1Effects of ubiquitin-protea-some pathway on mouse sperm capacitation, acrosome reaction and in vitro fertilization显示文摘Chlortetracycline (CTC) fluorescence patterns were used to study changes in the patterns B and AR of mouse sperm after incubation with reagents that would block the UPP. They were the monoclonal antibody againstubiquitinated proteins——UCPi; the polyclonal antibodyagainst ubiquitin-anti-Ub, and a special inhibitor againstproteasome——ALLN. Furthermore, we treated the capaci-tated sperm or the eggs with these reagents separately and tested whether the normal in vitro fertilization was blocked or not. 2Assessment of urinary concentration of cotinine in Chinese pregnant women exposed to environmental tobacco smoke显示文摘Environmental tobacco smoke(ETS)is a wellknown reason of many adverse health consequences in pregnant women.Exposure of ETS during pregnancy may increase the risk of some fetal diseases in pregnant women and/or birth defects in neonates.Many countries have implemented laws to control smoking and free of second hand smoke in all enclosed workplaces.In China,antismoking law was enforced in March 2010.We aimed to assess the exposure of pregnant women to cotinine after anti-smoking law was implemented in China.The urine samples were collected from 2,100 volunteers from Zhejiang Province,China and urinary concentration of cotinine was measured using gas chromatography and mass spectrometry.Cotinine was detected in 87%of the pregnant women.The GM and 95th percentile concentration inpregnant women were 4.28 and 44.00 lg/L,respectively.Similarly,the GM and 95th concentration of cotinine were significantly higher in smoking group than in non-smoking group(P\0.0001).Our results indicate that pregnant women in China are at the high risk of exposure ETS.Further attempts are required to make strategies to find out the sources of ETS exposure and prevent smoking at public places especially during pregnancy.Yajing Lei Wenhui Liu Lizheng Fang Muhammad Sajid Hamid Akash Kanwal Rehman Hua Narenmandura Weixing Shi Weixian Lu Yingchun Xu Shuqing Chen 2014Chinese Science Bulletin2014,59,13:0
3Application of a ddRT-PCR to quantify seasonal influenza virus for viral isolation显示文摘Viral isolation in cell cultures has been regarded for decades as the“gold standard”for the laboratory diagnosis of influenza viral infections.Not all viral strains could be isolated from clinical samples.This study aimed to quantify the viral load in the samples before isolation to save working time and improve working efficiency.Four hundred samples from patients with influenza-like cases were confirmed pdmH1N1 positive(200 cases)and B Victoria(BV)positive(200 cases)by whole-genome sequencing and analyzed by ddPCR for viral load in samples before isolation,and isolation results were verified by hemagglutination(HA)assay and hemagglutination-inhibition(HI)tests.Probit regression analysis was used to calculate the isolation viral load limit with a 95%probability level by SPSS 19.0 software.The results showed that the isolation limit of viral load was 4.9×10^(4)(95%CI:2.5×10^(4)–9.0×10^(4))copies/mL for pdmH1N1 and 1.9×10^(4)(95%CI:7.8×103–3.6×10^(4))copies/mL for BV.The isolation rate of clinical samples is positively correlated with the viral load in clinical samples,which can be used for viral culture,providing important guidance for daily work.Yimeng Liu Jiachen Zhao Xiaomin Peng Guilan Lu Weixian Shi Zhaomin Feng Hui Xu Shujuan Cui Yang Pan Daitao Zhang Peng Yang Quanyi Wang 2022Biosafety and Health2022,4,5:0
