| 1 | Successful management of postoperative recurrence of hepatocellular carcinoma with p53 gene therapy combining transcatheter arterial chemoembolization显示文摘Transcatheter arterial chemoembolization (TACE) has become the standard treatment for unresectable hepatocellular carcinoma (HCC). But this method has some shortages. p53 gene, which was found to be mutant in many human tumors, has been proved with broadspectrum anti-tumor effects. We reported a 23-year-old patient with recurrent HCC after irregular hepatectomy. The p53 gene was applied to this patient. We injected percutaneously and infused transcatheterally p53 gene (Gendicine, Shenzhen Sibiono Bentech, China) into his recurrent nodules in liver respectively and 4 d later, the patient received TACE therapy. In the 2 mo follow-up, the patient was in good clinical condition with normal liver function and no recurrence was identified. | 2 | Effect of Liuweibuqi capsule, a Chinese patent medicine, on the JAK1/STAT3 pathway and MMP9/TIMP1 in a chronic obstructive pulmonary disease rat model显示文摘OBJECTIVE: To observe effect of Liuweibuqi Capsule, a Traditional Chinese Medicine(TCM), on the janus kinase(JAK)/signal transducer and activator of transcription(STAT) pathway and matrix metalloproteinases(MMPs) in a chronic obstructive pulmonary disease(COPD) rat model with lung deficiency in terms of TCM's pattern differentiation.METHODS: Rats were randomly divided into a normal group, model group, Liuweibuqi group, Jinshuibao group, and spleen aminopeptidase group(n=10). Aside from the normal group, all rats were ex-posed to smoke plus lipopolysaccharide tracheal instillation to establish the COPD model with lung deficiency. Models were established after 28 days and then the normal and model groups were given normal saline(0.09 g/kg), Liuweibuqi group was given Liuweibuqi capsule(0.35 g/kg), Jinshuibao group was given Jinshuibao capsules(0.495 g/kg), and the spleen group was given spleen aminopeptidase(0.33 mg/kg), once a day for 30 days. Changes in symptoms, signs, and lung histology were observed. Lung function was measured with a spirometer. Serum cytokines were detected using enzyme-linked immunosorbent assay, and changes in the JAK/STAT pathway, MMP-9, and MMPs inhibitor1(TIMP1) were detected by immunohistochemistry, RT-PCR, and western blotting, respectively.RESULTS: Compared with the normal group, lung tissue was damaged, and lung function was reduced in the model control group. Additionally, the levels of interleukin(IL)-1β, γ interferon(IFN-γ), and IL-6 were higher, while IL-4 and IL-10 were lower in the model control group than those in the normal group. The expressions of JAK1, STAT3, p-STAT3,and MMP-9 m RNA and protein in lung tissue were higher, and TIMP1 m RNA and protein was lower in the model group compared with the normal group.After treatment, compared with the model group,the expression of inflammatory cytokines was lower in each treatment group, and expressions of JAK/STAT pathway, MMPs were lower. | 3 | Multidetector CT in evaluating blood supply of hepatocellular carcinoma after transcatheter arterial chemoembolization显示文摘AIM:To assess the value of multidetector-row computed tomography (MDCT) in choosing retreatment methods of hepatocellular carcinoma (HCC) through evaluating the blood supply of low-density area of HCC after transcatheter arterial chemoembolization (TACE).METHODS: Thirty-two patients with HCC after TACE treatment were examined by plain scanning and hepatic multidetector-row CT. The location of low-density area on plain scanning and the enhancement patterns on dynamic contrast-enhanced scanning were observed. At the same time, three-dimensional CT(3D CT) models of the volume rendering, curved multiplanar reformations, surface shaded display and maximum intensity projection reconstruction of the hepatic artery and portal vein were performed in 6 cases.RESULTS: In CT plain scanning data, low density areas of 32 cases of HCC after TACE treatment were divided into three types: peripheral, one-side-located and mixed types.In contrast-enhanced CT scans, the blood supply of low-density area was classified into four types: arterial blood supply (20 cases), portal blood supply (5 cases), arterial combined with portal blood supply (5 cases) and poor blood supply (2 cases). In 6 cases, the relationship between the low-density area and branches of hepatic artery as well asportal vein was shown by 3D CT.CONCLUSION: Hepatic MDCT is an effective method for evaluating the blood supply of low-density area and therapeutic effect of HCC after TACE treatment. | 4 | PELA microspheres loaded H.pylori lysates and their mucosal immune response显示文摘AIM: To prepare poly (D,L-lactide)-polyethylene glycol copolymer (PELA) microspheres loaded H.pylorilysates or CyStografin and observe their targeting