| 1 | Paraoxonase gene cluster variations associated with coronary heart disease in Chinese Han women显示文摘Background The oxidative modification of low-density lipoprotein in the artery wall is currently believed to be central to the pathogenesis of atherosclerosis. Paraoxonase (PON1), an enzyme located on high-density lipoprotein (HDL), can prevent low-density lipoprotein (LDL) from oxidation at a certain extent. Recent studies show two other members of paraoxonase gene family, PON2 and PON3, possess antioxidant properties similar to PON1. The aim of the present study was to explore the role of PON gene cluster on coronary heart disease (CHD) in Chinese Han women.Methods Seven polymorphisms including PON1 -107C>T, -162G>A, -831G>A, R160G, Q192R, PON2 S311C, and PON3 -133C>A were genotyped in 184 female patients with CHD and 239 female controls. The plasma PON1 activity toward phenylacetate was determined in 50 cases and 50 controls randomly selected. Results The plasma PON1 activities were significantly lower in cases than in controls. Individual SNP analysis showed that cases had significantly higher frequencies of PON1 -107T, -831G and PON2 311S alleles than controls. The genotype distributions of -107C>T were also significantly different between two groups. The odds ratios for the development of CHD were 1.66 for -107TC carriers and 2.0 for -107TT carriers, compared with (-107CC) carriers. Haplotype analyses showed that the distributions of haplotypes comprised of PON1 -107C>T and PON2 S311C were significantly different between cases and controls, with cases having higher frequency of T-S haplotype (44.8% vs. 36.3%, P=0.013). The T-S haplotype remained significantly associated with CHD after adjusting environmental risk factors (P=0.0069).Conclusions This association study suggested that lower plasma PON1 activity increased the risk of CHD in Chinese woman, which may be mediated by the higher frequency of -107T allele in cases. | 3 | FABRICATION OF Ti/SiC MMCs BY VACUUM HOT PRESSING WITH AID OF HYDROGEN显示文摘In order to alleviate the severe interface reaction, which is likely to occur during the fabrication of continuous SiC fiber reinforced titanium matrix composite, hydrogen was temporarily introduced in the fabrication to lower the consolidation temperature of Ti/SiC composites in the present work. Effects of different fabrication conditions, with or without hydrogen, on the bonding state and the matrix microstructure were investigated. In-situ hydrogenation method was found to larpely improve the interface bonding of the composite, and full consolidation could be achieved at 750 or even 700° C more than 100°C lower than the normal fabrication temperature of Ti/SiC composite, which consequently reduced the interfacial reaction layer between Ti matrix and SiC fiber. Different hydrogenation condition led to different matrix microstructure. In order to avoid the embrittling effect of residual hydrogen on the composite,Ti/SiC composite needs to be held in vacuum at the fabrication temperature for longer time after consolidation. | C.H. | 12 | Quantitative Evaluation of Aldo-keto Reductase Expression in Hepatocellular Carcinoma (HCC) Cell Lines显示文摘The involvement of aldo-keto reductases(AKRs) in tumorigenesis is widely reported,but their roles in the pathological process are not generally recognized due to inconsistent measurements of their expression.To overcome this problem,we simultaneously employed real-time PCR to examine gene expression and multiple reaction monitoring(MRM) of mass spectrometry(MS) to examine the protein expression of AKRs in fve different hepatic cell lines.These include one relatively normal hepatic cell line,L-02,and four hepatocellular carcinoma(HCC) cell lines,HepG2,HuH7,BEL7402 and SMMC7721.The results of real-time PCR showed that expression of genes encoding the AKR1C family members rather than AKR1A and AKR1B was associated with tumor,and most of genes encoding AKRs were highly expressed in HuH7.Similar observations were obtained through MRM.Different from HuH7,the protein abundance of AKR1A and AKR1B was relatively consistent among the other four hepatic cell lines,while protein expression of AKR1C varied signifcantly compared to L-02.Therefore,we conclude that the abundant distribution of AKR1C proteins is likely to be associated with liver tumorigenesis,and the AKR expression status in HuH7 is completely different from other liver cancer cell lines.This study,for the frst time,provided both overall and quantitative information regarding the expression of AKRs at both mRNA and protein levels in hepatic cell lines.Our observations put the previous use of AKRs as a biomarker into question since it is only consistent with our data from HuH7.Furthermore,the data presented herein demonstrated that quantitative evaluation and comparisons within a protein family at both mRNA and protein levels were feasible using current techniques. | Lei Yang Ju Zhang Shenyan Zhang Weiwei Dong Xiaomin Lou Siqi Liu | 2013 | Genomics, Proteomics & Bioinformatics2013,11,4: | 0 |
| 13 | Diagnostic Value of CYFRA21-1, CA19-9 and SCCAg Detection in Esophageal Cancer显示文摘Objectives To investigate the value of serum cytokeratin 19 fragment(CYFRA21-1),carbohydrate anti-gen 19-9(CA19-9)and squamous cell carcinoma antigen(SCCAg)in the diagnosis of esophageal cancer.Methods 100 cases of esophageal cancer(study group)and 50 healthy subjects(control group)were recruited in the study,the serum levels of CYFRA21-1,CA19-9 and SCCAg in these patients were detected.Subgroups analysis was performed in study group by clinical staging,and the diagnostic value of CYFRA21-1,CA19-9 and SCCAg in detecting esophageal cancer was analyzed by receiver operating characteristic curve(ROC).Results The serum levels of CYFRA21-1,CA19-9 and SCCAg in study group were significantly higher than those in control group,and those were significantly lower in patients with stageⅠ-Ⅱdisease than those with stageⅢandⅣdisease,indicating statistically significant difference(P<0.01);the sensitivity of serum CYFRA21-1,CA19-9 and SCCAg in the diagnosis of esophageal cancer was 82.26%,and specificity was 91.33%,with positive predictive value being 86.14%,and negative predictive value being 89.57%,and AUC value was 0.864,the combined detection was superior to each measure utilized alone,the difference was significantly significant(P<0.05).Conclusion The combined detection of serum CYFRA21-1,CA19-9,and SCCAg can significantly improve the diagnostic sensitivity and specificity in detecting the esophageal cancer. | Du Chunran Wang Yanming Hou Shenyan | 2018 | Proceedings of Anticancer Research2018,2,2: | 0 |

Both types of optimization problems involve multiple finite element models at each iteration step, which draws high demands in optimization methods. Considering the common characteristic for these two types of problems, which is that the design domain keeps the same no matter what the structure cases or payload cases are,both problems can be formulated into the unified expressions. A two-level multipoint approximation(TMA) method is firstly improved with the use of analytical sensitivity analysis for structural mass, and then this improved method is utilized to tackle these two types of problems.
