1Autophagy inhibition enhances apigenin-induced apoptosis in human breast cancer cells显示文摘Apigenin (4',5,7-trihydroxyflavone) is a member of the flavone subclass of flavonoids present in fruits and vegetables. The involvement of autophagy in the apigenin-induced apoptotic death of human breast cancer cells was investigated. Cell proliferation and viability were assessed by 3-(4,5-dimethylthiazol- 2-yl)-2,5-diphenyltetrazolium bromide (MTT) and clonogenic assays. Flow cytometry, fluorescent staining and Western blot analysis were employed to detect apoptosis and autophagy, and the role of autophagy was assessed using autophagy inhibitors. Apigenin dose- and time-dependently repressed the proliferation and clonogenic survival of the human breast cancer T47D and MDA-MB-231 cell lines. The death of T47D and MDA-MB-231 cells was due to apoptosis associated with increased levels of Caspase3, PARP cleavage and Bax/Bcl-2 ratios. The results from flow cytometry and fluorescent staining also verified the occurrence of apoptosis. In addition, the apigenin-treated cells exhibited autophagy, as characterized by the appearance of autophagosomes under fluorescence microscopy and the accumulation of acidic vesicular organelles (AVOs) by flow cytometry. Furthermore, the results of the Western blot analysis revealed that the level of LC3-II, the processed form of LC3-I, was increased. Treatment with the autophagy inhibitor, 3-methyladenine (3-MA), significantly enhanced the apoptosis induced by apigenin, which was accompanied by an increase in the level of PARP cleavage. Similar results were also confirmed by flow cytometry and fluorescence microscopy. These results indicate that apigenin has apoptosis-and autophagy-inducing effects in breast cancer cells. 2DNA methylation profile is associated with the osteogenic potential of three distinct human odontogenic stem cells显示文摘Among the various sources of human autologous stem cells,stem cells isolated from dental tissues exhibit excellent properties in tissue engineering and regenerative medicine.However,the distinct potential of these odontogenic cell lines remains unclear.In this study,we analyzed DNA methylation patterns to determine whether specific differences existed among three different odontogenic cell types.Using the HumanMethylation450 Beadchip,the whole genomes of human dental pulp stem cells(DPSCs),periodontal ligament stem cells(PDLSCs),and dental follicle progenitor cells(DFPCs)were compared.Then,the osteogenic potential of these cells was evaluated both in vitro and in vivo,and the methylation levels of certain genes related to bone formation differed among the three cell lines.P values less than 0.05 were considered to indicate statistical significance.The three cell types showed highly similar DNA methylation patterns,although specific differences were identified.Gene ontology analysis revealed that one of the most significantly different gene categories was related to bone formation.Thus,expression of cell surface epitopes and osteogenic-related transcription factors as well as the bone formation capacity were compared.The results showed that compared with DFPCs and DPSCs,PDLSCs had higher transcription levels of osteogenic-related factors,a higher in vitro osteogenic potential,and an increased new bone formation capacity in vivo.In conclusion,the results of this study suggested that the differential DNA methylation profiles could be related to the osteogenic potential of these human odontogenic cell populations.Additionally,the increased osteogenic potential of PDLSCs might aid researchers or clinicians in making better choices regarding tissue regeneration and clinical therapies.Tingting Ai Jieni Zhang Xuedong Wang Xiaowen Zheng Xueyan Qin Qian Zhang Weiran Li Wei Hu Jiuxiang Lin Feng Chen 2018Signal Transduction and Targeted Therapy2018,3,1:3
3Fabrication of lithium silicates from zeolite for CO2 capture at high temperatures显示文摘Li4Si O4 has been regarded as one of the most promising high-temperature CO2 sorbents.However,for practical applications,its CO2sorption kinetics,cycling stability and sorption properties at lower temperatures or lower CO2 concentrations have to be improved.In this contribution,four Li4Si O4 sorbents were synthesized from zeolite precursors MCM-41,MCM-48,TS-1,and ZSM-5.The CO2 uptake,cycling stability and the optimal CO2 sorption conditions were investigated.Among the samples,MCM-41-Li4Si O4 showed the best cycling stability at 650°C,with a stable reversible CO2 uptake of 29.1 wt%under 100 vol%CO2 during 20 cycles.But its sorption kinetics and CO2 uptakes at lower temperatures and lower CO2 concentrations need to be improved.We then demonstrated that the sorption kinetics can be improved by modifying the MCM-41 precursor with metals such as Al,Ti,Ca,and Na.The Na-MCM-41-Li4Si O4 sample exhibited the highest sorption rate,and reached the equilibrium sorption capacity close to the theoretical value of 36.7 wt%within 20 min.In addition,we proved that coating the MCM-41-Li4Si O4with Na2CO3and K2CO3can significantly increase the CO2uptakes at lower temperatures(e.g.550℃)and lower CO2concentrations(10–20 vol%).At 550℃ and under 20 vol%CO2,15 wt%K2CO3-MCM-41-Li4Si O4 and 10 wt%Na2CO3-MCM-41-Li4Si O4 sorbents resulted in a CO2 uptake of 32.2 wt%and 34.7 wt%,respectively,which are much higher than that of MCM-41-Li4Si O4(11.8 wt%).These two sorbents also showed good cycling stability.The promoiting mechasnim by alkali carbonate coating was discussed by a doubleshell model.Yu Zhang Yanshan Gao Benoit Louis Qiang Wang Weiran Lin 2019Journal of Energy Chemistry2019,28,6:2