in gastrointestinal mucous membrane or analyze the mucosal immune responses by oral administration. METHODS: PELA microspheres loaded H. Pylorilysates or Cystografin were preparedby double emulsion evaporation method. Their distribution in gastrointestinal mucous membrane was observed by CT. Balb/c mice orally immunized in mucosal immune responses, whose antibody production in salivary and gut washing and antibody secreting cells in Peyer's patches (PP) were estimated by ELISA and ELISPOT, respectively. The microspheres' physical properties, such as particle size, protein level and morphology were investigated.RESULTS: All prepared microspheres were found to have a smooth surface morphology from 3.20-4.05 μm in diameter and high encapsulation efficiency from 74.9-82.2 %. No significant correlation in their physical properties was shown, depending on their molecular weight at the similar composition ratio. Immunization with all types of PELA-Hp microspheres elevated the saliva sIgA level at week 3 by approximately 3-4 times that with soluble antigen, which was greatly enhanced after boosting. At one week after last immunization with all types of PELA-Hp microspheres (week 8), the specific sIgA-ASCs, IgG-ASCs and stgA in salivary rose obviously. In intestinal Peyer's patches, the specific sIgA-ASCs were 5.92-6.98× 104/ml cell and IgG-ASCs were 3.47-4.02 × 104/ml cell, about 5-9 times higher than those with soluble antigen (P<0.01). ASCs in intestine were more than those in stomach and the majority of the ASCs were sIgA-ASCs. The sIgA in gut washing fluid was 1.62-1.85 OD, about 3-6 times tthat of those with soluble antigen. There were significant differences of the ASCs and sIgA in gut washing fluid as compared with those of PBS and MS-0 (P<0.05). | 7 | Classification of the Gut Microbiota of Patients in Intensive Care Units During Development of Sepsis and Septic Shock显示文摘The gut microbiota of intensive care unit(ICU)patients displays extreme dysbiosis associated with increased susceptibility to organ failure,sepsis,and septic shock.However,such dysbiosis is difficult to characterize owing to the high dimensional complexity of the gut microbiota.We tested whether the concept of enterotype can be applied to the gut microbiota of ICU patients to describe the dysbiosis.We collected 131 fecal samples from 64 ICU patients diagnosed with sepsis or septic shock and performed 16S rRNA gene sequencing to dissect their gut microbiota compositions.During the development of sepsis or septic shock and during various medical treatments,the ICU patients always exhibited two dysbiotic microbiota patterns,or ICU-enterotypes,which could not be explained by host properties such as age,sex,and body mass index,or external stressors such as infection site and antibiotic use.ICU-enterotype I(ICU E1)comprised predominantly Bacteroides and an unclassified genus of Enterobacteriaceae,while ICU-enterotype II(ICU E2)comprised predominantly Enterococcus.Among more critically ill patients with Acute Physiology and Chronic Health Evaluation II(APACHE II)scores>18,septic shock was more likely to occur with ICU E1(P=0.041).Additionally,ICU E1 was correlated with high serum lactate levels(P=0.007).Therefore,different patterns of dysbiosis were correlated with different clinical outcomes,suggesting that ICU-enterotypes should be diagnosed as independent clinical indices.Thus,the microbial-based human index classifier we propose is precise and effective for timely monitoring of ICU-enterotypes of individual patients.This work is a first step toward precision medicine for septic patients based on their gut microbiota profiles. | Wanglin Liu Mingyue Cheng Jinman Li Peng Zhang Hang Fan Qinghe Hu Maozhen Han Longxiang Su Huaiwu He Yigang Tong Kang Ning Yun Long | 2020 | Genomics, Proteomics & Bioinformatics2020,18,6: | 7 |
| 8 | Effects of Mercury on the Structure and Activity of BLM642-1290 Recombinant Helicase显示文摘Objective Bloom's syndrome is an autosomal recessive disorder characterized by genomic instability and a predisposition to many cancers. Mutations of the BLM gene (encoding a BLM helicase) may form a structure of the etiology of this disease. As a global pollutant, mercury poses a major threat to human health. The current study was conducted to elucidate the effects of Hg 2+ on the structure and activity of BLM642‐1290 recombinant helicase, and to further explore the molecular mechanisms of mercury toxicity to the DNA helicase. Methods The effects of Hg 2+ on biological activity and structure of BLM642‐1290 recombinant helicase were determined by fluorescence polarized, ultraviolet spectroscopic, and free‐phosphorus assay technologies, respectively. Results The helicase activity, the DNA‐binding activity, and the ATPase activity of BLM642‐1290 recombinant helicase were inhibited