| 1 | Diversification of Hawaiian Cyrtandra (Gesneriaceae) under the influence of incomplete lineage sorting and hybridization显示文摘Cyrtandra(Gesneriaceae)is a genus of flowering plants with over 800 species distributed throughout Southeast Asia and the Pacific Islands.On the Hawaiian Islands,60 named species and over 89 putative hybrids exist,most of which are identified on the basis of morphology.Despite many previous studies on the Hawaiian Cyrtandra lineage,questions regarding the reconciliation of morphology and genetics remain,many of which can be attributed to the relatively young age and evidence of hybridization between species.We utilized targeted enrichment,high-throughput sequencing,and modern phylogenomics tools to test 31 Hawaiian Cyrtandra samples(22 species,two putative hybrids,four species with two samples each,one species with four samples)and two outgroups for species relationships and hybridization in the presence of incomplete lineage sorting(ILS).Both concatenated and species-tree methods were used to reconstruct species relationships,and network analyses were conducted to test for hybridization.We expected to see high levels of ILS and putative hybrids intermediate to their parent species.Phylogenies reconstructed from the concatenated and species-tree methods were highly incongruent,most likely due to high levels of incomplete lineage sorting.Network analyses inferred gene flow within this lineage,but not always between taxa that we expected.Multiple hybridizations were inferred,but many were on deeper branches of the island lineages suggesting a long history of hybridization.We demonstrated the utility of high-throughput sequencing and a phylogenomic approach using 569 loci to understanding species relationships and gene flow in the presence of ILS. | Joseph A.Kleinkopf Wade R.Roberts Warren L.Wagner Eric H.Roalson | 2019 | Journal of Systematics and Evolution2019，57，6： | 5 |
| 2 | A New Prognostic Model Covering All Stages of Intrahepatic Cholangiocarcinoma显示文摘Background and Aims:Intrahepatic cholangiocarcinoma(ICC)is the second most common primary hepatic malignancy that causes a poor survival.We aimed to identify its prognostic factors and to develop a nomogram that will predict survival of ICC patients among all stages.Methods:A total of 442 patients with pathology-proven ICC registered at the Fifth Medical Center of PLA General Hospital between July 2007 and December 2019 were enrolled.Subjects were followed for survival status until June 30,2020.A prognostic model visualized as a nomogram was constructed in the training cohort using multivariate cox model,and was then validated in the validation cohort.Results:The median age was 55 years.With a median follow-up of 50.4 months,337 patients died.The median survival was 11.6 months,with 1-,3-and 5-year survival rates of 48.3%,22.7%and 16.2%,respectively.Factors associated with overall survival were multiple tumors,lymph node involvement,vascular invasion,distant metastasis,decreased albumin,elevated lactate dehydrogenase(LDH),decreased iron,elevated fi-brinogen,elevated CA125 and elevated CA19-9.A nomo-gram predicting survival of ICC patients at the time of di-agnosis achieved a Harrel’s c-statistic of 0.758,significantly higher than the 0.582 of the TNM stage alone.Predicted median survivals of those within the low,mid and high-risk subgroups were 35.6,12.1 and 6.2 months,respectively.Conclusions:A nomogram based on imaging data and serum biomarkers at diagnosis showed good ability to predict survival in patients with all stages of ICC.Further studies are needed to validate the prognostic capability of our new model. | Shuang-Nan Zhou Shan-Shan Lu Da-Wei Ju Ling-Xiang Yu Xiao-Xiao Liang Xiao Xiang Suthat Liangpunsakul Lewis R.Roberts Yin-Ying Lu Ning Zhang | 2022 | Journal of Clinical and Translational Hepatology2022，10，2： | 2 |
