The above effects are fulfilled by regulation of cell cycle engine and other cell proliferation related proteins. | 2 | Effect of Silicon on Leaf Ultrastructure, Chlorophyll Content and Photosynthetic Activity of Barley Under Salt Stress显示文摘Two contrasting cultivars of barley (Horderm vykgare L.): Kepin No. 7 (salt sensitive), and Jian 4 (salt tolerant) were grown in a hydroponics system with 2 NaCl levels: 60 mmol NaCl L-1 and 120 mmol NaCl L-1, and 3 Si levels: 0 mmol Si L-1, 0.5 mmol Si L-1 and 1.0 mmol Si L-1 (as silicic acid). Compared with the plants treated with 60 mmol NaCl L-1 alone, the leaf chlorophyll contents of plants treated with salt and Si increased significantly for salt-sensitive cultivar at tillering stage, but for salt-tolerant cultivar,the addition of Si resulted in an obvious increase in the leaf chlorophyll content of plants exposed to 120 mmol NaCl L-1. However, this Si-enhancement of leaf chlorophyll content was also observed in the salttolerant plants at jointing stage, but not in the salt-sensitive plants. Moreover, leaf chlorophyll content was consistently higher for the salt-tolerant cultivar than for the salt-sensitive cultivar irrespective of salt and/or Si treatment. Compared with the plants treated with salt alone, net CO2 assimilation rate in plant leaves increased significantly for both cultivars when trested with salt and Si. The addition of Si to the salt treatment was found to improve the cell ultrastructure of leaves. Under salt stress condition, the double membranes of chloroplasts disappeared, but membrane integrity was markedly improved in the salt treatment supplemented with Si. Silicon was also found to ameliorate the damage to the ultrastructure of chloroplast granae which appeared to be disintegrated and vague in salt treatments without added Si. | 3 | Discovery of laryngeal carcinoma by serum proteomic pattern analysis显示文摘Laryngeal carcinoma is the most common malignancy among head and neck tu-mors. The purpose of this study is to find biomarkers for laryngeal carcinoma in patient blood serum using the Surface Enhanced Laser Desorption/Ionization (SELDI) technique. Serum samples from 33 laryngeal carcinoma (12 cases of glottis, 18 of supraglottis and 3 of subglottis) patients and 31 age- and sex-matched healthy people were analyzed by SELDI-TOF on a Pro-teinChip reader, PBSII-C. Protein profiles were generated using WCX2 protein chips. Protein peak clustering and classification analyses were performed utilizing the Biomarker Wizard and Biomarker Pattern software packages, respectively. The results showed that sixteen peaks had significant difference between laryngeal cancer patients and healthy group, eight of which were up-regulated in the patient samples, and the others were down-regulated. Two protein peaks 8153 Da and 2035 Da were automatically chosen for the system training and development of a classification tree. The analysis yielded a correct percentage of 96.9% for patients and 96.7% for control. The results suggest that serum is a useful resource for the detection of specific bio-markers for laryngeal carcinoma. | 5 | Effects of silica nanoparticles on growth and photosynthetic pigment contents of Scenedesmus obliquus显示文摘To assess the aquatic ecosystem safety for silica (SiO2) nanoparticles (NPs), the growth inhibition and photosynthetic pigment contents of Scenedesmus obliquus in logarithm growth phase exposed to SiO2 NPs and SiO2 bulk particles (BPs) suspensions were measured. SiO2 NPs with 10-20 nm diameters were found to be toxic. The 20% effective concentration (EC20) values for 72 and 96 hr were 388.1 and 216.5 mg/L, respectively. The contents of chlorophyll decreased significantly under moderate and high concentration (50, 100, and 200 mg/L) of SiO2 NPs after 96-hr exposure, but the carotenoids did not. SiO2 BPs were found to be nontoxic up to 200 mg/L. The toxicity of SiO2 NPs probablely due to their sorption to algal cells surface. | 6 | Relationship between osmotic stress and polyamines conjugated to the deoxyribonucleic acid-protein in wheat seedling roots显示文摘The contents of covalently conjugated polyamines (CC-PAs) and noncovalently conju- gated polyamines (NCC-PAs) to deoxyribonucleic acid-protein (DNP) isolated from wheat (Triticum aestivum L.) seedling roots under osmotic stress were detected. Results showed that after osmotic stress treatment for 7 d, the levels in NCC-spermine (NCC-Spm) and NCC-spermidine (NCC-Spd) of