| 1 | Hydrogen isotope composition of natural gases from the Tarim Basin and its indication of depositional environments of the source rocks显示文摘By measuring carbon and hydrogen isotope compositions for C1, C2 and C3 of 74 gas samples, natural gases from the Tarim Basin can be divided into six groups on the basis of their origins: (1) coal-type gas derived from coal measures; (2) coal-type gas generated from the T-J lacustrine mudstones; (3) oil-type gas derived from the Cambrian and low Ordovician marine source rocks; (4) oil-type gas from the source rocks deposited in the marine-transitional facies; (5) mixing gas between gas derived from the Carboniferous transitional source rocks and the Mesozoic humic gas, and (6) mixing gases of thermal genetic gas and little deep gas in the Southwest depression of the Tarim Basin. The δ D values of methane in natural gases originating from different type kerogens are affected by both palaeo-environments of the source rock formation (kerogen types) and thermal maturity, with sedimentary environment (kerogen type) as the main controlling factor. Under the similar thermal maturity, the hydrogen isotope composition of methane is more enriched in deuterium in marine environments than lacustrine one. With the increase of thermal maturity and the increase of carbon atomic numbers of gaseous alkanes, the hydrogen isotopes become enriched in deuterium. The δ D values of ethane and propane (δ D2, δ D3) are controlled mainly by thermal maturity and to a lesser degree by sedimentary environment of the source rock formation. The partial reversal of hydrogen isotopes for gaseous alkanes would be related to the microbial oxidation, mixing of sapropelic and humic gases and / or mixing of gases from similar kerogen sources with various thermal maturities. | 2 | Effects of Deep Fluids on Hydrocarbon Generation and Accumulation in Chinese Petroliferous Basins显示文摘Deep fluids in a petroliferous basin generally come from the deep crust or mantle beneath the basin basement, and they transport deep substances(gases and aqueous solutions) as well as heat to sedimentary strata through deep faults. These deep fluids not only lead to large-scale accumulations of CO_2, CH_4, H_2, He and other gases, but also significantly impact hydrocarbon generation and accumulation through organic-inorganic interactions. With the development of deep faults and magmatic-volcanic activities in different periods, most Chinese petroliferous basins have experienced strong impacts associated with deep fluid activity. In the Songliao, Bohai Bay, Northern Jiangsu, Sanshui, Yinggehai and Pearl Mouth Basins in China, a series of CO_2 reservoirs have been discovered. The CO_2 content is up to 99%, with δ^(13)C_(CO2) values ranging from-4.1‰ to-0.37‰ and ~3He/~4He ratios of up to 5.5 Ra. The abiogenic hydrocarbon gas reservoirs with commercial reserves, such as the Changde, Wanjinta, Zhaozhou, and Chaoyanggou reservoirs, are mainly distributed in the Xujiaweizi faulted depression of the Songliao Basin. The δ^(13)CCH4 values of the abiogenic alkane gases are generally >-30‰ and exhibit an inverse carbon isotope sequence of δ^(13)C_(CH4)>δ^(13)C_(C2H6)>δ^(13)C_(C3H8)>δ^(13)C_(C4H10). According to laboratory experiments, introducing external H_2 can improve the rate of hydrocarbon generation by up to 147% through the kerogen hydrogenation process. During the migration from deep to shallow depth, CO_2 can significantly alter reservoir rocks. In clastic reservoirs, feldspar is easily altered by CO_2-rich fluids, leading to the formation of dawsonite, a typical mineral in high CO_2 partial pressure environments, as well as the creation of secondary porosity. In carbonate reservoirs, CO_2-rich fluids predominately cause dissolution or precipitation of carbonate minerals. The minerals, e.g., calcite and dolomite, show some typical features, such as higher homogenization temperatures than the burial temperature, relatively high concentrations of Fe and Mn, positive Eu anomalies, depletion of 18 O and enrichment of radiogenic ^(87)Sr. Due to CO_2-rich fluids, the development of high-quality carbonate reservoirs is extended to deep strata. For example, the Well TS1 in the northern Tarim Basin revealed a high-quality Cambrian dolomite