1The impact of sample processing on the rapid antigen detection test for SARS-CoV-2: Virus inactivation, VTM selection, and sample preservation显示文摘Many factors have been identified as having the ability to affect the sensitivity of rapid antigen detection(RAD)tests for severe acute respiratory syndrome coronavirus 2(SARS-CoV-2).This study aimed to identify the impact of sample processing on the sensitivity of the RAD tests.We explored the effect of different inactivation methods,viral transport media(VTM)solutions,and sample preservation on the sensitivity of four RAD kits based on two SARS-CoV-2 strains.Compared with non-inactivation,heat inactivation significantly impacted the sensitivity of most RAD kits;however,β-propiolactone inactivation only had a minor effect.Some of the VTM solutions(VTM2,MANTACC)had a significant influence on the sensitivity of the RAD kits,especially for low viral-loads samples.The detection value of RAD kits was slightly decreased,while most of them were still in the detection range with the extension of preservation time and the increase of freeze–thaw cycles.Our results showed that selecting the appropriate inactivation methods and VTM solutions is necessary during reagent development,performance evaluation,and clinical application。Haiwei Zhou Conghui Wang Jian Rao Lan Chen Tingting Ma Donglai Liu Lili Ren Sihong Xu 2021Biosafety and Health2021,3,5:3

8The orbital characters of low-energy electronic structure in iron-chalcogenide superconductor K_xFe_(2-y)Se_2显示文摘The newly discovered iron-chalcogenide superconductorKxFe2ySe2exhibits a distinct electronic structure from other iron-based superconductors.Exploiting polarization-dependent angle-resolved photoemission spectroscopy,we have determined the orbital characters of band structure in aKxFe2ySe2superconductor.To a large extent,we find thatKxFe2ySe2superconductor shares similar orbital characters with other iron-based superconductors,but with its own characteristics.For example,we have resolved two highly degenerate electron cylinders around the zone corner in the s and p geometries,respectively,indicating negligible interactions between them.Moreover,in contrast to the band calculation results,the small electron pocket around Z is found to be mainly consisted of the d z 2 orbital.The determined orbital characters would help to construct a realistic model forKxFe2ySe2.CHEN Fei GE QingQin XU Min ZHANG Yan SHEN XiaoPing LI Wei MATSUNAMI Masaharu KIMURA Shin-ichi HU JiangPing FENG DongLai 2012Chinese Science Bulletin2012,57,30:0
9The repression of oncoprotein SET by the tumor suppressor p53 reveals a p53-SET-PP2A feedback loop for cancer therapy显示文摘The oncoprotein SET is frequently overexpressed in many types of tumors and contributes to malignant initiation and progression through multiple mechanisms,including the hijacking of the tumor suppressors p53 and PP2A.Targeting aberrant SET represents a promising strategy for cancer intervention.However,the mechanism by which endogenous SET is regulated in cancer cells remains largely unknown.Here,we identified the tumor suppressor p53 as a key regulator that transcriptionally repressed the expression of SET in both normal and cancer cells.In addition,p53 stimulated PP2A phosphatase activity via p53-mediated transcriptional repression of SET,whereby SET-mediated inhibition of PP2A was alleviated.Moreover,targeting the interaction between SET and PP2A catalytic subunit(PP2Ac)with FTY720 enhanced stress-induced p53 activation via PP2A-mediated dephosphorylation of p53 on threonine 55(Thr55).Therefore,our findings uncovered a previously unknown p53-SETPP2A regulatory feedback loop.To functionally potentiate this feedback loop,we designed a combined therapeutic strategy by simultaneously administrating a p53 activator and SET antagonist in cancer cells and observed a dramatic synergistic effect on tumor suppression.Our study reveals mechanistic insight into the regulation of the oncoprotein SET and raises a potential strategy for cancer therapy by stimulating the p53-SET-PP2A feedback loop.Han Yao Wenbin Xu Yajing Liu Zhijie Cao Jia Wen Mi Zhang Zhen Wu Xiaojun Yan Zishan Jiao Zijing Zhang Jianyuan Chen Meng Zhang Wei-Guo Zhu Donglai Wang 2023Science China(Life Sciences)2023,66,1:0
10Establishment and application of national reference panels for SARS-CoV-2 antigen detection kit显示文摘To develop a national reference panel for severe acute respiratory syndrome coronavirus 2(SARS-CoV-2)antigen detection kit and establish a quality standard.The cultures of SARS-CoV-2 and other pathogens were collected to establish a national reference panel for SARS-CoV-2 antigen detection.The stability and homogeneity of the reference panel were evaluated.Based on World Health Organization(WHO)guidance and nucleic acid quantitative results,a quality standard reference panel was established.Currently,three generations of SARS-CoV-2 antigen national reference materials with batch numbers 370095–202001,370095–202202,and 370095–202203 have been successfully established.These national reference panels comprised 8 positive samples,20 negative samples,1 repetitive sample,and 1 lower detection limit sample.The stability and homogeneity of the reference panel meet the requirements.The quality standards are as follows:the positive and negative coincidence rates are 8/8 and 20/20,respectively.The 10 test results of the medium and low-concentration repetitive reference materials should be positive,and the color rendering should be uniform(or the coefficient of variance should not be higher than 20.0%).The lower detection limit should be at least 5×105 U/mL(equivalent to copies/mL),and higher concentrations above the lower detection limit must be positive.A national reference panel for the SARS-CoV-2 antigen detection kit has been established.As the standard of SARS-CoV-2 antigen reagents,the reference panel has played a crucial role in the pre-marketing quality evaluation and post-marketing quality supervision in China.Tingting Ma Donglai Liu Keliang Lyu Tingting Gao Dawei Shi Lanqing Zhao Shu Shen Yabin Tian Sihong Xu Haiwei Zhou 2023Biosafety and Health2023,5,6:0
