based on the results,we suggest that the management strategies dealing with nitrogen pollution issue in the DRR should follow the specific spatiotemporal characteristics of NO3migration and transformation mechanisms. | Hao Jiang Wenjing Liu Jiangyi Zhang Li Zhou Xiaode Zhou Ke Pan Tong Zhao Yuchen Wang Zhifang Xu | 2020 | Journal of Environmental Sciences2020,32,7: | 5
Characteristics and origin of mafic and ultramafic xenoliths in trachyandesite lavas from Heikongshan volcano,Tengchong,Yunnan Province,China显示文摘The trachyandesite lavas from Heikongshan volcano of the Tengchong volcanic cluster, Yunnan Province contain relatively abundant mafic and ultramafic small xenoliths comprising mainly gabbro, pyroxenite and rare lherzolites, all less than 2.5 cm in size. Gabbro xenoliths are characterized by 'open' texture represented by relatively abundant vesicles and ground-mass fillings, while pyroxenite xenoliths are characterized by equigranular and poikilitic texture. Their mineral compositions (clinopyroxene ± orthopyroxene ± plagioclase) are similar to those of phenocrysts in lavas, and their equilibration temperature (1000-1125°C) is consistent with the crystallization temperature of the phenocrysts (998-1108°C). Thus, the textures and compositions of these xenoliths are different from those of lower crustal and mantle xenoliths, and were derived from the magma chamber. Among them, the gabbroic xenoliths come from the crystal-bubble-liquid zone at the top of the magma reservoir, while the pyroxenite xenoliths come from the cumulates in the bottom of magma reservoir. The studied single lherzolite xenolith with porphyroclastic texture has similar mineral compositions to the mantle xenoliths from eastern China, and is considered to be of upper mantle origin. It was brought into magma reservoir by replenishment magma, and might have experienced a cooling event before eruption. The formation of xenoliths in trachyandesite flows largely reflects the ascent, storage, evolution and eruption processes of magma. | YU HongMei, LIN ChuanYong, SHI LanBin, XU JianDong & CHEN XiaoDe Institute of Geology, China Earthquake Administration, Beijing 100029, China | 2010 | Science China Earth Sciences2010,53,9: | 5
Car-following behavior modeling driven by small data sets based on mnemonic extreme gradient boosting framework显示文摘Dear editor,In recent years,data-driven car-following models have been developed based on their ability to drill down to information in driving data and their flexibility.According to a study by Fleming et al.[1],the main limitation of existing methods is an insufficient amount of natural driving data.In addition, there are many situations during actual driving processes, such as those under extreme conditions[2]and in the early stages of car-following behavior modeling. In these cases, sparse data learning algorithms are indispensable. | Baichuan LOU Yufang LI Xiaoding LU Zhe XU | 2022 | Science China(Information Sciences)2022,65,6: | 2

Nanoparticles(NPs)-mediated systemic mRNA delivery to reverse trastuzumab resistance for effective breast cancer therapy显示文摘Monoclonal antibody-based therapy has achieved great success and is now one of the most crucial therapeutic modalities for cancer therapy. The first monoclonal antibody authorized for treating human epidermal growth receptor 2(HER2)-positive breast cancer is trastuzumab. However, resistance to trastuzumab therapy is frequently encountered and thus significantly restricts the therapeutic outcomes. To address this issue, tumor microenvironment(TME)pH-responsive nanoparticles(NPs)were herein developed for systemic mRNA delivery to reverse the trastuzumab resistance of breast cancer(BCa).This nanoplatform is comprised of a methoxyl-poly(ethylene glycol)-b-poly(lactic-co-glycolic acid)copolymer with a TME pH-liable linker(Meo-PEG-Dlinkm-PLGA)and an amphiphilic cationic lipid that can complex PTEN mRNA via electrostatic interaction. When the long-circulating mRNA-loaded NPs build up in the tumor after being delivered intravenously, they could be efficiently internalized by tumor cells due to the TME pH-triggered PEG detachment from the NP surface. With the intracellular mRNA release to up-regulate PTEN expression, the constantly activated PI3K/Akt signaling pathway could be blocked in the trastuzumab-resistant BCa cells, thereby resulting in the reversal of trastuzumab resistance and effectively suppress the development of BCa. | Zhihui Dong Zhuoshan Huang Senlin Li Ying Wang Yandan Yao Xianzhu Yang Xiaoding Xu | 2023 | Acta Pharmaceutica Sinica B2023,13,3: | 1
A Study on Dual-Load-Zone Model of Overlying Strata andEvolution Law of Mining Stress显示文摘The changeable structure and movement law of overlying strata are the maincontributor to the change of mining stress.Starting from the relevant theory of keystratum and particularly based on the theory of mine ground pressure and strata control, this research proposed a new solution to mining stress problems by establishing adual-load-zone stratum structural model.Elastic foundation beam theory was used tosolve the stress of overlying strata of the dual-load-zones with superposition method, which revised the traditional calculation method of mining stress. The abnormal increaseof lead abutment pressure in the mining area was explained effectively, through which theevolution law of mining stress in the case of hard rock was obtained. The results indicatethat mining stress experiences a drastic change within the range of 50 m ahead of the coalwall due to the collapse of main roof;under the influence of main key stratum andinferior key strata, the influence range of lead abutment pressure is extended up toapproximately 120 m in the working face;this remarkable increase can be attributed tothe excessive length of sagging zone. Results from both the dual-load-zone modelexperiment and field measurement demonstrate high consistency. The model can predictthe influence range of abutment pressure effectively and thus guide the safety productionof mining. | Yuejin Zhou Mingpeng Li Xiaoding Xu Meng Li | 2019 | Computers, Materials & Continua2019,,2: | 1

