1Statistical design and evaluation of a propranolol HCl gastric floating tablet显示文摘The purpose of this research was to apply statistical design for the preparation of a gastric floating tablet(GFT)of propranolol HCl and to investigate the effect of formulation variables on drug release and the buoyancy properties of the delivery system.The contents of polyethylene oxide(PEO)WSR coagulant and sodium bicarbonate were used as independent variables in central composite design of the best formulation.Main effects and interaction terms of the formulation variables were evaluated quantitatively using a mathematical model approach showing that both independent variables have significant effects on floating lag time,%drug release at 1 h(D_(1h))and time required to release 90%of the drug(t_(90)).The desired function was used to optimize the response variables,each with a different target,and the observed responses were in good agreement with the experimental values.FTIR and DSC studies of the statistically optimized formulation revealed there was no chemical interaction between drug and polymer.The statistically optimized formulation released drug according to first order kinetics with a non-Fickian diffusion mechanism.Evaluation of the optimized formulation in vivo in human volunteers showed that the GFT was buoyant in gastric fluid and that its gastric residence time was enhanced in the fed but not the fasted state.Meka Venkata Srikanth Nali Sreenivasa Rao Songa Ambedkar Sunil Battu Janaki Ram Venkata Ramana Murthy Kolapalli 2012Acta Pharmaceutica Sinica B2012,2,1:6
2Effects of disinhibition on spatiotemporal pattern of neuronal first recruitment in neuronal networks显示文摘The propagation of neuronal activities is a key feature to understanding information processing in networks. The analysis based on first-spikes of bursts in turn plays an important role in the research of neuronal activity propagation. Our focus here is to investigate how spatiotemporal patterns of neuronal first-spikes are affected by disinhibition. Multi-electrode arrays were used to record stimulation-evoked bursts of multiple neurons in randomly cultured neuronal networks. Both the precise timing of and the rank relationships between first-spikes were analyzed. Compared with evoked bursts in the network's native state, the precise first-spike latencies in its dis-inhibited state are more consistent and the propagation of its bursting activities is much faster. 3Optimization of microstructures and magnetic properties of Sm(Co_(bal)Fe_(0.227)Cu_(0.07)Zr_(0.023))_(7.6) magnets by sintering treatment显示文摘Magnetic properties and microstructures of Sm(Co_(bal)Fe_(0.227)Cu_(0.07)Zr_(0.023_)_(7.6) sintered magnets were optimized by sintering treatment. Results show that the knee-point magnetic field, Hknee, is twofold up and the intrinsic coercivity Hcjincreases by 40%, ranging from 21.64 to 30.39 kOe at the cost of a little decrease of Brfrom 10.84 to 10.31 kGs with sintering temperature decreasing from 1488 to 1473 K. And the average domain width is narrower and more uniform for the specimen sintered at 1473 K than that of the specimen sintered at 1488 K. It is impressive that the density of lamellar phase increases from ~0.050 to ~0.058 nm^(-1) with the sintering temperature decreasing from 1488 to 1473 K. Moreover, the average cellular size is about ~84 nm for the magnets sintered at 1473 K, which is 80% of that of the magnets sintered at 1488 K(~97 nm). And the cell boundary width of the magnets sintered at 1473 K(~7 nm) is only half average width of the magnets sintered at 1488 K(~14 nm). It is found that the Cu content in the cell boundaries is much higher(~17 at%) in the magnets sintered at 1473 K compared to that of the magnets sintered at 1488 K(~10 at%). 4Precolumn derivatization LC-MS/MS method for the determination and pharmacokinetic study of glucosamine in human plasma and urine显示文摘A selective precolumn derivatization liquid chromatography-tandem mass spectrometric (LC-MS/MS) method for the determination of glucosamine in human plasma and urine has been developed and validated. Glucosamine was derivatized by o-phthalaldehyde/3-mercaptopropionic acid. Chromatographic separation was performed on a Phenomenex ODS column (150 mm 4.6 mm, 5 mm) using linear gradient elution by a mobile phase consisting of methanol (A), and an aqueous solution containing 0.2% ammonium acetate and 0.1% formic acid (B) at a flow rate of 1 mL/min. Tolterodine tartrate was used as the internal standard (IS). With protein precipitation by acetonitrile and then the simple one-step derivatization, a sensitive bio-assay was achieved with the lower limit of quantitation (LLOQ) as low as 12 ng/mL for plasma. The standard addition calibration curves suitable for clinical sample analysis showed good linearity over the range of 0.012-8.27 mg/mL in plasma and 1.80-84.1 mg/mL in urine. 5Simultaneous determination of ivabradine and N-desmethylivabradine in human plasma and urine using a LC-MS/MS method: application to a pharmacokinetic study显示文摘A sensitive and specific liquid-chromatography tandem mass spectrometry(LC-MS/MS)assay has been developed and validated for the simultaneous quantification of ivabradine and its active metabolite N-desmethylivabradine in human plasma and urine.The assay employed a single liquid–liquid extraction of the analytes from plasma and urine samples,and diazepam was used as internal standard(IS).The chromatographic separation was achieved on a Diamonsil C18 column(150 mm4.6 mm,5 mm,Dikma)using a mixture of methanol and aqueous 5 mM ammonium acetate buffer containing 0.2%formic acid(80:20,v/v)as mobile phase.The assay for ivabradine and N-desmethylivabradine in plasma showed good linearity(r≥0.99)over the ranges 0.1013–101.3 ng/mL and 0.085–25.5 ng/mL,respectively.The assay for ivabradine and N-desmethylivabradine in urine showed good linearity(r≥0.99)over the ranges 10.13–6078 ng/mL and 8.5–850 ng/mL,respectively.The intra-and inter-day accuracy and precision values were found to be within the assay variability limits(RSD<15%)in accordance with FDA guidelines.The methods were successfully used for evaluating the pharmacokinetic properties of ivabradine and N-desmethylivabradine in human plasma and urine in Chinese healthy volunteers.Chengtao Lu Yanyan Jia Jing Yang Xin Jin Ying Songa Wenxing Liu Yi Ding Xiaoli Sun Aidong Wen 2012Acta Pharmaceutica Sinica B2012,2,2:3
