2Pancreatic microcirculatory impairment in experimental acute pancreatitis in rats显示文摘AIM: To study the feature of pancreatic microcirculatoryimpairment, especially the initial changes, in caerulein-induced experimental acute pancreatitis (AP).METHODS: The pancreatic microcirculation of caerulein-induced AP model was studied by intravital fluorescencemicroscopy with FITC-labeled erythrocytes (FITC-RBC),scanning electron microscopy of vascular corrosion casts,and light microscopy of Chinese ink-injected/cleared tissues.RESULTS: Animals in caerulein-treated group showedhyperamylemia (× 2), pancreatic oedema, infiltration ofinflammatory cells in pancreas. Constrictions of intralobulararteriolar sphincters, presence of vacuoles in all layers ofsphincter, and gross irregularity in capillary network of aciniwere found in the AP specimens. 3Estimation of internal nutrient release in large shallow Lake Taihu, China显示文摘Based on field investigation of wave, sediment suspension and the changes in nutrient concentration of the water column in Lake Taihu, China, we proposed two release models to quantify nutrient release under static and dynamic conditions, respectively. Under static conditions, nutrient release from sediments to the overlying water mainly depends on chemical diffusion induced by concentration gradient, in which the nutrient release is controlled by the temperature, dissolved oxygen concentration in the sediment-water interface, oxidation-reduction potential and the concentration difference between porewater and overlying water. Under dynamic condition (or disturbed condition),both dissolved and particulate nutrients in sediments are released into the water column because of wind-induced sediment suspension. The amount of nutrient release under dynamic conditions is larger than that under the static condition. The release of dissolved nutrients, however, does not increase because the wind induced turbulence made oxidation of metallic elements such as Fe (ferric iron), Mn which are capable of precipitating soluble reactive phosphate (SRP). Under dynamic conditions, therefore, the release of total phosphorus (TP) increases dramatically but the release of SRP is close to those under static conditions. In sediments of Lake Taihu, high Fe content leads to a high ratio of Fe to P contents in sediments (Fe:P ratio). Under dynamic conditions, therefore, nutrient release is controlled by the intensity of disturbance, sediment consolidation and nutrient content in sediments.As for dissolved nutrients, especially SRP, the release is also controlled by the intensity of dynamic re-oxidation, Fe content in sediments and nutrient concentration gradient between porewater and overlying water. Based on these two release modes, the release flux in Lake Taihu has been estimated. In the static condition (i.e. laboratory experimental condition), total release of NH4+-N for whole lake is ca. 10,000 ton/a, and PO43--P is ca. 900 ton/a. In the dynamic condition, nutrient release following sediment suspension was estimated according to three different intensities of wind forcing which were defined as 'calm' (wind speed is less than 2 m/s), 'gentle' (wind speed is greater than 2 m/s and less than 6 m/s) and 'gust' (wind speed is greater than 6 m/s). The release rate in the condition of 'calm' was estimated in terms of the nutrient release in the laboratory experimental static condition; whereas the release rate in conditions of 'gentle' and 'gust' was estimated in terms of measurement during sediment resuspension conducted in flume experiments. With the observation of wind velocity and frequency in 2001, each type of wind forcing took the frequency of 12%, 82% and 6% for 'calm', 'gentle' and 'gust', respectively. 4Effects of the tyrosine protein kinase inhibitor genistein on the proliferation,activation of cultured rat hepatic stellate cells显示文摘AIM: Hepatic stellate cell (HSC) plays a pivotal role inliver fibrosis and is considered as the therapeutic targetfor the treatment of hepatic fibrosis. Tyrosine proteinkinase plays an important role in the proliferation,activation of HSC. The purpose of the study is toinvestigate the effects of the tyrosine protein kinaseinhibitor genistein onthe proliferation and activationof cultured rat HscMETHODS: Rat HSC were isolated from Wistar rats by insitu perfusion of collagenase and pronase and single-stepdensity Nycodenz gradient. Culture-activated HSC wereserum-starved and incubated with 10-3 to 10-5 mol/Lconcentration of genistein for 24, 48 or 72 h. In PDGF-induced HSC proliferation, HSC were