1Enhanced fat degradation following the addition of a Pseudomonas species to a bioaugmentation product used in grease traps显示文摘The presence of fats, oils and greases(FOGs) in wastewater can lead to many problems including blockages. Investigation of a bioaugmentation product, consisting of Bacillus spp., to degrade butter(1%, W/V) and olive oil(1%, V/V) was performed in aerobic batch cultures for 13-day incubation. Gravimetric analysis of the remaining substrates showed slowly degradation of the oil after a 2-day lag, but no degradation of the butter. Addition of a Pseudomonas putida strain CP1 to the Bacillus spp. population promoted rapidly degradation of both fats after7 days of incubation. High fat accumulation revealed the potential use of the new bacterial mixture for production of added-value compounds. Lipase production only by the Bacillus spp.along with the analysis of the remaining lipids with thin layer chromatography and gas chromatography, suggested that the Bacillus spp. mainly only hydrolyzed the fat. The breakdown products were metabolized by the Pseudomonas putida CP1 performing preferential utilization of unsaturated fatty acids. Investigation of population dynamics using selective plating and a labeled Pseudomonas putida CP1::Tn7-gfp showed domination of the latter. 18Biotechnological valorization of biodiesel-derived glycerol: Trials with the non-conventional yeasts Yarrowia lipolytica and Rhodosporidium sp显示文摘Four non-conventional yeast strains belonging to the genera Yarrowia and Rhodosporidium were cultivated under nitrogen-limited conditions in shake flasks with biodiesel-derived glycerol employed as sole substrate.The strains R.toruloides DSM 4444 and Y.lipolytica ACA-DC 5033 displayed better performances and were further studied.At initial glycerol(Gly0)≈50 g/L,the strain ACA-DC 5033 produced dry cell weight(DCW)≈7.0 g/L,and also secondary extra-cellular metabolites as citric acid(CA)at≈16 g/L and polyols(Pol)(mannitol,erythritol and arabitol)at≈21 g/L.In double-media(besides nitrogen also Mg was limiting)for Gly0≈50 g/L,a shift towards CA production occurred(CAmax≈33 g/L).The strain DSM 4444 at Gly0≈50 g/L produced DCW=18.1 g/L containing lipids=30.3%in DCW.In single nitrogen-limited media with Gly0≈90 g/L,the strain ACA-DC 5033 produced Pol≈48 g/L and CA≈20 g/L,while the strain DSM 4444 produced DCW=27.3 g/L containing lipid=54.5%w/w.At the late growth phases,metabolites were re-consumed.Balanced growth phase(trophophase)and phase of secondary metabolite synthesis(idiophase)were successfully simulated with the aid of a modified Velhlust-Aggelis model.Lipid extraction process was studied for these two strains by using two different extraction methods.Yeast lipids contained mostly oleic acid,constituting suitable precursors for the synthesis of 2nd generation biodiesel.Sofia Sarantou Nikolaos G.Stoforos Ourania Kalantzi Seraphim Papanikolaou 2021Carbon Resources Conversion2021,4,1:0
19Sustainable arabitol production by a newly isolated Debaryomyces prosopidis strain cultivated on biodiesel-derived glycerol显示文摘The purpose of this research was the bioconversion of biodiesel-derived glycerol to arabitol through the adoption of sustainable bioprocessing.Arabitol production by Debaryomyces prosopidis FMCC Y69 was significantly affected by initial glycerol concentrations.The highest arabitol concentration of 45.5 g/L was achieved in the case of 120 g/L initial glycerol combined with the highest yield(0.379 g arabitol/g initial glycerol).The yeast strain showed a non-typical oleaginous behavior with low accumulation of lipids(up to 20.9%w/w)despite nitrogen-limited condition that prevailed in the fermentation media.Endopolysaccharides(IPs)were considerably high with a yield on the produced dry cell weight(YIPs/X)(=40.9%w/w)when 85 g/L of glycerol was utilized as the starting material.The fermentation efficiency was further improved when the optimal carbon to nitrogen(C/N)ratio was applied.More specifically,arabitol production was maximized(reaching 56.5 g/L)at a C/N ratio of 158 moles/moles while yield and productivity were also enhanced with respective values of 0.48 g arabitol/g consumed glycerol and 0.112 g/L.h.The increase in C/N ratios led to a decrease in cell dry weight(DCW)concentrations,which is rationally associated with decreased nitrogen availability in the growth medium.YIPs/X values were remarkably high(23.1–26.1%w/w)at the end of each fermentation with the highest concentration in absolute values of 7.3 g/L achieved at the lowest C/N ratio employed(i.e.,79 moles/moles).This study demonstrated the efficient arabitol production along with other value-added metabolites,such as IPs and cellular lipids,implementing biotechnology and renewable resources and thus contributing to the development of circular economy technologies.Rosanina Filippousi Erminta Tsouko Kerryane Mordini Dimitrios Ladakis Apostolis AKoutinas George Aggelis Seraphim Papanikolaou 2022Carbon Resources Conversion2022,5,1:0