reservoir with a porosity of 9.1% at 8408 m, and the Well ZS1 C in the central Tarim Basin revealed a large petroleum reserve in a Cambrian dolomite reservoir at ~6900 m. During the upward migration from deep to shallow basin strata, large volumes of supercritical CO_2 may extract petroleum components from hydrocarbon source rocks or deep reservoirs and facilitate their migration to shallow reservoirs, where the petroleum accumulates with the CO_2. Many reservoirs containing both supercritical CO_2 and petroleum have been discovered in the Songliao, Bohaiwan, Northern Jiangsu, Pearl River Mouth and Yinggehai Basins. | 3 | Presence of carboxylate salts in marine carbonate strata of the Ordos Basin and their impact on hydrocarbon generation evaluation of low TOC, high maturity source rocks显示文摘The total organic carbon(TOC)in the marine source rock of the Ordos Basin mostly ranges from 0.2%to 0.5%.The industrial standard commonly states that the TOC value has to be no less than 0.5%(0.4%for high mature or over-mature source rock)to form large petroleum reservoirs.However,gas source correlation indicates that the natural gas in the Jingbian gas field does receive contribution from marine source rocks.In order to determine the effect of carboxylate salts(or called as organic acid salts)on TOC in highly mature source rocks with low TOC value,we sampled the Ordovician marine source rock and the Permian transitional facies source rock in one drilled well in the southern Ordos Basin and performed infrared and GC-MS analysis.It is found that both kerogen-derived organic acids and carboxylate salt-conversed organic acids exist in both marine and transitional facies source rocks.The carboxylate salt-conversed organic acids mainly come from the complete acidification of carboxylate salts,which confirms the presence of carboxylate salts in the marine source rocks.Although the C16:O peak is the main peak for the organic acids both before and after acidification,the carboxylate salt-conversed organic acids have much less relative abundance ahead of C16:O compared with that of the kerogen-based and free organic acids.This observation suggests that the kerogen-based and free organic acids mainly decarboxylate to form lower carboxylic acids,whereas the carboxylate salt-conversed organic acids mainly break down into paraffins.By using calcium hexadecanoate as the reference to quantify the kerogen-derived and carboxylate salt-conversed organic acids,the high TOC(>2.0%)marine source rocks have low carboxylate salt content and the low TOC(0.2%–0.5%)marine source rocks contain high content of carboxylate salt.Therefore,for the marine source rocks with 0.2%–0.5%TOC,the carboxylate salts may be a potential gas source at high maturity stage. | LIU QuanYou JIN ZhiJun LIU WenHui LU LongFei MENG QianXiang TAO Ye HAN PinLong | 2013 | Science China Earth Sciences2013,56,12: | 9 |
| 4 | Geochemical Characteristics and Genetic Types of Natural Gas in the Xinchang Gas Field, Sichuan Basin, SW China显示文摘The molecular compositions and stable carbon and hydrogen isotopic compositions of natural gas from the Xinchang gas field in the Sichuan Basin were investigated to determine the genetic types. The natural gas is mainly composed of methane (88.99%–98.01%), and the dryness coefficient varies between 0.908 and 0.997. The gas generally displays positive alkane carbon and hydrogen isotopic series. The geochemical characteristics and gas-source correlation indicate that the gases stored in the 5^(th) member of the Upper Triassic Xujiahe Formation are coal-type gases which are derived from source rocks in the stratum itself. The gases reservoired in the 4^(th) member of the Xujiahe Formation and Jurassic strata in the Xinchang gas field are also coal-type gases that are derived from source rocks in the 3^(rd) and 4^(th) members of the Xujiahe Formation. The gases reservoired in the 2^(nd) member of the Upper Triassic Xujiahe Formation are mainly coal-type gases with small amounts of oil-type gas that is derived from source rocks in the stratum itself. This is accompanied by a small amount of contribution brought by source rocks in the Upper Triassic Ma'antang and Xiaotangzi formations. | 6 | The scientific connotation of oil and gas formations under deep