1Clinical observation of salvianolic acid B in treatment of liver fibrosis in chronic hepatitis B显示文摘AIM: To evaluate the clinical efficacy of salvianolic acidB (SA-B) on liver fibrosis in chronic hepatitis B.METHODS: Sixty patients with definite diagnosis of liverfibrosis with hepatitis B were included in the trial.Interferon-γ (IFN-γ) was used as control drug. Thepatients took orally SA-B tablets or received muscularinjection of IFN-γ in the double blind randomized test,The complete course lasted 6 months. The histologicalchanges of liver biopsy specimen before and after thetreatment were the main evidence in evaluation, incombination with the results of contents of serum HA,LN, Ⅳ-C, P-Ⅲ-P, liver ultrasound imaging, andsymptoms and signs.RESULTS: Reverse rate of fibrotic stage was 36.67 % inSA-B group and 30.0 % in IFN-γgroup. Inflammatoryalleviating rate was 40.0 % in SA-B group and 36.67 %in IFN-γ group. The average content of HA and Ⅳ-Cwas significantly lower than that before treatment. Theabnormal rate also decreased remarkably. Overallanalysis of 4 serological fibrotic markers showedsignificant improvement in SA-B group as comparedwith the IFN-γgroup. Score of liver ultrasound imagingwas lower in SA-B group than in IFN-γgroup (HA 36.7 %vs80 %,Ⅳ-C 3.3 % vs23.2 %). Before the treatment,ALT AST activity and total bilirubin content of patientswho had regression of fibrosis after oral administrationof SA-B, were significantly lower than those of patientswho had aggravation of fibrosis after oraladministration of SA-B. IFN-γ showed certain sideeffects (fever and transient decrease of leukocytes,occurrence rates were 50 % and 3.23 %), but SA-Bshowed no side effects.CONCLUSION: SA-B could effectively reverse liverfibrosis in chronic hepatitis B. SA-B was better than IFN-γ in reduction of serum HA content, overall decrease of4 serum fibrotic markers, and decrease of ultrasoundimaging score. Liver fibrosis in chronic hepatitis B withslight liver injury was more suitable to SA-B in anti-fibrotic treatment. 2Effects of Chinese Jianpi herbs on cell apoptosis and related gene expression in human gastric cancer grafted onto nude mice显示文摘AIM: To explore the mechanism of the Sijunzi decoction and another Chinese herbal recipe (SRRS) based mainly on the Sijunzi decoction in treatment of gastric cancer.METHODS: A human gastric adenocarcinoma cell line SGC7901 grafted onto nude mouse was used as the animal model. The mice were divided into 3 groups, one control and the two representative experimental conditions. Ahimals in the two experimental groups received either Sijunzi decoction or SRRS over a 40-day period starting at 1st day after grafting. Control animals received saline on an identical schedule. Animals were killed 41 days after being grafted.The effect of therapy was assessed by two ways: (1)tumor size was periodically measured during the life of the animals; (2) tumor weight was determined by a electron balance immediately after the animals killed. For detection of apoptotic cells, apoptotic indices(AI) were examined by the terminal deoxynucleotidyl transferase-mediated deoxyuddine tdphosphate fluorescence nick end labeling (TUNEL) method.Morphological alterations were observed with electron microscopy. S-P immunohistochemical method was used to detect the expression of Ki-67 in xenografts. Expression of bcl-2 and p53 was semiquantitatively detected using a reverse transcriptase-polymerase chain reaction (RT-PCR)technique.RESULTS: When compared with controls, tumor growth (size and weight) was significantly inhibited by treatment with the Sijunzi decoction (P<0.05) or SRRS (P<0.01). The tumor inhibitory rate in the Sijunzi decoction group was 34.33 % and SRRS group 46.53 %. AI of human gastric cancer xenografts in nude mice was significantly increased to 16.24±3.21% using TUNEL method and 11.38±6.46 % by FACScan in the Sijunzi decoction group compared with the controls (TUNEL: 2.63±1.03 %, P<0.01; FACScan: 7.15± 1.32 %, P<0.05). SRRS group was also found a significantly increased AI by using TUNEL method and flow cytometry analysis compared with the controls (TUNEL: 13.18±3.05 %,P<0.05; FACScan: 11.58±5.71% (P<0.05). Under electron microscope, cell shrinkage, nuclear chromatin condensation,formation of membrane blebs and apoptotic bodies were frequently observed in Sijunzi decoction group and SRRS group. The average labeling index (LI) for Ki-67 in SRRS group was significantly decreased to 8.43±2.22 % compared with the control group (10.37±t.91%) (P<0.05). The average labeling index for Ki-67 in sijunzi decoction group was 7.95±2.54 % which was lower than that of the control group, but showed no significance (P=0.07). The expression level of p53 mRNA was lower in both Sijunzi decoction group and SRRS group than that in control group (P<0.05; P<0.01). 