| 1 | Molecular Cloning and Expression Analysis of a Zinc Finger Protein Gene in Apple显示文摘A cDNA library was created from stem apex tissue from Jonathan apples (Malus domestica Borkh.), harvested in June to August, during which the plant transitions from vegetative growth to reproductive growth. From this library, we isolated an expressed sequence tag (EST) sequence containing a zinc finger motif, using this sequence, a 779 bp cDNA fragment was obtained by using 5' RACE, and a final full-length cDNA encoding an apple zinc finger protein (named MdZF1; GenBank accession number AB116545) was obtained by further PCR. This zinc finger motif of MdZF1 has high homology with INOETERMINATE1 (ID1) gene from maize which seemed to be involved in the transition to flowering. Northern blot and RT-PCR analyses showed that the MdZF1 expressed in the root, stem, leaves, shoot apex and floral organs of the apple, with expression levels higher in root, stem, leaves and floral shoot apex than that in floral organs (sepals, petals, stamens and pistils). | 2 | Crashworthiness simulation and improvement design of auto-body based on finite element mehtod显示文摘One developing commercial vehicle was simulated on crashworthiness using the nonlinear finite element method. The deformation of the auto-body, the movement of the steering wheel and the dynamic responses of the occupant at the initial velocity of 50 km/h were studied. The results show that the design of the vehicle could be improved on structure and material. The frontal longitudinal beam, the main energy-absorbing part of the auto-body, was optimized on structure. The data of the simulation predict that the hinge of the engine hood would fracture during the crash. The failure of the engine hood hinge would be a danger to both the driver and passengers. Then the problem was solved by changing the engine hood and hinge on structure and material. Simulation results also show that applying new material and new manufacture techniques could improve the crashworthiness of the vehicle greatly. | 3 | Certainties and uncertainties of land cover statistics in China显示文摘Vegetation or land cover maps have been made directly or indirectly available for the entire territory of China. The certainties and uncertainties of land cover statistics were analyzed by comparing three data sources: China’s Vegetation Map, IGPB DISCover, and University of Maryland Product. Great similarities in the statistics of 7 aggregated land cover types were found among the three data sources, particularly between the two global land cover maps. The per pixel agreement between any 2 of 3 maps was between 38.0%—51.4%; the per pixel agreement among all three maps was only 27.1%. Certainties were found in regions where vegetation types are typical and human land use practice is relatively homogenous; the uncertainties occurred to either vegetation transition zones or regions where land cover types and land use practice are relatively diversified. Systematic and multidisciplinary efforts are necessary to promote accurate mapping of nationwide land cover types in China. | 6 | Current status of radiation therapy for prostate cancer显示文摘Radiotherapy for the treatment of prostate cancer has been extensively explored in the past. Along with the comprehensive understanding of the biology of prostate cancer and rapid advances in terms of technology, the out- come of treatment for the patients with prostate cancer has improved. The authors review radiotherapy as the primary treatment for the disease, with particular emphasis on the technological advances from both the radiobiological and radiophysics aspects. Nonconventional fractionated irradiation like hyper- or hypo-fractionation has been imple- mented in the clinic, the final results still need to be confirmed in the future. Technological advances like IMRT, IGRT, in the last two decades have significantly improved the delivery of external radiotherapy to the prostate. This has re- sulted in an overall increase in the total dose that can be safely delivered to the prostate, which has led to modest im- provements in the biochemical outcome. However, establishing the standard therapy for prostate cancer remains con- troversial. | 12 | All-stage targeted therapy for the brain metastasis from triple-negative breast cancer显示文摘Brain metastasis is a common and serious complication of breast cancer,which is commonly associated with poor survival and prognosis.In particular,the treatment of brain metastasis from triplenegative breast cancer(BM-TNBC)has to face the distinct therapeutic challenges from tumor heterogeneity,circulating tumor cells(CTCs),blood-brain barrier(BBB)and blood-tumor barrier(BTB),which is in unmet clinical needs.Herein,combining with the advantages of synthetic and natural targeting moieties,we develop a“Y-shaped”peptide pVAP-decorated platelet-hybrid liposome drug delivery system to address the all-stage targeted drug delivery for the whole progression of BM-TNBC.Inherited from the activated platelet,the hybrid liposomes still retain the native affinity toward CTCs.Further,the peptide-mediated targeting to breast cancer cells and transport across BBB/BTB are demonstrated in vitro and in vivo.The resultant delivery platform significantly improves the drug accumulation both in orthotopic breast tumors and brain metastatic lesions,and eventually exhibits an outperformance in the inhibition of BM-TNBC compared with the free drug.Overall,this work provides a promising prospect for the comprehensive treatment of BMTNBC,which could be generalized to other cell types or used in imaging platforms in the future. | Zimiao Luo Sunyi Wu Jianfen Zhou Weixia Xu Qianzhu Xu Linwei Lu Cao Xie Yu Liu Weiyue Lu | 2023 | Acta Pharmaceutica Sinica B2023,13,1: | 0 |
