| 1 | N-acetylcysteine attenuates alcohol-induced oxidative stess in rats显示文摘AIM: To investigate free-radical scavenger effect of nacetylcysteine in rats intragastrically fed with ethanol.METHODS: Twenty-four rats divided into three groups were fed with ethanol (6 g/kg/day, Group 1), ethanol and nacetylcysteine (1 g/kg, Group 2), or isocaloric dextrose (control group, Group 3) for 4 weeks. Then animals were sacrificed under ether anesthesia, and intracardiac blood and liver tissues were obtained. Measurements were made in both serum and homogenized liver tissues.Malondialdehyde (MDA) level was measured by TBARS method. Glutathione peroxidase (GSH-Px) and superoxide dismutase (SOD) levels were studied by commercial kits.Kruskal-Wallis test was used for statistical analysis.RESULTS: ALT and AST in Group 1 (154 U/L and 302 U/L,respectively) were higher than those in Group 2 (94 U/L and 155 U/L) and Group 3 (99 U/L and 168 U/L) (P=0.001for both). Serum and tissue levels of MDA in Group 1 (1.84nmol/mL and 96 nmol/100 mg-protein) were higher than that in Group 2 (0.91 nmol/mL and 64 nmol/100 mg protein)and Group 3 (0.94 nmol/mL and 49 nmol/100 mg-protein)(P<0.001 for both). On the other hand, serum GSH-Px level in Group 1 (8.21 U/g Hb) was lower than that in Group 2(16 U/g Hb) and Group 3 (16 U/g-Hb) (P<0.001). | 2 | Role of immunotherapy in the treatment of allergic asthma显示文摘Allergen-specific immunotherapy(SIT) induces clinical and immunological tolerance as defined by persistence of clinical benefit and associated long-term immunological parameters after cessation of treatment. Although the efficacy of SIT has been shown in terms of reducing symptoms, medication consumption and ameliorating quality of life in both allergic rhinitis and asthma, there has long been some controversies about effectiveness of SIT in the treatment of allergic asthma. The type of allergen, the dose and protocol of immunotherapy, patient selection criteria, the severity and control of asthma, all are significant contributors to the power of efficacy in allergic asthma. The initiation of SIT in allergic asthma should be considered in case of coexisting of other allergic diseases such as allergic rhinitis, unacceptable adverse effects of medications, patient's preference to avoid long-term pharmacotherapy. Steroid sparing effect of SIT in allergic asthma is also an important benefit particularly in patients who have to use these drugs in high doses for a long-time. Symptomatic asthma is a risk factor for systemic reactions and asthma should be controlled at the time of administration of SIT. Both subcutaneous immunotherapy(SCIT) and sublingual immunotherapy(SLIT) have been found to be effective in patients with allergic asthma. | 4 | Effect of acupuncture therapy on fracture healing in rats with femur fractures显示文摘OBJECTIVE: To investigate the effect of acupuncture therapy on fracture healing in rats with femur fractures.METHODS: A total of 10 groups were formed;control group, groups sacrificed on 7 th, 14 th, and 21 st days of fracture formation, groups to which acupuncture was applied for 7, 14, and 21 d, groups to which fracture and acupuncture were applied for 7,14, and 21 d. A transverse fracture line was formed in femurs of rats by using a Gigli saw. The Kirschner wire was driven retrograde down from the fracture line to proximal part of the bone and then, the fracture was fixed towards distal part. Acupuncture was applied to the rats for 7, 14, and 21 d as 4 sessions per week after formation of the fracture.RESULTS: Malondialdehyde(MDA), reduced glutathione(GSH) levels, catalase(CAT), glutathione-Stransferase(GST), superoxide dismutase(SOD), and glucose-6-phosphate dehydrogenase(G6 PD) activities were measured. Despite the increased MDA levels, G6 PD and SOD activities reduced during the fracture healing. There was a statistically significant increase in MDA, GSH levels, and G6 PD activity in fracture groups compared to control group, but CAT, GST, and SOD activities decreased. The use of acupuncture enhanced callus development and bone mineralization during bone healing.CONCLUSION: The acupuncture therapy can affect suppression of the effects of free oxygen radicals and regulation of the antioxidant enzyme activity in fracture healing. | 5 | Protective effect of Nigella sativa oil against binge ethanolinduced oxidative stress and liver injury in rats显示文摘AIM: Nigella sativa L.