5A mixed target estimation fusion algorithm based on Gibbs-GLMB and federated filter显示文摘Mixed targets are composed of point targets,extended targets,and group targets.The point target can produce one measurement at most,the extended target and the group target can produce multiple measurements,but the sub-goals of the group target have a certain dependency relationship.At this time,the estimated fusion of the group target is converted to the estimated fusion of sub-targets with formation motion structure,and the distance among the sub-targets is very close,which brings difficulties to the estimated fusion of mixed targets.This paper combines the adjacency matrix in graph theory to dynamically model the discernible group target and introduces the concept of deformation.Also,it uses the finite mixture model method to dynamically model the extended target.Then the Gibbs-GLMB algorithm is used to estimate the state and number of the mixed targets.A dynamic detection federated filter fusion algorithm is proposed to fuse the mixed targets state estimates.The effectiveness of the algorithm is verified in the final simulation.Yu Liu Zhangming Peng Shibo Gao Jiangning Li 2022IET Cyber-Systems and Robotics2022,4,1:1
6A non-ACE2 competing human single-domain antibody confers broad neutralization against SARS-CoV-2 and circulating variants显示文摘The current COVID-19 pandemic has heavily burdened the global public health system and may keep simmering for years.The frequent emergence of immune escape variants have spurred the search for prophylactic vaccines and therapeutic antibodies that confer broad protection against SARS-CoV-2 variants.Here we show that the bivalency of an affinity maturated fully human singledomain antibody(n3113.1-Fc)exhibits exquisite neutralizing potency against SARS-CoV-2 pseudovirus,and confers effective prophylactic and therapeutic protection against authentic SARS-CoV-2 in the host cell receptor angiotensin-converting enzyme 2(ACE2)humanized mice.The crystal structure of n3113 in complex with the receptor-binding domain(RBD)of SARS-CoV-2,combined with the cryo-EM structures of n3113 and spike ecto-domain,reveals that n3113 binds to the side surface of up-state RBD with no competition with ACE2.The binding of n3113 to this novel epitope stabilizes spike in up-state conformations but inhibits SARS-CoV-2 S mediated membrane fusion,expanding our recognition of neutralization by antibodies against SARS-CoV-2.Binding assay and pseudovirus neutralization assay show no evasion of recently prevalent SARS-CoV-2 lineages,including Alpha(B.1.1.7),Beta(B.1.351),Gamma(P.1),and Delta(B.1.617.2)for n3113.1-Fc with Y58L mutation,demonstrating the potential of n3113.1-Fc(Y58L)as a promising candidate for clinical development to treat COVID-19.Zhenlin Yang Yulu Wang Yujia Jin Yuanfei Zhu Yanling Wu Cheng Li Yu Kong Wenping Song Xiaolong Tian Wuqiang Zhan Ailing Huang Shanshan Zhou Shuai Xia Xiaoxu Tian Chao Peng Cuicui Chen Yibing Shi Gaowei Hu Shujuan Du Yuyan Wang Youhua Xie Shibo Jiang Lu Lu Lei Sun Yuanlin Song Tianlei Ying 2021Signal Transduction and Targeted Therapy2021,6,12:1

8Particle shape effect on hydrodynamics and heat transfer in spouted bed:A CFD-DEM study显示文摘Spouted bed has drawn much attention due to its good heat and mass transfer efficiency in many chemical units.Investigating the flow patterns and heat and mass transfer inside a spouted bed can help optimize the spouting process.Therefore,in this study,the effects of particle shape on the hydrodynamics and heat transfer in a spouted bed are investigated.This is done by using a validated computational fluid dynamics-discrete element method(CFD-DEM)model,considering volume-equivalent spheres and oblate and prolate spheroids.The results are analysed in detail in terms of the flow pattern,microstruc-ture,and heat transfer characteristics.The numerical results show that the prolate spheroids(Ar=2.4)form the largest bubble from the beginning of the spouting process and rise the highest because the fluid drag forces can overcome the interlocking and particle-particle frictional forces.Compared with spherical particles,ellipsoidal spheroids have better mobility because of the stronger rotational kinetic energy resulting from the rough surfaces and nonuniform torques.In addition,the oblate spheroid system exhibits better heat transfer performance benefiting from the larger surface area,while prolate spheroids have poor heat transfer efficiency because of their orientation distribution.These findings can serve as a reference for optimizing the design and operation of complex spouted beds.Dianyu E Peng Zhou Suya Guo Jia Zeng Jiaxin Cui Youyuan Jiang Yuanxiang Lu Zeyi Jiang Zhengquan Li Shibo Kuang 2022Particuology2022,20,10:0

