2Anti-SARS-CoV-2 activities in vitro of Shuanghuanglian preparations and bioactive ingredients显示文摘Human infection with severe acute respiratory syndrome coronavirus 2(SARS-CoV-2)causes coronavirus disease 2019(COVID-19)and there is no cure currently.The 3CL protease(3CLpro)is a highly conserved protease which is indispensable for CoVs replication,and is a promising target for development of broad-spectrum antiviral drugs.In this study we investigated the anti-SARS-CoV-2 potential of Shuanghuanglian preparation,a Chinese traditional patent medicine with a long history for treating respiratory tract infection in China.We showed that either the oral liquid of Shuanghuanglian,the lyophilized powder of Shuanghuanglian for injection or their bioactive components dose-dependently inhibited SARS-CoV-23CLpro as well as the replication of SARS-CoV-2 in Vero E6 cells.Baicalin and baicalein,two ingredients of Shuanghuanglian,were characterized as the first noncovalent,nonpeptidomimetic inhibitors of SARS-CoV-23CLpro and exhibited potent antiviral activities in a cell-based system.Remarkably,the binding mode of baicalein with SARS-CoV-23CLpro determined by X-ray protein crystallography was distinctly different from those of known 3CLpro inhibitors.Baicalein was productively ensconced in the core of the substrate-binding pocket by interacting with two catalytic residues,the crucial S1/S2 subsites and the oxyanion loop,acting as a“shield”in front of the catalytic dyad to effectively prevent substrate access to the catalytic dyad within the active site.Overall,this study provides an example for exploring the in vitro potency of Chinese traditional patent medicines and effectively identifying bioactive ingredients toward a specific target,and gains evidence supporting the in vivo studies of Shuanghuanglian oral liquid as well as two natural products for COVID-19 treatment.Hai-xia Su Sheng Yao Wen-feng Zhao Min-jun Li Jia Liu Wei-juan Shang Hang Xie Chang-qiang Ke Hang-chen Hu Mei-na Gao Kun-qian Yu Hong Liu Jing-shan Shen Wei Tang Lei-ke Zhang Geng-fu Xiao Li Ni Dao-wen Wang Jian-ping Zuo Hua-liang Jiang Fang Bai Yan Wu Yang Ye Ye-chun Xu 2020Acta Pharmacologica Sinica2020,41,9:32

5The rice OsV4 encoding a novel pentatricopeptide repeat protein is required for chloroplast development during the early leaf stage under cold stress显示文摘Pentatricopeptide repeat(PPR)proteins,characterized by tandem arrays of a 35 amino acid motif,have been suggested to play central and broad roles in modulating the expression of organelle genes in plants.However,the molecular mechanisms of most rice PPR genes remains unclear.In this paper,we isolated and characterized a temperature-conditional virescent mutant,OsV4,in rice(Oryza sativa cultivar Jiahua1(WT,japonica rice variety)).The mutant displays albino phenotype and abnormal chloroplasts at the three leaf stage,which gradually turns green after the four leaf stage at a low temperature(20°C).But the mutant always develops green leaves and well-developed chloroplasts at a high temperature(32°C).Genetic and molecular analyses uncovered that OsV4 encodes a novel chloroplast-targeted PPR protein including four PPR motifs.Further investigations show that the mutant phenotype is associated with changes in chlorophyll content and chloroplast development.The OsV4transcripts only accumulate to high levels in young leaves,indicating that its expression is tissue-specific.In addition,transcript levels of some ribosomal components and plastid-encoded polymerase-dependent genes are dramatically reduced in the albino mutants grown at 20°C.These findings suggest that OsV4 plays an important role during early chloroplast development under cold stress in rice.Xiaodi Gong Qianqian Su Dongzhi Lin Quan Jiang Jianlong Xu Jianhui Zhang Sheng Teng Yanjun Dong 2014Journal of Integrative Plant Biology2014,56,4:19