by Hg 2+ treatment. The LMCT (ligand‐to‐metal charge transition) peaks of the helicase were enhanced with the increase of the Hg 2+ level. The LMCT peaks of the same concentration of helicase gradually increased over time. Conclusions The biological activity of BLM642‐1290 recombinant helicase is inhibited by Hg 2+ treatment. The conformation of the helicase is significantly altered by Hg 2+ . | 9 | The Form of Sulfate in Pseudo-Boehmite and Its Effect on Properties of Pseudo-Boehmite显示文摘A series of pseudo-boehmite samples with their sulfate radicals(SO2-4) concentration ranging from 0.9% to 3.0% were prepared by the reaction of Na Al O2 solution on Al2(SO4)3 solution. The existing form of sulfate radicals was investigated. Results have shown that sulfates in pseudo-boehmite included two parts, the soluble sulfate radicals and the insoluble sulfate radicals, which accounted for 99% of the total amount of sulfate radicals. XRD, low-temperature N2-adsorption, and TEM were used to characterize the properties of these pseudo-boehmite samples. Results have shown that the relative crystallinity and crystal size of pseudo-boehmite decreased with the increase of sulfate radicals in the support. In the meanwhile, the bound water content in pseudo-boehmite increased. The TEM images of pseudo-boehmite indicated that the pseudoboehmite was prone to become amorphous hydrated alumina. This results in collection of larger amount of raw data(big data) that is generated at different levels of network architecture and is typically underutilized. To unleash its full value, innovative machine learning algorithms need to be utilized in order to extract valuable insights which can be used for improving the overall network's performance. Additionally, a major challenge for network operators is to cope up with increasing number of complete(or partial) cell outages and to simultaneously reduce operational expenditure. This paper contributes towards the aforementioned problems by exploiting big data generated from the core network of 4 G LTE-A to detect network's anomalous behavior. We present a semi-supervised statistical-based anomaly detection technique to identify in time: first, unusually low user activity region depicting sleeping cell, which is a special case of cell outage; and second, unusually high user traffic area corresponding to a situation where special action such as additional resource allocation, fault avoidance solution etc. may be needed. | 12 | SuperDARN CUTLASS Finland radar observations of high-latitude magnetic reconnections under northward interplanetary magnetic field (IMF) conditions显示文摘A number of backscatter power enhancement events with 'equatorward-moving radar auroral forms' in the high-latitude ionosphere were observed by SuperDARN CUTLASS Finland radar when the IMF was northward during 09:00 -10:00 UT on 26 March 2004. These events were also associated with sunward flow enhancements at each location in the Northern Hemisphere which were shown in ionospheric convections measured by the SuperDARN radars. These are typical features of high-latitude (lobe) magnetic reconnections. The durations of the velocity enhancements imply that the evolution time of the lobe reconnections is about 8-16 min from their origin at the reconnection site to their addition to the magnetotail lobe again. In additional, the Double Star TC-1 spacecraft was moving from magnetosheath into magnetosphere, and crossing the magnetopause near the subsolar region during this interval, and observed typical low-latitude magnetic reconnection signatures. In order to overcome the time delay, a novel feed-forward compensation active disturbance rejection control(FFC-ADRC) approach is proposed. It combines advantages of the Smith predictor and the traditional active disturbance rejection control(ADRC). The tracking differentiator(TD) is designed to predict the control signal, which adds an anticipatory control to the control signal and allows a higher observer bandwidth to obtain better disturbance rejection. The modified extended state observer(ESO) is designed to estimate both system states and the total disturbances(internal disturbance, uncertainties and delayed disturbance). Then the Lyapunov theory and the theory of the input-output stability are applied to prove the asymptotic stability of the closed-loop control system. | 15 | Quantitative analysis on the dynamic characteristics of megadunes around the Crescent Moon Spring, China显示文摘The Crescent Moon Spring is a precious natural heritage. However, the dynamic characteristics of megadunes around the Crescent Moon Spring are not well known. This paper quantitatively studied the character- istics and changes of megadunes around the Crescent Moon Spring by interpreting aerial photographs taken in 1985 and 2004 and analysing the