10Gp350-anchored extracellular vesicles: promising vehicles for delivering therapeutic drugs of B cell malignancies显示文摘Although chimeric antigen receptor-modified(CAR)T cell therapy has been successfully applied in the treatment of acute B lymphocytic leukemia,its effect on Burkitt lymphoma(BL)and chronic B lymphocytic leukemia(B-CLL)is unsatisfactory.Moreover,fatal side effects greatly impede CAR T cell application.Extracellular vesicles(EVs)are excellent carriers of therapeutic agents.Nevertheless,EVs mainly accumulate in the liver when administered without modification.As an envelope glycoprotein of Epstein–Barr viruses,gp350 can efficiently bind CD21 on B cells.Here,gp350 was directly anchored onto red blood cell EVs(RBC-EVs)via its transmembrane region combined with low-voltage electroporation.The results showed that gp350 could anchor to RBC-EVs with high efficiency and that the resulting gp350-anchored RBC-EVs(RBC-EVs/gp350^(Etp))exhibited increased targeting to CD21+BL and B-CLL relative to RBC-EVs.After the loading of doxorubicin or fludarabine,RBC-EVs/gp350^(Etp) had powerful cytotoxicity and therapeutic efficacy on CD21+BL or B-CLL,respectively.Moreover,RBC-EVs/gp350^(Etp) loaded with a drug did not exhibit any apparent systemic toxicity and specifically induced the apoptosis of tumor B cells but not normal Bcells.Therefore,our findings indicate that drug-loaded RBC-EVs/gp350^(Etp) may be adopted in the treatment of CD21+B cell malignancies.Huiqing Xiu Xi Nan Danfeng Guo JiaoliWang Jiahui Li Yanmei Peng Guirun Xiong Shibo Wang Changjun Wang Gensheng Zhang Yunshan Yang Zhijian Cai 2022Asian Journal of Pharmaceutical Sciences2022,17,3:0
11Effect on proliferation and apoptosis of T24 cell lines via silencing DNMT1 with RNA interference显示文摘Expression of DNA methyltransferase 1(DNMT1),which plays an important role on aberrantly methylated CpG in the promoter regions of tumor sup-pressor genes(TSGs),is higher in bladder cancer cells than in normal bladder cells.Therefore,its overexpression is closely related to tumor formation.In this study,the eukaryotic vector pshRNA-DNMT1 was constructed and transfected into T24 cells.Levels of DNMT1 mRNA and protein were detected by reverse transcription-polymerase chain reaction(RT-PCR)and western blot.Relative to the blank control at the 24th,48th and 72nd hour after transfection of pshRNA-DNMT1,the inhibitory rates of DNMT1 mRNA levels in T24 cells were 28.44%,52.48%,70.91%,respectively.Those of DNMT1 proteins were 24.27%,57.79%,and 77.74%,respectively.Proliferation and apoptosis were assayed by MTT and flow cytometry with Annexin-V-FITC/PI staining.The growth inhibition rates of pshRNA-DNMT1 at the 24th,48th and 72nd hour after transfection of pshRNA-DNMT1 were(4.34¡0.76)%,(9.87¡1.54)%and(13.78¡1.93)%,respectively.There were statistically significant differ-ences between pshRNA-DNMT1 and the control blank at each time points(P,0.01);24,48 and 72 hours after T24 cells were transfected by pshRNA-DNMT1,the apoptosis rates of pshRNA-DNMT1 were(3.87¡0.81)%,(8.69¡1.23)%and(11.46¡1.24)%,respectively(P,0.01 vs blank control).Based on this case,our conclu-sion is that the recombinant plasmid pshRNA-DNMT1 can silence the expression of gene DNMT1 mRNA and protein effectively,and to some extent,it also can inhibit the proliferation of bladder cancer cell and promote the cellular apoptosis.Shilong ZHANG Fuqing ZENG Shibo PENG Liang WANG 2008Frontiers of Medicine2008,2,4:0

13Time Domain Full Waveform Inversion Based on Gradient Preconditioning with an Angle-Dependent Weighting Factor显示文摘There are lots of low wavenumber noises in the gradients of time domain full waveform inversion(FWI),which can seriously reduce the accuracy and convergence speed of FWI.Thus,we introduce an angle-dependent weighting factor to precondition the gradients so as to suppress the low wavenumber noises when the multi-scale FWI is implemented in the high frequency.Model experiments show that the FWI based on the gradient preconditioning with an angle-dependent weighting factor has faster convergence speed and higher inversion accuracy than the conventional FWI.The tests on real marine seismic data show that this method can adapt to the FWI of field data,and provide high-precision velocity models for the actual data processing.XIA Dongming SONG Peng LI Xishuang TAN Jun XIE Chuang WANG Shaowen LIU Kai ZHAO Bo MAO Shibo 2022Journal of Ocean University of China2022,21,6:0
