1Conservation metagenomics: a new branch of conservation biology显示文摘Multifaceted approaches are required to monitor wildlife populations and improve conservation efforts. In the last decade,increasing evidence suggests that metagenomic analysis offers valuable perspectives and tools for identifying microbial communities and functions. It has become clear that gut microbiome plays a critical role in health, nutrition, and physiology of wildlife, including numerous endangered animals in the wild and in captivity. In this review, we first introduce the human microbiome and metagenomics, highlighting the importance of microbiome for host fitness. Then, for the first time, we propose the concept of conservation metagenomics, an emerging subdiscipline of conservation biology, which aims to understand the roles of the microbiota in evolution and conservation of endangered animals. We define what conservation metagenomics is along with current approaches, main scientific issues and significant implications in the study of host evolution, physiology,nutrition, ecology and conservation. We also discuss future research directions of conservation metagenomics. 2Effect of China's rapid development on its iconic giant panda显示文摘Anthropogenic factors affect biodiversity and have led to the contraction or extinction of animal populations worldwide.Here,we use historical demographic data,spanning the past 300 years,to show that a rapid distributional contraction of giant pandas took place during the 18th and 19th centuries alongside the increase in human population.Land-use also underwent a significant change across the areas where giant pandas were found because of government agricultural policy and the introduction of new crops.The impact of social development on giant pandas includes habitat loss and fragmentation,and range reductions.Our findings would facilitate the design of effective conservation strategies that seek to conserve and increase current habitats of this iconic species,especially in areas that our analysis has identified as places where pandas have suffered from high human pressure.ZHU LiFeng HU YiBo ZHANG ZeJun WEI FuWen 2013Chinese Science Bulletin2013,58,17:8
3GENETIC DIVERSITY AMONG CHINESE SIKA DEER (CERVUS NIPPON) POPULATIONS AND RELATIONSHIPS BETWEEN CHINESE AND JAPANESE SIKA DEER显示文摘Sika deer (Cervus nippon) is a cervid endemic to mainland and insular Asia and endan-gered. We analyzed variation in the mitochondrial DNA (mtDNA) control region for four subspecies to understand the genetic diversity, population structure and evolutionary history in China. 335 bp were se-quenced and eight haplotypes were identified based on 25 variable sites among the populations. Sika deer in China showed lower genetic diversity, sug-gesting a small effective population size due to habi-tat fragmentation, a low number of founder individu-als, or the narrow breeding program. AMOVA analy-sis indicated that there was significant genetic subdi-vision among the four populations, but no correlation between the genetic and geographic distance. Phy-logenetic analyses also revealed that Chinese sika deer may be divided into three genetic clades, but the genetic structure among Chinese populations was inconsistent with subspecies designations and pre-sent geographic distribution. 4Diet drives convergent evolution of gut microbiomes in bamboo-eating species显示文摘Gut microbiota plays a critical role in host physiology and health.The coevolution between the host and its gut microbes facilitates animal adaptation to its specific ecological niche.Multiple factors such as host diet and phylogeny modulate the structure and function of gut microbiota.However,the relative contribution of each factor in shaping the structure of gut microbiota remains unclear.The giant(Ailuropoda melanoleuca)and red(Ailurus styani)pandas belong to different families of order Carnivora.They have evolved as obligate bamboo-feeders and can be used as a model system for studying the gut microbiome convergent evolution.Here,we compare the structure and function of gut microbiota of the two pandas with their carnivorous relatives using 16S rRNA and metagenome sequencing.We found that both panda species share more similarities in their gut microbiota structure with each other than each species shares with its carnivorous relatives.This indicates that the specialized herbivorous diet rather than host phylogeny is the dominant driver of gut microbiome convergence within Arctoidea.Metagenomic analysis revealed that the symbiotic gut microbiota of both pandas possesses a high level of starch and sucrose metabolism and vitamin B12 biosynthesis.These findings suggest a diet-driven convergence of gut microbiomes and provide new insight into host-microbiota coevolution of these endangered species.Guangping Huang Xiao Wang Yibo Hu Qi Wu Yonggang Nie Jiuhong Dong Yun Ding Li Yan Fuwen Wei 2021Science China(Life Sciences)2021,64,1:6
