1Soil physical properties and maize root growth under different tillage systems in the North China Plain显示文摘The standard cultivation system in the North China Plain is double cropping of winter wheat and summer maize. The main effects of this cultivation system on root development and yield are decreases in soil nutrient content and depth of the plow layer under either long-term no-tillage or rotary tillage before winter wheat sowing and no tillage before summer maize sowing. In this study, we investigated the combined effects of tillage practices before winter wheat and summer maize sowing on soil properties and root growth and distribution in summer maize. Zhengdan 958(ZD958) was used as experimental material, with three tillage treatments: rotary tillage before winter wheat sowing and no tillage before summer maize sowing(RTW + NTM), moldboard plowing before winter wheat sowing and no tillage before summer maize sowing(MPW + NTM), and moldboard plowing before winter wheat sowing and rotary tillage before summer maize sowing(MPW + RTM).Tillage practice showed a significant(P < 0.05) effect on grain yield of summer maize. Grain yields under MPW + RTM and MPW + NTM were 30.6% and 24.0% higher, respectively, than that under RTW + NTM. Soil bulk density and soil penetration resistance decreased among tillage systems in the order RTW + NTM > MPW + NTM > MPW + RTM. Soil bulk densities were 3.3% and 515% lower in MPW + NTM and MPW + RTM, respectively, than that in RTW + NTM, and soil penetration resistances were respectively 17.8% and 20.4% lower,across growth stages and soil depths. Root dry matter and root length density were highest under MPW + RTM, with the resulting increased root activity leading to a yield increase of summer maize. 2The Epitope Study on the SARS-CoV Nucleocapsid Protein显示文摘The nucleocapsid protein (N protein) has been found to be an antigenic protein in a number of coronaviruses. Whether the N protein in severe acute respiratory syndrome-associated coronavirus (SARS-CoV) is antigenic remains to be elucidated. Using Western blot and Enzyme-linked Immunosorbent Assay (ELISA),the recombinant N proteins and the synthesized peptides derived from the N protein were screened in sera from SARS patients. All patient sera in this study displayed strong positive immunoreactivities against the recombinant N proteins,whereas normal sera gave negative immunoresponses to these proteins, indicating that the N protein of SARS-CoV is an antigenic protein. Furthermore, the epitope sites in the N protein were determined by competition experiments, in which the recombinant proteins or the synthesized peptides competed against the SARS-CoV proteins to bind to the antibodies raised in SARS sera. One epitope site located at the C-terminus was confirmed as the most antigenic region in this protein. A detailed screening of peptide with ELISA demonstrated that the amino sequence from Codons 371 to 407 was the epitope site at the C-terminus of the N protein. 3Characterization of a Mg_(95.5)Zn_(1.5)Y_(3)alloy both containing W phase and LPSO phase with or without heat treatment显示文摘The influence of solid solution treatment on the microstructure and corrosion resistance of as-cast Mg_(95.5)Zn_(1.5)Y_(3) alloy is characterized.The microstructure of the as-cast Mg_(95.5)Zn_(1.5)Y_(3) alloy mainly consisted ofα-Mg,W(Mg_(3)Zn_(3)Y_(2))phase,and the long period stacking ordered(LPSO)(Mg_(12)ZnY)phase.After solid solution treatment,most of the W phase disappears gradually with increasing solution treatment time,with only a small amount of W phase distributed as particle.The LPSO phase slightly dissolved into substrate,and its morphology transitions from blocky shape to rod shape.Solid solution treatment of Mg_(95.5)Zn_(1.5)Y_(3) exhibits excellent corrosion resistance,because the Y and Zn atoms became enriched in the matrix and the changed morphologies of the LPSO and W phases were modified through heat treatment.The alloy created with solid solution treatment at 520 ℃ for 10 hours exhibits corrosion potential of−1.419 V,suggesting a significant improvement in corrosion performance.Li Zhumin Wan Diqing Huang Yi Ye Shuting Hu Yinglin 2017Journal of Magnesium and Alloys2017,5,2:8