3The effect of folic acid on the development of stomach and other gastrointestinal cancers显示文摘Objective To evaluate the roles of folic acid and β-carotene in the chemoprevention of gastric and other gastrointestinal (GI) cancers. Methods In a randomized, double-blind, placebo-controlled trial, a total of 216 patients with atrophic gastritis were randomly assigned to one of the four groups: ①folate (FA, 20 mg per day plus vitamin B 12 1 mg, intramuscularly, per month for one year, then 20 mg two times a week plus 1 mg per three months for the next year); ②natural β-carotene (N-βC, 30 mg per day for first year, then 30mg two times a week for the next); ③synthetic β-carotene (S-βC, administered as in N-βC); and ④placebo. Follow-ups continued from 1994 to 2001. Results A total of 7 new cases of gastrointestinal cancers were diagnosed with 3 stomach, 1 colon and 1 esophageal cancers occurring in the placebo group; 1 stomach cancer in both of the N-βC and S-βC groups, and no cancer occurring in FA group. In terms of GI cancers, there was a significant reduction in the FA group, compared with the placebo group (P= 0.04). A similar trend was observed in both N-βC and S-βC groups (P=0.07-0.08). Taken together, the three intervention groups displayed a highly significant decrease in occurrence (P=0.004, vs placebo), and a lower risk for GI cancers (OR=0.12; 95% confidence interval, 0.03-0.51). For development of gastric cancer, any one of the three active-treated groups did not reach statistically significant reduction. The FA group showed obvious improvement of the gastric mucosal lesions with more patients displaying lesions reversed or stable atrophy and inflammation (P=0.04), reversed intestinal metaplasia (P=0.06) at the end of follow-up, and reversed displasia (P=0.017) at 12 months. Two cases of false jaundice were found in β-carotene groups with no influence on administration, and no side-effects were reported in FA group. Conclusions This trial revealed the interventional effect of folic acid on the development of GI cancers, a similar effect of β-carotene was also detected. 5The expression of hTERT mRNA and cellular immunity in gastric cancer and precancerosis显示文摘AIM:To observe the expression of Human telomerasereverse transcriptase (hTERT) in gastric carcinomas andprecancerosis lesions, to evaluate the immune state ofsuch patients, and to then study the clinical significanceof hTERT and immune state for the diagnosis, treat-ment and prognosis of gastric cancer METHODS: In situ hybridization was used to detect theexpression of hTERT mRNA in 116 endoscopic of gas-tric mucosa. Analyzed tissue samples were as follows:30 cases of chronic superficial gastritis (CSG), 44 ofprecancerosis lesions (including 27 of chronic atrophicgastritis, 8 of adenomatous polyp and 9 of gastric ulcer)and 42 of gastric cancer (GC). In addition, the T lym-phocyte subsets (CD3+, CD4+, CD8+, CD4+/CD8+ ) andnatural killer cells (NK) in peripheral blood were deter-mined by flow cytometric analysis (FCM) in 30 cases ofCSG, 27 of precancorosis (chronic atrophic gastritis,CAG), and 42 of GC. The data were compared with thoseof normal control (NC).RESULTS:The detected positive rate of hTERT varied asfollows: 86 % (36/42) in GC, 36 % (16/44) inprecancerosis lesions and 0 % (0/30) in CSG. The ex-pression of hTERT mRNA was not associated with pa-tient gender, tumor location, macroscopic type, lymphnode metastasis, or degree of differentiation, It wasfound that the CD3+, CD4+ of the CSG group were lowerthan that of NC (P<0.05). Meanwhile, the T lympho-cyte subsets (CD3+,CD4+, CD4+/CD8+ ratio) and NK cellsof CAG were remarkably lower than that of NC and CSGgroups (P<0.05-0.01). Values ofT cells and NK