fluids and organic-inorganic interaction显示文摘As a relatively stable craton block in the earth system, the petroliferous basin is influenced by the evolution of the earth system from the early development environment of source rocks, hydrocarbon formation, and reservoir dissolution to hydrocarbon accumulation or destruction. As a link between the internal and external factors of the basin, deep fluids run through the whole process of hydrocarbon formation and accumulation through organic-inorganic interaction. The nutrients carried by deep fluids promote the bloom of hydrocarbon-generating organisms and extra addition of carbon and hydrogen source, which are beneficial to the development of high-quality source rock and enhancement of the hydrocarbon generation potential. The energy carried by the deep fluid promotes the early maturation of the source rock and facilitates the hydrocarbon generation by activation and hydrogenation in high-mature hydrocarbon sources. The dissolution alteration of carbonate rocks and clastic reservoirs by CO_2-rich deep fluids improves the deep reservoir space, thus extending the oil and gas reservoir space into greater depth. The extraction of deeply retained crude oil by deep supercritical CO_2 and the displacement of CH_4 in shale have both improved the hydrocarbon fluidity in deep and tight reservoirs. Simultaneously, the energy and material carried by deep fluids(C, H, and catalytic substances) not only induce inorganic CH_4 formation by Fischer-Tropsch(F-T) synthesis and 'hydrothermal petroleum' generation from organic matter by thermal activity but also cause the hydrothermal alteration of crude oil from organic sources. | 7 | New Method for Monitoring Tire Pressure of Cars Based on the Tire Radial Deformation显示文摘Monitoring tire pressure of cars and signaling abnormal conditions is an important means to prevent deadly accidents.Large achievements have been gained,especially in direct tire pressure monitoring system(TPMS).But there has been rarely research on indirect TPMS in the world.In China,the research on indirect TPMS is almost lacking.The international research on the indirect monitoring tire pressure method is mainly based on measuring and comparing the rotating speed of wheels.But it is very difficult to measure wheel rotating speed accurately because of the influence of many random factors.In this paper,the authors propose a new method in which the tire pressure can be monitored indirectly.This method can be used for tire calibration,wheel speed frequency standardization,wheel speed frequency comparison,and abnormal tire pressure determination.The pulse frequencies from wheel speed sensors of ABS are used to indicate tire deformation.Because the frequency has a relationship with tire deformation,the tire deformation reflects the tire pressure.Small sample statistics is used in the new method to increase the accuracy,and the experimental samples using the principle of the new method have been made and tested.The result of vehicle tests on road demonstrates that the method is efficient and accurate to monitor tire pressure.The research has positive potential for developing products. | HAN Zongqi WANG Liqiang LIU Quanyou JU Xuekun | 2010 | Chinese Journal of Mechanical Engineering2010,23,2: | 7 |
| 8 | Potential petroleum sources and exploration directions around the Manjar Sag in the Tarim Basin显示文摘Since the discovery of the Tahe oilfield,it has been controversial on whether the main source rock is in the Cambrian or Middle-Upper Ordovician strata.In this paper,it is assumed that the crude oil from the Wells YM 2 and TD 2 was derived from the Middle-Upper Ordovician and Cambrian source rocks,respectively.We analyzed the biomarkers of the crude oil,asphalt-adsorbed hydrocarbon and saturated hydrocarbon in bitumen inclusions from the Lunnan and Hade areas in the North Uplift of the Tarim Basin.Results show that the ratios of tricyclic terpane C_(21)/C_(23)in the crude oil,asphalt-adsorbed hydrocarbon and saturated hydrocarbon in bitumen inclusions are less than 1.0,indicating that they might be from Upper Ordovician source rocks;the ratios of C_(28)/(C_(27)+C_(28)+C_(29))steranes in the saturated hydrocarbon from reservoir bitumen and bitumen inclusions are higher than 