| 1 | Plankton community composition in the Three Gorges Reservoir Region revealed by PCR-DGGE and its relationships with environmental factors显示文摘To explore the relationships between community composition and the environment in a reservoir ecosystem, plankton communities from the Three Gorges Reservoir Region were studied by PCR-denaturing gradient gel electrophoresis fingerprinting. Bacterial and eukaryotic operational taxonomic units (OTUs), generated by DGGE analysis of the PCR-amplified 16S and 18S rRNA genes, were used as surrogates for the dominant 'biodiversity units'. OTU composition among the sites was heterogeneous; 46.7% of the total bacterial OTUs (45) and 64.1% of the eukaryotic OTUs (39) were identified in less than half of the sampling sites. Unweighted pair group method with arithmetic averages (UPGMA) clustering of the OTUs suggested that the plankton communities in the Xiangxi Rive sites were not always significantly different from those from the Yangtze River sites, despite clear differences in their environmental characterizations. Canonical correspondence analysis (CCA) was applied to further investigate the relationships between OTU composition and the environmental factors. | 2 | Arsenic dynamics in the rhizosphere and its sequestration on rice roots as affected by root oxidation显示文摘A pot experiment was conducted to investigate the effects of root oxidation on arsenic(As) dynamics in the rhizosphere and As sequestration on rice roots. There were significant differences(P < 0.05) in pH values between rhizosphere and non-rhizosphere soils, with pH 5.68–6.16 in the rhizosphere and 6.30–6.37 in non-rhizosphere soils as well as differences in redox potentials(P < 0.05). Percentage arsenite was lower(4%–16%) in rhizosphere soil solutions from rice genotypes with higher radial oxygen loss(ROL) compared with genotypes with lower ROL(P < 0.05). Arsenic concentrations in iron plaque and rice straw were significantly negatively correlated(R =-0.60, P < 0.05). Genotypes with higher ROL(TD71 and Yinjingruanzhan) had significantly(P < 0.001) lower total As in rice grains(1.35 and 0.96 mg/kg, respectively) compared with genotypes with lower ROL(IAPAR9, 1.68 mg/kg; Nanyangzhan 2.24 mg/kg) in the As treatment, as well as lower inorganic As(P < 0.05). The present study showed that genotypes with higher ROL could oxidize more arsenite in rhizosphere soils, and induce more Fe plaque formation, which subsequently sequestered more As. This reduced As uptake in aboveground plant tissues and also reduced inorganic As accumulation in rice grains. | 4 | Biodegradation of nicosulfuron by a novel Alcaligenes faecalis strain ZWS11显示文摘A bacterial strain ZWS11 was isolated from sulfonylurea herbicide-contaminated farmland soil and identified as a potential nicosulfuron-degrading bacterium. Based on morphological and physicochemical characterization of the bacterium and phylogenetic analysis of the 16 S r RNA sequence, strain ZWS11 was identified as Alcaligenes faecalis. The effects of the initial concentration of nicosulfuron, inoculation volume, and medium p H on degradation of nicosulfuron were investigated. Strain ZWS11 could degrade 80.56% of the initial nicosulfuron supplemented at 500.0 mg/L under the conditions of p H 7.0, 180 r/min and30°C after incubation for 6 days. Strain ZWS11 was also capable of degrading rimsulfuron,tribenuron-methyl and thifensulfuron-methyl. Four metabolites from biodegradation of nicosulfuron were identified, which were 2-aminosulfonyl-N, N-dimethylnicotinamide(M1), 4, 6-dihydroxypyrimidine(M2), 2-amino-4, 6-dimethoxypyrimidine(M3) and2-(1-(4,6-dimethoxy-pyrimidin-2-yl)-ureido)-N,N-dimethyl-nicotinamide(M4). Among the metabolites detected, M2 was reported for the first time. Possible biodegradation pathways of nicosulfuron by strain ZWS11 were proposed. The degradation proceeded mainly via cleavage of the sulfonylurea bridge, O-dealkylation, and contraction of the sulfonylurea bridge by elimination of a sulfur dioxide group. | 8 | Shaking table comparative test and associated study of a stepped wall-frame structure显示文摘A new structural system c alled a stepped wall-frame structure is proposed in this study to solve the bottom yielding problem of RC frames, which widely occurred during previous earthquakes such as the Wenchuan and Yushu earthquakes in China. A 1/5 scale ordinary RC frame model and a stepped wall-frame model were subjected to shake table motions together to study the seismic behavior of the new structural system. This paper presents the dynamic characteristics, the seismic responses and the failure and collapse mechanism of the two models under low, moderate and high intensity shaking. The test results and further analysis demonstrate that the seismic performance of stepped wall-frame structures is superior to ordinary RC frames in terms of the well-controlled deformation pattern and more uniformly distributed damage. The operating system as the manager of the computing resources, is also facing new challenges.For IoE systems and applications, an innovative operating system is missing to support services, collect data, and manage the things. However, IoE applications are all around us and increasingly becoming a necessity rather than a luxury. Therefore, it is important that the process of configuring and adding devices to the IoE is not a complex one. The ease of installation, operation, and maintenance of devices on the network unarguably plays an important role in the wide spread use of IoE devices in smart homes and everywhere else. In this paper, we propose Sofie, which is a smart operating system for the IoE. We also give the design of Sofie. Sofie can be implemented via different IoT systems, such as Home Assistant, openHAB, and so on. In order to implement Sofie to get some early experience, we leverage Home Assistant to build a prototype for the smart home.
