1Parallel selection of distinct Tof5 alleles drove the adaptation of cultivated and wild soybean to high latitudes显示文摘Photoperiod responsiveness is a key factor limiting the geographic distribution of cultivated soybean and its wild ancestor.In particular,the genetic basis of the adaptation in wild soybean remains poorly understood.In this study,by combining whole-genome resequencing and genome-wide association studies we identified a novel locus,Time of Flowering 5(Tof5),which promotes flowering and enhances adaptation to high latitudes in both wild and cultivated soybean.By genomic,genetic and transgenic analyses we showed that Tof5 en-codes a homolog of Arabidopsis thaliana FRUITFULL(FUL).Importantly,further analyses suggested that different alleles of Tof5 have undergone parallel selection.The Tof5H1 allele was strongly selected by humans after the early domestication of cultivated soybean,while Tof5H2 allele was naturally selected in wild soybean,and in each case facilitating adaptation to high latitudes.Moreover,we found that the key flowering repressor E1 suppresses the transcription of Tof5 by binding to its promoter.In turn,Tof5 physically associates with the promoters of two important FLOWERING LOCUS T(FT),FT2a and FT5a,to upregulate their transcription and promote flowering under long photoperiods.Collectively,ourfindings provide insights into how wild soybean adapted to high latitudes through natural selection and indicate that cultivated soybean underwent changes in the same gene but evolved a distinct allele that was artificially selected after domestication.Lidong Dong Qun Cheng Chao Fang Lingping Kong Hui Yang Zhihong Hou Yongli Li Haiyang Nan Yuhang Zhang Qingshan Chen Chunbao Zhang Kun Kou Tong Su Lingshuang Wang Shichen Li Haiyang Li Xiaoya Lin Yang Tang Xiaohui Zhao Sijia Lu Baohui Liu Fanjiang Kong 2022Molecular Plant2022,15,2:5
2Systematic analysis of intron size and abundance parameters in diverse lineages显示文摘All eukaryotic genomes have genes with introns in variable sizes.As far as spliceosomal introns are concerned,there are at least three basic parameters to stratify introns across diverse eukaryotic taxa:size,number,and sequence context.The number parameter is highly variable in lower eukaryotes,especially among protozoan and fungal species,which ranges from less than4%to 78%of the genes.Over greater evolutionary time scales,the number parameter undoubtedly increases as observed in higher plants and higher vertebrates,reaching greater than 12.5 exons per gene in average among mammalian genomes.The size parameter is more complex,where multiple modes appear at work.Aside from intronless genes,there are three other types of intron-containing genes:half-sized,minimal,and size-expandable introns.The half-sized introns have only been found in a limited number of genomes among protozoan and fungal lineages and the other two types are prevalent in all animal and plant genomes.Among the size-expandable introns,the sizes of plant introns are expansion-limited in that the large introns exceeding 1000 bp are fewer in numbers and transposon-free as compared to the large introns among animals,where the larger introns are filled with transposable elements and appear expansion-flexible,reaching several kilobasepairs(kbp)and even thousands of kbp in size.Most of the intron parameters can be studied as signatures of the specific splicing machineries of different eukaryotic lineages and are highly relevant to the regulation of gene expression and functionality.In particular,the transcription-splicing-export coupling of eukaryotic intron dispensing leads to a working hypothesis that all intron parameters are evolved to be efficient and function-related in processing and routing the spliced transcripts.WU JiaYan XIAO JingFa WANG LingPing ZHONG Jun YIN HongYan WU ShuangXiu ZHANG Zhang YU Jun 2013Science China(Life Sciences)2013,56,10:4
3A universal pipeline for mobile mRNA detection and insights into heterografting advantages under chilling stress显示文摘Heterografting has long been used to enhance the chilling tolerance of temperature-sensitive crops,including watermelon,whose mechanism is known to involve bidirectional long-distance mRNA movements.Despite several studies reporting on mobile mRNA(mb-mRNA)profiles in plants,accurate identification of mb-mRNAs is challenging owing to an array of technical problems.Here,we developed a bioinformatical pipeline that took most of the known technical concerns into consideration and is considered to be a universal tool for mb-mRNA detection in heterografts.By applying this pipeline to a commercial watermelon–bottle gourd heterografting system,we detected 130 and 1144 mb-mRNAs upwardly and 167 and 1051 mb-mRNAs downwardly transmitted under