1Characteristics of Obesity and Its Related Disorders in China显示文摘Obesity is a medical condition with excess body fat accumulation to the extent which leads to serious health consequences.Abdominal obesity,also known as central obesity,refers to the presence of excess fat in the abdominal area.Obesity,especially abdominal obesity,contributes to many metabolic disorders including metabolic syndrome (MetS),type 2 diabetes (T2DM) and cardiovascular diseases (CVD).The incidence of obesity has increased dramatically in recent years worldwide.In China,more than one-third of adults are overweight or obese and 10%-20% of all adults are affected by MetS.The pathogenesis underlying the abdominal obesity remains unclear.The ultimate health outcome of obesity and its related metabolic disorders have prompted physicians to take aggressive treatments (lifestyle changes,pharmacological interventions and surgical therapies) before a serious consequence becomes clinically apparent.In this review,we discuss the prevalence,pathogenesis and clinic features of obesity in China.WEI-PING JIA ,CHEN WANG,SHAN JIANG,AND JIE-MIN PAN Department of Endocrinology and Metabolism,Shanghai Jiao Tong University Affiliated Sixth People’s Hospital,Shanghai Diabetes Institute,Shanghai Clinical Center of Diabetes,Shanghai 200233,China 2010Biomedical and Environmental Sciences2010,23,1:46
2Symmetric-key cryptosystem with DNA technology显示文摘DNA cryptography is a new field which has emerged with progress in the research of DNA computing. In our study, a symmetric-key cryptosystem was designed by applying a modern DNA biotechnology, microarray, into cryptographic technologies. This is referred to as DNA symmetric-key cryptosystem (DNASC). In DNASC, both encryption and decryption keys are formed by DNA probes, while its ciphertext is embedded in a specially designed DNA chip (microarray). The security of this system is mainly rooted in difficult biology processes and problems, rather than conventional computing technology, thus it is unaffected by changes from the attack of the coming quantum computer. The encryption process is a fabrication of a specially designed DNA chip and the decryption process is the DNA hybridization. In DNASC, billions of DNA probes are hybridized and identified at the same time, thus the decryption process is conducted in a massive, parallel way. 3Metabolomics of gastric cancer metastasis detected by gas chromatography and mass spectrometry显示文摘AIM:To elucidate the underlying mechanisms of metastasis and to identify the metabolomic markers of gastric cancer metastasis.METHODS:Gastric tumors from metastatic and nonmetastatic groups were used in this study.Metabolites and different metabolic patterns were analyzed by gas chromatography,mass spectrometry and principal components analysis (PCA),respectively.Differentiation performance was validated by the area under the curve (AUC) of receiver operating characteristic curves.RESULTS:Twenty-nine metabolites were differentially expressed in animal models of human gastric cancer.Of the 29 metabolites,20 were up-regulated and 9 were down-regulated in metastasis group compared to non-metastasis group.PCA models from the metabolite profiles could differentiate the metastatic from the nonmetastatic specimens with an AUC value of 1.0.These metabolites were mainly involved in several metabolic pathways,including glycolysis (lactic acid,alaline),serine metabolism (serine,phosphoserine),proline metabolism (proline),glutamic acid metabolism,tricarboxylic acid cycle (succinate,malic acid),nucleotide metabolism (pyrimidine),fatty acid metabolism (docosanoic acid,and octadecanoic acid),and methylation(glycine).The serine and proline metabolisms were highlighted during the progression of metastasis.CONCLUSION:Proline and serine metabolisms play an important role in metastasis.The metabolic profiling of tumor tissue can provide new biomarkers for the treatment of gastric cancer metastasis.Jin-Lian Chen,Jun-Duo Hu,Jing Fan,Jing Hong,Department of Gastroenterology,Shanghai Sixth People’s Hospital,Shanghai Jiao Tong University,Shanghai 200233,China Hui-Qing Tang,Jian-Zhong Gu,Shanghai Laboratory Animal Center,Chinese Academy of Sciences,Shanghai 201615,China 2010World Journal of Gastroenterology2010,16,46:11
4Germline mutation analysis of hPMS2 gene in Chinese families with hereditary nonpolyposis colorectal cancer显示文摘AIM: To study the germline mutation of hPMS2 gene in 26 unrelated Chinese hereditary nonpolyposis colorectal cancer (HNPCC) probands and to fulfill the screening strategy for HNPCC in Chinese. METHODS: Genomic DNA was extracted from the peripheral blood. To avoid the interference of pseudogene in detection of the remaining 11 exons (exon 1-5, 9, 11-15), long-range polymerase chain reaction (PCR) was conducted to amplify the complete coding region of hPMS2 gene firstly. Then 1/8 of the PCR productswere used as template to amplify the individual exon respectively and DNA sequencing was done. Direct DNA sequencing of the conventional PCR products of exon 6, 7, 8 and 10 of hPMS2 gene was performed. The