stimulatel with L0μg@L-1PDGF-BB for 15 min, and then treaeed with genisteinfor the same time. Cell proliferation was measured byMTr assay and based on flow cytometric analysis of cellcycle. The a-smooth muscle actin (α-SMA) expression inHSC was studied with confocal laser microscopy and flowcytometry. c-fos c-jun and cyclin D1 expression in HSCwas also detected by flow cytometry.RESULTS: Genistein inhibited basal and PDGF-inducedproliferation of HSC at the concentration of 10-8 to 10-5mol/L, and treatment with 10-7 mol/L concentration ofgenistein for 48 h inhibited the HSC proliferationsignificantly (the inhibition rate was 70.3 %, P<0.05).Immunofluorescence detected by confocal lasermicroscopy and flow cytometry showed that treatmentwith 10-7 mol/L genistein for 48 h suppressed theexpression of α-SMA significantly in HSC (the specificfluorescence intensity were 60.2±21.5 vs 35.3±11.6and 12.8± 10.4 vs9.54±6.39, respectively, both P<0.05).The intensity of c-fos, c-jun and cydin D1 expression ofHSCs treated with 10-7 mol/L genistein for 48 h wasalso significantly decreased compared with the controls.CONCLUSION: Genistein influences proliferation of HSC,suppresses the expression of α-SMA in HSC and tinhibits the intensity of c-fos, c-jun and cyclin D1expression of HSCs. 5HLA class Ⅰ expression in primary hepatocellular carcinoma显示文摘AIM:To investigate whetherCTL vaccine therapy is suitable for primary hepatocellular carcinoma(HCC)from the viewpoint of HLA classIantigens expression.METHODS:The immunocytochemistry,image analysis,flow cytometry,and labeled streptavidin bioti(LSAB)method of immunohistochemistry were applied respectively to study 4HCCcell lines(e.g.Alexander,HepG2,SMMC-7721,andQGY-7703)cultured in vitro and 6frozen tissue specimens of HCC.RESULTS:The positive control cell line Raji had very strong positive staining,Most mitotic and nonmitotic cells of the 4HCCcell lines had various intensity of HLAclassⅠantigens expression.The negative control cell K562 and the control slides of all the cell lines had no positive staining,In the 6HCCspecimens immunohistochemically studied,histological normal hepatocytes had no or very weak positive staining and the liver sinus had very strong positive staining.Most HCCcells in the sections from the 6HCCspecimens had strong positive HLAclassⅠantigens staining.The positive staining was located in the cytoplasm,the perinuclear area,and at the cell membrane of the liver cancer cells.Flow cytometry also revealed that Raji and those 4HCCcell lines had strong HLAclassⅠantigens expression.which was confirmed quantitatively by the image analysis.It showed that the objective grayscale values of Raji and those 4HCCcell lines were significantly different from that of K562(Raji114.04±10.94,Alexander165.97±5.35,HepG2167.02±12.60,QGY-7703161.46±7.13,SMMC-7721 165.93±5.21,K562244.89±4.60,P<0.01).Significant differences were also found ebtween Raji and the 4HCCcell lines.CONCLUSION:HCC cells express HLA classI antigens strongly,Fromthis point of view.the active specific immunotherapy of CTL vaccine is suitable and practicable for HCC.Jian Huang Mei-Ying Cai Da-Peng Wei Immunology Department,West China University of Medical Sciences,Chengdu 610044,Sichuan Province,China 2002World Journal of Gastroenterology2002,8,4:33
6Laparoscopic total mesorectal excision of low rectal cancer with preservation of anal sphincter:A report of 82 cases显示文摘AIM: To assess the feasibility and efficacy of laparoscopic total mesorectal excision (LTME) of low rectal cancer with preservation of anal sphincter.METHODS: From June 2001 to June 2003, 82 patients with low rectal cancer underwent laparoscopic total mesorectal excision with preservation of anal sphincter. The lowest edge of tumors was below peritoneal reflection and 1.5-7 cm from the dentate line (1.5-5 cm in 48 cases, 5-7 cm in 34 cases).RESULTS: LTME with anal sphincter preservation was performed on 82 randomized patients with low rectal cancer, and 100 % sphincter preservation rate was achieved. There were 30 patients with laparoscopic low anterior resection (LLAR) at the level of the anastomosis below peritoneal reflection and 2 cm above from the dentate line; 27 patients with laparoscopic ultralow anterior resection (LULAR) at the level of anastomoses 2 cm below from the dentate line; and 25 patients with laparoscopic coloanal anastomoses (LC AA) at the level of the anastomoses at or below the dentate line. No defunctioning ileostomy was created in any case. The mean operating time was 120 minutes (ranged from 110-220 min), and the mean operative blood loss was 20 mL (ranged from 5-120 mL).Bowel function was restored and diet was resumed on day 1 or 2 after operation. The mean hospital stay was 8 days (ranged from 5-14). Postoperative analgesics were used in 45 patients. After surgery, 2 patients had urinary retention, one had anastomotic leakage, and another 2patients had local recurrence one year later. 