| 2 | Redundant CAMTA Transcription Factors Negatively Regulate the Biosynthesis of Salicylic Acid and N-Hydroxypipecolic Acid by Modulating the Expression of SARD1 and CBP60g显示文摘Two signal molecules, salicylic acid (SA) and N-hydroxypipecolic acid (NHP), play critical roles in plant immunity. The biosynthetic genes of both compounds are positively regulated by master immune-regulating transcription factors SARD1 and CBP60g. However, the relationship between the SA and NHP pathways is unclear. CALMODULIN-BINDING TRANSCRIPTION FACTOR 1 (CAMTA1), CAMTA2, and CAMTA3 are known redundant negative regulators of plant immunity, but the underlying mechanism also remains largely unknown. In this study, through chromatin immunoprecipitation and electrophoretic mobility shift assays, we uncovered that CBP60g is a direct target of CAMTA3, which also negatively regulates the expression of SARD1, presumably via an indirect effect. The autoimmunity of camta3-1 is suppressed by sard1 cbp60g double mutant as well as ald1 and fmo1, two single mutants defective in NHP biosynthesis. Interestingly, a suppressor screen conducted in the camta1/ 2/ 3 triple mutant background yielded various mutants blocking biosynthesis or signaling of either SA or NHP, leading to nearly complete suppression of the extreme autoimmunity of camta1/ 2/ 3, suggesting that the SA and NHP pathways can mutually amplify each other. | 3 | 脂肪乳对内毒素诱导急性肺损伤小鼠的干预作用显示文摘目的比较以橄榄油为主的脂肪乳Clinoleic20%○R和豆油为主的脂肪乳Lipoven20%○R对脂多糖(LPS)诱导急性肺损伤(ALI)小鼠炎症反应的作用。方法雄性Balb/C小鼠按随机数字表法分为生理盐水(NaCl)、Clino和Lipo3组。用微量泵分别向3组小鼠泵入质量分数为0.9%的NaCl、20%Clinoleic和20%Lipoven,并在进行取血和肺泡灌洗前8h和24h向气管内喷洒LPS制备ALI模型。观察3组小鼠LPS诱导8h和24h时的存活率、肺组织湿/干重比、支气管肺泡灌洗液(BALF)中白细胞计数、肿瘤坏死因子-α(TNF-α)、巨噬细胞炎性蛋白-2(MIP-2)、肺组织髓过氧化物酶(MPO)活性及血清花生四烯酸(AA)、油酸、亚油酸含量。结果LPS诱导后可引起BALF中白细胞计数、TNF-α、MIP-2,肺组织MPO活性、W/D比例及血清AA水平显著升高。Lipoven显著降低ALI小鼠24h后的存活率(P均<0.01),而Clino组与NaCl组之间差异无显著性;肺组织MPO活性在24h后两个脂肪乳组明显高于NaCl组(P均<0.01),而两个脂肪乳之间差异无显著性。注射ALI8h后,Lipo组BALF中MIP-2和血清AA均较NaCl组和Clino组升高,24h后BALF中白细胞、TNF-α、AA水平也均显著高于NaCl组和Clino组(P均<0.01);而肺组织水肿程度和油酸、亚油酸含量在3组间比较差异均无显著性。结论Lipoven20%加重ALI小鼠的炎症反应,降低实验动物的存活率,可能与其增加AA产生有关;而Clinoleic20%对炎症反应的影响较轻。 | 毕铭华 王宝恩 Martina Schafer Konstantin Mayer 张淑文 李敏 王惠吉 | 2006 | 中国危重病急救医学2006,18,12: | 5 |

| 10 | Degradation of intestinal mRNA:A matter of treatment显示文摘AIM: To characterize the influence of location, species and treatment upon RNA degradation in tissue samples from the gastrointestinal tract. METHODS: The intestinal samples were stored in different medium for different times under varyingconditions: different species(human and rat), varying temperature(storage on crushed ice or room temperature), time point of dissection of the submucous-mucous layer from the smooth muscle(before or after storage), different rinsing methods(rinsing with Medium, PBS, RNALater or without rinsing at all) and different regions of