| 2 | Decreased fragile histidine triad expression in colorectal cancer and its association with apoptosis inhibition显示文摘AIM: To detect the expression of fragile histidine triad (FHIT) in normal colorectal tissue, colorectal adenoma and colorectal cancer (CRC) tissue, and to analyze its relationship with the clinicopathological features of CRC, and apoptosis-associated proteins (Bcl-2, Bax, survivin) and apoptosis in colorectal cancer. METHODS: FHIT mRNA analysis was performed by nested reverse transcription-polymerase chain reaction (RT-PCR) assay. Tissue microarray (TMA) was established to detect the expression of FHIT, Bcl-2, Bax and survivin genes in 80 CRC tissue specimens, 16 colorectal adenoma tissue specimens and 16 hemorrhoid (PPH) tissue specimens during the same period of time as the control. Citrate-microwave-SP was used as immunohistochemical method. The relationship between clinicopathological factors, such as differentiation grades and 5-year survival rate was observed. TUNEL assay was used to detect the apoptosis index in 80 CRC tissue specimens. RESULTS: Ten out of 26 (38.5%) CRC tissue specimens expressed aberrant FHIT transcripts, none of the aberrant FHIT transcripts was observed in the matchednormal tissue and colorectal adenoma tissue by nested RT-PCR assay. The positive rate of FHIT gene expression in normal colorectal tissue, colorectal adenoma and carcinoma tissue was 93.75%, 68.75% and 46.25%, respectively. Clinicopathological analysis of patients showed that the decreased FHIT gene expression was not associated with age, sex, serum CEA levels, tumor site and size, histological classification. However, the expression of FHIT was correlated with differentiation grades, pathological stages, lymph node metastases and 5-year survival rate after operation. The positive rate of apoptosis-associated proteins (Bax, Bcl-2 and survivin) in CRC tissue was 72.50%, 51.25% and 77.50%, respectively. The expression of these apoptosis- associated proteins in CRC tissue was correlated with the expression of FHIT. The mean apoptosis index in FHIT negative tumors was significantly lower than that in FHIT positive tumors (5.41 ± 0.23 vs 0.56 ± 0.10, P < 0.01). | 3 | Identification and quality analysis of Panax notoginseng and Panax vietnamensis var.fuscidicus through integrated DNA barcoding and HPLC显示文摘Objective:Root or rhizome of Panax notoginseng(Sanqi)is known for its eutherapeutic effects.Panax vietnamensis var.fuscidicus,called Yesanqi or Yuenan sanqi by local residents,is also commercially available.They are similar in morphology,leading to serious safety problems in clinical medication.It is necessary to find the rapid and efficient methods to identify them.Methods:P.notoginseng and P.vietnamensis var.fuscidicus were identified by DNA barcoding based on the ITS2 sequence.Notoginsenoside R1 and ginsenosides(Rb1,Rg1,Re,Rd,Rc,and Rb2)were analyzed in the roots,fibrils,stems,leaves,and flowers of P.notoginseng and P.vietnamensis var.fuscidicus using high-performance liquid chromatography(HPLC).Results:P.notoginseng and P.vietnamensis var.fuscidicus were separated into branches of divergent clusters,and P.vietnamensis var.fuscidicus and Panax vietnamensis were clustered into a clade with 98%similarity according to DNA barcoding analysis.The chemical compositions of P.notoginseng and P.vietnamensis var.fuscidicus were similar in roots;while their compositions and contents of notoginsenoside R1 and ginsenosides in flowers,leaves,stems,and fibrils were different.Conclusion:ITS2 is a rapid and efficient method to identify P.notoginseng and P.vietnamensis var.fuscidicus.HPLC analysis indicated that pharmacological action might be different between P.notoginseng and P.vietnamensis var.fuscidicus. | Juan Yang Lin-lin Dong Guang-fei Wei Hao-yu Hu Guang-wei Zhu Jie Zhang Shi-lin then | 2018 | Chinese Herbal Medicines2018，10，2： | 7 |

METHODS:We enrolled a total of 1308 male workers aged from 22 to 63 years.Data was extracted from the workers'periodic health check-ups in hospitals.All cases were from the community of northern Taiwan. This was a cross-sectional observational study from July to September in 2004.We grouped all cases into four groups,based on the quartile of homeostasis model assessment.The top fourth quartile group was defined as the group with insulin resistance.We performed multivariate logistic regression analysis for the odds ratio of the risk factors for insulin resistance. RESULTS:Compared with metabolic syndrome,the coexistence of both factors had a 4.3-fold(95%CI: 2.7-6.8)increased risk,which was more than metabolic syndrome with a 3.6-fold(95%CI:2.6-5.0)increased risk.The two factors had a synergistic effect.The synergistic index of obesity and elevated alanine aminotransferase(ALT)was 2.1(95%CI:1.01-4.3).CONCLUSION:Obesity and elevated ALT are associatedwith insulin resistance.The effects are synergistic.