9A new scoring system for assessment of liver function after successful hepatectomy in patients with hepatocellular carcinoma显示文摘BACKGROUND:Whether a major liver resection is safe has been judged mainly from the patient's hepatic reserve.However,a safe limit for liver resection does not exist yet.This study aimed to construct a new scoring system as a guide to determine a safe limit for liver resection and avoid liver dysfunction after hepatectomy.METHODS:Eighty-six patients with hepatocellular carcinoma who had undergone hepatectomy in West China Hospital from March 2007 to June 2010 were reviewed.The patients were classified according to the levels of total bilirubin after hepatectomy and the parameters in the perioperative period were compared.Receiver operating characteristic (ROC) analysis was made to assess the liver function compensatory (LFC) value to predict liver dysfunction of the patients after hepatectomy.LFC value is defined as the preoperative KICG value×22.487+standard remnant liver volume (SRLV)×0.020.RESULTS:Patients were classified into group Ⅰ (normal group,n=69) and group Ⅱ (with total bilirubin >85.5 μmol/L for 7 days after hepatectomy,n=17) based on the levels of total bilirubin after hepatectomy.Group II was further divided into two subgroups:recovered subgroup (n=14) and fatal subgroup (n=3).There were no significant differences in preoperative data or intraoperative findings except the indocyanine green test parameters (KICG and ICG R15) and SRLV.ROC analysis showed that the sensitivity and specificity of an LFC value ≤13.01 were 94.1% and 82.6% respectively for predicting liver dysfunction of the patients after hepatectomy.CONCLUSIONS:The LFC value appears to be a good predictor of postoperative liver dysfunction in patients who undergo hepatectomy for HCC.An expected LFC value of 13.01 seems to be a safe limit for liver resection.Zheng-Gui Du,Bo Li,Yong-Gang Wei,Jie Yin,Xi Feng and Xi Chen Department of Liver and Vascular Surgery,West China Hospital,Sichuan University,Chengdu 610041,China 2011Hepatobiliary & Pancreatic Diseases International2011,10,3:23
10Organ fibrosis inhibited by blocking transforming growth factor-β signaling via peroxisome proliferator-activated receptor γ agonists显示文摘BACKGROUND:Organ fibrosis has been viewed as one of the major medical problems, which can lead to progressive dysfunction of the liver, lung, kidney, skin, heart, and eventually death of patients. Fibrosis is initiated by a variety of pathological, physiological, biochemical, and physical factors. Regardless of their different etiologies, they all share a common pathogenetic process: excessive activation of the key profibrotic cytokine, transforming growth factor-β (TGF-β). Peroxisome proliferator-activated receptor γ (PPARγ), a ligand-activated transcription factor of the nuclear receptor superfamily, has received particular attention in recent years, because the activation of PPARγ by both natural and synthetic agonists could effectively inhibit TGF-β-induced profibrotic effects in many organs. DATA SOURCES: The English-language medical databases, PubMed, Elsevier and SpringerLink were searched for articles on PPARγ, TGF-β, and fibrosis, and related topics. RESULTS: TGF-β is recognized as a key profibrotic cytokine. Excessive activation of TGF-β increases synthesis of extracellular matrix proteins and decreases their degradation, associated with a gradual destruction of normal tissue architecture and function, whereas PPARγ agonists inhibit TGF-β signal transduction and are effective antifibrogenic agents in many organs including the liver, lung, kidney, skin and heart. CONCLUSIONS: The main antifibrotic activity of PPARγ agonists is to suppress the TGF-β signaling pathway by so-called PPARγ-dependent effect. In addition, PPARγ agonists, especially 15d-PGJ2, also exert potentially antifibrotic activity independent of PPARγ activation. TGF-β1/Smads signaling not only plays many essential roles in multiple developmental processes, butalso forms cross-talk networks with other signal pathways, and their inhibition by PPARγ agonists certainly affects the cytokine networks and causes non-suspected side-effects. 