4Dual roles of the Arabidopsis PEAT complex in histone H2A deubiquitination and H4K5 acetylation显示文摘Histone H2A monoubiquitination is associated with transcriptional repression and needs to be removed by deubiquitinases to facilitate gene transcription in eukaryotes.However,the deubiquitinase responsible for genome-wide H2A deubiquitination in plants has yet to be identified.In this study,we found that the previously identified PWWP-EPCR-ARID-TRB(PEAT)complex components interact with both the ubiquitin-specific protease UBP5 and the redundant histone acetyltransferases HAM1 and HAM2(HAM1/2)to form a larger version of PEAT complex in Arabidopsis thaliana.UBP5 functions as an H2A deubiquitinase in a nucleosome substrate-dependent manner in vitro and mediates H2A deubiquitination at the whole-genome level in vivo.HAM1/2 are shared subunits of the PEAT complex and the conserved NuA4 histone acetyltransferase com-plex,and are responsible for histone H4K5 acetylation.Within the PEAT complex,the PWWP components(PWWP1,PWWP2,and PWWP3)directly interact with UBP5 and are necessary for UBP5-mediated H2A deu-biquitination,while the EPCR components(EPCR1 and EPCR2)directly interact with HAM1/2 and are required for HAM1/2-mediated H4K5acetylation.Collectively,our study not onlyidentifies dual roles of thePEAT com-plex in H2A deubiquitination and H4K5 acetylation but also illustrates how these processes collaborate at the whole-genome level to regulate the transcription and development in plants.Si-Yao Zheng Bin-Bin Guan Dan-Yang Yuan Qiang-Qiang Zhao Weiran Ge Lian-Mei Tan Shan-Shan Chen Lin Li She Chen Rui-Ming Xu Xin-Jian He 2023Molecular Plant2023,16,11:1

7miR398b negatively regulates cotton immune responses to Verticillium dahliae via multiple targets显示文摘MicroRNAs(miRNAs)play essential roles in plant defense responses,although such roles have not been identified in cotton in response to the plant pathogenic fungus Verticillium dahliae.In this study,the functions of miR398b and its target genes in cotton-V.dahliae interaction were investigated.The transcript levels of miR398b were down-regulated by V.dahliae infection and miR398b overexpression in cotton made the plants more susceptible to V.dahliae.The results suggest that miR398b negatively regulates cotton resistance to V.dahliae.This may occur by miR398b repression of some CC-NBS-LRR genes via translational inhibition,interfering with defense responses and leading to cotton susceptibility to V.dahliae.Alternatively,miR398b may guide the cleavage of the mRNAs of GhCSD1,GhCSD2 and GhCCS,each of which functions in reactive oxygen species(ROS)regulation and homeostasis,thereby causing excessive ROS accumulation in miR398b-overexpressing plants in response to V.dahliae infection.This study suggests conserved and novel roles of miR398b in the cotton–V.dahliae interaction.These discoveries may be coupled with new strategies in cotton breeding programs to improve resistance to V.dahliae.Yuhuan Miao Kun Chen Jinwu Deng Lin Zhang Weiran Wang Jie Kong Steven J.Klosterman Xianlong Zhang Alifu Aierxi Longfu Zhu 2022The Crop Journal2022,10,4:0
8Sexual Risk Behavior and Satisfaction Among Men Who Have Sex With Men Living With Detectable HIV Viral Loads:A Nationwide Online Survey in China显示文摘Little is known about sexual behavior and satisfaction among men who have sex with men(MSM)with detectable HIV viral loads(VL)in China.This study aims to investigate sexual risk behavior and sexual satisfaction among MSM living with HIV in China,as well as poten-tial predictors.This cross-sectional study was conducted in February 2021 using an anonymous online survey based on WeChat.Data were collected on sociodemographics,sexual behavior,HIV clinical information,sexual satisfaction and mental health.Descriptive sta-tistics,logistic regression and multivariable logistic regression were performed.MSM with detectable HIV VL(VL+)or VL not sure were more likely to report inconsistent condom use,even with HIV-negative/unknown-status regular male partners.Having an HIV-positive regular partner and recreational drug use were positively associated with inconsistent condom use during anal sex among MSM with undetectable VL(VL-).Alcohol use before or during sexual activities was positively associated with inconsistent condom use during anal sex among VL+or VL not sure MSM.A total of 58.3%of participants reported being sexually satisfied.MSM living with HIV who were currently employed,had experienced anal sex in the prior 6 months,had VL-HIV status,or had more frequent ejaculation during sex/masturbation were more likely to report sexual satisfaction.The development of targeted education and interventions to minimize sexual risk behavior among MSM living with HIV is in dire need.Treating sexual dysfunction and developing a reasonable antiretroviral therapy regimen to achieve viral suppression can help promote sexual satisfaction in MSM living with HIV.Leiwen Fu Bingyi Wang Tian Tian Xinyi Zhou Zhen Lu Yinghui Sun Weiran Zheng Yanxiao Gao Yi-Fan Lin Hui Li Huachun Zou 2023Infectious Microbes & Diseases2023,5,3:0
