Mesenchymal stem cells: Molecular characteristics and clinical applications显示文摘Mesenchymal stem cells (MSCs) are non-hematopoietic stem cells with the capacity to differentiate into tissues of both mesenchymal and non-mesenchymal origin. MSCs can differentiate into osteoblastic, chondrogenic, and adipogenic lineages, although recent studies have demonstrated that MSCs are also able to differentiate into other lineages, including neuronal and cardiomyogenic lineages. Since their original isolation from the bone marrow, MSCs have been successfully harvested from many other tissues. Their ease of isolation and ex vivo expansion combined with their immunoprivileged nature has made these cells popular candidates for stem cell therapies. These cells have the potential to alter disease pathophysiology through many modalities including cytokine secretion, capacity to differentiate along various lineages, immune modulation and direct cell-cell interaction with diseased tissue. Here we first review basic features of MSC biology including MSC characteristics in culture, homing mechanisms, differentiation capabilities and immune modulation. We then highlight some in vivo and clinical evidence supporting the therapeutic roles of MSCs and their uses in orthopedic, autoimmune, and ischemic disorders. | Farbod Rastegar Deana Shenaq Eric R Wagner Stephanie H Kim Russell R Reid Hue H Luu Rex C Haydon | 2010 | World Journal of Stem Cells2010,2,4: | 35
Sensing for advancing mining automation capability:A review of underground automation technology development显示文摘This paper highlights the role of automation technologies for improving the safety, productivity, and environmental sustainability of underground coal mining processes. This is accomplished by reviewing the impact that the introduction of automation technology has made through the longwall shearer automation research program of Longwall Automation Steering Committee(LASC). This result has been achieved through close integration of sensing, processing, and control technologies into the longwall mining process. Key to the success of the automation solution has been the development of new sensing methods to accurately measure the location of longwall equipment and the spatial configuration of coal seam geology. The relevance of system interoperability and open communications standards for facilitating effective automation is also discussed. Importantly, the insights gained through the longwall automation development process are now leading to new technology transfer activity to benefit other underground mining processes. | Ralston Jonathon Reid David Hargrave Chad Hainsworth David | 2014 | International Journal of Mining Science and Technology2014,24,3: | 37
Longwall automation: Delivering enabling technology to achieve safer and more productive underground mining显示文摘The ongoing need to deliver improved safety, productivity and environmental benefit in coal mining presents an open challenge as well as a powerful incentive to develop new and improved solutions. This paper assesses the critical role that enabling technologies have played in the delivery of remote and automated capability for longwall mining. A brief historical account is given to highlight key technical contributions which have influenced the direction and development of present-day longwall technology. The current state of longwall automation is discussed with particular attention drawn to the technologies that enable automated capability. Outcomes are presented from an independently conducted case study that assessed the impact that CSIRO's LASC longwall automation research has made to the longwall mining industry in Australia. Importantly, this study reveals how uptake of this innovative technology has significantly benefitted coal mine productivity, improved working conditions for personnel and enhanced environmental outcomes. These benefits have been widely adopted with CSIRO automation technology being used in 60 per cent of all Australian underground operations.International deployment of the technology is also emerging. The paper concludes with future challenges and opportunities to highlight the ongoing scope for longwall automation research and development. | Ralston Jonathon C. Reid David C. Dunn Mark T. | 4 | Molecular Analysis of Legume Nodule Development and Autoregulation显示文摘Legumes are highly important food, feed and biofuel crops. With few exceptions, they can enter into an intricate symbiotic relationship with specific soil bacteria called rhizobia. This interaction results in the formation of a new root organ called the nodule in which the rhizobia convert atmospheric nitrogen gas into forms of nitrogen that are useable by the plant. The plant tightly controls the number