1Replacement of Dietary Fish Oil with Vegetable Oils Improves the Growth and Flesh Quality of Large Yellow Croaker(Larmichthys crocea)显示文摘We investigated the effect of the replacement of dietary fish oil with vegetable oils on the growth and flesh quality of large yellow croaker(Larmichthys crocea). The basal diet(FO) was formulated to contain 66.5% fish meal and 6.4% menhaden fish oil; whereas the other 3 experimental diets were formulated by replacing the fish oil with 50% soybean oil(SO50), 100% soybean oil(SO100) and 100% palm oil(PO100), respectively. The 4 diets were randomly assigned to 4 floating sea cages(3.0 m × 3.0 m × 3.0 m), and each was stocked with 250 fish individuals with an initial average weight of 245.29 g ± 7.45 g. The fish were fed to apparent satiation twice a day at 5:00 and 17:00, respectively, for 12 weeks. Experimental analysis showed that the specific growth rate of fish fed SO50 or PO100 were significantly higher than that of fish fed FO or SO100(P<0.05), and crude lipid contents of ventral muscle and viscera were significantly lower in fish fed FO than in those fed the other 3 diets(P<0.05). No significant differences in condition factor, viscerosomatic index, hepatosomatic index, gutted yield and colorimetric values of fish among the dietary treatments were observed(P>0.05). Compared to FO diet, SO50, SO100 and PO100 diets led to substantial decreases in the liquid loss and water loss from fresh fillets(1 d, 4℃)(P<0.05). Similarly, thiobarbituric acid reactive substance(TBARS) values of fillets under different storage conditions(1 d, 4℃; 7 d, 4℃; 4 weeks,-20℃; 8 weeks,-20℃) decreased significantly after partial or complete replacement of fish oil with vegetable oils. 2Microbial activity and community diversity in a variable charge soil as affected by cadmium exposure levels and time显示文摘Effects of cadmium(Cd) on microbial biomass,activity and community diversity were assessed in a representative variable charge soil(Typic Aquult) using an incubation study.Cadmium was added as Cd(NO3)2 to reach a concentration range of 0~16 mg Cd/kg soil.Soil extractable Cd generally increased with Cd loading rate,but decreased with incubation time.Soil microbial biomass was enhanced at low Cd levels(0.5~1 mg/kg),but was inhibited consistently with increasing Cd rate.The ratio of microbial biomass C/N varied with Cd treatment levels,decreasing at low Cd rate(<0.7 mg/kg available Cd),but increasing progressively with Cd loading.Soil respiration was restrained at low Cd loading(<1 mg/kg),and enhanced at higher Cd levels.Soil microbial metabolic quotient(MMQ) was generally greater at high Cd loading(1~16 mg/kg).However,the MMQ is also affected by other factors.Cd contamination reduces species diversity of soil microbial communities and their ability to metabolize different C substrates.Soils with higher levels of Cd contamination showed decreases in indicator phospholipids fatty acids(PLFAs) for Gram-negative bacteria and actinomycetes,while the indicator PLFAs for Gram-positive bacteria and fungi increased with increasing levels of Cd contamination.Jia-li SHENTU Zhen-li HE Xiao-e YANG Ting-qiang LI 2008Journal of Zhejiang University-Science B(Biomedicine & Biotechnology)2008,9,3:4
3Chinese family with atypical granular corneal dystrophy type I caused by the typical R555W mutation in TGFBI显示文摘AIM:To investigate the clinical features and genetic defects in four generations of a Chinese family affected with atypical granular corneal dystrophy type I(GCD type I).·METHODS:Family history and clinical data were recorded.Genomic DNA samples were obtained from peripheral blood leukocytes of all participated.Exons of the transforming growth factor-β-induced(TGFBI) gene were directly sequenced after being amplified by polymerase chain reaction(PCR),and multi-point linkage analysis using microsatellite makers flanking the gene was applied to identify the disease-causing mutation.