Nanomedicine targeting ferroptosis to overcome anticancer therapeutic resistance显示文摘A potential reason for the failure of tumor therapies is treatment resistance.Resistance to chemotherapy, radiotherapy, and immunotherapy continues to be a major obstacle in clinic, resulting in tumor recurrence and metastasis. The major mechanisms of therapy resistance are inhibitions of cell deaths, like apoptosis and necrosis, through drug inactivation and excretion, repair of DNA damage, tumor heterogeneity,or changes in tumor microenvironment, etc.Recent studies have shown that ferroptosis play a major role in therapies resistance by inducing phospholipid peroxidation and iron-dependent cell death. Some ferroptosis inducers in combination with clinical treatment techniques have been used to enhance the effect in tumor therapy.Notably, versatile ferroptosis nanoinducers exhibit an extensive range of functions in reversing therapy resistance, including directly triggering ferroptosis and feedback regulation.Herein,we provide a detailed description of the design, mechanism, and therapeutic application of ferroptosis-mediated synergistic tumor therapeutics. We also discuss the prospect and challenge of nanomedicine in tumor therapy resistance by regulating ferroptosis and combination therapy. | Jing Cai Xiaoding Xu Phei Er Saw | 2024 | Science China(Life Sciences)2024,67,1: | 0

One-dimensional and two-dimensional nanomaterials for the detection of multiple biomolecules显示文摘The complexity of biological samples determines that the detection of a single biomolecule is unable to satisfy actual needs. Moreover, the 'false positives' results caused by a single biomolecule detections easily leads to erroneous clinical diagnosis and treatment. Thus, it is important for the homogenous quantification of multiple biomolecules in not only basic research but also practical application. As a consequent, a large number of literatures have been exploited to monitor multiple biomolecules in homogenous solution, enabling facilitating the development of the disease diagnosis, treatment as well as drug discovery. One-dimensional nanomaterials and two-dimensional nanomaterials have special physical and chemical properties, such as good electrochemical properties, stable structure, large specific surface area, and biocompatibility, which are widely used in electrochemical and fluorescent detection of biomolecules. This tutorial review highlights the recent development for the detection of multiple biomolecules by using nanomaterials including one-dimensional materials(1DMs) as well as twodimensional materials(2DMs). | Quan Wang Xudong Wang Min Xu Xiaoding Lou Fan Xia | 2019 | Chinese Chemical Letters2019,30,9: | 0
Nanoparticles (NPs)-mediated Siglec15 silencing and macrophage repolarization for enhanced cancer immunotherapy显示文摘cell infiltration and proliferation in tumor tissues are the main factors that significantly affect the therapeutic outcomes of cancer immunotherapy.Emerging evidence has shown that interferon-gamma(IFN)could enhance CXCL9 secretion from macrophages to recruit T cells, but Siglec15 expressed on TAMs can attenuate T cell proliferation. Therefore, targeted regulation of macrophage function could be a promising strategy to enhance cancer immunotherapy via concurrently promoting the infiltration and proliferation of T cells in tumor tissues. We herein developed reductionresponsive nanoparticles(NPs)made with poly(disulfide amide)(PDSA)and lipid-poly(ethylene glycol)(lipid-PEG)for systemic delivery of Siglec15 siRNA(siSiglec15)and IFN for enhanced cancer immunotherapy. After intravenous administration, these cargo-loaded could highly accumulate in the tumor tissues and be efficiently internalized by tumor-associated macrophages(TAMs).With the highly concentrated glutathione(GSH)in the cytoplasm to destroy the nanostructure, the loaded IFN and si-Siglec15 could be rapidly released, which could respectively repolarize macrophage phenotype to enhance CXCL9 secretion for T cell infiltration and silence Siglec15 expression to promote T cell proliferation, leading to significant inhibition of hepatocellular carcinoma(HCC)growth when combining with the immune checkpoint inhibitor. The strategy developed herein could be used as an effective tool to enhance cancer immunotherapy. | Xiaodi Liu Qi Zhang Yixia Liang Shiyu Xiong Yan Cai Jincheng Cao Yanni Xu Xiaolin Xu Ye Wu Qiang Lu Xiaoding Xu Baoming Luo | 2023 | Acta Pharmaceutica Sinica B2023,13,12: | 0
Nanoparticles(NPs)-mediated lncBCMA silencing to promote eEF1A1 ubiquitination and suppress breast cancer growth and metastasis显示文摘Long non-coding RNAs(lncRNAs)play an important role in cancer metastasis.Exploring metastasis-associated lncRNAs and developing effective strategy for targeted regulation of lncRNA function in vivo are of utmost importance for the treatment of metastatic cancer, which however remains a big challenge.Herein,we identified a new functional lncRNA(denoted lncBCM A),which could stabilize the expression of eukaryotic translation elongation factor 1A1(eEF1A1)via antagonizing its ubiquitination to promote triple-negative breast cancer(TNBC)growth and metastasis.Based on this regulatory mechanism,an endosomal pH-responsive nanoparticle(NP)platform was engineered for systemic lncBCMA siRNA(siBCMA)delivery. This NPs-mediated siBCMA delivery could effectively silence lncBCMA expression and promote eEF1A1 ubiquitination, thereby leading to a significant inhibition of TNBC tumor growth and metastasis. These findings show that lncBCMA could be used as a potential biomarker to predict the prognosis of TNBC patients and NPs-mediated lncBCMA silencing could be an effective strategy for metastatic TNBC treatment. | Ke Yang Lei Xu Ying Xu Qian Shen Tao Qin Yunfang Yu Yan Nie Herui Yao Xiaoding Xu | 2023 | Acta Pharmaceutica Sinica B2023,13,8: | 0