of nodules it forms, via a complex root-to-shoot-to-root signaling loop called autoregulation of nodulation (AON). This regulatory process involves peptide hormones, receptor kinases and small metabolites. Using modern genetic and genomic techniques, many of the components required for nodule formation and AON have now been isolated. This review addresses these recent findings, presents detailed models of the nodulation and AON processes, and identifies gaps in our understanding of these process that have yet to be fully explained. | Brett J. | 5 | Platelets in hemostasis and thrombosis:Novel mechanisms of fibrinogen-independent platelet aggregation and fibronectin-mediated protein wave of hemostasis显示文摘Platelets are small anucleate cells generated from megakaryocytes in the bone marrow.Although platelet generation, maturation, and clearance are still not fully understood, significant progress has been made in the last 1-2 decades. In blood circulation, platelets can quickly adhere and aggregate at sites of vascular injury, forming the platelet plug(i.e.the first wave of hemostasis).Activated platelets can also provide negatively charged phosphatidylserinerich membrane surface that enhances cell-based thrombin generation, which facilitates blood coagulation(i.e.the second wave of hemostasis).Platelets therefore play central roles in hemostasis. However, the same process of hemostasis may also cause thrombosis and vessel occlusion, which are the most common mechanisms leading to heart attack and stroke following ruptured atherosclerotic lesions. In this review,we will introduce the classical mechanisms and newly discovered pathways of platelets in hemostasis and thrombosis, including fibrinogen-independent platelet aggregation and thrombosis, and the plasma fibronectin-mediated 'protein wave' of hemostasis that precedes the classical first wave of hemostasis.Furthermore,we briefly discuss the roles of platelets in inflammation and atherosclerosis and the potential strategies to control atherothrombosis. | Yan Hou Naadiya Carrim Yiming Wang Reid C.Gallant Alexandra Marshall Heyu Ni | 2015 | The Journal of Biomedical Research2015,29,6: | 21
The wonders of BMP9:From mesenchymal stem cell differentiation, angiogenesis, neurogenesis, tumorigenesis, and metabolism to regenerative medicine显示文摘Although bone morphogenetic proteins(BMPs)initially showed effective induction of ectopic bone growth in muscle,it has since been determined that these proteins,as members of the TGF-b superfamily, play a diverse and critical array of biological roles. These roles include regulating skeletal and bone formation, angiogenesis, and development and homeostasis of multiple organ systems.Disruptions of the members of the TGF-b/BMP superfamily result in severe skeletal and extra-skeletal irregularities, suggesting high therapeutic potential from understanding this family of BMP proteins.Although it was once one of the least characterized BMPs,BMP9 has revealed itself to have the highest osteogenic potential across numerous experiments both in vitro and in vivo, with recent studies suggesting that the exceptional potency of BMP9 may result from unique signaling pathways that differentiate it from other BMPs.The effectiveness of BMP9 in inducing bone formation was recently revealed in promising experiments that demonstrated efficacy in the repair of critical sized cranial defects as well as compatibility with bone-inducing bio-implants,revealing the great translational promise of BMP9.Furthermore,emerging evidence indicates that, besides its osteogenic activity,BMP9 exerts a broad range of biological functions, including stem cell differentiation, angiogenesis, neurogenesis, tumorigenesis, and metabolism. This review aims to summarize our current understanding of BMP9 across biology and the body. | Sami Mostafa Mikhail Pakvasa Elam Coalson Allen Zhu Alex Alverdy Hector Castillo Jiaming Fan Alex Li Yixiao Feng Di Wu Elliott Bishop Scott Du Mia Spezia Alissa Li Ofir Hagag Alison Deng Winny Liu Mingyang Li Sherwin S·Ho Aravind Athiviraham Michael J·Lee Jennifer Moriatis Wolf Guillermo A·Ameer Hue H·Luu Rex C·Haydon Jason Strelzow Kelly Hynes Tong-Chuan He Russell R·Reid | 2019 | Genes & Diseases2019,6,3: | 15