·RESULTS:Clinical features were quite variable in patients,some patients only had opacities in the epithelium,and others revealed multiple bilateral circular,discrete,crumb-like opacities mainly in the epithelium,with several in different depths of corneal stroma,and the performance was different bilaterally,even in the same patient.Directly nucleotide sequencing revealed a heterozygous p.R555W mutation in the coding sequence of the TGFBI gene in all affected individuals of the family,but was not found in all unaffected.The maximum logarithm of odds(LOD) score obtained by multi-point analysis was detected at marker locus D5S393(LOD =2.740;α=1.000).·CONCLUSION:Our case presented with clinical futures and the pathogenic mutations in TGFBI gene,the phenotype of the pedigree was quite different from typical GCD type I,so we suggested that this phenotypewas a variant of GCD type I.These findings expand the knowledge about GCD type I,and demonstrate that molecular genetic analysis is important to make an accurate diagnosis of patients with variable corneal dystrophies in clinic.Su-Juan Zhao Ya-Nan Zhu Xing-Chao Shentu Qi Miao 2013International Journal of Ophthalmology(English edition)2013,6,4:4
4Hydrogen peroxide-induced apoptosis of human lens epithelial cells is inhibited by parthenolide显示文摘AIM: To explore the effect of parthenolide on hydrogen peroxide(H_2O_2)-induced apoptosis in human lens epithelial(HLE) cells. METHODS: The morphology and number of apoptotic HLE cells were assessed using light microscopy and flow cytometry. Cell viability was tested by MTS assay. In addition, the expression of related proteins was measured by Western blot assay. RESULTS: Apoptosis of HLE cells was induced by 200 μmol/L H_2O_2, and the viability of these cells was similar to the half maximal inhibitory concentration(IC50), as examined by MTS assay. In addition, cells were treated with either different concentrations(6.25, 12.5, 25 and 50 mol/L) of parthenolide along with 200 μmol/L H_2O_2 or only 50 μmol/L parthenolide or 200 mol/L H_2O_2 for 24 h. Following treatment with higher concentrations of parthenolide(50 μmol/L), fewer HLE cells underwent H_2O_2-induced apoptosis, and cell viability was increased. Further, Western blot assay showed that the parthenolide treatment reduced the expression of caspase-3 and caspase-9, which are considered core apoptotic proteins, and decreased the levels of phosphorylated nuclear factor-κB(NF-κB), ERK1/2 [a member of the mitogen-activated protein kinase(MAPK) family], and Akt proteins in HLE cells. 5Dystrophia canthorum in Waardenburg syndrome with a novel MITF mutation显示文摘AIM:To reveal a novel MITF gene mutation in Waardenburg syndrome(WS),which is an autosomal dominant inherited neurogenic disorder that consists of various degrees of sensorineural deafness and pigmentary abnormalities in the eyes,hair and skin.METHODS:The genetic analysis of the Chinese family was conducted by whole-exome sequencing,then the results were confirmed by Sanger sequencing.RESULTS:WS is classified into type I to IV,which are identified by the W index,clinical characteristics and additional features.The MITF gene mostly accounts for WS type II.In this study,a de novo heterozygous mutation in the MITF gene,c.638 A>G in exon 7,was identified in the patient diagnosed with WS type I features,as the W index was 2.17(over 2.10),with dystrophia canthorum,congenital bilateral profound hearing loss,bilateral heterochromia irides,premature greying of the hair,and excessive freckling on the face at birth.She also underwent refractive errors and esotropia,reduced pigmentation of the choroid and visible choroid vessels.The mutation was not found in previous studies or mutation databases.CONCLUSION:The novel mutation in the MITF gene,which altered the protein in amino acids 213 from the glutamic acid to glycine,is the genetic pathological cause for WS features in the patient.Those characteristics of this family revealed a novel genetic heterogeneity of MITF in WS,which expanded the database of MITF mutations and offered a possible in correcting the W index value of WS in distinct ethnicities.Moreover,ocular symptoms should be emphasized in all types of WS patients.Xia-Jing Tang Xi-Yuan Ping Chen-Qi Luo Xiao-Ning Yu Ye-Lei Tang Xing-Chao Shentu 2020International Journal of Ophthalmology(English edition)2020,13,7:3