5Seasonal and reproductive variation in chemical constituents of scent signals in wild giant pandas显示文摘Seasonally reproducing animals show many behavioral and physiological changes during the mating period, including increased signaling for intrasexual competition and mate attraction. We collected 102 anogenital gland secretions(AGS) from marking trees in Foping Nature Reserve, and used gas chromatography mass spectrometry to analyze these chemical composition. Of these marks, all but one were from males, confirmed with DNA analysis. We found that several chemical constituents, especially volatile compounds, were present only during the mating season and that the relative abundance of many compounds changed as a function of breeding season, whereas nonvolatile compounds were lower in the mating season. This seasonal variation in chemical composition of AGS most likely plays an important role in governing giant panda reproduction, including mate location, attraction, and male-male competition. The chemical properties of many of these putative chemosignals—such as volatility and longevity—are suggestive of these roles, and undoubtedly contribute to successful reproduction for this species with a characteristically sophisticated chemical communication system. 7First evidence of prey capture and meat eating by wild Yunnan snub-nosed monkeys Rhinopithecus bieti in Yunnan,China显示文摘Most extant nonhuman primates occasionally prey on fast-moving, warm-blooded animals; however, Indriidae, Lepilemuridae,and Colobinae either scavenged for meat or did not eat meat at all. Here we report six cases of animal consumptionby the snub-nosed monkey Rhinopithecus bieti in a wild, habituated group between 2004 and 2009 in Yunnan, China. At present,only males in an all-male unit within the study group were involved in active hunting. Such a male-biased activity may be relatedto the group structure and spatial spread of R. bieti. Two females were observed eating freshly killed birds. The findings confirmedthat R. bieti engaged in scavenging and, when hunting, employed a cranio-cervical bite to kill their prey. Meat eating islikely a nutrient maximization feeding strategy in R. bieti, especially in males. A begging behavior occurred after successful preycapture. Although begging was observed, no sharing of the meat was seen. 9Home range and seasonality of Yunnan snub-nosed monkeys显示文摘Home range studies are essential for understanding an animal’s behavioral ecology and for making wildlife conservation efficient.The home range of a group of Yunnan snub-nosed monkeys(Rhinopithecus bieti Milne-Edwards,1897)was estimated using a global positioning system collar from December 2003 to October 2004 in northern Yunnan Province,China.To measure the ranging area of the study group 1291 animal locations were fixed.Based on the 100%minimum convex polygon method,the home range size was approximately 32.8 km2;based on the grid-cell method it was approximately 17.8 km2 using a 250-m grid and 23.3 km2 using a 500-m grid.We assessed seasonal changes in the home range of this monkey population and found a different ranging pattern of from earlier studies.Daily range size was calculated as 4.80±5.81 ha(mean±standard deviation),with a range of 45.66 ha(0.01–45.67 ha).Baoping REN Ming LI Yongcheng LONG Ruidong WU Fuwen WEI 2009Integrative Zoology2009,4,2:3

11Characteristics of night-time sleeping places selected by golden monkeys(Rhinopithecus bieti)in the Samage Forest,Baima Snow Mountain Nature Reserve,China显示文摘We examined the criteria for sleeping place selection in a social band of Rhinopithecus bieti(black-and-white snubnosed or golden monkeys)living in the mountainous Samage Forest,Baima Snow Mountain Nature Reserve,Yunnan,China.We performed principal component analysis and found that slope aspect,tree height and trunk diameter were likely key variables influencing selection of sleeping places.Sleeping sites were preferentially located in mixed deciduous/conifer forest.The monkeys slept exclusively in evergreen trees,of which 82%were conifers(mostly Picea likiangensis and Tsuga dumosa)and 18%evergreen oaks(Cyclobalanopsis oxyodon and Quercus spp.).Sleeping trees were tall(mean 30.5 m),had high boles(mean 18.4 m),large diameters(mean 62.6 cm)and large crown areas(mean 57.9 m^(2)).A comparative analysis of phytological and architectural features between trees in“sleeping site plots”(n=18)and trees in“non-sleeping-site plots”(n=66)revealed that diameter,crown surface area and tree height were significantly(P<0.01)larger in the former compared with the latter.All investigated roosting sites were situated on steep mountain slopes.Valleys and mountain ridges were avoided.We also detected re-use of roosting sites on several occasions,but not on consecutive nights.It is most likely that a mix of factors(stability of trees,access to food,unit cohesion,monitoring potential)explains the pattern of sleeping site preference,but predation at night seems to be only slightly important.Climate appears to have a profound influence on patterns of sleeping site selection in the monkeys’harsh temperate habitat.This is demonstrated by the monkeys’preference for mixed forest at medium elevations over montane fir forest at high elevations and slopes instead of ridges,with reduced exposure to wind and precipitation inherent in the former.We also emphasize the possibly substantial role that non-environmental factors(the nature of social organization and socio-behavioral strategies)play in determining sleeping site use in R.bieti and other primates.Dayong LI Cyril C.GRUETER Baoping REN Qihai ZHOU Ming LI Zhengsong PENG Fuwen WEI 2006Integrative Zoology2006,1,4:2