cells ofthe GC group were significantly abnormal when com-pared with the CAG group (P<0.05-0.01). Furthermore,with tumor progression, the function of T cells wasweakened gradually.CONCLUSION: The expression of telomerase may be acrucial step in gastric carcinogenesis and increasedhTERT mRNA may serve as a novel marker for diagno-sis of GC. 7Influence of Different Extraction and Purification Methods on Astragalus Polysaccharides and Pharmacological Evaluation显示文摘Objective To clarify the influence on component and pharmacological action of Astragalus polysaccharides(APS) as complementary therapeutic agents prepared by different extraction and purification techniques.Methods Components of APS prepared by different extraction and purification techniques were analyzed,and these APS were used for synergy and attenuation of chemotherapy,radiotherapy treatment with H22 liver cancer and Lewis lung cancer of tumor-bearing mice,and also used for the regulation of immune function to immunosuppression mice.Results Experimental data were analyzed by means of statistical method to get pharmaco-result:A3(extracted by microwave assistance and purified by membrane separation)>A4(extracted by refluxing and purified by membrane separation)>A1(extracted by refluxing and no purification)≈A2(extracted by microwave assistance and no purification).There were no significant differences on pharmacodynamic action between A1 and A2.However,compared with A1 and A2,it was worth noting that A3 and A4 exhibited good pharmacodynamic action.Then A3-in and A4-in,the samples in dialyzer after dialysis,were separated and purified to get homogeneous APS,which were the principal constituents of APS in dialyzer,with the molecular weight(Mw) of 7669 and 14142 determined by HPGPC,respectively.The average Mw of APS outside of the dialyzer,A3-out was 3102 and A4-out 3256,which were the main compositions of A3 and A4,accounted for 79.63% and 53.92%,respectively.Conclusion APS with Mw about 5000 Da exhibit better antitumor effect and immunological activity.Refluxing,microwave assistance extractions,and membrane enrichment techniques bring different cases on Mw distribution,components and pharmacodynamic action,and obviously exhibit relationship among component,Mw distribution,and pharmacological action.YANG Yi-fang1,FENG Jing-qian1,XU Hai-yan1,PENG Dan-ming2,HU Jian-ping2 1.Shanghai Institute of Pharmaceutical Industry,Shanghai 200040,China 2.Jiangxi Provincial Academy of Traditional Chinese Medicine,Nanchang 330077,China 2010Chinese Herbal Medicines2010,2,1:24
8Investigation of the effect of berberines alkaloids in Coptis chinensis Franch on Bacillus shigae growthby microcalorimetry显示文摘The inhibitory effects of five berberines alkaloids (BAs) from rhizoma of Coptis chinensis Franch, a traditional Chinese medicinal (TCM) herb, on Bacillus shigae (B. shigae) growth were investigated by microcalorimetry. The power-time curves of B. shigae with and without BAs were acquired; meanwhile, the extent and duration of inhibitory effects on the metabolism were evaluated by growth rate constants (k1, k2), half inhibitory ratio (IC50), maximum heat output (Pmax), and peak time (tp). The values of k1 and k2 of B. shigae in the presence of the five BAs decreased with the increasing concentrations of BAs. Moreover, Pmax was reduced, and the value of tp increased with increasing concentrations of the five drugs. The inhibitory activity varied with different drugs. IC50 of the five BAs was respectively 75 μg/mL for berberine, 90 μg/mL for coptisine, 115 μg/mL for palmatine, 220 μg/mL for epiberberine, and 400 μg/mL for jatrorrhizine. The sequence of antimicrobial activity of the five BAs: berberine > coptisine > palmatine > epiberberine > jatrorrhizine. The functional groups methylenedioxy at C2 and C3 on phenyl ring improve antimicrobial activity more strongly than methoxyl at C2 and C3 on phenyl ring. 9Effect of Fuzhenghuayu decoction on vascular endothelial growth factor secretion in hepatic stellate cells显示文摘Objective: To investigate the effect of Fuzhenghuayudecoction on autocrine activation of hepatic stellatecell (HSC).Methods: The drug serum containing Fuzhenghuayudecoction was collected from normal rats, and cul-tured with activated HSC in vitro. The conditionedmedium from the drug serum treated HSC was addedto primary cultured quiescent HSC. Cell prolifera-tion was assayed by tetrazolium colorimetric test,and the contents of type Ⅰ collagen and vascular endo-thelial growth factor (VEGF) in the supernatantwere measured with ELISA.Results: The conditioned medium from activatedHSC could stimulate the quiescent HSC proliferationand type Ⅰ collagen secretion. 