25,suggesting that they might come from the Cambrian source rocks,however,the ratios of C_(28)/(C_(27)+C_(28)+C_(29))steranes in oil from the North Uplift are less than 25,suggesting that they might be sourced from the Upper Ordovician source rocks.These findings demonstrate that the sources of crude oil in the Tarim Basin are complicated.The chemical composition and carbon isotopes of Ordovician reservoired oil in the Tarim Basin indicated that the crude oil in the North Uplift(including the Tahe oilfield)and Tazhong Depression was within mixture areas of crude oil from the Wells YM 2 and TD 2 as the end members of the Cambrian and Middle-Upper Ordovician sourced oils,respectively.This observation suggests that the crude oil in the Ordovician strata is a mixture of oils from the Cambrian and Ordovician source rocks,with increasing contribution from the Cambrian source rocks from the southern slope of the North Uplift to northern slope of the Central Uplift of the Tarim Basin.Considering the lithology and sedimentary facies data,the spatial distribution of the Cambrian,Middle-Lower Ordovician and Upper Ordovician source rocks was reconstructed on the basis of seismic reflection characteristics,and high-quality source rocks were revealed to be mainly located in the slope belt of the basin and were longitudinally developed over the maximum flooding surface during the progressive-regressive cycle.Affected by the transformation of the tectonic framework in the basin,the overlays of source rocks in different regions are different and the distribution of oil and gas was determined by the initial basin sedimentary structure and later reformation process.The northern slope of the Central Uplift-Shuntuo-Gucheng areas would be a recent important target for oil and gas exploration,since they have been near the slope area for a long time. | JIN ZhiJun LIU QuanYou YUN JinBiao Tenger | 2017 | Science China Earth Sciences2017,60,2: | 7 |
| 9 | Geochemical characteristics and origin of gases from the Upper,Lower Paleozoic and the Mesozoic reservoirs in the Ordos Basin,China显示文摘The Ordos Basin,the second largest sedimentary basin in China,contains the broad distribution of natural gas types.So far,several giant gas fields have been discovered in the Upper and Lower Paleozoic in this basin,each having over 1000×10 8 m 3 of proven gas reserves,and several gas pools have also been discovered in the Mesozoic.This paper collected the data of natural gases and elucidated the geochemical characteristics of gases from different reservoirs,and then discussed their origin.For hydrocarbons preserved in the Upper Paleozoic,the elevatedδ13C values of methane,ethane and propane indicate that the gases would be mainly coal-formed gases;the singular reversal in the stable carbon isotopes of gaseous alkanes suggests the mixed gases from humic sources with different maturity.In the Lower Paleozoic,theδ13C 1 values are mostly similar with those in the Upper Paleozoic,but theδ13C 2 andδ13C 3 values are slightly lighter,suggesting that the gases would be mixing of coal-type gases as a main member and oil-type gases.There are multiple reversals in carbon isotopes for gaseous alkanes,especially abnormal reversal for methane and ethane(i.e.δ13C 1 >δ13C 2 ),inferring that gases would be mixed between high-mature coal-formed gases and oil-type gases.In the Mesozoic,the δ13Cvalues for gaseous alkanes are enriched in 12C,indicating that the gases are mainly derived from sapropelic sources;the carbon isotopic reversal for propane and butane in the Mesozoic is caused by microbial oxidation and mixing of gases from sapropelic sources with different maturity.In contrast to the Upper Paleozoic gases,the Mesozoic gases are characterized by heavier carbon isotopes of iso-butane than normal butane,which may be caused by gases generated from different kerogen types. | 10 | Controls of Late Jurassic-Early Cretaceous tectonic event on source rocks and seals in marine sequences,South China显示文摘Thermal evolution of source rocks and dynamic sealing evolution of cap rocks are both subjected to tectonic evolution.The marine sequences in South China have experienced superposed structural deformation from multiple tectonic events.To investigate the effectiveness of preservation