1Potential antioxidant bioactive peptides from camel milk proteins显示文摘Camel milk is traditionally considered to have medicinal characteristics that it has potential health benefits and could help to treat several illnesses. Particularly, it is closest to human breast milk and has high levels of nutrients and bioactive components. The aim of this study was to explore the antioxidant peptides derived from protein fractions of camel milk. Camel milk proteins(CMP) were fractionated into camel casein protein(CCP) and camel whey protein(CWP), which were hydrolyzed with pepsin to produce peptic digests P-CCP and P-CWP, respectively. RP-HPLC was used for fractionation of the peptides from the P-CCP and P-CWP. The antioxidant activities were evaluated using superoxide anion generating system of xanthine oxidase(XOD)and 2,2-diphenyl-1-picrylhydrazyl(DPPH) scavenging assay. Active peptides were analyzed using matrixassisted laser desorption ionization time of flight mass spectrometry(MALDI-TOF-MS) whereas a number of antioxidant peptides, with masses ranging from 913 to 2,951 Da, derived mainly from alpha-casein, lactophorin and lactoferrin, were identified. When yeast cells are used as a system for modeling mitochondrial disease, the peptides in caseins and whey fractions significantly enhanced the tolerance of yeast cells against peroxide-induced oxidative stress. 2Changing paradigms in lower extremity reconstruction in war-related injuries显示文摘Background: Ballistic high-energy trauma has substantially increased the severity of non-fatal extremity injuries incurred in modern warfare. Expedient medical care, refinement in surgical techniques, and soft tissue coverage have brought about a paradigm shift in the management of lower extremity wounds during the last decade with an increased emphasis on limb salvage.Methods: A literature-based study was conducted to analyze reconstructive modalities based on the location, depth, and severity of wounds, as well as mechanism of injury, concomitant vascular injuries and open fractures, choice of flap, timing of definitive reconstruction, and complications.Results: Extremity injuries account for over 60% of injuries in the recent conflicts in Iraq and Afghanistan, with the majority secondary to explosive devices. The severity of these injuries is profound compared with civilian registries, and conventional injury scoring systems have failed to accurately predict outcomes in combat trauma. The mainstay of treatment is serial debridement, negative pressure therapy, fracture stabilization, and treatment of concomitant injuries by the forward medical teams with subsequent definitive reconstruction after transport to an advanced military treatment facility. Autologous reconstruction with free tissue transfer and pedicled flaps remains the primary modality for soft tissue coverage in limb salvage. Adjunct innovative modalities, such as external tissue expansion, dermal substitutes, and regenerative matrices, have also been successfully utilized for limb salvage.Conclusion: Lower extremity injuries account for the vast majority of injuries in modern warzones. Explosive devices represent the most common mechanism of injury, with blast impact leading to extensive soft tissue injuries necessitating complex reconstructive strategies. 3Potent antioxidant peptides derived from honey major protein enhance tolerance of eukaryotic cells toward oxidative stress显示文摘Honey is known for its medicinal benefits and receiving renewed attention as natural medicine.Studies on health benefits of honey attributed its antioxidant activity to phenolic compounds,but the contribution of proteins and peptides to the antioxidant activity of honey is lacking.The aim of this study was to explore the contribution of proteins and peptides to the antioxidant activity of honey,which remained obscure for decades.Total honey proteins(THP)were isolated by dialysis method and hydrolyzed through simulated gastrointestinal digestion.The hydrolysates were fractionated using size-exclusion chromatography.The antioxidant activity was determined by using superoxide radical-scavenging,DPPH reduction and intracellular ROS assays.THP was shown to exhibit superoxide-scavenging activity but its pepsin-hydrolysate(HP-p)showed superior scavenging activity.The HP-p produced five peptide fractions(P1~P5)when fractionated on Sephacryl S-100 size-exclusion column.The five fractions showed superoxide-scavenging activities and DPPH reducing activities,whereas the slow-eluting peptide fractions(P3 and P4)were the most potent.MALDI-TOF/MS analysis identified a pentapeptide(TSNTF)as the dominant peptide in the active fractions P3 and P4.Human colonic epithelial cells treated with P3 and P4 peptides exhibited lower intracellular ROS,when oxidative stress was induced by H2O2 or diethyl maleate(DEM),indicating strong tolerance to oxidative stress.The viabilities of human cells or yeast cells were largely decreases under oxidative stress,but treated cells with P3 and P4 showed higher viability compared with the untreated cells.The results are the first to describe a novel antioxidant peptide from honey that confer ex vivo anti-oxidative function within a complicated milieu of eukaryotic cells and pave the way for its potential as nutraceutical or therapeutic peptide for risk-reduction of oxidative-stress and related diseases.Hisham R.Ibrahim Fukiko Nanbu Takeshi Miyata 2021Food Production, Processing and Nutrition2021,3,1:0