Epidemiology of perforated peptic ulcer:Age-and gender-adjusted analysis of incidence and mortality显示文摘AIM:To investigate the epidemiological trends in inci-dence and mortality of perforated peptic ulcer(PPU)in a well-defined Norwegian population. METHODS:A retrospective,population-based,single-center,consecutive cohort study of all patients diag-nosed with benign perforated peptic ulcer.Included were both gastric and duodenal ulcer patients admitted to Stavanger University Hospital between January 2001 and December 2010.Ulcers with a malignant neoplasia diagnosis, verified by histology after biopsy or resection, were excluded.Patients were identified from the hospitals administrative electronic database using pertinent ICD-9 and ICD-10 codes(K25.1,K25.2,K25.5, K25.6,K26.1,K26.2,K26.5,K26.6).Additional searches using appropriate codes for relevant laparoscopic and open surgical procedures(e.g.,JDA 60,JDA 61,JDH 70 and JDH 71)were performed to enable a complete identification of all patients.Patient demographics, presentation patterns and clinical data were retrieved from hospital records and surgical notes.Crude and adjusted incidence and mortality rates were estimated by using national population demographics data. RESULTS:In the study period,a total of 172 patients with PPU were identified. The adjusted incidence rate for the overall 10-year period was 6.5 per 100 000 per year(95%CI:5.6-7.6)and the adjusted mortality rate for the overall 10-year period was 1.1 per 100 000 per year(95%CI:0.7-1.6).A non-significant decline in ad-justed incidence rate from 9.7 to 5.6 occurred during the decade. The standardized mortality ratio for the whole study period was 5.7(95%CI:3.9-8.2),while the total 30-d mortality was 16.3%.No difference in in-cidence or mortality was found between genders. However, for patients≥60 years, the incidence increased over 10-fold,and mortality more than 50-fold,compared to younger ages. The admission rates outside office hours were high with almost two out of three(63%) admissions seen at evening/night time shifts and/or during weekends. The observed seasonal variations in admissions were not statistically significant. CONCLUSION:The adjusted incidence rate, seasonal distribution and mortality rate was stable.PPU fre-quently presents outside regular work-hours.Increase in incidence and mortality occurs with older age. | Kenneth Thorsen Jon Arne Sreide Jan Terje Kvaly Tom Glomsaker Kjetil Sreide | 2013 | World Journal of Gastroenterology2013,19,3: | 14
Overexpression of an ABA biosynthesis gene using a stress-inducible promoter enhances drought resistance in petunia显示文摘The response of plants to drought stress includes reduced transpiration as stomates close in response to increased abscisic acid(ABA)concentrations.Constitutive overexpression of 9-cis-epoxycarotenoid dioxygenase(NCED),a key enzyme in ABA biosynthesis, increases drought resistance, but causes negative pleiotropic effects on plant growth and development. We overexpressed the tomato NCED(LeNCED1)in petunia plants under the control of a stress-inducible promoter,rd29A.Under water stress, the transgenic plants had increased transcripts of NCED mRNA,elevated leaf ABA concentrations, increased concentrations of proline, and a significant increase in drought resistance. The transgenic plants also displayed the expected decreases in stomatal conductance, transpiration, and photosynthesis. After 14 days without water, the control plants were dead, but the transgenic plants, though wilted, recovered fully when re-watered.Well-watered transgenic plants grew like non-transformed control plants and there was no effect of the transgene on seed dormancy. | Alejandro C Estrada-Melo Chao Ma Michael S Reid Cai-Zhong Jiang | 2015 | Horticulture Research2015,2,1: | 12
Transcriptomic landscape regulated by the 14 types of bone morphogenetic proteins(BMPs)in lineage commitment and differentiation of mesenchymal stem cells(MSCs)显示文摘Mesenchymal stem cells(MSCs)are ubiquitously-existing multipotent progenitors that can self-renew and differentiate into multiple lineages including osteocytes, chondrocytes, adipocytes, tenocytes and myocytes.MSCs represent one of the most commonly-used adult progenitors and serve as excellent progenitor cell models for investigating lineagespecific differentiation regulated by various cellular signaling pathways, such as bone morphogenetic proteins(BMPs).As members of TGFb superfamily,BMPs play diverse and important roles in development and adult tissues. At least 14 BMPs have been identified in mammals.Different BMPs exert distinct but overlapping biological functions.Through a comprehensive analysis of 14 BMPs in MSCs,we demonstrated that BMP9 is one of the most potent BMPs in inducing osteogenic differentiation of MSCs.Nonetheless,a global mechanistic view of BMP signaling in regulating the proliferation and differentiation of MSCs remains to be fully elucidated. Here,we conducted a comprehensive transcriptomic profiling in the MSCs stimulated by 14 types of BMPs.Hierarchical clustering analysis classifies 14 BMPs into three subclusters:an osteo/chondrogenic/adipogenic cluster,a tenogenic cluster, and BMP3 cluster. We also demonstrate that six BMPs(e.g.,BMP2,BMP3,BMP4,BMP7,BMP8,and BMP9)can induce ISmads