| 1 | Enforced PGC-1α expression promotes CD8 T cell fitness, memory formation and antitumor immunity显示文摘Memory CD8 T cells can provide long-term protection against tumors,which depends on their enhanced proliferative capacity,self-renewal and unique metabolic rewiring to sustain cellular fitness.Specifically,memory CD8 T cells engage oxidative phosphorylation and fatty acid oxidation to fulfill their metabolic demands.In contrast,tumor-infiltrating lymphocytes(TILs)display severe metabolic defects,which may underlie their functional decline.Here,we show that overexpression of proliferator-activated receptor gamma coactivator 1-alpha(PGC-1α),the master regulator of mitochondrial biogenesis(MB),favors CD8 T cell central memory formation rather than resident memory generation.PGC-1α-overexpressing CD8 T cells persist and mediate more robust recall responses to bacterial infection or peptide vaccination.Importantly,CD8 T cells with enhanced PGC-1αexpression provide stronger antitumor immunity in a mouse melanoma model.Moreover,TILs overexpressing PGC-1αmaintain higher mitochondrial activity and improved expansion when rechallenged in a tumor-free host.Altogether,our findings indicate that enforcing mitochondrial biogenesis promotes CD8 T cell memory formation,metabolic fitness,and antitumor immunity in vivo. | Nina Dumauthioz Benjamin Tschumi Mathias Wenes Bastien Marti Haiping Wang Fabien Franco Wenhui Li Isabel CLopez-Mejia Lluis Fajas Ping-Chih Ho Alena Donda Pedro Romero Lianjun Zhang | 2021 | Cellular & Molecular Immunology2021,18,7: | 3 |
| 2 | Three-dimensional numerical simulation of flow in trapezoidal cutthroat flumes based on FLOW-3D显示文摘To solve the common problem of flumes flowmeasurement accuracy without sacrificing water head, a new type of trapezoidal cutthroat flume to measure the discharge in terminal trapezoidal channels is presented.Using the computational fluid dynamic method, threedimensional flow fields in trapezoidal cutthroat flumes were simulated using the RNG k-ε three-dimensional turbulence model along with the Tru VOF technique.Simulations were performed for 12 working conditions,with discharges up to 0.075 m3$s–1 to determine hydraulic performance. Experimental data for the trapezoidal cutthroat flume in terminal trapezoidal channel were also obtained to validate the simulation results. Velocity distribution of the flume obtained from simulation analyses were compared with observed results based on timeaveraged flow field and comparison yielded a solid agreement between results from the two methods, with relative error below 10%. The results indicated that the Froude number and the longitudinal average velocity increased along the convergence section and decreased in the divergent section. In the upper throat, the Froude number was less than 0.5, which meets the water measurement requirement, and the critical flow appeared near the throat section. The maximum water head loss of the trapezoidal cutthroat flume was less than 9% of the total head, compared to the rectangular cutthroat flume,and head loss of trapezoidal cutthroat flume was significantly less. Regression models developed for upstream depth versus discharge under different working conditions were satisfactory, with a relative error of less than 2.06%, which meets the common requirements of flow measurement in irrigation areas. | 3 | AIMP1 promotes multiple myeloma malignancy through interacting with ANP32A to mediate histone H3 acetylation显示文摘Background:Multiple myeloma(MM)is the second most common hematological malignancy.An overwhelming majority of patients with MM progress to serious osteolytic bone disease.Aminoacyl-tRNA synthetase-interacting multifunctional protein 1(AIMP1)participates in several steps during cancer development and osteoclast differentiation.This study aimed to explore its role in MM.Methods:The gene expression profiling cohorts of MM were applied to determine the expression of AIMP1 and its association with MM patient prognosis.Enzyme-linked immunosorbent assay,immunohistochemistry,and Western blotting were used to detect AIMP1 expression.Protein chip analysis,RNA-sequencing,and chromatin immunoprecipitation and next-generation sequencing were employed to screen the interacting proteins and key downstream targets of AIMP1.The impact of AIMP1 on cellular proliferation was determined using 3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyltetrazolium bromide(MTT)assay in vitro and a xenograft model in vivo.Bone lesions were evaluated using tartrate-resistant acid phosphatase staining in vitro.A NOD/SCID-TIBIA mouse model was used to evaluate the effect of siAIMP1-loaded exosomes on bone lesion formation in vivo.Results:AIMP1 expression was increased in MM patients and strongly associated with unfavorable outcomes.Increased AIMP1 expression promoted MM cell proliferation in vitro and in vivo via activation of the mitogen-activated protein kinase(MAPK)signaling pathway.Protein chip assays and subsequent experiments revealed that AIMP1 interacted with acidic leucine-rich nuclear phosphoprotein 32 family member A(ANP32A)to regulate histone H3 acetylation.In addition,AIMP1 increased histone H3 acetylation enrichment function of GRB2-associated and regulator of MAPK protein 2(GAREM2)to increase the phosphorylation of extracellular-regulated kinase 1/2(p-ERK1/2).Furthermore,AIMP1 promoted osteoclast differentiation by activating nuclear factor of activated T cells c1(NFATc1)in vitro.In contrast,exosome-coated small interfering RNA of AIMP1 effectively suppressed MM progression and osteoclast differentiation in vitro and in vivo.Conclusions:Our data demonstrate that AIMP1 is a novel regulator of histone H3 acetylation interacting with ANP32A in MM,which accelerates MM malignancy via activation of the MAPK signaling pathway. | Rongfang Wei Yan Zhu Yuanjiao Zhang Wene Zhao Xichao Yu Ling Wang Chunyan Gu Xiaosong Gu Ye Yang | 2022 | Cancer Communications2022,42,11: | 2 |