12Sex-related gene and sex identification of Crested Ibis Nipponia nippon (Ciconiifor-mes: Threskiornithidae)显示文摘The Crested Ibis (Nipponia nippon) is a critical endangered species of the world. At present, a reintroduction program will be conducted to save this species essentially. However, because the Crested Ibise is a sexual alike bird, it is very difficult to identify the sex from the morphological character. In order to identify the sex easily and select the right individuals for captive breeding and reintroduction, the sex related gene on W chromosome was amplified and the sex of three Crested Ibises were also identified in the present study. The 262bp fragment was also sequenced, and we found that there were 13 different nucleotide sites with 2.25 of transition/transversion based on the comparison with that of the Oriental White Stork. 13Lineage-specific evolution of bitter taste receptor genes in the giant and red pandas implies dietary adaptation显示文摘Taste 2 receptors(TAS2R)mediate bitterness perception in mammals,thus are called bitter taste receptors.It is believed that these genes evolved in response to species-specific diets.The giant panda(Ailuropoda melanoleuca)and red panda(Ailurus fulgens styani)in the order Carnivora are specialized herbivores with an almost exclusive bamboo diet(>90%bamboo).Because bamboo is full of bitter tasting compounds,we hypothesized that adaptive evolution has occurred at TAS2R genes in giant and red pandas throughout the course of their dietary shift.Here,we characterized 195 TAS2R genes in 9 Carnivora species and examined selective pressures on these genes.We found that both pandas harbor more putative functional TAS2R genes than other carnivores,and pseudogenized TAS2R genes in the giant panda are different from the red panda.The purifying selection on TAS2R1,TAS2R9 and TAS2R38 in the giant panda,and TAS2R62 in the red panda,has been strengthened throughout the course of adaptation to bamboo diet,while selective constraint on TAS2R4 and TAS2R38 in the red panda is relaxed.Remarkably,a few positively selected sites on TAS2R42 have been specifically detected in the giant panda.These results suggest an adaptive response in both pandas to a dietary shift from carnivory to herbivory,and TAS2R genes evolved independently in the 2 pandas.Our findings provide new insight into the molecular basis of mammalian sensory evolution and the process of adaptation to new ecological niches.Lei SHAN Qi WU Le WANG Lei ZHANG Fuwen WEI 2018Integrative Zoology2018,13,2:2
14Alternative polyadenylation of mRNA and its role in cancer显示文摘Alternative polyadenylation(APA)is a molecular process that generates diversity at the 3′end of RNA polymeraseⅡtranscripts from over 60%of human genes.APA is derived from the existence of multiple polyadenylation signals(PAS)within the same transcript,and results in the differential inclusion of sequence information at the 3′end.While APA can occur between two PASs allowing for generation of transcripts with distinct coding potential from a single gene,most APA occurs within the untranslated region(3′UTR)and changes the length and content of these non-coding sequences.APA within the 3′UTR can have tremendous impact on its regulatory potential of the mRNA through a variety of mechanisms,and indeed this layer of gene expression regulation has profound impact on processes vital to cell growth and development.Recent studies have particularly highlighted the importance of APA dysregulation in cancer onset and progression.Here,we review the current knowledge of APA and its impacts on mRNA stability,translation,localization and protein localization.We also discuss the implications of APA dysregulation in cancer research and therapy.Fuwen Yuan William Hankey Eric J.Wagner Wei Li Qianben Wang 2021Genes & Diseases2021,8,1:2
15Habitat use and locomotion of the François’langur(Trachypithecus francoisi)in limestone habitats of Nonggang,China显示文摘We collected data on habitat use and locomotion of the François’langur(Trachypithecus francoisi)between August 2003 and July 2004 at Nonggang Nature Reserve,China.A total of 739 h of behavioral data were collected during this study.We tested 2 predictions:(1)that the langurs may have special patterns of habitat use and locomotion adaptive to the limestone habitat,and(2)the langurs may exhibit different patterns of habitat use and locomotion among different zones of limestone hill.Our results indicated that François’langurs spent more time in the low-risk,relatively food-poor cliff-hilltop areas.When young leaves and fruit were scarce in the dry season,the langurs increased their time in the high-risk,food-rich valley basin.François’langurs were semi-terrestrial,and leaping and climbing were their main locomotor modes.These behavioral patterns are considered to be related to characteristics of topography and vegetation in limestone habitat,such as large areas of cliff and discontinuous canopy.Our results also supported Prediction 2.The langurs confined locomotion to the main canopy and frequently adopted leaping while traveling in the hillside and valley basin.While traveling in cliff-hilltop areas,they tended to stay in the lower stratus(≤5 m)or move on the ground,and walking and climbing were their dominant traveling modes.Qihai ZHOU Bang LUO Fuwen WEI Chengming HUANG 2013Integrative Zoology2013,8,4:2