same analysis was made in 130 healthy persons without family histories of HNPCC to further investigate the pathological effects of the detected missense mutation. RESULTS: One HNPCC proband fulf illed Bethesda guidelines and was found to carry the germline mutation of hPMS2 gene, which has not been reported in Chinese HNPCC families. It was a missense mutation at c.1532C>T of exon 11. It was detected in three controls as well with an occurrence rate of 2.3% (3/130). Since it could not be found in the PMS2-single nucleotide polymorphism (SNP) database, this missense mutation is a new SNP unreported up to date. Meanwhile, 260 reported SNPs of hPMS2 gene were detected in the 26 HNPCC probands. The 2nd and 5th exons were probably the hot SNP regions of hPMS2 gene in Chinese HNPCC families involving 53.1% of all reported SNP. CONCLUSION: The germline mutation of hPMS2 gene may be rare in Chinese HNPCC families. 5Evaluation of the colorectal cancer risk conferred by rare UNC5C alleles显示文摘AIM:To evaluate the risk associated with variants of the UNC5C gene recently suspected to predispose to familial colorectal cancer(CRC).METHODS:We screened patients with familial CRC forms as well as patients with sporadic CRCs.In a first time,we analyzed exon 11 of the UNC5C gene in 120unrelated patients with suspected hereditary CRC,58patients with suspected Lynch-associated cancer or polyposis,and 132 index cases of Lynch syndrome families with a characterized mutation in a DNA mismatch repair(MMR).Next,1023 patients with sporadic CRC and1121 healthy individuals were screened for the variants identified in patients with familial cancer.RESULTS:Of 120 patients with familial CRC of unknown etiology,one carried the previously reported mis-sense mutation p.Arg603Cys(R603C)and another exhibited the unreported variant of unknown significance p.Thr617Ile(T617I).The p.Ala628Lys(A628K)mutation previously described as the main UNC5C risk variant for familial CRC was not detected in any cases of familial CRC of unknown etiology,but was present in a patient with familial gastric cancer and in two Lynch syndrome patients in co-occurrence with MMR mutations.A statistically non-significant increase in cancer risk was identified in familial CRC and/or other Lynchassociated cancers(1/178 patients vs 2/1121 healthy controls,OR=3.2,95%CI:0.29-35.05,P=0.348)and in sporadic CRCs(4/1023 patients vs 2/1121 healthy controls,OR=2.2,95%CI:0.40-12.02,P=0.364).CONCLUSION:We confirm that UNC5C mutations are very rare in familial and sporadic CRCs,but further investigations are needed to justify routine UNC5C testing for diagnostic purposes.Sébastien Küry Céline Garrec Fabrice Airaud Flora Breheret Virginie Guibert Cécile Frenard Shuo Jiao Dominique Bonneau Pascaline Berthet Céline Bossard Olivier Ingster Estelle Cauchin Stéphane Bezieau 2014World Journal of Gastroenterology2014,20,1:4

7Short-segment Barrett's esophagus and cardia intestinal metaplasia:A comparative analysis显示文摘AIM:To investigate the endoscopy and histology of short-segment Barrett's esophagus (SSBE) and cardia intestinal metaplasia (CIM),and their correlation with Helicobacter pylori (H. pylori) gastritis and gastroesophageal reflux disease (GERD). METHODS:Biopsy specimens were taken from 32 SSBE patients and 41 CIM patients with normal appearance of the esophagogastric junction. Eight biopsy specimens from the lower esophagus,cardia,and gastric antrum were stained with hematoxylin/eosin,Alcian blue/periodic acid-Schiff,Alcian blue/high iron diamine and Gimenez dye. Results were graded independently by one pathologist. RESULTS:The SSBE patients were younger than the CIM patients (P < 0.01). The incidence of dysplasia and incomplete intestinal metaplasia subtype was higher in SSBE patients than in CIM patients (P < 0.01). H. pylori infection was correlated with antral intestinal metaplasia (P < 0.05),but not with reflux symptomatic,endoscopic,or histological markers of GERD in CIM patients. SSBE was correlated with reflux symptomatic and endoscopic esophagitis (P < 0.01),but not with H. pylori infection and antral intestinal metaplasia. CONCLUSION:Dysplasia risk is significantly greater in SSBE patients than in CIM patients. 10Thermal oxide synthesis and characterization of Fe_3O_4 nanorods and Fe_2O_3 nanowires显示文摘Fe3O4 nanorods and Fe2O3 nanowires have been synthesized through a simple thermal oxide reaction of Fe with C2H2O4 solution at 200-600℃ for 1 h in the air. The morphology and structure of Fe3O4 nanorods and Fe2O3 nanowires were detected with powder X-ray diffraction, scanning electron microscopy and transmission electron microscopy. The influence of temperature on the morphology development was experimentally investigated. The results show that the polycrystals Fe3O4 nanorods with cubic structure and the average diameter of 0.5-0.8 μm grow after reaction at 200-500℃ for 1 h in the air. When the temperature was 600℃, the samples completely became Fe2O3 nanowires with hexagonal structure. It was found that C2H2O4 molecules had a significant effect on the formation of Fe3O4 nanorods.