effectively, while BMP2,BMP3,BMP4,BMP7,and BMP11 up-regulate Smad-independent MAP kinase pathway.Furthermore,we show that many BMPs can upregulate the expression of the signal mediators of Wnt,Notch and PI3K/AKT/mTOR pathways. While the reported transcriptomic changes need to be further validated, our expression profiling represents the first-of-its-kind to interrogate a comprehensive transcriptomic landscape regulated by the 14 types of BMPs in MSCs. | Linghuan Zhang Qing Luo Yi Shu Zongyue Zeng Bo Huang Yixiao Feng Bo Zhang Xi Wang Yan Lei Zhenyu Ye Ling Zhao Daigui Cao Lijuan Yang Xian Chen Bin Liu William Wagstaff Russell R*Reid Hue H*Luu Rex C*Haydon Michael J*Lee Jennifer Moriatis Wolf Zhou Fu Tong-Chuan He Quan Kang | 2019 | Genes & Diseases2019,6,3: | 11
Multiple cells of origin in cholangiocarcinoma underlie biological, epidemiological and clinical heterogeneity显示文摘Recent histological and molecular characterization of cholangiocarcinoma(CCA) highlights the heterogeneity of this cancer that may emerge at different sites of the biliary tree and with different macroscopic or morphological features.Furthermore, different stem cell niches have been recently described in the liver and biliarytree, suggesting this as the basis of the heterogeneity of intrahepatic(IH)-and extrahepatic(EH)-CCAs,which are two largely different tumors from both biological and epidemiological points of view. The complexity of the organization of the liver stem cell compartments could underlie the CCA clinical-pathological heterogeneity and the criticisms in classifying primitive liver tumors. These recent advances highlight a possible new classification of CCAs based on cells of origin and this responds to the need of generating homogenous diagnostic, prognostic and, hopefully, therapeutic categories of IH-and EH-CCAs. | Vincenzo Cardinale Guido Carpino Lola Reid Eugenio Gaudio Domenico Alvaro | 2012 | World Journal of Gastrointestinal Oncology2012,4,5: | 10

A long-form α-neurotoxin from cobra venom produces potent opioid-independent analgesia显示文摘Aim:In light of the antinociceptive activity of the short-chain neurotoxin, cobrotoxin, and other acetylcholine antagonists, the antinociceptive activity andmechanisms of cobratoxin(CTX),a long-chain postsynaptic α-neurotoxin,wasinvestigated in rodent pain models. Methods:CTX was administered intraperito-neally(30,45,68 μg/kg),intra-cerebral ventricularly(4.5 μg/kg)or microinjectedinto periaqueductal gray(PAG;4.5 μg/kg).The antinociceptive action was testedusing the hot-plate and acetic acid writhing tests m mice and rats. The involve-ment of the cholinergic system and opioid system in CTX-induced analgesia wasexamined by pretreatment of animals with atropine(0.5 mg/kg,im;or 10 mg/kg,ip)or naloxone(1 and 5 mg/kg,ip).The effect of CTX on motor activity was testedusing the Animex test. Results:CTX exhibited a dose-dependent analgesic ac-tion in mice as determined by both the hot-plate and acetic acid writhing tests. The peak effect of analgesia was seen 3 h after administration. In the mouse aceticacid writhing test, the intra-cerebral ventricular administration of CTX at 4.5 μg/kg(1/12th of a systemic dose)produced marked analgesic effects.Microinjection ofCTX(4.5 μgkg)into the PAG region did not elicit an analgesic action in rats in thehot-plate test.Atropine at 0.5 mg/kg(im)and naloxone at 1 and 5 mg/kg(ip)bothfailed to block the analgesic effects of CTX,but atropine at 10 mg/kg(ip)didantagonize the analgesia mediated by CTX in the mouse acetic acid writhing test.Acetylsalicylic acid(300 mg/kg)did not enhance the analgesic effects of CTX.Atthe highest effective dose of 68 μg/kg the neurotoxin did not change the sponta-neous mobility of mice. Conclusion:CTX has analgesic effects, which are medi-ated in the central nervous system though not through the PAG.The centralcholinergic system but not opioid system appears to be involved in theantinociceptive action of CTX. | Zhi-xin CHEN Hui-ling ZHANG Zhen-lun GU Bo-wen CHEN Rong HAN Paul F REID Laurence N RAYMOND Zheng-hong QIN | 2006 | Acta Pharmacologica Sinica2006,27,4: | 7