(Ranunculaceae) is considered as a therapeutic plant-based medicine for liver damage. In this study, the aim was to study the effect of Nigella sativa oil(NSO) pretreatment on ethanol-induced hepatotoxicity in rats. METHOD: Rats were given Nigella sativa oil at doses of 2.5 and 5.0 mL·kg-1, orally for 3 weeks, followed by oral ethanol(EtOH) administration(5 g·kg-1) every 12 h three times(binge model). RESULTS: Binge ethanol application caused significant increases in plasma transaminase activities and hepatic triglyceride and malondialdehyde(MDA) levels. It decreased hepatic glutathione(GSH) levels, but did not change vitamins E and vitamin C levels and antioxidant enzyme activities. NSO(5.0 mL·kg-1) pretreatment significantly decreased plasma transaminase activities, hepatic MDA, and triglyceride levels together with amelioration in hepatic histopathological findings. | 11 | Oxidative stress in portal hypertension-induced rats with particular emphasis on nitric oxide and trace metals显示文摘AIM: To investigate the oxidative-stress-related changes in rats with portal hypertension with particular emphasis on nitric oxide (NO) and trace metals. METHODS: Cirrhosis was induced by partial portal vein ligation (PVL) in Wistar rats. The lipid peroxidation marker (malondialdehyde, MDA), antioxidant defense enzymes including superoxide dismutase (SOD), catalase (CAT), glutathione (GSH), and agents known to have antioxidant features including nitric oxide (NO), zinc (Zn), copper (Cu) were determined both in serum and in liver tissue at 4 wk after surgery in PVL and sham-operated rats. Portal pressure of all experimental animals was measured. MDA was detected by thiobarbituric acid reactivity assay.SOD activity was determined by inhibition of nitroblue tetrazolium reduction with xanthine/xanthine oxidase used asa superoxide generator. CAT activity was determined by the breakdown of hydrogen peroxide. GSH concentrations were measured by using metaphosphoric acid for protein precipitation and 5'-5'-dithio-bis-2-nitrobenzoic acid for color development. NO was detected by the Griess method after reduction of nitrate to nitrite with nitrate reductase, and the concentrations of Zn and Cu were measured by a Shimadzu 680 AA atomic absorption spectrometer. Histopathological confirmation was done under light microscope. Statistical analyses were done by Student's t-test, and significance of the difference was tested by the unpaired Mann-Whitney test. P<0.05 was considered statistically significant.RESULTS: Histopathological studies confirmed PVL induced cirrhotic changes. There was a statistically significant difference in portal pressure between PVL and control groups (P<0.001). The results showed significant increases in the levels of MDA and NO in both tissue and serum (P<0.05 and P<0.001, respectively in tissue; P<0.001 for each in serum), and Zn only in tissue (P<0.001)in rats with PVL compared with sham-operated rats. Besides, PVL rats exhibited reduced plasma and tissue GSH, CAT, SOD (P<0.001 for each). | 15 | Potential effects of L-NAME on alcohol-induced oxidative stress显示文摘AIM: Nitric oxide (NO) is a highly reactive oxidant synthesized from L-arginine by nitric oxide synthase (NOS). NO may cause injury through the generation of potent radicals. Nwnitro-L-arginine methyl ester (L-NAME) is a non-selective inhibitor of NOS. We aimed to evaluate whether L-NAME treatment had protective effects against oxidative stress in rats intragastrically fed with ethanol during a 4 wk-period.METHODS: Thirty-six male Wistar rats were divided into 3 equal groups: group 1 (control group-isocaloric dextrose was given), group 2 (6 g/kg.d ethanol-induced group) and group 3 (both ethanol 6 g/kg.d and L-NAME 500 mg/L in drinking water-given group). Animals were sacrificed at the end of 4 wk-experimental period, and intracardiac blood and liver tissues were obtained. Biochemical measurements were performed both in plasma and in homogenized liver tissues. Alanine amino transferase (ALT), aspartate amino transferase (AST), malondialdehyde (MDA), NO, superoxide dismutase (SOD), catalase (CAT) and glutathione (GSH) levels were measured by spectrophotometry.RESULTS: ALT and AST in group 2 (62 U/L and 128 U/L,respectively) were higher than those in group 1 (24 U/L and 38 U/L) and group 3 (37 U/L and 81 U/L) (P<0.001 for both).Plasma and tissue levels of MDA in group 2 (4.66 μmol/L and 0.55 nmol/mg protein) were higher than in group 1 (2.65 μmol/L and 0.34 nmol/mg protein) and group 3 (3.43 μmol/L and 0.36 nmol/mg protein) (P<0.001 for both). Plasma and liver tissue levels of NO in group 2 (54.67 μmol/L and 586.50 nmol/mg protein) were higher than in group 1 (34.67 μmol/L and 435.33 nmol/mg protein)and group 3 (27.50 μmol/L and 412.75 nmol/mg protein )(P<0.001 for both). Plasma and liver tissue SOD activities in group 2 (15.25 U/mL and 5.38 U/ mg protein,respectively) were lower than in group 1 (20.00 U/mL and 8.13 U/mg protein) and group 3 (19.00 U/mL and 6.93 U/mg protein) (P<0.001 for both). Plasma and liver tissue CAT activities in group 2 (145 U/mL and 37 U/ mg protein,respectively) were lower than in group 1 (176 U/mL and 73 U/mg protein) and group 3 (167 U/mL and 61 U/mg protein) (P<0.001 for both).