drought-tolerant Yumai No. 18 cv. increased more markedly than that of drought-sensitive Yangmai No. 9 cv., while the NCC-putrescine (NCC-Put) could not be statistically detected in two cultivars. Exogenous Spm treatment alleviated osmotic stress injury to Yangmai No. 9 cv. seedlings, coupled with marked increases of NCC-Spm and NCC-Spd levels of this cultivar. Under PEG osmotic stress, the concomitant treatment of drought-tolerant Yumai No. 18 cv.seedlings with methylglyoxyl-bis (guanylhydrazone) (MGBG), an inhibitor of S-adenosylmethionine decarboxylase (SAMDC), aggravated osmotic stress injury to this cultivar, coupled with market decreases of the NCC-Spm and NCC- Spd levels. Owing to their unique features, stem cells provide excellent opportunities for experimental analyses of basic developmental processes such as pluripotency control and cell fate decision and for regenerative medicine by stem cell-based therapy. Stem cell cultures have been best studied in 3 vertebrate organisms. These are the mouse, human and a small laboratory fish called medaka. Specifically, medaka has given rise to the first embryonic stem (ES) cells besides the mouse, the first adult testis-derived male stem cells spermatogonia capable of test-tube sperm production, and most recently, even haploid ES cells capable of producing Holly, a semi-cloned fertile female medaka from a mosaic oocyte created by microinjecting a haploid ES cell nucleus directly into a normal oocyte. | 8 | Discovery and identification of Serum Amyloid A protein elevated in lung cancer serum显示文摘Two hundred and eighteen serum samples from 175 lung cancer patients and 43 healthy individuals were analyzed by using Surface Enhaced Laser Desorption/Ionization Time of Flight Mass Spectrome- try (SELDI-TOF-MS). The data analyzed by both Biomarker Wizard? and Biomarker Patterns? software showed that a protein peak with the molecular weight of 11.6 kDa significantly increased in lung cancer. Meanwhile,the level of this biomarker was progressively increased with the clinical stages of lung cancer. The candidate biomarker was then obtained from tricine one-dimensional sodium dodecyl sul- fate-polyacrylamide gel electrophoresis by matching the molecular weight with peaks on WCX2 chips and was identified as Serum Amyloid A protein (SAA) by MALDI/MS-MS and database searching. It was further validated in the same serum samples by immunoprecipitation with commercial SAA antibody. To confirm the SAA differential expression in lung cancer patients, the same set of serum samples was measured by ELISA assay. The result showed that at the cutoff point 0.446(OD value)on the Receiver Operating Characteristic (ROC) curve, SAA could better discriminate lung cancer from healthy indi- viduals with sensitivity of 84.1% and specificity of 80%. We explored this issue using NIH3T3 cells with inducible p21Waf1/cip1 expression. Induc-tion of p21Waf1/Cip1 triggered G1 growth arrest, and NIH3T3-p21 cells exhibited morphologic features,such as enlarged and flattened cellular shape, specific to the senescence phenotype. The results showed that under NSDB<0 (CK), nitrogen translocation and senescence of the top 4leaves was normal. Nitrogen exported from the top leaves (LFT) especially the top 4 leaves decreased remar-kably with increase of NSDB, thus, their MDA contents and differences in color (SPAD value) decreased,while the activities of SOD and CAT increased. | 12 | Effects of Panicle Nitrogen Fertilization on Non-Structural Carbohydrate and Grain Filling in Indica Rice显示文摘Grain filling, a crucial stage of grain yield formation in rice, is usually affected by the panicle nitrogen (N) fertilization. Field and pot culture experiments were conducted to explore the underlying mechanisms of N effect. Two rice cultivars with high lodging resistance were grown in the field and pot. Four panicle N fertilization treatments were conducted in 2006 and repeated in 2007. The result showed that medium level of panicle N fertilization treatment (NM) enhanced the accumulation and translocation of non-structural carbohydrate (NSC) in the stem and sheath. Compared with non-nitrogen treatment (N0), NM promoted the translocation of labeled 13C from stem and sheath to grain. But, low level of panicle N fertilization treatment (NL) and high level of panicle N fertilization treatment (NH) showed the negative effect. The endosperm cell, grain length, and grain width of NM increased more quickly