6Relative bioavailability and pharmacokinetic comparison of two different enteric formulations of omeprazole显示文摘In order to comply with the requirements for a drug listed in China, the study was developed to compare the pharmacokinetics and relative bioavailability of two different enteric formulations of omeprazole (OPZ) in healthy Chinese subjects. A total of 32 volunteers participated in the study. Plasma concentrations were analyzed by non- stereospecific liquid chromatography/tandem mass spectrometric (LC-MS/MS) method. After administration of a single 40-mg dose of the two OPZ formulations, the comparative bioavailability was assessed by calculating individual AUC0-t (the area under the concentration-time curve from time zero to the last measurable concentration), AUC0-∞ (the area under the concentration-time curve extrapolated to infinity), Cmax (the maximum observed concentration), and Tpeak (the time to Cmax) values of OPZ, 5-hydroxyomeprazole (OH-OPZ), and omeprazole sulfone (OPZ-SFN), respectively. The 90% confidence intervals (CIs) of AUC0-t, AUC0-∞, and Cmax were 85.4%-99.0%/88.8%-98.6%/87.6%-99.4%, 85.5%-99.2%/89.0%-98.6%/88.5%-101.3%, and 72.3%-87.6%/79.6%-91.1%/88.4%-99.1% for OPZ/OH-OPZ/ OPZ-SFN, respectively, and Tpeak values did not differ significantly. 8Microbial Biomass and PLFA Profile Changes in Rhizosphere of Pakchoi(Brassica chinensis L.) as Affected by External Cadmium Loading显示文摘The effects of root activity on microbial response to cadmium(Cd) loading in the rhizosphere are not well understood.A pot experiment in greenhouse was conducted to investigate the effects of low Cd loading and root activity on microbial biomass and community structure in the rhizosphere of pakchoi(Brassica chinensis L.) on silty clay loam and silt loamy soil.Cd was added into soil as Cd(NO_3)_2 to reach concentrations ranging from 0.00 to 7.00 mg kg^(-1).The microbial biomass carbon(MBC) and community structure were affected by Cd concentration,root activity,and soil type.Lower Cd loading rates(<1.00 mg kg^(-1)) stimulated the growth of pakchoi and microorganisms,but higher Cd concentrations inhibited the growth of microorganisms.The content of phospholipid fatty acids(PLFAs) was sensitive to increased Cd levels.MBC was linearly correlated with the total PLFAs.The content of general PLFAs in the fungi was positively correlated with the available Cd in the soil,whereas those in the bacteria and actinomycetes were negatively correlated with the available Cd in the soil.These results indicated that fungi were more resistant to Cd stress than bacteria or actinomycetes,and the latter was the most sensitive to Cd stress.Microbial biomass was more abundant in the rhizosphere than in the bulk soil.Root activity enhanced the growth of microorganisms and stabilized the microbial community structure in the rhizosphere.PLFA analysis was proven to be sensitive in detecting changes in the soil microbial community in response to Cd stress and root activity.SHENTU Jia-Li HE Zhen-Li ZENG Yan-Yan HE Shan-Ying DU Shao-Ting SHEN Dong-Sheng 2014Pedosphere2014,24,4:3
9Overexpression of the CmJAZ1-like gene delays flowering in Chrysanthemum morifolium显示文摘Chrysanthemum(Chrysanthemum morifolium)is one of the four major cut-flower plants worldwide and possesses both high ornamental value and cultural connotation.As most chrysanthemum varieties flower in autumn,it is costly to achieve annual production.JAZ genes in the TIFY family are core components of the jasmonic acid(JA)signaling pathway;in addition to playing a pivotal role in plant responses to defense,they are also widely implicated in regulating plant development processes.Here,we characterized the TIFY family gene CmJAZ1-like from the chrysanthemum cultivar‘Jinba’.CmJAZ1-like localizes in the nucleus and has no