conditions,it is of great importance to understand the controls of key tectonic events on the dynamic evolution of cap rocks.This paper discusses the controls of Late Jurassic-Early Cretaceous(J3-K1) tectonic event on source and cap rocks in marine sequences in South China based on the relationships between J3-K1 tectonic event and the burial history types of the marine sequences,the hydrocarbon generation processes of marine source rocks,the sealing evolution of cap rocks,the preservation of hydrocarbon accumulations,and the destruction of paleo-oil pools.The study has the following findings.In the continuously subsiding and deeply buried areas during the J3-K1 period,marine source rocks had been generating hydrocarbons for over a long period of time and hydrocarbon generation ended relatively late.At the same time,the sealing capacity of the overburden cap rocks had been constantly strengthened so that hydrocarbons could be preserved.In the areas which suffered compressional deformation,folding and thrusting,uplifting and denudation in J3-K1,the burial history was characterized by an early uplifting and the hydrocarbon generation by marine source rocks ended(or suspended) during the J3-K1 period.The sealing capacity of the cap rocks was weakened or even vanished.Thus the conditions for preserving the hydrocarbon accumulations were destroyed.The continuously subsiding and deeply buried areas during the J3-K1 period are the strategic precincts of the petroleum exploration in marine sequences in South China. | JIN ZhiJun YUAN YuSong LIU QuanYou WO YuJin | 2013 | Science China Earth Sciences2013,56,2: | 6 |
| 11 | Pore Size Distribution of a Tight Sandstone Reservoir and its Effect on Micro Pore-throat Structure: A Case Study of the Chang 7 Member of the Xin’anbian Block, Ordos Basin, China显示文摘Pore distribution and micro pore-throat structure characteristics are significant for tight oil reservoir evaluation, but their relationship remains unclear. This paper selects the tight sandstone reservoir of the Chang 7 member of the Xin’anbian Block in the Ordos Basin as the research object and analyzes the pore size distribution and micro pore-throat structure using field emission scanning electron microscopy(FE-SEM), high-pressure mercury injection(HPMI), highpressure mercury injection, and nuclear magnetic resonance(NMR) analyses. The study finds that:(1) Based on the pore size distribution, the tight sandstone reservoir is characterized by three main patterns with different peak amplitudes. The former peak corresponds to the nanopore scale, and the latter peak corresponds to the micropore scale. Then, the tight sandstone reservoir is categorized into three types: type 1 reservoir contains more nanopores with a nanopore-to-micropore volume ratio of 82:18;type 2 reservoir has a nanopore-to-micropore volume ratio of 47:53;and type 3 reservoir contains more micropores with a nanopore-to-micropore volume ratio of 35:65.(2) Affected by the pore size distribution, the throat radius distributions of different reservoir types are notably offset. The type 1 reservoir throat radius distribution curve is weakly unimodal, with a relatively dispersed distribution and peak ranging from 0.01 μm to 0.025 μm. The type 2 reservoir’s throat radius distribution curve is single-peaked with a wide distribution range and peak from 0.1 μm to 0.25 μm. The type 3 reservoir’s throat radius distribution curve is single-peaked with a relatively narrow distribution and peak from 0.1 μm to 0.25 μm. With increasing micropore volume, pore-throat structure characteristics gradually improve.(3) The correlation between micropore permeability and porosity exceeds that of nanopores, indicating that the development of micropores notably influences the seepage capacity. In the type 1 reservoir, only the mean radius and effective porosity have suitable correlations with the nanopore and micropore porosities. The pore-throat structure parameters of the type 2 and 3 reservoirs have reasonable correlations with the nanopore and micropore porosities, indicating that the development of these types of reservoirs is affected by the pore size distribution. This study is of great significance for evaluating lacustrine tight sandstone reservoirs in China. | 13 | Mercury concentration in natural gas and its distribution in the Tarim Basin显示文摘Here I collected natural gas samples from 41 industrial gas wells in the Tarim Basin,and studied the mercury distribution in the area.My data show that there is certain regularity in the distribution of mercury in the Tarim Basin.Generally,the mercury concentration is high at the edge of the basin and low in the central basin.The highest mercury concentration occurs in the Southwest Depression,ranging from 15428.5 to 296763.0ng/m3 with an average of 156095.7ng/m3,followed by the Kuqa Depression ranging from 15.0 to 56964.3ng/m3 with an average of 11793.7ng/m3,and the Hade oil and gas field in the North Depression has the lowest mercury concentration ranging from 17.7 to 3339.5ng/m3 with an average of 1678.6ng/m3.The mercury concentrations in the natural gases of different structural units are various,with the highest mercury concentration in the zone of strong structural activity of Southwest Depression.It is profitable of Hg accumulation in the self sourced and self accumulated gas reservoirs or volcanic existence;in contrast,the low Hg concentration exists in the secondary gas reservoir.The mercury concentration in the natural gas generated under continental depositional environment is higher than that in marine gas.Therefore,the mercury concentration in the natural gases is constrained by gas genesis,depositional environment of source rocks,tectonic activity,and volcanic activity,and the tectonic activity is the main factor for the mercury concentration in the natural gas,followed by volcanic activity and depositional environments. | LIU QuanYou | 2013 | Science China Earth Sciences2013,56,8: | 3 |
| 14 | The extraction of potassium from K-feldspar ore by low temperature molten salt method显示文摘The low temperature molten salt method was used to extract potassium from K-feldspar ore, and some related factors including mass ratio between NaNO_3, NaOH, H_2O and K-feldspar ore, particle size of K-feldspar ore,reaction temperature and time were investigated, respectively. In addition, the optimum condition for this method was determined by a series of condition experiments. What was more, the K-feldspar ore and the leach residue after reaction based on the above optimum condition were analyzed by XRD, SEM and EDS,separately. | 15 | Cgr1,a ripe rot resistance QTL in Vitis amurensis‘Shuang Hong’grapevine显示文摘Ripe rot is a serious grapevine disease in Vitis L.and Muscadinia(Planch.)Small.However,resistance to this disease has been reported in some oriental Vitis species.To identify resistance-related Quantitative Trait Loci(QTLs)from the Chinese grape species V.amurensis,an F1 population of V.vinifera‘Cabernet Sauvignon’×V.amurensis‘Shuang Hong’was used to map the ripe rot resistance loci expected in‘Shuang Hong’grape.A total of 7598 single nucleotide polymorphisms(SNPs)between the parents were identified in our previous study,and 934 SNPs were selected for genetic map construction.These SNPs are distributed across the 19 chromosomes covering a total of 1665.31 cM in length,with an average of 1.81 cM between markers.Ripe rot resistance phenotypes among the hybrids were evaluated in vitro using excised leaves for three consecutive years from 2016 to 2018;a continuous variation was found among the F1 hybrids,and the Pearson correlation coefficients of the phenotypes scored in all three years were significant at the 0.01 level.Notably,the first QTL reported for resistance to grape ripe rot disease,named Cgr1,was identified on chromosome 14 of‘Shuang Hong’grapevine.Cgr1 could explain up to 19.90%of the phenotypic variance.In addition,a SNP named‘np19345’was identified as a molecular marker closely linked to the peak of Cgr1 and has the potential to be developed as a marker for the Cgr1 resistance haplotype. | Peining Fu Quanyou Tian Gongti Lai Rongfang Li Shiren Song Jiang Lu | 2019 | Horticulture Research2019,6,1: | 3 |