4Two selenium tolerant Lysinibacillus sp. strains are capable of reducing selenite to elemental Se efficiently under aerobic conditions显示文摘Microbes play important roles in the transport and transformation of selenium(Se) in the environment, thereby influencing plant resistance to Se and Se accumulation in plant. The objectives are to characterize the bacteria with high Se tolerance and reduction capacity and explore the significance of microbial origins on their Se tolerance, reduction rate and efficiency. Two bacterial strains were isolated from a naturally occurred Se-rich soil at tea orchard in southern Anhui Province, China. The reduction kinetics of selenite was investigated and the reducing product was characterized using scanning electron microscopy and transmission electron microscopy-energy dispersive spectroscopy. The bacteria were identified as Lysinibacillus xylanilyticus and Lysinibacillus macrolides,respectively, using morphological, physiological and molecular methods. The results showed that the minimal inhibitory concentrations(MICs) of selenite for L. xylanilyticus and L. macrolides were 120 and 220 mmol/L, respectively, while MICs of selenate for L.xylanilyticus and L. macrolides were 800 and 700 mmol/L, respectively. Both strains aerobically reduced selenite with an initial concentration of 1.0 mmol/L to elemental Se nanoparticles(SeNPs) completely within 36 hr. Biogenic SeNPs were observed both inside and outside the cells suggesting either an intra-or extracellular reduction process. Our study implied that the microbes from Se-rich environments were more tolerant to Se and generally quicker and more efficient than those from Se-free habitats in the reduction of Se oxyanions. 5Loss of stat3 function leads to spine malformation and immune disorder in zebrafish显示文摘STAT(Signal Transducers and Activators of Transcription) gene family members have been revealed to be involved in cell growth and differentiation in vertebrates. Despite their physiological importance,their functions are poorly studied at organ and systemic levels. In this study, we performed a genome-wide analysis using data from invertebrates to vertebrates to identify STAT genes and analyze their evolutionary history. Interestingly, the STAT gene family undergoes genome duplications during the evolutionary history with STAT3 homologues firstly appearing in the basal extant vertebrate, sea lamprey, suggesting its possible roles in spine formation. To investigate the functions of stat3 in fish species, TALEN technology was performed to generate mutant zebrafish lines. Stat3 mutant zebrafish showed no obvious defects at early developmental stage but displayed severe lateral and vertical curvature of the spine(scoliosis), spine fracture and the incomplete bone joints with narrower junction between vertebrae at early juvenile stage, as indicated by Alizarin red and Alcian blue staining,radiography and micro-computed tomography(Micro CT) analysis. Transcriptome analysis reveals dramatic alterations in a number of genes involved in immune and infection response, skeletal development and somatic growth, especially downregulated expression of collagen gene family, in the juvenile stat3 mutant zebrafish. Moreover, most of the collagen genes were detected to have abnormal expression pattern during the formation of spine deformities in stat3 mutants. 6Experimental Study on Interaction Between Degrees of Freedom in a Wave Buoy显示文摘Increasing degrees of freedom(DOFs)is a useful way to raise the power capture efficiency of oscillating wave energy converters.Thus,this study proposes a buoy with three DOFs,which are surge,heave,and pitch.The hydrodynamic performance and power capture efficiency of the buoy is physically modeled.Amplitudes of unidirectional and coupled motions are compared to analyze the interaction effect between freedoms under conditions with and without power take-off damping.The capture width ratio and corresponding growth rates are also calculated.Results show that the buoy makes a periodic sinusoidal(or approximate)movement in every DOF.Coupling effect can cause an increase in the amplitude in one DOF and a decrease in the amplitudes of the others.This phenomenon shows that the kinematic energy of the buoy redistributes to all DOFs compared with the unidirectional conditions.Adding DOFs can improve the power absorption of the buoy in most cases,but the number of DOFs is not the more the better.HUANG Shuting SHI Hongda CAO Feifei TAN Junzhe CHENG Haixun LI Demin LIU Shangze GONG Haoxiang TAO Ji 2019Journal of Ocean University of China2019,18,6:5