10Infrared thermoimages display of body surface temperature reaction in experimental cholecystitis显示文摘AIM:To display the thermoimages of the body surface inexperimental cholecystitis,to observe the body surfacetemperature reaction in visceral disorders,and to study ifthe theory of body surface-viscera correlation is true and themechanism of temperature changes along the meridians.METHODS:By injecting bacteria suspension into the stricturebile duct and gallbladder,21 rabbits were prepared as acutepyogenic cholangiocholecystitis models,with another 8rabbits prepared by the same process except withoutinjection of bacteria suspension as control.The body surfaceinfrared thermoimagas were continuously observed on thehair shaven rabbit skin with AGA-782 thermovision 24 hbefore,1-11 d after and(2,3 wk)4 wk after the operation witha total of over 10 records of thermoimeges.RESULTS:Twelve cases out of 21 rabbits with cholecystitisrevealed bi-lateml longitudinal high temperature lines in itstrunk;with negative findings in the control group.The high-temperature line appeared on d 1-d 2,first in the right trunk,after the preparation of the model,about 7 d after the modelpreparation,the lines appeared at the left side too,persisting for 4 wk.The hyper-temperature line revealed 1.1-2.7 ℃ higher than before the model preparation,0.7-2.5℃higher than the surrounding skin.The length of the hightemperature line might reach a half length of the body trunk,or as long as the whole body itself.CONCLUSION: The appearance of the longitudinal high temperature lines at the lateral aspects of the trunk in the experimental group is directly bound up with the experimental animals pyogenic cholecystitis, with its running course quite similar to that of the Gallbladder Channel of Foot Shaoyang, but different to the zones of hyperalgesia and site of referred pain in cholecystitis.Dong Zhang Yuan-Gen Zhu Shu-You Wang Hui-Min Ma Yan Yan Ye Wei-Xing Fu Wei-Guo Hu,Institute of Acupuncture,China Academy of Traditional Chinese Medicine,Beijng 100700,China 2002World Journal of Gastroenterology2002,8,2:19
11Metabonomic phenotype and identification of “heart blood stasis obstruction pattern” and “qi and yin deficiency pattern” of myocardial ischemia rat models显示文摘The traditional Chinese medicine concepts of 'Xinxueyuzuzheng (heart blood stasis obstruction pattern)' and 'Qiyinliangxuzheng (qi and yin deficiency pattern)' for myocardial ischemia rat models were constructed in the present study. Endogenous metabolites in rat plasma were analyzed using the GC/TOF-MS-based metabonomic method. Significant metabolic differences were observed between the control and two model groups, and the three groups were distinguished clearly by pattern recognition. Compared with those of the control, the levels of hydroxyproline, threonic acid, glutamine and citric acid were strikingly up or down-regulated in model rats. The metabolites contributing most to the classification between the two 'pattern' rats were identified, such as valine, serine, threonine, ornithine, hydroxyproline, lysine, 2-hydroxybutanoic acid, 3-hydroxybutanoic acid, galactofuranose and inositol. These compounds were indicated as the potential biomarkers. The results suggested that the two 'patterns' are involved in dysfunction in oxidative stress, energy metabolism and amino acid metabolism. 14Progress in research into the mechanism of Radix salviae miltiorrhizae in treatment of acute pancreatitis显示文摘BACKGROUND: Acute pancreatitis (AP) is a common acute abdomen with high mortality, multiple complications and complicated causes. There is no effective therapy for AP. Radix salviae miltiorrhizae (Danshen), a traditional herbal medicine, has a low price and a wide range of clinical applications. It is effective to promote blood flow, eliminate stagnancy, and relieve pain. It is also found to be effective in treating AP. We reviewed the progress in research into the mechanism of Radix salviae miltiorrhizae in treatment of AP. DATA SOURCES: An english-language literature search was conducted using MEDLINE (1988-2005) on Radix salviae miltiorrhizae (Danshen) and acute pancreatitis. 