1Biological function of Presenilin and its role in AD pathogenesis显示文摘Presenilins(PSs)are the catalytic core of γ-secretase complex.However,the mechanism of FAD-associated PS mutations in AD pathogenesis still remains elusive.Here we review the general biology and mechanism of γ-secretase and focus on the catalytic components–presenilins and their biological functions and contributions to the AD pathogenesis.The functions of presenilins are divided into γ-secretase dependent and γ-secretase independent ones.The γ-secretase dependent functions of presenilins are exemplified by the sequential cleavages in the processing of APP and Notch;the γ-secretase independent functions of presenilins include stabilizing β-catenin in Wnt signaling pathway,regulating calcium homeostasis and their interaction with synaptic transmission.Shuting Zhang Mingming Zhang Fang Cai Weihong Song 2013Translational Neurodegeneration2013,2,1:1

3The genomic characteristics and pathogenicity of a mammalian orthoreovirus within a new lineage from wild pika in plateau显示文摘Emerging and re-emerging viruses from wild animals have seriously threatened the health of humans and domesticated animals in recent years.Herein,we isolated a new mammalian orthoreovirus(MRV),Pika/MRV/GCCDC7/2019(PMRV-GCCDC7),in the Qinghai-Tibet Plateau wild pika(Ochotona curzoniae).Though the PMRVGCCDC7 shows features of a typical reovirus with ten gene segments arranged in 3:3:4 in length,the virus belongs to an independent evolutionary branch compared to other MRVs based on phylogenetic tree analysis.The results of cellular susceptibility,species tropism,and replication kinetics of PMRV-GCCDC7 indicated the virus could infect four human cell lines(A549,Huh7,HCT,and LoVo)and six non-human cell lines,including Vero-E6,LLCMK2,BHK-21,N2a,MDCK,and RfKT cell,derived from diverse mammals,i.e.monkey,mice,canine and bat,which revealed the potential of PMRV-GCCDC7 to infect a variety of hosts.Infection of BALB/c mice with PMRVGCCDC7 via intranasal inoculation led to relative weight loss,lung tissue damage and inflammation with the increase of virus titer,but no serious respiratory symptoms and death occurred.The characterization of the new reovirus from a plateau-based wild animal has expanded our knowledge of the host range of MRV and provided insight into its risk of trans-species transmission and zoonotic diseases.Kexin Zong Yuanyuan Guo Jingdong Song Maoshun Liu Junfeng Hao Jie Zhang Xin Li Shiyan Zhu Shuting Huo Ziqian Xu Peipei Liu Yingze Zhao Yuhai Bi Jiapeng Qu George F.Gao Jun Liu 2023Virologica Sinica2023,38,6:0

5Vagal-mAChR4 signaling promotes Friend virus complex(FV)-induced acute erythroleukemia显示文摘Erythroleukemia belongs to acute myeloid leukemia(AML)type 6(M6),and treatment remains difficult due to the poor prognosis of the disease.Friend virus(FV)is a complex of two viruses:Friend murine leukemia virus(F-MuLV)strain along with a defective spleen focus-forming virus(SFFV),which can induce acute eryth-roleukemia in mice.We have previously reported that activation of vagalα7 nicotinic acetylcholine receptor(nAChR)signaling promotes HIV-1 transcription.Whether vagal muscarinic signaling mediates FV-induced erythroleukemia and the underlying mechanisms remain unclear.In this study,sham and vagotomized mice were intraperitoneally injected with FV.FV infection caused anemia in sham mice,and vagotomy reversed this change.FV infection increased erythroblasts ProE,EryA,and EryB cells in the spleen,and these changes were blocked by vagotomy.In bone marrow,FV infection reduced EryC cells in sham mice,an effect that was coun-teracted by vagotomy.FV infection increased choline acetyltransferase(ChAT)expression in splenic CD4^(+)and CD8þT cells,and this change was reversed by vagotomy.Furthermore,the increase of EryA and EryB cells in spleen of FV-infected wild-type mice was reversed after deletion of ChAT in CD4^(+)T cells.In bone marrow,FV infection reduced EryB and EryC cells in sham mice,whereas lack of ChAT in CD4^(+)T cells did not affect this change.Activation of muscarinic acetylcholine receptor 4(mAChR4)by clozapine N-oxide(CNO)significantly increased EryB in the spleen but decreased the EryC cell population in the bone marrow of FV-infected mice.Thus,vagal-mAChR4 signaling in the spleen and bone marrow synergistically promotes the pathogenesis of acute erythroleukemia.We uncover an unrecognized mechanism of neuromodulation in erythroleukemia.Shuting Song Zhekai Lin Caiqi Zhao Jing Wen Jie Chen Shitao Xie Huaxin Qi Jianhua Wang Xiao Su 2023Virologica Sinica2023,38,3:0

7Parallel isolation of calicivirus and reovirus from lethal co-infected mink during a potential epidemic of farmed mink infections显示文摘Mink has been identified as an animal with susceptibility to SARS-CoV-2 and also as the only animal with evidence to transmit the virus back to humans.Thus,the surveillance of viruses among high-density farmed minks has a significant meaning for the control of zoonotic emerging diseases in humans.Within anal swabs of minks that died of unknown causes in a mink farm,mink calicivirus(MCV)and mammalian orthoreovirus(MRV)were detected and simultaneously observed within MDCK cell culture from the sample of the same lethal mink.The parallel isolation was successfully performed by utilizing cell lines from different host sources with distinct viral sensitivities,i.e.Mv.1.Lu and Vero-E6 and the two viruses were independently separated.The prevalence of the virus among the minks and its genomic characteristics were investigated through deep sequencing technology.Phylogenetic analysis of the viral genome showed a close relationship of the newly isolated MCV-GCCDC8-2020 with MCV strains belonging to the genus Vesivirus,but with unique mutations derived from the major structural protein(VP1).The reovirus MRV-GCCDC9-2020 isolated from the same mink belongs to serotype 3 mammalian orthoreovirus and genome analysis showed a potential reassortment derived from reoviruses in different species.This study provides a beneficial reference on viral co-infection within disease investigation in farmed minks and raises the concern for the virus surveillance among the high-density fed animal farms.Yuanyuan Guo William J.Liu Jingdong Song Kexin Zong Hao Lin Xin Li Shuting Huo Sai Liu Hongli Ran Hongmei Li Peipei Liu He Huang George F.Gao Guizhen Wu 2021Biosafety and Health2021,3,5:0