the gut(proximal and distal small intestine, caecum, colon and rectum). The total RNA from different parts of the gut(rat: proximal and distal small intestine, caecum, colon and rectum, human: colon and rectum) and individual gut layers(muscle and submucosal/mucosal) was extracted. The quality of the RNA was assessed by micro capillary electrophoresis. The RNA quality was expressed by the RNA integrity number which is calculated from the relative height and area of the 18 S and 28 S RNA peaks. From rat distal small intestine q PCR was performed for neuronal and glial markers.RESULTS: RNA obtained from smooth muscle tissue is much longer stable than those from submucosal/mucosal tissue. At RT muscle RNA degrades after one day, on ice it is stable at least three days. Cleaning and separation of gut layers before storage and use of RNALater, maintains the stability of muscle RNA at RT for much longer periods. Different parts of the gut show varying degradation periods. RNA obtained from the submucosal/mucosal layer always showed a much worse amplification rate than RNA from muscle tissue. In general RNA harvested from rat tissue, either smooth muscle layer or submucosal/mucosal layer is much longer stable than RNA from human gut tissue, and RNA obtained from smooth muscle tissue shows an increased stability compared to RNA from submucosal/mucosal tissue. At RT muscle RNA degrades after one day, while the stability on ice lasts at least three days. Cleaning and separation of gut layers before storage and use of RNALater, maintains the stability of muscle RNA at RT for much longer periods. Different parts of the gut show varying degradation periods. The RNA from muscle and submucosal/mucosal tissue of the proximal small intestine degrades much faster than the RNA of distal small intestine, caecum or colonwith rectum. RNA obtained from the submucosal/mucosal layer always showed a much more reduced amplification rate than RNA from muscle tissue [β-Tubulin Ⅲ for muscle quantification cycle(Cp): 22.07 ± 0.25, for β-Tubulin Ⅲ submucosal/mucosal Cp: 27.42 ± 0.19].CONCLUSION: Degradation of intestinal m RNA depends on preparation and storage conditions of the tissue. | 12 | 用于脂肪移植及脂肪干细胞分离获取与处理技术的对比显示文摘背景与目的脂肪获取和处理技术的差异可能会影响脂肪移植的成功率。本研究利用裸鼠模型,对比不同处理方法获取的脂肪移植物的性质,并评估移植脂肪组织中存活组织的体积。方法第一阶段,通过负压吸引辅助脂肪抽吸或超声辅助脂肪抽吸2种方式获取脂肪,并利用2种孔隙尺寸的筛网进行过滤。对移植材料的如下方面进行分析:平均团块大小,油脂、脂肪和水溶性成分的构成比,以及基质血管细胞群(stromalvascularfraction,SVF)的产量。将过滤物及滤液注入无胸腺的裸鼠皮下。第二阶段,将仅通过负压吸引吸脂术中获取脂肪抽吸物,进行离心、纱布摇匀或过滤处理,然后利用相似的规范进行研究。结果与负压吸引脂肪抽吸术相比,超声辅助的脂肪抽吸术获取的脂肪在SVF含量和脂肪移植体内研究成活量情况均相似。通过超声辅助的脂肪抽吸术仅比负压脂肪抽吸术产生稍微多的油脂;通过500μm或800μm孔径的滤网进行过滤均可有效去除液体和油脂。利用纱布法处理的脂肪,其体内移植成活率和SVF产量均最高。对所有移植物的组织学分析均显示为完整的脂肪组织。结论通过超声辅助及负压吸引行脂肪抽吸术获取的脂肪,其在异种移植模型中具有相似的成活率。对于少量填充移植而要求最佳成活率的身体敏感区域而言,利用纱布摇匀的处理方式可能是用于美容性脂肪移植的最佳方法。过滤法和离心法均可有效去除液体成分,其处理过的脂肪具有相似的移植成活率,当所需移植体积较大时,这2种方法均可行。 | Christine Fisher Tara L.