15Rb1 protects endothelial cells from hydrogen peroxide-induced cell senescence by modulating redox status显示文摘Objectives Endothelial senescence has been proposed to be involved in endothelial dysfunction and atherogenesis. This study investigates the effects of ginsenoside Rbl, a major constituent of ginseng,on H2O2-induced endothelial senescence.Methods Primary human umbilical vein endothelial cells(HUVECs) senescence was induced by H2O2 as judged by senescence-associated P-galactosidase assay (SA-P-gal).Fntracellur superoxide dismutase(S0D1) activity and malondialdehyde(MDA) level were determined by commercial kit.S0D1 mRNA and protein expression were analyzed by real time PCR and Western blot.Reactive oxygen species(ROS) were determined by flow cytometry.Results Rb1 was found to reverse endothelial senescence,as witnessed by a significant decrease of senescent cell numbers. Rbl could markedly increase intracellular SOD activity, decrease the MDA level,and suppress the generation of intracellular ROS in H2O2-treated HUVECs.Consistent with these findings,Rbl can effectively restore SOD1 mRNA and protein expression which decreased in H2O2 treated cells. 16Function and regulation of cholecystokinin-octapeptide, β-endorphin and gastrin in anorexic infantile rats treated with ErBao Granules显示文摘AIM To study the role of cholecystokinin octapeptide ( CCK-8), β-endorphin ( β-EP), and gastrin in an anorexic infantile rat model and no subsequent regulation of nose peptides by the Yunpi complex prescription ErBao Granule.METHODS We fed infantile rats with special prepared forage. A liquid extract of ErBao Granule was administered to the rats daily for 3weeks, CCK-8, β-EP, and gastrin concentrations in hypothalamus, gastric antrum, and plasma of the rats were measured by radioimmunoassay,and were compared with controls.RESULTS Treatment of rats with ErBao Granule inhibited CCK-8 secretion and increased β-EP and gastrin secretion. CCK-8 concentration in hypothalamus and plasma of model control group increased significantly and correlated negatively with food intake of models.respectively. β-EP concentration in gastric antrum and plasma of model control group decreased significantly and showed a positive correlation with food intake of models,respectively. Hypothalamus concentration of β-EP was similar in models and controls. Gastrin concentration in gastric antrum of models was lower than in the blank control group, and correlated positively to food intake of models.Finally, CCK-8 concentrations in plasma of rats showed a positive correlation with plasma β-EP(r- 0.68, P<0.05).CONCLUSION The increased plasma and hypothalamus concentration of CCK-8,decreased gastric antrum and plasma level of β-EP. and decreased gastric antrum concentration of gastrin are associated significantly with the anorexia of infantile anorexic rat models produced by special forage. 19Down-Regulation of CD44 Contributes to the Differentiation of HL-60 Cells Induced by ATRA or HMBA显示文摘CD44 is highly expressed in human acute myeloid leukemia (AML) cells. Some experiments had shown that it was possible to reverse differentiation blockage in AML cells by CD44 ligation with specific antibodies, indicating that CD44 was closely related to the differentiation of leukemia cells. The differentiation of acute promyelocytic leukemia cell line HL-60 cells could be induced by all trans-retinoic acid (ATRA) and hexamethylene bisacetamide (HMBA), but so far the mechanism was not demonstrated clearly. In the present study, we investigated whether ATRA or HMBA induced the growth arrest of HL-60 cells by down-regulating the expression of CD44. The results indicated that the proliferation of HL-60 cells was obviously inhibited and the differentiation was induced by both ATRA and HMBA. The decreased expression of CD44 and cyclin E mRNA, and the increased expression of p27 and p21 at mRNA levels were observed. Furthermore, there was a negative correlation between the expression of CD44 and p27.