11Pathomorphological study on location and distribution of Kupffer cells in hepatocellular carcinoma显示文摘AIM: To clarify the location and distribution of Kupffer cells in hepatocellular carcinoma (HCC), and to investigate their role in hepatocarcinogenesis.METHODS: Kupffer cells were immunohistochemically stained by streptavadin-peroxidase conjugated method (S-P). The numbers of Kupffer cells in cancerous, para-cancerous and adjacent normal liver tissues of 48 HCCs were comparatively examined.RESULTS: The mean number of Kupffer cells in cancerous,para-cancerous and adjacent normal liver tissues was 12.7±6.8, 18.1±8.2 and 18.9±7.9 respectively. The number of Kuppfer cells in cancerous tissues was significantly lower than that in para-cancerous tissues (t=2.423, P<0.05) and adjacent normal liver tissues (t=2.52t, P<0.05). As tumor size increased, the number of Kupffer cells in cancerous tissues significantly decreased (F=4.61, P<0.05). 12Improved two-cuff technique for orthotopic liver transplantation in rat显示文摘BACKGROUND: The first orthotopic liver transplantatio in rat (ROLT) was reported by Lee in 1973. Kamada inno vatively applied cuff technique to ROLT in 1979. However the operative procedures were highly demanding and th operative mortality was relatively high. The purpose of thi study was to improve the model of ROLT, simplify opera tive procedures, and enhance the successful rate of opera tion. METHODS: Orthotopic liver transplantation was per formed in 160 Wistar rats by improved two-cuff technique The portal vein between donor and recipient was anasto mosed with the cuff technique. The same method was use to anastomose the infrahepatic vena cava. The suprahepati vena cava and the hepatic artery were anastomosed by m crovascular suturing and the bile duct was anastomosed en to end by a Teflon catheter. RESULTS: The average time for donor operation, graf preparation and anhepatic phase was 31 minutes, 14 mi nutes and 13 minutes, respectively. The anastomosis tim for the suprahepatic vena cava, portal vein, infrahepatic ve na cava, hepatic artery and bile duct was 7 minutes, 2 mi nutes, 2 minutes, 8 minutes and 1 minute, respectively The main causes for operative mortality were pneumotho rax, anesthesia, air embolism and massive bleeding, an the successful rate of operation was 92.5%. The causes fo death after operation were stoma bleeding, infection, b liary obstruction and graft failure. 13Transforming growth factor-β1 gene polymorphisms associated with chronic obstructive pulmonary disease in Chinese population显示文摘Aim: To determine the frequencies of polymorphism and haplotype in the transforming growth factor-beta 1 (TGF-β1) gene promoter in the Chinese population and to investigate the susceptibility of this population to chronic obstructive pulmonary disease (COPD). Methods: The target fragments of the TGF-β1 gene promoter were amplified and analyzed by polymerase chain reaction-restriction fragment length polymorphism technique in 84 COPD patients and 97 age-and sex-matched healthy controls. The test for Hardy-Weinberg equilibrium was performed using HWE program of the LINKUTIL package and statistical analysis was carded out with the SPSS statistical package. An expectation maximization algorithm was used for the pairwise linkage disequilibrium test and haplotype analysis. Results: More carriers of the -800A allele, or fewer carders of the -509T allele, were detected in the COPD patients compared with the non-symptomatic control subjects [for the -800A allele, 29.8% vs 14.4%, respectively, χ^2=6.257,degrees of freedom (df)=l, P=0.012; for the-509T allele, 27.3% vs 44.3%, respectively, χ^2=5.582, df=1, P=0.018]. The prevalence of the-800A allele was significantly higher in the COPD patients than in control subjects (P=0.009), whereas the frequency of the-509T allele was significantly higher in control subjects than in the COPD patients (P=0.008). In addition, this distribution tendency for the-800A or-509T allele was similar in heavy smokers (smoking history≥20 pack years);(number of packs of cigarettes per day multiplied by the number of years of smoking)χ^2=7.235, P=0.007, and χ^2=5.636, P=0.018, respectively). The linkage disequilibrium was found between-800 G→A and-509 C→T (D>0.60, P<0.0001), and the frequency of the AC haplotype, consisting of the least common base at -800 and the most common base at -509, was significantly higher in patients with COPD than in controls (0.056 vs 0.021, P<0.05). 