normal and chilling-stress conditions,respectively.Quantitative real-time PCR indicated a high accuracy rate(88.2%)of mb-mRNA prediction with our pipeline.We further revealed that the mobility of mRNAs was not associated with their abundance.Functional annotation and classification implied that scions may convey the stress signal to the rootstock,subsequently triggering energy metabolism reprogramming and abscisic acid-mediated stress responses by upward movement of effective mRNAs,ultimately leading to enhanced chilling tolerance.This study provides a universal tool for mb-mRNA detection in plant heterografting systems and novel insights into heterografting advantages under chilling stress.Ying Wang Lingping Wang Nailin Xing Xiaohua Wu Xinyi Wu Baogen Wang Zhongfu Lu Pei Xu Ye Tao Guojing Li Yuhong Wang 2020Horticulture Research2020,7,1:3
4Agronomical selection on loss-of-function of GIGANTEA simultaneously facilitates soybean salt tolerance and early maturity显示文摘Salt stress and flowering time are major factors limiting geographic adaptation and yield productivity in soybean(Glycine max).Although improving crop salt tolerance and latitude adaptation are essential for efficient agricultural production,whether and how these two traits are integrated remains largely unknown.Here,we used a genome-wide association study to identify a major salt-tolerance locus controlled by E2,an ortholog of Arabidopsis thaliana GIGANTEA(GI).Loss of E2 function not only shortened flowering time and maturity,but also enhanced salt-tolerance in soybean.E2 delayed soybean flowering by enhancing the transcription of the core flowering suppressor gene E1,thereby repressing Flowering Locus T(FT)expression.An E2 knockout mutant e2^(CR) displayed reduced accumulation of reactive oxygen species(ROS)during the response to salt stress by releasing peroxidase,which functions in ROS scavenging to avoid cytotoxicity.Evolutionary and population genetic analyses also suggested that loss-of-function e2 alleles have been artificially selected during breeding for soybean adaptation to high-latitude regions with greater salt stress.Our findings provide insights into the coupled selection for adaptation to both latitude and salt stress in soybean;and offer an ideal target for molecular breeding of early-maturing and salt-tolerant cultivars.Lidong Dong Zhihong Hou Haiyang Li Zhaobo Li Chao Fang Lingping Kong Yongli Li Hao Du Tai Li Lingshuang Wang Milan He Xiaohui Zhao Qun Cheng Fanjiang Kong Baohui Liu 2022Journal of Integrative Plant Biology2022,64,10:3
5Neuroprotective effects of human telomerase reverse transcriptase on beta-amyloid fragment 25-35-treated human embryonic cortical neurons显示文摘BACKGROUND:Numerous current studies have suggested that human telomerase reverse transcriptase(hTERT) gene has neuroprotective effects and can inhibit apoptosis induced by various cytotoxic stresses;however,the mechanism of action remains unknown. OBJECTIVE:To evaluate the neuroprotective effects and possible mechanism of action of hTERT gene transfection in human embryonic cortical neurons treated with beta-amyloid fragment 25-35 (Aβ_(25-35)). DESIGN,TIME AND SETTING:The randomized,controlled and molecular biological studies were performed at the Department of Anatomy and Brain Research,Zhongshan School of Medicine,Sun Yat-sen University,China,from September 2005 to June 2008. MATERIALS:AdEasy-1 Expression System was gifted by Professor Guoquan Gao from Sun Yat-Sen University,China.Human cortical neurons were derived from 12-20 week old aborted fetuses,obtained from the Guangzhou Maternal and Child Health Hospital,China.Mouse anti-Cdk5 and mouse anti-p16 monoclonal antibodies(Lab Vision,USA),and mouse anti-hTERT monoclonal antibody(Epitomics,USA),were used in this study. METHODS:(1) Recombinant adenovirus vectors,encoding hTERT(Ad-hTERT) and green fluorescent protein(Ad-GFP),were constructed using the AdEasy-1 Expression System.Human embryonic cortical neurons in the Ad-hTERT group were transfected with Ad-hTERT for 1-21 days. Likewise,human embryonic cortical neurons in the Ad-GFP group were transfected with Ad-GFP for 1-21 days.Human embryonic cortical neurons in the control group were cultured as normal.