1胎盘生长因子和色素上皮衍生因子在非小细胞肺癌中的表达及其与预后的关系显示文摘目的探讨胎盘生长因子(PlGF)和色素上皮衍生因子(PEDF)在非小细胞肺癌中的表达特点及与肿瘤新生血管形成的关系,了解这两种因子在判断非小细胞肺癌患者预后中的意义。方法采用超敏过氧化酶免疫组织化学法检测手术切除的81例非小细胞肺癌标本中PlGF和PEDF的表达水平,并利用CD31染色进行微血管计数,统计分析这两个因子与微血管密度(MVD)的关系,及其在非小细胞肺癌患者预后中的意义。结果PlGF和PEDF在81例非小细胞肺癌患者中的阳性率分别为43·2%(35例)和53·1%(43例)。PlGF阳性表达与高值MVD相关;PEDF阳性表达与低值MVD相关;PlGF和PEDF的表达状况存在负相关。单因素和多因素分析均提示PlGF是非小细胞癌患者独立的预后指标。结论PlGF和PEDF均可在非小细胞肺癌组织中表达,二者在血管生成的过程中具有相互拮抗的作用。与PEDF相比,PlGF在非小细胞肺癌发展以及预测患者预后方面具有更大的作用。张力建 陈晋峰 陆爱萍 柯杨 Robert E Mansel Wen G Jiang 2005中华医学杂志2005,85,47:10
2Co-effect of Demand-control-support Model and Effort-reward Imbalance Model on Depression Risk Estimation in Humans: Findings from Henan Province of China显示文摘Objective To investigate the co-effect of Demand-control-support(DCS) model and Effort-reward Imbalance(ERI) model on the risk estimation of depression in humans in comparison with the effects when they are used respectively. Methods A total of 3 632 males and 1 706 females from 13 factories and companies in Henan province were recruited in this cross-sectional study. Perceived job stress was evaluated with the Job Content Questionnaire and Effort-Reward Imbalance Questionnaire(Chinese version). Depressive symptoms were assessed by using the Center for Epidemiological Studies Depression Scale(CES-D). Results DC(demands/job control ratio) and ERI were shown to be independently associated with depressive symptoms. The outcome of low social support and overcommitment were similar. High DC and low social support(SS), high ERI and high overcommitment, and high DC and high ERI posed greater risks of depressive symptoms than each of them did alone.ERI model and SS model seem to be effective in estimating the risk of depressive symptoms if they are used respectively. Conclusion The DC had better performance when it was used in combination with low SS. The effect on physical demands was better than on psychological demands. 3基质细胞衍化因子-1的表达及其与乳腺癌患者预后的关系显示文摘目的探讨基质细胞衍化因子-1(SDF-1)在不同细胞系及乳腺癌组织和正常乳腺组织中的表达,并分析SDF-1的表达水平与乳腺癌患者预后的关系。方法选择乳腺癌细胞系、成纤维细胞系、血管内皮细胞系,利用RT-PCR方法分析SDF-1mRNA的表达。采用免疫组织化学染色和实时定量PCR测定120例乳腺癌组织和32例正常乳腺组织中SDF-1的表达。通过SPSS统计软件对所得结果行统计学分析。结果SDF-1的表达见于人胚胎肺成纤维细胞系、某些乳腺癌细胞系(MDA-MB435s、MDA-MB436、MCF7)及乳腺癌和正常乳腺组织。淋巴结有转移者SDF-1mRNA的表达水平为399.00±210.00,淋巴结无转移者为0.89±0.47(P=0.048)。预后不良者(出现肿瘤局部复发、远处转移和因乳腺癌死亡者)较无疾病存活者SDF-1mRNA水平明显增高,为670.00±346.00vs0.83±0.35(P=0.01)。SDF-1mRNA的表达水平与患者的总存活率和无瘤存活率有关,P值分别为0.01和0.035。结论SDF-1mRNA的表达既可见于基质细胞也可见于乳腺癌细胞。SDF-1的表达水平与乳腺癌患者有无淋巴结转移、预后和存活密切相关。SDF-1可作为乳腺癌患者预后判断的一项指标。康骅 Robert EMansel Wen G Jiang 2005中国普外基础与临床杂志2005,12,5:7