4Numerical analysis of hemodynamic parameters in stenosed arteries under pulsatile flow conditions显示文摘This research studies the changes in flow patterns and hemodynamic parameters of diverse shapes and sizes of stenosis.Six different shapes and sizes of stenosis are constructed to investigate the variations in hemodynamics as the morphology changes.Changes in shape(trapezoidal and bell-shaped)and sizes of stenosis change the stresses on the walls and their flow patterns.TAWSS and OSI results specify that trapezoidal stenosis exerts greater stress than bell-shaped stenosis.Also,as the length of the trapezoidal stenosis increases,the TAWSS increases,whereas the trend is the opposite for bell-shaped stenosis.Later,this paper also studies different degrees of stenosis extracted from real images.Changes in velocity flow patterns,wall shear stress(WSS),Time-averaged wall shear stress(TAWSS)and Oscillatory shear index(OSI)have been studied for these images.Results illustrate that the peak velocity rises drastically as the stenosis percentage increases.Negative velocity is seen close to the artery's walls,indicating flow separation.This flow separation region is seen throughout the cycle except in the accelerating flow region.An increase in stenosis also increases WSS and TAWSS drastically.Negative WSS is seen downstream of stenosis,indicating flow recirculation.Such negative WSS in the blood vessels also promotes endothelial dysfunction.OSI values greater than 0.2 are seen near the stenosis region,indicating atherosclerosis growth.Regions of high OSI and low TAWSS are also identified,indicating probable regions of plaque development.Priyambada Praharaj Chandrakant Sonawane Anand Pandey Vikas Kumar Arundhati Warke Hitesh Panchal R.Ibrahim Chander Prakash 2023Medicine in Novel Technology and Devices2023,,4:0

6Neuroprotective potential of Indigofera oblongifolia leaf methanolic extract against lead acetate-induced neurotoxicity显示文摘Lead(Pb) is one of the most common environmental toxicants,exposure to which can cause significant neurotoxicity and an associated decline in brain function.This study investigated the possible neuroprotective role of Indigofera oblongifolia leaf methanolic extract(IOLME) against lead-induced neurotoxicity.Rats were intraperitoneally injected with lead acetate,with or without IOLME(intragastric administration for 5 days),and the neuroprotective effect of IOLME was assessed by measuring the lead concentration,redox status(lipid peroxidation,nitric oxide and glutathione),enzymatic antioxidant activities(superoxide dismutase,catalase,glutathione peroxidase and reductase),PCR assays of apoptosis markers(Bax and Bcl-2)and histopathology of the brain.The increases in the lipid peroxidation,nitric oxide,and apoptosis,the decreases in the glutathione level and the activity of antioxidant enzymes,and the altered histology of the brain induced by lead acetate were mitigated in the brain of rats pre-treated with IOLME.These findings indicate that IOLME has beneficial effects and it mitigates lead acetate-induced neurotoxicity via its antioxidant and anti-apoptotic activities.Saleh Al-Quraishy Mohamed A.Dkhil Shaimaa R.Ibrahim Ahmed E.Abdel Moneim 2016Neural Regeneration Research2016,11,11:0
7ActivityNET:Neural networks to predict public transport trip purposes from individual smart card data and POIs显示文摘Predicting trip purpose from comprehensive and continuous smart card data is beneficial for transport and city planners in investigating travel behaviors and urban mobility.Here,we propose a framework,ActivityNET,using Machine Learning(ML)algorithms to predict passengers’trip purpose from Smart Card(SC)data and Points-of-Interest(POIs)data.The feasibility of the framework is demonstrated in two phases.Phase I focuses on extracting activities from individuals’daily travel patterns from smart card data and combining them with POIs using the proposed“activity-POIs consolidation algorithm”.Phase II feeds the extracted features into an Artificial Neural Network(ANN)with multiple scenarios and predicts trip purpose under primary activities(home and work)and secondary activities(entertainment,eating,shopping,child drop-offs/pick-ups and part-time work)with high accuracy.As a case study,the proposed ActivityNET framework is applied in Greater London and illustrates a robust competence to predict trip purpose.The promising outcomes demonstrate that the cost-effective framework offers high predictive accuracy and valuable insights into transport planning.Nilufer Sari Aslam Mohamed R.Ibrahim Tao Cheng Huanfa Chen Yang Zhang 2021Geo-Spatial Information Science2021,24,4:0