7A novel selective autophagy receptor,CCDC50,delivers K63 polyubiquitination-activated RIG-I/MDA5 for degradation during viral infection显示文摘Autophagy is a con served process that delivers cytosolic substa nces to the lysosome for degradatio n,but its direct role in the regulation of antiviral inn ate immu nity remains poorly un derstood.Here,through high-throughput screeni ng,we discovered that CCDC50 functions as a previously unknown autophagy receptor that negatively regulates the type I interferon(IFN)signaling pathway initiated by RIG-I-like receptors(RLRs),the sensors for RNA viruses.The expression of CCDC50 is enhanced by viral infection,and CCDC50 specifically recognizes K63-polyubiquitinated RLRs,thus delivering the activated RIG-I/MDA5 for autophagic degradation.The associatio n of CCDC50 with phagophore membrane protei n LC3 is con firmed by crystal structure analysis.In contrast to other known autophagic cargo receptors that associate with either the LIR-docking site(LDS)or the UIMdocking site(UDS)of LC3,CCDC50 can bind to both LDS and UDS,representing a new type of cargo receptor.In mouse models with RNA virus infection,CCDC50 deficiency reduces the autophagic degradation of RIG-I/MDA5 and promotes type I IFN responses,resulting in enhaneed viral resistance and improved survival rates.These results reveal a new link between autophagy and antiviral innate immune resp on ses and provide additional in sights into the regulatory mecha nisms of RLR-mediated antiviral signaling.Panpan Hou Kongxiang Yang Penghui Jia Lan Liu Yuxin Lin Zibo Li Jun Li Shuliang Chen Shuting Guo Ji'An Pan Junyu Wu Hong Peng Weijie Zeng Chunmei Li Yingfang Liu Deyin Guo 2021Cell Research2021,31,1:5

10Essential roles of stat5.1/stat5b in controlling fish somatic growth显示文摘Signal transducer and activator of transcription 5b(STAT5b) has been identified as a key downstream mediator of growth hormone(GH) signaling in somatic growth of mammalian. However, the corresponding homologue gene of Stat5b is unknown in fish species. In this study, we generated loss-offunction mutants in stat5.1 and stat5.2, two stat5 homologues existing in zebrafish. In stat5.1-deficient zebrafish, a significant reduction of body length and body weight was detected in the embryos/larvae and adults compared with the wild-type control fish, and sexual size dimorphism in adult zebrafish was also eliminated. However, the stat5.2-deficient zebrafish displayed a normal developmental phenotype during all lifespan. Chromatin immunoprecipitation combined with deep sequencing(Ch IP-seq) method was adopted to further investigate the potential transcriptional targets of Stat5 protein and cast much light upon the biological function of Stat5. We identified more than 800 genes as transcriptional targets of Stat5 during zebrafish embryogenesis. KEGG analysis indicated that the Stat5 target gene network is predominantly linked to the metabolic pathways, neuroactive ligand-receptor interaction and JAK-STAT signaling pathways. Further validation studies suggested that Stat5.1 protein could directly regulate the expression of gh1, and stat5.1-mutated zebrafish showed a reduction of gh1 mRNA level. In the present study, stat5.1 was revealed as the corresponding homologue gene of Stat5b in fish species. 13Construction of robust coupling interface between MoS2 and nitrogen doped graphene for high performance sodium ion batteries显示文摘As a layered inorganic material,MoS2 has recently attracted intensive attention as anode for sodium ion batteries(SIBs).However,this anode is plagued with low electronic conductivity,serious volume expansion and sluggish kinetics,resulting in capacity fading and poor rate performance.Herein,we develop an interface engineering strategy to substantially enhance the sodium storage performance of MoS2 by incorporating layered MoS2 into three dimensional N-doped graphene scaffold.The strong coupling-interface between MoS2 and N-doped graphene scaffold can not only stabilize the MoS2 structure during sodium insertion/extraction processes,but also provide plenty of anchor sites for additional surface sodium storage.The 3D MoS2@N-doped graphene composite as anode for SIBs performs an outstanding specific capacity of 667.3 mA h g^-1 at 0.2 A g^-1,a prolonged stability with a capacity retention of 94.4%after 140cycles and excellent rate capability of 445 mA h g^-1 even at a high rate of 10 A g^-1.We combined experiment and theoretical simulation to further disclose the interaction between MoS2 and N-doped graphene,adsorption and diffusion of sodium on the composite and the corresponding sodium storage mechanism.This study opens a new door to develop high performance SIBs by introducing the interface engineering technique.Yun Qiao Jiawei Wu Xiaoguang Cheng Yudong Pang Zhansheng Lu Xiangdong Lou Qingling Li Jin Zhao Shuting Yang Yang Liu 2020Journal of Energy Chemistry2020,29,9:3