16Ranging behavior of the François’langur(Trachypithecus francoisi)in limestone habitats of Nonggang,China显示文摘Ranging behavior is an important aspect of animal behavior that researchers use to investigate ecological influences on individual behavior.We collected data on the ranging behavior of one group of François’langurs(Trachypithecus francoisi)between August 2003 and July 2004 in a limestone habitat within Nonggang Nature Reserve,China.We evaluated the influences of food availability,water resources and sleeping sites on ranging behavior.During the study period,the total home range size for the study group was 64.5 or(69.3 ha if the lacunae within the borders in which langurs were not observed were included),and the mean daily path length was 541 m.These are well within the range of variation reported in Trachypithecus species.The monthly range size was between 9.8 and 23.3 ha,and monthly range size correlated negatively with young leaf availability.The langurs tended to use a larger range size during young leaf-lean periods.The langurs did not use their home range uniformly,and 74%of their activities occurred within 35%of their home range.The most heavily used quadrats in the home range were located near the most frequently used sleeping sites,suggesting that sleeping sites have a significant influence on the ranging behavior of François’langurs in limestone habitats.Qihai ZHOU Chengming HUANG Ming LI Fuwen WEI 2011Integrative Zoology2011,6,2:2

18Molecular mechanisms of adaptive evolution in wild animals and plants显示文摘Wild animals and plants have developed a variety of adaptive traits driven by adaptive evolution,an important strategy for species survival and persistence.Uncovering the molecular mechanisms of adaptive evolution is the key to understanding species diversification,phenotypic convergence,and inter-species interaction.As the genome sequences of more and more non-model organisms are becoming available,the focus of studies on molecular mechanisms of adaptive evolution has shifted from the candidate gene method to genetic mapping based on genome-wide scanning.In this study,we reviewed the latest research advances in wild animals and plants,focusing on adaptive traits,convergent evolution,and coevolution.Firstly,we focused on the adaptive evolution of morphological,behavioral,and physiological traits.Secondly,we reviewed the phenotypic convergences of life history traits and responding to environmental pressures,and the underlying molecular convergence mechanisms.Thirdly,we summarized the advances of coevolution,including the four main types:mutualism,parasitism,predation and competition.Overall,these latest advances greatly increase our understanding of the underlying molecular mechanisms for diverse adaptive traits and species interaction,demonstrating that the development of evolutionary biology has been greatly accelerated by multi-omics technologies.Finally,we highlighted the emerging trends and future prospects around the above three aspects of adaptive evolution.Yibo Hu Xiaoping Wang Yongchao Xu Hui Yang Zeyu Tong Ran Tian Shaohua Xu Li Yu Yalong Guo Peng Shi Shuangquan Huang Guang Yang Suhua Shi Fuwen Wei 2023Science China(Life Sciences)2023,66,3:1
19Ecological scale and seasonal heterogeneity in the spatial behaviors of giant pandas显示文摘We report on the first study to track the spatial behaviors of wild giant pandas(Ailuropoda melanoleuca)using high-resolution global positioning system(GPS)telemetry.Between 2008 and 2009,4 pandas(2 male and 2 fe-male)were tracked in Foping Reserve,China for an average of 305 days(±54.8 SE).Panda home ranges were larger than those of previous very high frequency tracking studies,with a bimodal distribution of space-use and distinct winter and summer centers of activity.Home range sizes were larger in winter than in summer,although there was considerable individual variability.All tracked pandas exhibited individualistic,unoriented and multi-phasic movement paths,with a high level of tortuosity within seasonal core habitats and directed,linear,large-scale movements between habitats.Pandas moved from low elevation winter habitats to high elevation(>2000 m)summer habitats in May,when temperatures averaged 17.5°C(±0.3 SE),and these large-scale movements took<1 month to complete.The peak in panda mean elevation occurred in Jul,after which they began slow,large-scale movements back to winter habitats that were completed in Nov.An adult female panda made 2 long-distance movements during the mating season.Pandas remain close to rivers and streams during winter,possi-bly reflecting the elevated water requirements to digest their high-fiber food.Panda movement path tortuosity and first-passage-time as a function of spatial scale indicated a mean peak in habitat search effort and patch use of approximately 700 m.Despite a high degree of spatial overlap between panda home ranges,particularly in winter,we detected neither avoidance nor attraction behavior between conspecifics.Zejun ZHANG James K.SHEPPARD Ronald R.SWAISGOOD Guan WANG Yonggang NIE Wei WEI Naxun ZHAO Fuwen WEI 2014Integrative Zoology2014,9,1:1