15Interventional treatments for hepatocellular carcinoma显示文摘BACKGROUND: Hepatocellular carcinoma (HCC) is one of the most frequent primary malignant tumors in the world. Hepatic resection and liver transplantation are considered optimal for potential treatment of HCC. However, only 20% of HCCs can be surgically treated. And most of surgically-noneligible patients have to receive interventional managements including local ablation and transarterial chemoembolization (TACE). In this paper, we review the interventional treatments of HCC. DATA SOURCES: A literature search of PubMed database was conducted and research articles were reviewed. RESULTS: Percutaneous ethanol injection (PEI) is usually applied to small HCC for a complete necrosis. Radiofrequency ablation, an alternative to PEI, also causes tumor necrosis and needs fewer times of ablation. Other methods such as acetic acid injection, laser, microwave, etc have enriched local ablation for HCC. High intensity focus ultrasound (HIFU) is thought to be promising. TACE, another common modality, can improve the survival rate of patients with HCC. The newly developed embolic agents and adjuvant rAd-p53 gene therapy are well reported. CONCLUSIONS: Surgically-noneligible HCC can be treated with interventional procedures. Each method has its advantages and disadvantages. 16Apoptosis of rat hepatic stellate cells induced by anti-focal adhesion kinase antibody显示文摘AIM: To explore the role of focal adhesion kinase (FAI)in the apoptosis in culture-activated rat hepatic stellatecells (HSCs) using a specific anti-FAK antibody.METHODS: Rat HSCs were prepared from Wistar rats byin situ perfusion of collagenase and pronase and single-step density Nycodenze gradient. Culture-activatedHSCs were serum-starved and treated with the anti-FAK antibodies for 24, 48 or 72 h. The apoptosis of HSCwas detected by DNA-fragment assay, flow cytometryand caspase-3 activity determination. The expressionof tissue inhibitor of metalloproteinase-1 (TIMP-1)mRNA was assessed by reverse transcriptionpolymerase chain reaction (RT-PCR).RESULTS: The experiment showed that anti-FAKantibodies induced apoptosis of culture-activated ratHSCs. This phenomenon displayed the classical featuresof apoptotic cell death (DNA fragmentation, cell cycleanalysis) after treated with 30 mg@L-1 FAK antibody for72 h, and accompanied by a significant increase ofcespase-3 activity(1208±76) vs (309±28) nmol@min-1@ g-1, t=-208.5, P<0.05. 17Infantile hepatic hemangioendothelioma:A clinicopathologic study in a Chinese population显示文摘AIM:To investigate whether the clinicopathologic features of infantile hemangioendothelioma(IHE) of the liver in a Chinese population are similar to the features observed in other races.METHODS:The clinical data,radiological findings,histopathological changes and outcome of 12 cases of IHE diagnosed by the Department of Pathology,West China Hospital over the last 10 years were analyzed retrospectively.Immunohistochemical studies were carried out using antibodies against CD31,CD34,Factor Ⅷ,cytokeratin 8 and cytokeratin 18.RESULTS:The 12 patients were aged from fetal to 5 years(three males and nine females).The tumor was presented with different clinical manifestations,mainly as an asymptomatic,palpable,upper abdominal mass,except for the two fetuses who were detected antena-tally by ultrasound.In one patient,this presentation was accompanied by an initial severe pneumothorax.No symptoms of congestive heart failure were present and neither congenital abnormalities nor vascular tumors in the skin or other organs were found.Laboratory abnormalities included leukocytosis(40%),anemia(60%),thrombocytosis(60%),hyperbilirubinemia(16.7%),abnormal liver function(50%) and increased α-fetoprotein(80%).Based on radiological findings and gross specimens,the tumor presented as a solitary lesion or a multifocal space-occupying lesion.The tumor size ranged from 5.0 cm × 3.5 cm × 2.0 cm to 13.8 cm × 9.0 cm × 7.7 cm,and the 0.2-1.1 cm nodules were diffusely distributed within the multifocal tumor.Seven cases were surgically resected,three cases underwent biopsy and the two fetuses were aborted.Histologically,nine cases were classified as typeⅠ and three as type Ⅱ,presenting aggressive morphologic features,immature vessels,active mitosis and necrosis.An inflammatory component,predominantly eosinophilic granulocytes,sometimes obscured the nature of the tumor.Ten patients are alive after a follow-up of 1-9 years.Based on immunohistochemistry,the endothelial cells in all cases were positive for CD31,CD34 and polyclonal factor Ⅷ antigen,whereas the scattered hyperplasia bile ducts were positive for cytokeratin 8 and cytokeratin 18.CONCLUSION:The clinical manifestations of IHE are non-specific.There is no significant correlation between histological type and prognosis.The clinicopathologic features of IHE in Chinese patients may provide a clue to further evidence-based studies.Zhang Zhang,Hui-Jiao Chen,Wen-Juan Yang,Hong Bu,Bing Wei,Xiao-Yu Long,Jing Fu,Rui Zhang,Yun-Bi Ni,Hong-Ying Zhang,Department of Pathology,West China Hospital,Sichuan University,Guoxuexiang 37,Chengdu 610041,Sichuan Province,China 2010World Journal of Gastroenterology2010,16,36:16