transcriptional activity in yeast.Tissue expression pattern analysis indicated that CmJAZ1-like was most active in the root and shoot apex.Overexpressing CmJAZ1-like with Jas domain deletion in chrysanthemum resulted in late flowering.RNA-Seq analysis of the overexpression lines revealed some differentially expressed genes(DEGs)involved in flowering,such as the homologs of the flowering integrators FT and SOC1,an FUL homolog involved in flower meristem identity,AP2 domain-containing transcription factors,MADS box genes,and autonomous pathway-related genes.Based on KEGG pathway enrichment analysis,the differentially transcribed genes were enriched in carbohydrate metabolic and fatty acid-related pathways,which are notable for their role in flowering in plants.This study preliminarily verified the function of CmJAZ1-like in chrysanthemum flowering,and the results can be used in molecular breeding programs aimed at flowering time regulation of chrysanthemum.Yunxiao Guan Lian Ding Jiafu Jiang Yuanyue Shentu Wenqian Zhao Kunkun Zhao Xue Zhang Aiping Song Sumei Chen Fadi Chen 2021Horticulture Research2021,8,1:3
10Inhibition of NF-kB and Wnt/β-catenin/GSK3p Signaling Pathways Ameliorates Cardiomyocyte Hypertrophy and Fibrosis in Streptozotocin (STZ)-induced Type 1 Diabetic Rats显示文摘Type 1 diabetes mellitus(T1DM)is associated with an increased risk of diabetic cardiomyopathy(DCM).Nuclear factor kappa B(NF-kB)and Wnt/β-catenin/GSK3p have been demonstrated to play pathogenic roles in diabetes.In this study,we evaluated the roles of these two pathways in T1 DM-induced cardiomyopathy in rats.Streptozotocin(STZ)-induced type 1 diabetic rats were treated with pyrrolidine dithiocarbamate(PDTC)or meisoindigo(Me)to inhibit NF-kB and Wnt/β-catenin/GSK3P respectively for 4 or 8 weeks.As compared with untreated diabetic rats,treatment with either PDTC or Me partly attenuated the myocardial hypertrophy and interstitial fibrosis,improved cardiac function,and exhibited reduction in inflammatory reaction.In addition,we found that inhibiting NF-κB and Wnt/β-catenin/GSK3β pathways could regulate glucose and lipid metabolism.The effects were associated with the decrease of NF-κB activity and the downregulation of some proinflammatory cytokines,including tumor necrosis factor-alpha(TNF-α)and interleukin(IL)-2.Our data suggested that the activities of NF-κB and Wnt/β-catenin/GSK3β pathways were both increased and inhibiting NF-κB and Wnt/β-catenin/GSK3β signaling pathways might improve myocardial injury in T1DM rats.Jing-jing LIU Lu-mei SHENTU Ning MA Li-ying WANG Gui-min ZHANG Ying SUN Yan WANG Jun LI Yan-ling MU 2020Current Medical Science2020,40,1:2

12超声靶向击碎微泡技术介导P-选择素靶向阳离子超声微泡改善人血管内皮生长因子165基因转染缺血心肌的实验性研究(英文)显示文摘目的:构建一种靶向阳离子微泡,探讨其提高超声靶向击碎微泡技术(TCUMGT)介导的体内基因转染效率及治疗效果。创新点:提出运用TCUMGT介导的基因转染技术,利用微泡的携基因和靶向定位释放双项功能来上调缺血区的人血管内皮生长因子165(hVEGF_(165))的表达水平,发挥其成血管作用,从而改变缺血心肌的存活性。方法:通过聚乙二醇40硬脂酸酯、二硬脂酰基磷脂酰乙醇胺-聚乙二醇2000、pcDNA_(3.1)-hV EGF165和抗P-选择素单克隆抗体等制备P-选择素靶向阳离子微泡。微泡分成四组:(1)仅微泡(MB);(2)微泡+P-选择素(MB+P);(3)微泡+pcDNA_(3.1)-hVEGF_(165)质粒(MB+VEGFp);(4)微泡+P-选择素+pcDNA_(3.1)-hVEGF_(165)质粒(MB+P+VEGFp)。逆转录聚合酶链反应(RT-PCR)和酶联免疫吸附试验(ELISA)结果显示:TCUMGT成功转染hVEGF_(165)基因,并且通过P-选择素为靶点可以提高转染效率。另外与其他组相比,MB+P+VEGFp组的心肌血管密度增加和心功改善最为明显。结论:本研究表明,通过TCUMGT技术,靶向超声微泡可以有效识别缺血心肌,释放pcDNA_(3.1)-hVEGF_(165)重组质粒,提高心肌微环境,促进心肌功能的恢复。Wei-hui SHENTU Cao-xin YAN Chun-mei LIU Rui-xiang QI Yao WANG Zhao-xu HUANG Li-ming ZHOU Xiang-dong YOU 2018Journal of Zhejiang University-Science B(Biomedicine & Biotechnology)2018,19,9:2
