| 1 | Non-viral and viral delivery systems for CRISPR-Cas9 technology in the biomedical field显示文摘The clustered regularly interspaced short palindromic repeats(CRISPR)-associated protein 9(CRISPR-Cas9) system provides a novel genome editing technology that can precisely target a genomic site to disrupt or repair a specific gene. Some CRISPR-Cas9 systems from different bacteria or artificial variants have been discovered or constructed by biologists, and Cas9 nucleases and single guide RNAs(sgRNA) are the major components of the CRISPR-Cas9 system. These Cas9 systems have been extensively applied for identifying therapeutic targets, identifying gene functions, generating animal models, and developing gene therapies.Moreover, CRISPR-Cas9 systems have been used to partially or completely alleviate disease symptoms by mutating or correcting related genes. However, the efficient transfer of CRISPR-Cas9 system into cells and target organs remains a challenge that affects the robust and precise genome editing activity. The current review focuses on delivery systems for Cas9 mRNA, Cas9 protein, or vectors encoding the Cas9 gene and corresponding sgRNA. Non-viral delivery of Cas9 appears to help Cas9 maintain its on-target effect and reduce off-target effects, and viral vectors for sgRNA and donor template can improve the efficacy of genome editing and homology-directed repair. | 3 | Brake wear-derived particles:Single-particle mass spectral signatures and real-world emissions显示文摘Brake wear is an important but unregulated vehicle-related source of atmospheric particulate matter(PM).The single-particle spectral fingerprints of brake wear particles(BWPs)provide essential information for understanding their formation mechanism and atmospheric contributions.Herein,we obtained the single-particle mass spectra of BWPs by combining a brake dynamometer with an online single particle aerosol mass spectrometer and quantified real-world BWP emissions through a tunnel observation in Tianjin,China.The pure BWPs mainly include three distinct types of particles,namely,Bacontaining particles,mineral particles,and carbon-containing particles,accounting for 44.2%,43.4%,and 10.3%of the total BWP number concentration,respectively.The diversified mass spectra indicate complex BWP formation pathways,such as mechanical,phase transition,and chemical processes.Notably,the mass spectra of Ba-containing particles are unique,which allows them to serve as an excellent indicator for estimating ambient BWP concentrations.By evaluating this indicator,we find that approximately 4.0%of the PM in the tunnel could be attributable to brake wear;the real-world fleet-average emission factor of 0.28 mg km1 veh1 is consistent with the estimation obtained using the receptor model.The results presented herein can be used to inform assessments of the environmental and health impacts of BWPs to formulate effective emissions control policies. | Jiayuan Liu Jianfei Peng Zhengyu Men Tiange Fang Jinsheng Zhang Zhuofei Du Qijun Zhang Ting Wang Lin Wu Hongjun Mao | 2023 | Environmental Science and Ecotechnology2023,,3: | 0 |
| 4 | Mannogalactoglucan from mushrooms protects pancreatic islets via restoring UPR and promotes insulin secretion in TIDM mice显示文摘Type 1 diabetes mellitus(T1DM) lacks insulin secretion due to autoimmune deficiency of pancreaticβ-cells.Protecting pancreatic islets and enhancing insulin secretion has been therapeutic approaches.Mannogalactoglucan is the main type of polysaccharide from natural mushroom,which has potential medicinal prospects.Nevertheless,the antidiabetic property of mannogalactoglucan in T1DM has not been fully elucidated.In this study,we obtained the neutral fraction of alkali-soluble Armillaria mellea polysaccharide(AAMP-N) with the structure of mannogalactoglucan from the fruiting body of A.mellea and investigated the potential therapeutic value of AAMP-N in T1DM.We demonstrated that AAMP-N lowered blood glucose and improved diabetes symptoms in T1DM mice.AAMP-N activated unfolded protein response(UPR) signaling pathway to maintain ER protein folding homeostasis and promote insulin secretion in vivo.Besides that,AAMP-N promoted insulin synthesis via upregulating the expression of transcription factors,increased Ca^(2+) signals to stimulate intracellular insulin secretory vesicle transport via activating calcium/calmodulin-dependent kinase Ⅱ(CamkⅡ) and cAMP/PKA signals,and enhanced insulin secretory vesicle fusion with the plasma membrane via vesicle-associated membrane protein 2(VAMP2).Collectively,these studies demonstrated that the therapeutic potential of AAMP-N on pancreatic islets function,indicating that mannogalactoglucan could be natural nutraceutical used for the treatment of T1DM. | Ting Liu Si Chen Yunhe Qu Lujuan Zheng Xiaoxuan Yang Shuhan Men Yuanning Wang Hanrui Ma Yifa Zhou Yuying Fan | 2024 | Food Science and Human Wellness2024,13,3: | 0 |
| 5 | Single-shot compressed optical field topography显示文摘Femtosecond lasers are powerful in studying matter's ultrafast dynamics within femtosecond to attosecond time scales.Drawing a three-dimensional(3D)topological map of the optical field of a femtosecond laser pulse including its spatiotemporal amplitude and phase distributions,allows one to predict and understand the underlying physics of light interaction with matter,whose spatially resolved transient dielectric function experiences ultrafast evolution.However,such a task is technically challenging for two reasons:first,one has to capture in single-shot and squeeze the 3D information of an optical field profile into a two-dimensional(2D)detector;second,typical detectors are only sensitive to intensity or amplitude information rather than phase.Here we have demonstrated compressed optical field topography(COFT)drawing a 3D map for an ultrafast optical field in single-shot,by combining the coded aperture snapshot spectral imaging(CASSI)technique with a global 3D phase retrieval procedure.COFT can,in single-shot,fully characterize the spatiotemporal coupling of a femtosecond laser pulse,and live stream the light-speed propagation of an air plasma ionization front,unveiling its potential applications in ultrafast sciences. | HAOCHENG TANG TING MEN XIANGLEI LIU YAODAN HU JINGQIN SU YANLEI ZUO PING LI JINYANG LIANG MICHAEL C.DOWNER ZHENGYAN LI | 2022 | Light(Science & Applications)2022,11,9: | 0 |