14Design and experimental performance verification of a thermal property test-bed for lunar drilling exploration显示文摘Chinese Chang'e lunar exploration project aims to collect and return subsurface lunar soil samples at a minimum penetration depth of 2 m in 2017. However, in contrast to those on the Earth, automated drilling and sampling missions on the Moon raise the risk of burning bits.Test-beds are required for testing the thermal properties of drill tools in a lunar environment. In this paper, a novel temperature measuring method based on thermocouples and a slip ring was proposed. Furthermore, a data acquisition system for a drilling process was designed. A vacuous,cryogenic, and anhydrous soil environment simulating the lunar surface was established. A drilling test-bed that can reach a depth of 2.2 m was developed. A control strategy based on online monitoring signals was proposed to improve the drilling performance. Vacuum and non-vacuum experiments were performed to test the temperature rising effect on drill tools. When compared with the non-vacuum experiment, the vacuum temperature rise resulted in a 12 °C increase. 15A chromosome-scale genome assembly of Artemisia argyi reveals unbiased subgenome evolution and key contributions of gene duplication to volatile terpenoid diversity显示文摘Artemisia argyi Le´vl.et Vant.,a perennial Artemisia herb with an intense fragrance,is widely used in traditional medicine in China and many other Asian countries.Here,we present a chromosome-scale genome assembly of A.argyi comprising 3.89 Gb assembled into 17 pseudochromosomes.Phylogenetic and comparative genomic analyses revealed that A.argyi underwent a recent lineage-specificwhole-genomeduplication(WGD)event after divergence fromArtemisia annua,resulting in two subgenomes.Wedeciphered the diploid ancestral genome of A.argyi,and unbiased subgenome evolution was observed.The recent WGD led to a large number of duplicated genes in the A.argyi genome.Expansion of the terpene synthase(TPS)gene family through various types of gene duplication may have greatly contributed to the diversity of volatile terpenoids in A.argyi.In particular,we identified a typical germacrene D synthase gene cluster within the expanded TPS gene family.The entire biosynthetic pathways of germacrenes,(+)-borneol,and(+)-camphor were elucidated in A.argyi.In addition,partial deletion of the amorpha-4,11-diene synthase(ADS)gene and loss of function of ADS homologs may have resulted in the lack of artemisinin production in A.argyi.Our study provides newinsights into the genome evolution of Artemisia and lays a foundation for further improvement of the quality of this important medicinal plant.Hongyu Chen Miaoxian Guo Shuting Dong Xinling Wu Guobin Zhang Liu He Yuannian Jiao Shilin Chen Li Li Hongmei Luo 2023Plant Communications2023,4,3:2
16GENERATOR VIBRATION FAULT DIAGNOSIS METHOD BASED ON ROTOR VIBRATION AND STATOR WINDING PARALLEL BRANCHES CIRCULATING CURRENT CHARACTERISTICS显示文摘Rotor vibration characteristics are first analyzed, which are that the rotor vibration of fundamental frequency will increase due to rotor winding inter-turn short circuit fault, air-gap dynamic eccentricity fault, or imbalance fault, and the vibration of the second frequency will increase when the air-gap static eccentricity fault occurs. Next, the characteristics of the stator winding parallel branches circulating current are analyzed, which are that the second harmonics circulating current will increase when the rotor winding inter-turn short circuit fault occurs, and the fundamental circulating current will increase when the air-gap eccentricity fault occurs, neither being strongly affected by the imbalance fault. Considering the differences of the rotor vibration and circulating current characteristics caused by different rotor faults, a method of generator vibration fault diagnosis, based on rotor vibration and circulating current characteristics, is developed. 