16卵内免疫接种不同重组新城疫病毒Clone30变异株的致病性及免疫原性研究显示文摘虽然新城疫病毒(NDV)活疫苗株可以应用于1日龄雏鸡,但是在孵化前3 d卵内注射该活疫苗株后,可对鸡胚有致病性。以反向遗传学为基础,通过在疫苗株Clone30内引入特定突变的融合(F)蛋白和血凝素神经氨酸酶(HN)蛋白,作者获得了重组NDV(rNDV),Mast等(2006)的研究建议采用该方法减弱目的疫苗变异株的毒力,进而获得rNDV49。另一个重组株(rNDVGu)是通过改变rNDV和NDV疫苗株Clone30之间的差异序列而生成的。与NDVClone30相比,rNDV、rNDV49和rNDVGu重组病毒已经降低了毒力,且脑内接种致病指数降低,平均死亡时间升高。卵内注射后,所有感染组的孵化率差异很大。但是经过21 d的观察期,NDV Clone30组只有一只鸡存活;然而,接种重组NDV组的孵化鸡的存活率为40%~80%,且rNDVGu是最高致病性的病毒。另外,孵化后21 d内,重组病毒可诱导产生抵抗NDV强毒感染的保护力。重组病毒诱发不同抗体反应结果显示,免疫原性与毒力相关。总之,研究结果表明,点突变可以降低NDV的毒力,然而通过卵内注射后对鸡致病力的研究显示,rNDV的F和HN蛋白的特定氨基酸的改变并没有引起其毒力进一步的减弱。Ramp K 吴艳 2012中国畜牧兽医2012,39,8:0
