1Time-domain Spectral Finite Element Method for Wave Propagation Analysis in Structures with Breathing Cracks显示文摘Guided waves are generally considered as a powerful approach for crack detection in structures,which are commonly investigated using the finite element method(FEM).However,the traditional FEM has many disadvantages in solving wave propagation due to the strict requirement of mesh density.To tackle this issue,this paper proposes an efficient time-domain spectral finite element method(SFEM)to analyze wave propagation in cracked structures,in which the breathing crack is modeled by definiiig the spectral gap element.Moreover,novel orthogonal polynomials and Gauss-Lobatto-Legendre quadrature rules are adopted to construct the spectral element.Meanwhile,a separable hard contact is utilized to simulate the breathing behavior.Finally,a comparison of the numerical results between the FEM and the SFEM is conducted to demonstrate the high efficiency and accuracy of the proposed method.Based on the developed SFEM,the nonlinear features of waves and influence of the incident mode are also studied in detail,which provides a helpful guide for a physical understanding of the wave propagation behavior in structures with breathing cracks.Zexing Yu Chao Xu Fei Du Shancheng Cao Liaiigxian Gu 2020Acta Mechanica Solida Sinica2020,33,6:2
2Glycolytic potential enhanced by blockade of pyruvate influx into mitochondria sensitizes prostate cancer to detection and radiotherapy显示文摘Objective:This study aimed to evaluate the effects of mitochondrial pyruvate carrier(MPC)blockade on the sensitivity of detection and radiotherapy of prostate cancer(PCa).Methods:We investigated glycolysis reprogramming and MPC changes in patients with PCa by using metabolic profiling,RNASeq,and tissue microarrays.Transient blockade of pyruvate influx into mitochondria was observed in cellular studies to detect its different effects on prostate carcinoma cells and benign prostate cells.Xenograft mouse models were injected with an MPC inhibitor to evaluate the sensitivity of 18F-fluorodeoxyglucose positron emission tomography with computed tomography and radiotherapy of PCa.Furthermore,the molecular mechanism of this different effect of transient blockage towards benign prostate cells and prostate cancer cells was studied in vitro.Results:MPC was elevated in PCa tissue compared with benign prostate tissue,but decreased during cancer progression.The transient blockade increased PCa cell proliferation while decreasing benign prostate cell proliferation,thus increasing the sensitivity of PCa cells to 18F-PET/CT(SUVavg,P=0.016;SUVmax,P=0.03)and radiotherapy(P<0.01).This differential effect of MPC on PCa and benign prostate cells was dependent on regulation by a VDAC1-MPC-mitochondrial homeostasis-glycolysis pathway.Conclusions:Blockade of pyruvate influx into mitochondria increased glycolysis levels in PCa but not in non-carcinoma prostate tissue.This transient blockage sensitized PCa to both detection and radiotherapy,thus indicating that glycolytic potential is a novel mechanism underlying PCa progression.The change in the mitochondrial pyruvate influx caused by transient MPC blockade provides a critical target for PCa diagnosis and treatment.Huan Xu Junyi Chen Zhi Cao Xi Chen Caihong Huang Jin Ji Yalong Xu Junfeng Jiang Yue Wang Guowang Xu Lina Zhou Jingyi He Xuedong Wei Jason Boyang Wu Zhong Wang Shancheng Ren Fubo Wang 2022Cancer Biology & Medicine2022,19,9:1

4Full-field out-of-plane vibration displacement acquisition based on speckle-projection digital image correlation and its application in damage localization显示文摘Stereo-digital image correlation(Stereo-DIC)has been widely explored for modal analysis in plate-type structures due to its noncontact and full-field advantages.However,when the traditional stereo-DIC is adopted to capture the out-of-plane displacements,several challenging issues exist such as the development of surface speckles,asynchronous camera recording,and efficiency and accuracy degradation due to high computation costs.Moreover,with the captured out-of-plane displacements,effective and efficient evaluation of the high spatial resolution mode shapes and their application to damage localization are also critical problems.To tackle these issues,a speckle-projection DIC technique using a single high‐speed camera is proposed to obtain the out-of-plane vibration displacements.Moreover,an enhanced peak-picking modal analysis method is adopted to enhance the estimation accuracy and efficiency of mode shapes.In addition,the low‐rank property of mode shapes in an intact state and the spatial sparse property of damage locations are harnessed for the detection of damage positions without requiring reference data on the healthy state.Finally,the modal analysis and damage localization results based on the proposed speckle-projection DIC are compared with those of the traditional two-camera stereo-DIC technique to verify its feasibility and effectiveness.It is found that the differences in the identified resonant frequencies between these two methods are smaller than 1%for higher modes.Moreover,the proposed speckle-projection DIC has the same accuracy as the traditional two-camera stereo-DIC in terms of measurement accuracy,mode shape estimation,and damage localization.Shancheng Cao Jinwei Yan Haibo Nian Chao Xu 2022International Journal of Mechanical System Dynamics2022,2,4:0