| 18 | Organic-rich formation and hydrocarbon enrichment of lacustrine shale strata:A case study of Chang 7 Member显示文摘Lacustrine shale is an important target for the exploration of unconventional oil and gas in China beyond marine shale gas.However,the formation environment of lacustrine shale differs from that of marine shale,resulting in a different reservoir composition,organic matter,oil and gas content,and hydrocarbon mobility.In this study,the Chang 7 shale of the Yanchang Formation in the Ordos Basin was used to analyze the effect of volcanic activity on the paleoproductivity and preservation conditions during the formation of lacustrine shale.The results show that algae and bacteria were developed before the eruption.After the eruption,the number of bacteria declined,but the increased prosperity of algae reflects that the volcanic activity enhanced ancient productivity.The sulfate generated by volcanic activity promotes bacterial sulfate reduction,and the produced H_(2)S leads to a strong reducing environment in the waterbody,which is conducive to the preservation of organic matter.Organic geochemical analysis shows that the black shale in the shale strata has a high total organic carbon(TOC)content and strong hydrocarbon generation potential,whereas the tuff has a low TOC content and can scarcely generate hydrocarbons,indicating that the tuff deposited by volcanic activity cannot be considered as effective source rock.In terms of storage space,shale is mainly laminar and dispersed,and it includes organic and inorganic pores.The development of organic pores is affected by thermal maturity,whereas inorganic pores mainly occur between detrital particles and crystals.Tuff is mainly supported by heterogeneous matrix and associated with alteration.Its pores include inter-and intragranular mineral pores.The development of tight sandstone pores is affected by compaction,cementation,and dissolution,which mainly consist of intra-and intergranular pores.The Chang 7 lacustrine shale generally contains oil,but different lithologies have different oil drainage efficiencies.Sandstone and shale exhibit the best and worst oil drainage efficiency,respectively.It is mainly affected by the pore size distribution,fluid properties,and rock wettability.Therefore,the development of shale oil should mainly focus on lacustrine shale formations with interbeds.The mutual dissolution of organic matter and hydrocarbons in the shale section leads to the poor mobility and difficult development of hydrocarbons. | Quanyou LIU Peng LI Zhijun JIN Yuewu SUN Guang HU Dongya ZHU Zhenkai HUANG Xinping LIANG Rui ZHANG Jiayi LIU | 2022 | Science China Earth Sciences2022,65,1: | 2 |

| 20 | Substrate stiffness-dependent regulatory volume decrease and calcium signaling in chondrocytes显示文摘The pericellular matrix stiffness is strongly associated with its biochemical and structural changes during the aging and osteoarthritis progress of articular cartilage.However,how substrate stiffness modulates the chondrocyte regulatory volume decrease(RVD)and calcium signaling in chondrocytes remains unknown.This study aims to investigate the effects of substrate stiffness on the chondrocyte RVD and calcium signaling by recapitulating the physiologically relevant substrate stiffness.Our results showed that substrate stiffness induces completely different dynamical deformations between the cell swelling and recovering progresses.Chondrocytes swell faster on the soft substrate but recovers slower than the stiff substrate during the RVD response induced by the hypo-osmotic challenge.We found that stiff substrate enhances the cytosolic Ca^(2+)oscillation of chondrocytes in the iso-osmotic medium.Furthermore,chondrocytes exhibit a distinctive cytosolic Ca^(2+)oscillation during the RVD response.Soft substrate significantly improves the Ca^(2+)oscillation in the cell swelling process whereas stiff substrate enhances the cytosolic Ca^(2+)oscillation in the cell recovering process.Our work also suggests that the TRPV4 channel is involved in the chondrocyte sensing substrate stiffness by mediating Ca^(2+)signaling in a stiffness-dependent manner.This helps to understand a previously unidentified relationship between substrate stiffness and RVD response under the hypo-osmotic challenge.A better understanding of substrate stiffness regulating chondrocyte volume and calcium signaling will aid the development of novel cell-instructive biomaterial to restore cellular functions. | Min Zhang Xiaoan Wu Genlai Du Weiyi Chen Quanyou Zhang | 2022 | Acta Biochimica et Biophysica Sinica2022,54,1: | 2 |