13Metabolic syndrome risk factors and dry eye syndrome:a Meta-analysis显示文摘AIM:To explore the relationship between metabolic risk factors and dry eye syndrome(DES).METHODS:Retrieved studies on the association of metabolic syndrome risk factors(hypertension,hyperglycemia,obesity,and hyperlipidemia) and DES were collected from PubMed,Web of Science,and the Cochrane Library in December 2015.Odds ratio(OR) with 95% confidence interval(Cl) were pooled to evaluate the final relationship.Subgroup analyses were conducted according to diagnostic criteria of DES.RESULTS:Nine cross-sectional studies and three case-control studies were included in this Meta-analysis.The pooled results showed that people with hypertension,hyperglycemia,and hyperlipidemia had a higher risk of suffering from DES(P<0.05),especially the typical DES symptoms.On the other hand,obesity did not increase the risk of DES.CONCLUSION:The present Meta-analysis suggests that all metabolic risk factors except obesity were risk factors for DES.Ye-Lei Tang Ya-Lan Cheng Yu-Ping Ren Xiao-Ning Yu Xing-Chao Shentu 2016International Journal of Ophthalmology(English edition)2016,9,7:2
14Signaling pathways in Parkinson’s disease:molecular mechanisms and therapeutic interventions显示文摘Parkinson’s disease(PD)is the second most common neurodegenerative disease worldwide,and its treatment remains a big challenge.The pathogenesis of PD may be related to environmental and genetic factors,and exposure to toxins and gene mutations may be the beginning of brain lesions.The identified mechanisms of PD includeα-synuclein aggregation,oxidative stress,ferroptosis,mitochondrial dysfunction,neuroinflammation,and gut dysbiosis.The interactions among these molecular mechanisms complicate the pathogenesis of PD and pose great challenges to drug development.At the same time,the diagnosis and detection of PD are also one of obstacles to the treatment of PD due to its long latency and complex mechanism.Most conventional therapeutic interventions for PD possess limited effects and have serious side effects,heightening the need to develop novel treatments for this disease.In this review,we systematically summarized the pathogenesis,especially the molecular mechanisms of PD,the classical research models,clinical diagnostic criteria,and the reported drug therapy strategies,as well as the newly reported drug candidates in clinical trials.We also shed light on the components derived from medicinal plants that are newly identified for their effects in PD treatment,with the expectation to provide the summary and outlook for developing the next generation of drugs and preparations for PD therapy.Xu Dong-Chen Chen Yong Xu Yang ShenTu Chen-Yu Peng Li-Hua 2023Signal Transduction and Targeted Therapy2023,8,3:2
15Determination of risperidone in human plasma by HPLC-MS/MS and its application to a pharmacokinetic study in Chinese volunteers显示文摘This study presents a rapid, specific and sensitive liquid chromatography/tandem mass spectrometry (LC-MS/MS) assay for determination of risperidone (RIS) in human serum using paroxetine as an internal standard (IS). An Alltima-C18 column (2.1 mm×100 mm, 3 μm) and a mobile phase consisting of 0.1% formic acid-acetonitrile (40:60, v/v) were used for separation. The analysis was performed by selected reaction monitoring (SRM) method, and the peak area of the m/z 411.3→191.1 transition for RIS was measured versus that of the m/z 330.1→192.1 transition for IS to generate the standard curves. The assay linearity of RIS was confirmed over the range 0.25~50.00 ng/ml and the limit of quantitation was 0.05 ng/ml. The linear range corresponds well with the serum concentrations of the analytes obtained in clinical pharmacokinetic studies. Intraday and interday relative standard deviations were 1.85%~9.09% and 1.56%~4.38%, respectively. The recovery of RIS from serum was in the range of 70.20%~84.50%. The method was successfully applied to investigate the bioequivalence between two kinds of tablets (test versus reference products) in 18 healthy male Chinese volunteers. 