than that of N0 from 4 to 10 d after anthesis. During the early period of grain filling, sucrose-phosphate synthase (EC 2.4.1.14, SPS) activity were significantly higher for the NM treatment than those of the NL and NH treatments. Sucrose synthase (EC 2.4.1.13, SuSase) activity in the grains was substantially enhanced by NM, with the duration of higher activity being longer than those of the other treatments. At maturing stage, NM significantly increased the filled grain number, the seed-setting rate, and the grain weight compared with NL and NH. | 13 | Monitoring Protein and Starch Accumulation in Wheat Grains with Leaf SPAD and Canopy Spectral Reflectance显示文摘The research was conducted to determine the relationships of protein and starch accumulation dynamics in grains of wheat to post-heading leaf SPAD values and canopy spectral reflectance. The results showed that leaf nitrogen accumulation was exponentially related to leaf SPAD values and linearly related to canopy spectral reflectance, and that there was negative linear relationship between leaf nitrogen accumulation and grain protein accumulation, but positive linear relationship between post-heading leaf nitrogen translocation and grain protein accumulation at maturity. In addition, leaf SPAD values were parabolically related with and ratio indices R(1 500,610) and R(1 220,560) were exponentially related with protein and starch accumulation in grains. | 14 | Effects of nitrogen ion implantation on Ca^(2+) concentration and membrane potential of pollen cell显示文摘The effects of low energy nitrogen ion implantation on Ca2+ concentration and membrane potential of illy (Mum davidii Duch) pollen cell have been studied. The results showed that the Ca2+ concentration was increased when pollen grain was implanted by nitrogen ion with energy 100 keV and dose 1013 ions/cm2. However, the increase of Ca2+ concentration was partly inhibited by the addition of Ca2+ channel inhibitor depending on dose. And nitrogen ion implantation caused depolarization of pollen cell membrane potential. In other words, membrane potential was increased, but the effect decreased by adding Ca2+ channel inhibitor. However, it was still significantly higher than the membrane potential of control cells. It was indicated that the depolarization of cell membrane potential opened the calcium channel on the membrane that caused the increasing of intracel-lular calcium concentration. | 19 | Subtropical Modern Greenhouse Cucumber Canopy Transpiration Under Summer Climate Condition显示文摘Greenhouse canopy transpiration not only has effects on greenhouse air temperature and humidity, but also is important for determining the set-point of fertigation. In this study, Penman-Monteith equation was used to calculate the greenhouse cucumber canopy transpiration under summer climate condition.The effects of greenhouse environmental factors on canopy transpiration were analyzed based on the measurements of greenhouse microclimate factors and canopy transpiration. The results showed that Penman-Monteith equation was reliable and robust in estimating greenhouse cucumber canopy transpiration under summer climate condition. Greenhouse cucumber canopy transpiration rate increased linearly with the increase of net radiation and water vapor pressure deficit (VPD) above the canopy. But the maximum value of the canopy transpiration rate occurred at the same time as that of VPD whereas about two hours later than that of net radiation. | 20 | A Knowledge Model-and Growth Model-Based Decision Support System for Wheat Management显示文摘By applying the system analysis principle and mathematical modeling technique to knowledge expression system for crop cultural management, the fundamental relationships and quantitative algorithms of wheat growth and management indices to variety types, ecological environments and production levels were analysed and extracted, and a dynamic knowledge model with temporal and spatial characters for wheat management (WheatKnow) was developed. By adopting the soft component characteristics as non language rele vance, re-utilization and portable system maintenance, and by further integrating the wheat growth simulation model (WheatGrow) and intelligent system for wheat management, a comprehensive and digital knowledge model, growth model and component-based decision support system for wheat management (MBDSSWM) was established on the platforms of Visual C++ and Visual Basic.