6Formation mechanisms of megadunes and lakes in the Badain Jaran Desert,Inner Mongolia显示文摘Field observations demonstrate that calc-sinters occurred in the lakes of Badain Jaran Desert. 87Sr/86Sr ratios,14C,δ 13C and mineral com-positions of calc-sinters,and 3He/4He,4He/20Ne,δ 18O,δ D,pH and TDS of water from springs and lakes are analyzed in detail. The results indicate that the lake water is supplied through deep fault zone. The “ker-nel” of stabilized dunes in the Badain Jaran Desert perhaps consists of calc-sinters and calcareous ce-mentation layers. Deep-seated groundwater effuses from this “kernel” and recharges to lakes in desert. Precipitation and snowmelt water from the Qing-hai-Tibet Plateau is fed into the Badain Jaran Desert,Gurinai,Wentougaole and Ejinqi areas through the Xigaze-Langshan Fault zone. The isotopic composi-tions of groundwater in the Alax Plateau are abnor-mal due to the strong evaporation of the Gyaring and Ngoring lake water in the headstream of the Yellow River. Groundwater dissolves dissoluble fractions of rocks during its transportation through the fault zone and flows out of the mouth of spring in the Badan Jaran Desert. The dissoluble fractions are finally de-veloped into calc-sinters and calcareous cementation layers around the spring. Calci-sinters are gradually largened and eventually emerge on the surface of lake water. Eolian sands accumulate on the surfaces of calc-sinters and calcareous cementation layers,and eventually develop into dunes. Invasion of magma causes an increase in the temperature of groundwater within the faults. Groundwater evapora-tion provides water vapor for the formation of humid stabilized dunes during its upwelling. Rhizoconcre-tions found in Yihejigede indicate that the dune was formed and remained immovable 4700 years ago. 7A perspective on carbon materials for future energy application显示文摘Nanocarbon materials play a critical role in the development of new or improved technologies and devices for sustainable production and use of renewable energy. This perspective paper defines some of the trends and outlooks in this exciting area, with the effort of evidencing some of the possibilities offered from the growing level of knowledge, as testified from the exponentially rising number of publications, and putting bases for a more rational design of these nanomaterials. The basic members of the new carbon family are fullerene, graphene, and carbon nanotube. Derived from them are carbon quantum dots, nanohorn, nanofiber, nano ribbon, nanocapsulate, nanocage and other nanomorphologies. Second generation nanocarbons are those which have been modified by surface functionalization or doping with heteroatoms to create specific tailored properties. The third generation of nanocarbons is the nanoarchitectured supramolecular hybrids or composites of the first and second generation nanocarbons, or with organic or inorganic species. 9Research advances in the detection of miRNA显示文摘MicroRNAs (miRNAs) are a family of endogenous, small (approximately 22 nucleotides in length), noncoding, functional RNAs. With the development of molecular biology, the research of miRNA biological function has attracted significant interest, as abnormal miRNA expression is identified to contribute to serious human diseases such as cancers. Traditional methods for miRNA detection do not meet current demands. In particular, nanomaterial-based methods, nucleic acid amplification-based methods such as rolling circle amplification (RCA), loop-mediated isothermal amplification (LAMP), strand-displacement amplification (SDA) and some enzyme-free amplifications have been employed widely for the highly sensitive detection of miRNA. MiRNA functional research and clinical diagnostics have been accelerated by these new techniques. Herein, we summarize and discuss the recent progress in the development of miRNA detection methods and new applications. 