A basic helix-loop-helix transcription factor, PhFBH4, regulates flower senescence by modulating ethylene biosynthesis pathway in petunia显示文摘The basic helix-loop-helix(bHLH)transcription factors(TFs)play important roles in regulating multiple biological processes in plants. However, there are few reports about the function of bHLHs in flower senescence. In this study,a bHLH TF,PhFBH4,was found to be dramatically upregulated during flower senescence.Transcription of PhFBH4 is induced by plant hormones and abiotic stress treatments.Silencing of PhFBH4 using virus-induced gene silencing or an antisense approach extended flower longevity, while transgenic petunia flowers with an overexpression construct showed a reduction in flower lifespan.Abundance of transcripts of senescence-related genes(SAG12,SAG29)was significantly changed in petunia PhFBH4 transgenic flowers.Furthermore, silencing or overexpression of PhFBH4 reduced or increased, respectively, transcript abundances of important ethylene biosynthesis-related genes,ACS1 and ACO1,thereby influencing ethylene production. An electrophoretic mobility shift assay showed that the PhFBH4 protein physically interacted with the G-box cis-element in the promoter of ACS1,suggesting that ACS1 was a direct target of the PhFBH4 protein. In addition, ectopic expression of this gene altered plant development including plant height, internode length, and size of leaves and flowers, accompanied by alteration of transcript abundance of the gibberellin biosynthesis-related gene GA2OX3.Our results indicate that PhFBH4 plays an important role in regulating plant growth and development through modulating the ethylene biosynthesis pathway. | Jing Yin Xiaoxiao Chang Takao Kasuga Mai Bui Michael S Reid Cai-Zhong Jiang | 2015 | Horticulture Research2015,2,1: | 7
Adipose derived stem cells and nerve regeneration显示文摘Injuries to peripheral nerves are common and cause life-changing problems for patients alongside high social and health care costs for society. Current clinical treatment of peripheral nerve injuries predominantly relies on sacrificing a section of nerve from elsewhere in the body to provide a graft at the injury site. Much work has been done to develop a bioengineered nerve graft, precluding sacrifice of a functional nerve. Stem cells are prime candidates as accelerators of regeneration in these nerve grafts. | 17 | 3D printing of silk fibroin-based hybrid scaffold treated with platelet rich plasma for bone tissue engineering显示文摘3D printing/bioprinting are promising techniques to fabricate scaffolds with well controlled and patient-specific structures and architectures for bone tissue engineering. In this study,we developed a composite bioink consisting of silk fibroin(SF),gelatin(GEL),hyaluronic acid(HA),and tricalcium phosphate(TCP)and 3D bioprinted the silk fibroin-based hybrid scaffolds. The 3D bioprinted scaffolds with dual crosslinking were further treated with human platelet-rich plasma(PRP)to generate PRP coated scaffolds.Live/Dead and MTT assays demonstrated that PRP treatment could obviously promote the cell growth and proliferation of human adipose derived mesenchymal stem cells(HADMSC).In addition, the treatment of PRP did not significantly affect alkaline phosphatase(ALP)activity and expression, but significantly upregulated the gene expression levels of late osteogenic markers. This study demonstrated that the 3D printing of silk fibroin-based hybrid scaffolds,in combination with PRP post-treatment,might be a more efficient strategy to promote osteogenic differentiation of adult stem cells and has significant potential to be used for bone tissue engineering. | Liang Wei Shaohua Wu Mitchell Kuss Xiping Jiang Runjun Sun Reid Patrick Xiaohong Qin Duan Bin | 2019 | Bioactive Materials2019,4,1: | 6

| 20 | Geochemistry of the Rare Earth Elements in Natural Terrestrial Waters:A Review of What Is Currently Known显示文摘The range of observed Chemical compositions of natural terrestrial waters varies greatly especially when compared to the essentially constant global composition of the oceans. The concentrations of the REEs in natural terrestrial waters also exhibit more variation than what Was reported in seawater. In terrestrial waters, pH values span the range from acid up to alkaline. In addition, terrestrial waters can range from very dilute waters through to highly concentrated brines. The REE concentrations and their behavior. in natural terrestrial waters reflect these compositional ranges. Chemical weathering of rocks represents the source of the REEs to natural terrestrial waters and, consequently, the REE signature of rocks can impart their REE signature to associated waters. In addition, because of the typical low solubilities of the REEs, both surface and solution complexation can be important in fractionating REEs in aqueous solution. Both of these processes are important in all natural terrestrial waters, howev-er, their for relative importance varies as a function of the overall solution composition. In alkaline waters, for example, solution complexation of the REEs with carbonate ions appears to control their aqueous distributions whereas in acid waters, the REE signature of the labile fraction of the REEs is readily leached from the rocks. In circumneutral pH waters, both processes appear to be important and their relative significance has not yet been determined. | KEVIN, H.