4色素上皮衍生因子基因的克隆以及与肺癌细胞生长和肿瘤新生血管形成影响的研究显示文摘目的克隆并在肺癌细胞SKMES1中表达色素上皮衍生因子(PEDF)蛋白;在体实验中观察PEDF蛋白过表达后对肺癌细胞生长和肿瘤相关性新生血管形成的影响。方法通过PCR技术扩增出可以表达PEDF蛋白的全长基因;利用基因重组技术与pEF6/v5-His质粒连接后构建出PEDF基因过表达载体PEDF^exp;将PEDF^exp载体转染SKMES1肺癌细胞,建立稳定过表达PEDF蛋白的肺癌细胞株;将过表达PEDF蛋白的肺癌细胞接种于鸡胚,在体实验中明确PEDF蛋白过表达后对肺癌细胞生长和肿瘤新生血管形成的影响。结果从人体组织的cDNA中成功扩增出PEDF全长序列,并正确构建了可以表达PEDF蛋白的载体,基因测序证实所构建的质粒载体中正确连接了没有核苷酸突变的目的片段;RT—PCR和Western印迹实验在RNA和蛋白水平证实,稳定转染PEDF^exp质粒的肺癌细胞与对照组肺癌细胞相比可以过表达PEDF蛋白。鸡胚实验证实PEDF基因过表达后,SKMES1^PEDFexp肺癌细胞形成病灶的体积明显小于对照组[(0.10±O.05)cm^3vs(0.17±0.07)cm^3](P=0.016);SKMES1^PEDFexp肺癌细胞形成病灶的质量也明显小于对照组细胞[(0.008±0.004)mgvs(0.024±0.009)mg](P=0.006)。肿瘤相关新生血管计数结果显示,PEDF基因的过表达后,肿瘤病灶中一级血管的数量明显少于对照组细胞(15±3vs41±9)(P〈0.001);二级血管的数量也明显少于对照组细胞(75±22vs175±39)(P=0.001)。结论成功构建出含有PEDF基因表达载体,为研究PEDF基因在肺癌中的功能奠定了基础。PEDF蛋白在肺癌生长以及肿瘤血管形成过程中的抑制作用说明PEDF蛋白是一种非常有前途的具有肺癌治疗作用的生物药物。陈晋峰 赵威 张建芝 Wen G Jiang 张力建 2009中华医学杂志2009,89,7:6
5荧光实时定量PCR检测NSCLC中PEDF基因表达状态的研究显示文摘背景与目的SYBRGreenⅠ是一种较为常用的非探针类定量PCR的方法,其主要优点在于操作过程简单,结果具有一定特异性。本研究的目的是建立检测色素上皮衍生因子(pigmentepitheliumderivedfactor,PEDF)mRNA的SYBRGreenⅠ实时定量PCR的方法,了解肺癌组织及其远癌肺组织中PEDF的表达水平,研究肺癌组织中PEDF的表达水平与各种临床病理特征之间的关系。方法利用RTPCR的方法扩增并纯化PEDF以及内对照GAPDHmRNA的目的片段,倍比稀释后作为定量模板。利用相对定量的方法,检测21例肺癌及相应远癌肺组织中PEDFmRNA相对于GAPDHmRNA的表达量,进行相对定量分析。结果21例肺癌组织及相应的远癌肺组织中均可检出PEDF的表达,肺癌组织中PEDF的相对表达量为0.5505±0.3590(0.11~1.11),远癌肺组织中为0.7219±0.2582(0.29~1.31)(P=0.024)。早期(Ⅰ~Ⅱ期)肺癌患者和肿瘤较小时(T1)PEDF的表达量显著高于晚期(Ⅲ~Ⅳ期,P=0.010)和肿瘤较大者(T2~4,P=0.007)。结论所建立的SYBRGreenⅠ实时定量PCR方法可以成功地检测PEDF基因的表达量。初步检测结果显示PEDF可能是一种肺癌发生发展过程中的保护因素。张力建 陈晋峰 柯杨 Robert E Mansel Wen G Jiang 2006中国肺癌杂志2006,9,2:4
6mRNA expression of DOK1-6 in human breast cancer显示文摘AIM:To examine the expression of downstream of tyrosine kinase(DOK)1-6 genes in normal and breast cancer tissue and correlated this with several clinicopathological and prognostic factors.METHODS:DOK1-6 m RNA extraction and reverse transcription were performed on fresh frozen breast cancer tissue samples(n = 112) and normal background breast tissue(n = 31). Tissues were collected between 1991 and 1996 at two centres and all patients underwent mastectomy and ipsilateral axillary node dissection. All tissues were randomly numbered and the details were only made known after all analyses were completed. Transcript levels of expression were determined using real-time polymerase chain reaction and analyzed against TNM stage, tumour grade and clinical outcome over a 10-year follow-up period.RESULTS:DOK-2 and DOK-6 expression decreased with increasing TNM stage. DOK-6 expression decreased with increasing Nottingham Prognostic Index(NPI) [NPI-1 vs NPI-3(mean copy number 15.4 vs 0.22, 95%CI:2.7-27.6, P = 0.018) and NPI-2 vs NPI-3(mean copy number 7.6 vs 0.22, 95%CI:0.1-14.6, P = 0.048)]. After a median follow up period of 10 years, higherlevels of DOK-2 expression were found among patients who remained disease-free compared to those who developed local or distant recurrence(mean copy number 3.94 vs 0.0000096, 95%CI:1.0-6.85, P = 0.0091), and distant recurrence(mean copy number 3.94 vs 0.0025, 95%CI:1.0-6.84, P = 0.0092).