(2) Human embryonic cortical neurons in the Ad-hTERT group were treated with 10μmol/L Aβ_(25-35) for 24 hours.Normal human embryonic cortical neurons treated with 10μmol/LAβ_(25-35) for 24 hours served as a model group.Human embryonic cortical neurons in the Ad-GFP and control groups were not treated with Aβ_(25-35). MAIN OUTCOME MEASURES:Expression of hTERT in human embryonic cortical neurons was evaluated by immunocytochemical staining and Western blot assay.Telomerase activity was measured using a PcR-based telomeric repeat amplification protocol(TRAP) ELISA kit.Neural activity in human embryonic cortical neurons was examined by MTT assay;apoptosis was measured using TUNEL assay;and Cdk5 and p16 protein expressions were measured by Westem blot. RESULTS:Expression of hTERT protein was significantly increased and peaked at day 3 post-transfection in the Ad-hTERT group.No hTERT expression was detected in the Ad-GFP and control groups.Telomerase activity was significantly greater in the Ad-hTERT group compared with the Ad-GFP and control groups(P<0.01).Compared with the control group,cell activity was significantly decreased(P<0.05),and cell apoptotic rate,Cdk5 and p16 expression were significantly increased(P<0.01) in the model group.Compared with the model group,cell activity was increased in the Ad-hTERT group,and peaked at day 3 post-transfection(P<0.05). Neuroprotective effects also peaked at day 3 post-transfection;and the apoptotic rate,Cdk5 and p16 expression significantly decreased(P<0.01). 6Anion Exchange Membrane Based on Interpenetrating Polymer Network with Ultrahigh Ion Conductivity and Excellent Stability for Alkaline Fuel Cell显示文摘A high-performance anion exchange membrane(AEM)is critical for the development of alkaline fuel cell.In this work,AEMs with an interpenetrating polymer network(IPN)are synthesized.An electron microscope clearly reveals a highly efficient“ion channel”network,which is constructed with a small amount of cation exchange groups.This specially designed ion channel leads to extraordinary hydroxide conductivity(e.g.,257.8 mS cm^(-1) at 80℃)of IPN AEMs at moderate ion exchange capacity(IEC=1:75 mmol g^(-1)),as well as excellent long-term alkaline stability at harsh condition which showed that 81%of original conductivity can be retained after a long time for 1248 hours.Moreover,a remarkable peak power density of 1.20 Wcm^(-2)(0.1 MPa backpressure)with nonprecious metal(FeNx-CNTs)as oxygen reduction reaction(ORR)catalyst in a fuel cell test was achieved.This work offers a general strategy to prepare high-performance AEMs based on IPN structure design.Lingping Zeng Qian He Yunchuan Liao Shangyi Kuang Jianchuan Wang Wei Ding Qiang Liao Zidong Wei 2020Research2020,,1:2
7GIS-based Risk Zonation of Rainstorm Disaster in Guizhou Province显示文摘[Objective] This paper aimed to study the risk zoning of rainstorm in Guizhou based on GIS. [Method] Taking Guizhou as study area, 1 km×1 km grid data as evaluation unit, and by dint of daily precipitation in meteorological station in Guizhou from 1961 to 2008, the rainstorm risk zoning system was constructed from the aspects of disaster-stricken dangers, suffering flexibility, disaster environment sensitivity and disaster prevention or mitigation; based on the level analysis method to determine factor weight, the risk assessment model based on GIS was set up to evaluate the four sub-indicators and risks and to get the rainstorm disaster in Guizhou in the end. [Result] The risk assessment and zonation results showed a general trend that the risk level decreased from the central to all around. The low risk area distributed in the northwest of Guizhou province because of less heavy rains and high capacity of rainstorm disaster resistant, while high risk area mainly distributed in the west-central of Guizhou due to concentration rainstorms, large terrain undulation and low coverage rate of forest. Especially, according to Anshun, the high risk area took up 98.02% of the city, and the Gangwu County, where a super-large geological disaster concurred in 2010 is located at the high risk area, which showed that the risk assessment coincided with the actual situation. 13Development and validation of InDel markers for identification of QTL underlying flowering time in soybean显示文摘Soybean [Glycine max(L.) Merrill] is a major plant source of protein and oil. An accurate and well-saturated molecular linkage map is a prerequisite for forward genetic studies of gene function and for modern breeding for many useful agronomic traits. Next-generation sequence data available in public databases provides valuable information and offers new insights for rapid and efficient development of molecular markers. In this study, we attempted to show the feasibility and facility of using genomic resequencing data as raw material for identifying putative In Del markers. First, we identified 17,613 In Del sites among 56 soybean accessions and obtained 12,619 primer pairs. Second, we constructed a genetic map with a random subset of 2841 primer pairs and aligned 300 polymorphic markers with the 20 consensus linkage groups(LG). The total genetic distance was 2347.3 c M and the number of mapped markers per LG ranged from 10 to 23 with an average of 15 markers. The largest and smallest genetic distances between adjacent markers were 52.3 c M and 0.1 cM, respectively. Finally, we validated the genetic map constructed by newly developed In Del markers by QTL analysis of days to flowering(DTF) under different environments. One major QTL(qDTF4) and four minor QTL(qDTF20, qDTF13, qDTF12,and q DTF11) on 5 LGs were detected. 