18Liver transplantation for patients with hepatolithiasis显示文摘Because of its complicated pathological features such as repeated cholangitis and multiple operations, hepatolithiasis is difficult to treat and often lead to portal hypertension, and liver failure. The aim of this study was to investigate the indications of orthotopic liver transplantation ( OLT) for patients with hepatolithiasis and the improvement of operative techniques and the treatment of postoperative complications. METHODS:The data of 4 patients with hepatolithiasis who had undergone OLT at our department in recent years were reviewed retrospectively. RESULTS:The 4 patients with hepatolithiasis complicated with secondary biliary cirrhosis in end-stage liver disease included 1 woman and 3 men. One patient underwent OLT using veno-venous bypass prior to the mobilization of the liver. Two patients were subjected to liver transplantation with improved piggyback technique without bypass. One patient received OLT without bypass. All patients were operated on successfully and recovered uneventfully. Patient 1 had bleeding from the digestive tract on the postoperative day 6. Patient 2 had hemorrhage from the digestive tract and a leakage of end-to-side intestinal anastomosis on the postoperative day 44. Patient 4 was reoperated on because of hemorrhage from the anastomotic stoma of the hepatic artery on the postoperative days 8 and 10. In the 4 patients the mean operative time was 7.9 hours and blood loss was 910 ml. Postoperative pathological analysis revealed biliary liver cirrhosis. Follow-up of all patients showed good conditions. CONCLUSIONS:Hepatolithiasis with secondary biliary liver cirrhosis in patients with end-stage liver disease is indicated for liver transplantation. 19Subcellular daunorubicin distribution and its relation to multidrug resistance phenotype in drug-resistant cell line SMMC-7721/R显示文摘AIM: To investigate the correlation between subcellulardaunorubicin distribution and the multidrug resistancephenotype in drug-resistant cell line SMMC-7721/R.METHODS: The multidrug resistant cell line SMMC-7721/R, a human hepatocellular carcinoma cell line,was established. Antisense oligonudeotides (AS-ODN)were used to obtain different multidrug resistancephenotypes by inhibiting the expression of mdr1 geneand/or multidrug resistance-related protein gene(mrp)using Lipofectamine as delivery agent. Expression ofmdr1 and mrp genes was evaluated by RT-PCR andWestern blotting. Intracellular daunorubicin (DNR)concentration was measured by flow cytometry.Subcellular DNR distribution was analyzed by confocallaser scanning microscopy. Adriamycin (ADM) and DMRsensitivity was examined by MTT method.RESULTS: Low level expression of mdr1 and mrp mRNAsand no expression of P-Glycoprotein(P-gp) andmultidrug resistance-related protein (P190) weredetected in parental sensitive cells SMMC-7721/S, butover-expression of these two genes was observed indrug-resistant cell SMMC-7721/R. The expression ofmdr1 and mrp genes in SMMC-7721/R cells was downregulated to the level in the SMMC-7721/S cells by AS-ODN. Intracellular DNR concentration in SMMC-7721/S cells was 10 times higher than that in SMMC-7721/Rcells. In SMMC7721/S cells intracellular DNRdistributed evenly in the nucleus and cytoplasm, whilein SMMC-7721/R cells DNR distributed in a punctatepattern in the cytoplasm and was reduced in thenucleus. DNR concentration in SMMC-7721/R cells co-transfected with AS-ODNs targeting to mdr1 and mrpmRNAs recovered to 25 percent of that in SMMC7721/Scells. Intracellular DNR distribution pattern in drug-resistant cells treated by AS-ODN was similar to drug-sensitive cell, and the cells resistance index (RI) to DNRand ADM decreased at most from 88.0 and 116.0 to 4.0and 2.3, respectively. Co-Transfection of two AS-ODNsshowed a stronger synergistic effect than separatetransfection.CONCLUSIONS: P-gp and P190 are two membersmediating MDR in cell line SMMC7721/R. Intracellulardrug concentration increase and subcellular distributionchange are two important factors in multidrugresistance (MDR) formation.