17Effects of sludge characteristics on electrical resistance and energy consumption during electro-dewatering process显示文摘The increase of electrical resistance(ER)and energy consumption(EC)during the later stage of dewatering is a major problem hindering the development of electro-dewatering(EDW)technology.As the variations of sludge characteristics are significant during the EDW process,the relationships between sludge characteristics and ER and EC during EDW remain unclear.In this study,the effects of moisture content(MC),thickness,pH,conductivity,zeta potential,temperature,and gas volume on the ER and EC during the EDW process were statistically investigated using correlation and multiple linear regression analyses.Herein,the results showed that the ER of the sludge near the anode was primarily affected by pH,whereas the sludge near the cathode was primarily affected by the MC and conductivity.Further,sludge temperature and conductivity were the most reliable indicators to predict the EC near the anode and cathode,respectively.The results of this study provide theoretical guidance useful for solving the increase of ER and EC during the later stage of the EDW process.Li Sha Xiaoyan Yu Zhangxiang Wu Xingxin Liu Shuting Zhang 2021Journal of Environmental Sciences2021,33,3:2
18Genomic evidence of lung carcinogenesis associated with coal smoke in Xuanwei area, China显示文摘Xuanwei area, in southwestern China,harbors the highest female lung cancer rate in the country(Supplementary Table 1) [1]. 20Alterations in intestinal microbiota composition coincide with impaired intestinal morphology and dysfunctional ileal immune response in growing-finishing pigs under constant chronic heat stress显示文摘Background:Previous studies had shown that short-term acute heat stress(HS)affected the host’s metabolism and intestinal microbiota independent of feed intake(FI)reduction,and long-term calorie restriction caused intestinal morphological injuries and gut microbial alterations.However,research on the effects of constant chronic HS on intestinal microbial composition and the roles of FI reduction played in is limited.This study aimed to investigate the effects of 7-day constant chronic HS on the composition of intestinal microbes in growing-finishing pigs,and its relationship with pigs’performance,intestinal morphology,and ileal immune response.Twenty-four growingfinishing pigs(Duroc×Large White×Landrace,30±1 kg body weight)were randomly assigned to three treatments(n=8),1)thermal neutral(TN)conditions(25±1℃)with ad libitum FI,2)HS conditions(35±1℃)with ad libitum FI,3)pair-fed(PF)with HS under TN conditions to discriminate the confounding effects of dissimilar FI,and the FI was the previous day’s average FI of HS.The small intestinal segments(duodenum,jejunum,and ileum)and feces were collected on d 8.Results:Results indicated that HS drastically declined(P<0.05)average daily gain(ADG)and average daily feed intake(ADFI)(about 61%)in comparison with TN,and caused hyperpyrexia,meanwhile PF caused hypothermia.Morphological observation by light and electron microscopes showed that both HS and PF treatment decreased(P<0.05)the villus and microvillus height compared with TN.Additionally,HS increased(P<0.05)protein expression of heat shock protein 70 in the duodenum,jejunum,and ileum.Furthermore,the expression of tight junction protein zonula occluden-1(ZO-1)in the duodenum and ileum,and Occludin in the ileum were enhanced(P<0.05)compared with TN and PF.Moreover,HS significantly enhanced(P<0.05)the mRNA relative expression of inflammatory cytokines(TLR-2,TLR-4,and tumor necrosis factor-α(TNF-α),IL-6,IL-8,PG1–5,β-defensin 2(pBD-2)),mucins(mucin-1 and mucin-2)and P65 protein level in the ileal mucosa tissue.Intestinal microbiota analysis by 16S rRNA sequencing showed lower(P<0.10)αdiversity in both HS and PF,and a separated cluster ofβdiversity among groups.Compared with TN,HS but not PF mainly reduced(FDR<0.05)Bacteroidetes(phylum),Bacteroidia(class)and elevated the proportions of Proteobacteria(phylum,FDR<0.05),Bacillales(order,FDR<0.05),Planococcaceae(family,FDR<0.05),Kurthia(genus,FDR<0.05),Streptococcaceae(family,FDR<0.10)and Streptococcus(genus,FDR<0.10).Notably,Lactobacillales(order)was decreased(FDR<0.05)by PF alone.Furthermore,the Spearman correlation analysis indicated that the microbes prevalent in HS were positively(P<0.05)associated with intestinal morphological injuries indicators and ileal immune response parameters,and the microbes reduced in HS were negatively(P<0.05)with the performance data.Conclusions:Intestinal morphological injuries and ileal immune response caused by constant chronic HS independent of FI showed close connections with alterations in intestinal microbiota in growing-finishing pigs.Yunxia Xiong Shuting Cao Hao Xiao Qiwen Wu Hongbo Yi Zongyong Jiang Li Wang 2022Journal of Animal Science and Biotechnology2022,13,3:2