15Stent-assisted coiling of intracranial carotid ophthalmic segment aneurysm segment aneurysms:Long-term follow-up from a single center显示文摘Background:To evaluate the efficacy of stent-assisted coiling(SAC)for the treatment of carotid ophthalmic segment aneurysm segment aneurysms(OSAs)of the internal carotid artery(ICA)through detailed long-term follow-up of a large patient cohort.Methods:We retrospectively analyzed 88 consecutive patients with OSAs between January 2009 and January 2020 at our center.Angiographic results were evaluated using the modified Raymond grading system and clinical outcomes were evaluated using the mRS scale.The primary endpoints were major aneurysm recurrence and poor clinical outcomes for at least 18 months of follow-up.The patients were asked to attend clinical follow-up assessments and possibly undergo DSA or MR via telephone.Results:We enrolled 88 patients with 99 OSAs treated with coiling,of whom 76 were treated with SAC.The coiling procedures were successful in all 88 patients.Overall,complications occurred in 8 patients(9.1%).No procedure-related mortality was observed.67(76.1%)experienced immediate aneurysm occlusion at the end of the procedure.Long-term angiographic follow-up(18 months)was available in 45/88 aneurysms(51%)(average 18.7±5.2 months).Four patients continued their follow-up for 5 years after initial aneurysm treatment.After a clinical follow-up time of 28.7 months(range,12-51 months),85 patients(95.5%)achieved favorable clinical outcomes(mRS scores of 0-2).Conclusions:This study indicates that SAC treatment is a safe and effective therapeutic alternative for ruptured and unruptured OSAs.The procedural risks are low with relatively long-term effectiveness.Wenquan Gu Geng Zhou Aizada Aldiyarova Tengyue Liu Yi Zhang Weidong Liu Lingping Meng Binxian Gu MingHua Li Ming Su Chen Su Aihua Liu Wu Wang 2023Journal of Interventional Medicine2023,6,3:0
16A New Post-hoc Flat Field Measurement Method for the Solar X-Ray and Extreme Ultraviolet Imager Onboard the FengYun-3E Satellite显示文摘Extreme ultraviolet(EUV)observations are widely used in solar activity research and space weather forecasting since they can observe both the solar eruptions and the source regions of the solar wind.Flat field processing is indispensable to remove the instrumental non-uniformity of a solar EUV imager in producing high-quality scientific data from original observed data.FengYun-3E(FY-3E)is a meteorological satellite operated in a Sunsynchronous orbit,and the routine EUV imaging data from the Solar X-ray and Extreme Ultraviolet Imager(X-EUVI)onboard FY-3E has the characteristic of concentric rotation.Taking advantage of the concentric rotation,we propose a post-hoc flat field measurement method for its EUV 195A channel in this paper.This method removes the small-scale and time-varying component of coronal activities by taking the median value for each pixel along the time axis of a concentric rotation data cube,and then derives the large-scale and invariable component of the quiet coronal radiation,and finally generates a flat field image.The flat field can be generated with cadences from hundreds of minutes(one orbit)to several days.Higher flat field accuracy can be achieved by employing more data.Further analysis shows that our method is able to measure the instrumental spot-like nonuniformity possibly caused by contamination on the detector,which mostly disappears after the in-orbit selfcleaning process.It can also measure the quasi-periodic grid-like non-uniformity,possibly from the obscuration of the support mesh on the rear filter.After flat field correction,these instrumental non-uniformities from the original data are effectively removed.Moreover,the X-EUVI 195A data after dark and flat field corrections are consistent with the 193A imaging data from the Atmospheric Imaging Assembly onboard the Solar Dynamics Observatory,verifying the suitability of the method.The post-hoc method does not occupy observation time,which is advantageous for space weather operations.Our method is not only suitable for FY-3E/X-EUVI but also a candidate method for the flat field measurement of future solar EUV telescopes.Qiao Song Xianyong Bai Bo Chen Xiuqing Hu Yajie Chen Zhenyong Hou Xiaofan Zhang Lingping He Kefei Song Peng Zhang Jing-Song Wang Xiaoxin Zhang Weiguo Zong Jinping Dun Hui Tian Yuanyong Deng 2022Research in Astronomy and Astrophysics2022,22,10:0