12Optimization of the Assembly Efficiency for Lidamycin Chromophore Bound to its Apoprotein:A Case Study using Orthogonal Array显示文摘Objective Lidamycin (LDM) can be dissociated to an apoprotein (LDP) and an active enediyne chromophore (AE).The detached AE can reassemble with its LDP‐containing fusion protein to endow the latter with potent antitumor activity.However,the reassembly of AE with LDP is affected by several factors.Our aim was to optimize the assembly efficiency of the AE with a LDP‐containing fusion protein and investigate the influence of several factors on the assembly efficacy.Methods A method based on RP‐HPLC was developed to analyze the assembly rate,and an orthogonal experimental design L9 (3 4 ) was used to investigate the effects of temperature,assembly time,pH and molecular ratio of LDP‐containing fusion protein to AE on the assembly rate.Furthermore,the determined optimum conditions for the assembly rate of the LDP‐containing fusion protein with AE were applied and evaluated.Results A calibration curve based on the LDM micromolar concentration against the peak‐area of AE by HPLC was obtained.The order in which individual factors in the orthogonal experiment affected the assembly rate were temperature>time>pH>molar ratio of AE to protein and all were statistically significant (P<0.01).The optimal assembly conditions were temperature at 10°C,time of 12 h,pH 7.0,and the molar ratio of AE: protein of 5:1.The assembly rate of AE with a LDP‐containing fusion protein was improved by 23% after condition optimization.Conclusion The assembly rate of chromophore of lidamycin with its LDP‐containing fusion protein was improved after condition optimization by orthogonal design,and the optimal conditions described herein should prove useful for the development of this type of LDP‐containing fusion protein.ZHONG Gen Shen GUO Xiao Fang ZHANG Sheng Hua ZHEN Yong Su 2011Biomedical and Environmental Sciences2011,24,6:9

16MicroRNA in vivo precipitation identifies miR-151-3p as a computational unpredictable miRNA to target Stat3 and inhibits innate IL-6 production显示文摘MicroRNAs(miRNAs)function as important regulators in the immune response and inflammation.Several approaches have been reported to computationally predict miRNAs and their potential targets.However,there are still many miRNA–target interactions that are unpredictable by using the current computational algorithms.We established a miRNA in vivo precipitation method(miRIP)to identify unpredictable miRNAs with definite targets in these cells.Because Stat3 is a well-known transcription factor involved in innate immunity and inflammation,we utilized the miRIP method to identify miRNAs that bind Stat3 mRNA in macrophages.Among the captured miRNAs,miR-151-3p was confirmed to interact with Stat3 mRNA 3′-UTR and downregulate the Stat3 protein levels.LPS stimulation decreased miR-151-3p expression,thereby increasing IL-6 production.Therefore,we found that miR-151-3p inhibited LPS-induced IL-6 production by targeting Stat3.These data further confirmed miRIP as an efficient method to identify unpredictable miRNAs and explore miRNAs-mediated regulation in innate immunity and inflammation.Xiang Liu Xiaoping Su Sheng Xu Huamin Wang Dan Han Jiangxue Li Mingyan Huang Xuetao Cao 2018Cellular & Molecular Immunology2018,15,2:8

18BRIF and CARIF progress显示文摘China Institute of Atomic Energy (CIAE) is currently constructing Beijing rare ion beam facility (BRIF) and is proposing China advanced rare ion beam facility (CARIF). This paper is aiming at introducing the progress of BRIF project and the con ceptual design CARIF. The ISOL type facility BRIF under construction is composed of a 100 MeV 300 ?A proton cyclotron, an ISOL with mass resolution of 20000, and a super-conducting LINAC of 2 MeV/q, and will be commissioned in 2013. CARIF facility proposed is planned to use both ISOL and PF techniques. It is based on a China advanced research reactor CARR that was critical, with ISOL separation of fission fragment, post acceleration to 150 MeV/u, and fragmentation of neu tron-rich fission fragment beam like 132Sn. 20Determination of the ^(12)C(p, γ)^(13)N reaction rates fromthe ^(12)C(~7Li,~6He)^(13)N reaction显示文摘The angular distribution of the 12C(7Li,6He)13N reaction at E(7Li) = 44.0 MeV was measured at the HI-13 tandem accelerator of Beijing, China. The asymptotic normalization coefficient (ANC) of 13N → 12C + p was derived to be (1.64 ± 0.11) fm-1/2 through the distorted wave Born approximation (DWBA) analysis.