16Effectiveness of stocking Sparus macrocephalus fry in situ in the East China Sea显示文摘The sea area surrounding Baishishan Island has abundant fishery resources because of its plentiful food and rocky reefs that provide refuge from predators.However,overfishing has occurred in many areas and some species’local populations need to be replenished through stocking programs.This case study,using Sparus microcephalus as the test species,designed and analyzed detailed stocking strategies to improve the survival ratio and control the active range of the released fishes.Controlled variables included release location,release time,and tidal level.Two stocking strategies were designed,executed,and evaluated for comparison.A traditional stocking method was applied to the high flow area of Baishishan Island in Xiangshan Bay,East China Sea.The released fry were transported by boat and released to the sea using buckets.Released fish were traced using plastic tags.The average distance of the 19 recaptured fry from the release point was 7.6 km with 52.9%mortality rate.The distance at recapture and mortality rate among recaptured individuals showed that the traditional releasing method was not successful.An in situ stocking method was used in a low tide area and natural process was imitated to decrease the stress response and promote physiological adaptation.The purpose of this method was to prolong the release process applying temporary net cages at the release location,which resulted in a regional distribution of the released fish and possibly improving the food availability.A new Sr+tagging technique was adopted to evaluate the release effect and quantify comparative release data.Only 1 fish among the 354 caught was found farther than 1 km from the release point in the following 6 months.The results validated the promotion of the new stocking technique.Qingsong Hu Ning Bao Hafiz Abdur Rahman Yazhou Jiang Shouyu Zhang Jun Li Jikang Shentu 2019Aquaculture and Fisheries2019,4,2:2
17Fuzzy Optimization of an Elevator Mechanism Applying the Genetic Algorithm and Neural Networks显示文摘Considering the indefinite character of the value of design parameters and being satisfied with load-bearing capacity and stiffness, the fuzzy optimization mathematical model is set up to minimize the volume of tooth corona of a worm gear in an elevator mechanism. The method of second-class comprehensive evaluation was used based on the optimal level cut set, thus the optimal level value of every fuzzy constraint can be attained; the fuzzy optimization is transformed into the usual optimization. The Fast Back Propagation of the neural networks algorithm are adopted to train feed-forward networks so as to fit a relative coefficient. 19An ε-domination based two-archive 2 algorithm for many-objective optimization显示文摘The two-archive 2 algorithm(Two_Arch2) is a manyobjective evolutionary algorithm for balancing the convergence,diversity,and complexity using diversity archive(DA) and convergence archive(CA).However,the individuals in DA are selected based on the traditional Pareto dominance which decreases the selection pressure in the high-dimensional problems.The traditional algorithm even cannot converge due to the weak selection pressure.Meanwhile,Two_Arch2 adopts DA as the output of the algorithm which is hard to maintain diversity and coverage of the final solutions synchronously and increase the complexity of the algorithm.To increase the evolutionary pressure of the algorithm and improve distribution and convergence of the final solutions,an ε-domination based Two_Arch2 algorithm(ε-Two_Arch2) for many-objective problems(MaOPs) is proposed in this paper.In ε-Two_Arch2,to decrease the computational complexity and speed up the convergence,a novel evolutionary framework with a fast update strategy is proposed;to increase the selection pressure,ε-domination is assigned to update the individuals in DA;to guarantee the uniform distribution of the solution,a boundary protection strategy based on I_(ε+) indicator is designated as two steps selection strategies to update individuals in CA.To evaluate the performance of the proposed algorithm,a series of benchmark functions with different numbers of objectives is solved.The results demonstrate that the proposed method is competitive with the state-of-the-art multi-objective evolutionary algorithms and the efficiency of the algorithm is significantly improved compared with Two_Arch2.WU Tianwei AN Siguang HAN Jianqiang SHENTU Nanying 2022Journal of Systems Engineering and Electronics2022,33,1:1