dune crestlines and the wind data collected from 2011 to 2012. Results revealed that pyramid dunes were formed by a complex wind regime. The Crescent Moon Spring was not buried by shifting sands because of the stable wind regime and relative stability of pyramid dunes. The crestlines of the dunes around the spring moved northward between 1985 and 2004. The south-facing slip faces were also exposed to wind ero- sion, whereas the other faces were under deposition, thus indicating that the southerly wind was relatively en- hanced. | 16 | Computational fluid dynamics evaluation of the effect of different city designs on the wind environment of a downwind natural heritage site显示文摘Disturbance in wind regime and sand erosion deposition balance may lead to burial and eventual vanishing of a site.This study conducted 3D computational fluid dynamics(CFD)simulations to evaluate the effect of a proposed city design on the wind environment of the Crescent Spring,a downwind natural heritage site located in Dunhuang,Northwestern China.Satellite terrain data from the Advanced Spaceborne Thermal Emission and Reflection Radiometer(ASTER)Digital Elevation Model(DEM)were used to construct the solid surface model.Steady-state Reynolds Averaged Navier-Stokes equations(RANS)with shear stress transport(SST)k-ωturbulence model were then applied to solve the flow field problems.Land-use changes were modeled implicitly by dividing the underlying surface into different areas and by applying corresponding aerodynamic roughness lengths.Simulations were performed by using cases with different city areas and building heights.Results show that the selected model could capture the surface roughness changes and could adjust wind profile over a large area.Wind profiles varied over the greenfield to the north and over the Gobi land to the east of the spring.Therefore,different wind speed reduction effects were observed from various city construction scenarios.The current city design would lead to about 2 m/s of wind speed reduction at the downwind city edge and about 1 m/s of wind speed reduction at the north of the spring at 35-m height.Reducing the city height in the north greenfield area could efficiently eliminate the negative effects of wind spee.By contrast,restricting the city area worked better in the eastern Gobi area compared with other parts of the study area.Wind speed reduction in areas near the spring could be limited to 0.1 m/s by combining these two abatement strategies.The CFD method could be applied to simulate the wind environment affected by other land-use changes over a large terrain. | BenLi LIU JianJun QU QingHe NIU JunZhan WANG KeCun ZHANG | 2014 | Journal of Arid Land2014,6,1: | 4 |
| 17 | Process optimization of reaction of acid leaching residue of asbestos tailing and sodium hydroxide aqueous solution显示文摘Silica is the major component of the acid leaching residue of asbestos tailing. The waterglass solution can be prepared by the reaction of the residue with sodium hydroxide aqueous solution. Compared to the high temperature reaction method, this process is environmental friendly and low cost. In this paper, the reaction process of the residue and the sodium hydroxide aqueous solution is optimized. The optimum reaction process parameters are as follows: the usage of sodium hydroxide is 26.4 g/100 g acid leaching residue, the reaction temperature is 90℃, the reaction time is 1 h, and the ratio of the liquid/solid is 2.0. The significance sequence of the process parameters to the alkali leaching reaction effect is the usage of sodium hydroxide > the ratio of the liquid/solid > the reaction time > the reaction temperature. The significance sequence to the leaching ratio of SiO2 is the ratio of the liquid/solid > the usage of sodium hydroxide > the reaction time > the reaction temperature. The significance sequence to the modulus of the sodium silicate is the ratio of the liquid/solid > the usage of sodium hydroxide > the reaction time > the reaction temperature. Under the optimum conditions, the leaching ratio of the SiO2 is 77.5%, and the modulus of the sodium silicate is 3.15. | 18 | Oxygen‑Deficient β‑MnO_(2)@Graphene Oxide Cathode for High‑Rate and Long‑Life Aqueous Zinc Ion Batteries显示文摘Recent years have witnessed a booming interest in grid-scale electrochemical energy storage,where much attention has been paid to the aqueous zinc ion batteries(AZIBs).Among various cathode materials for AZIBs,manganese oxides have risen to prominence due to their high energy density and low cost.However,sluggish reaction kinetics and poor cycling stability dictate against their practical application.Herein,we demonstrate the combined use of defect engineering and interfacial optimization that can simultaneously promote rate capability and cycling stability of MnO_(2) cathodes.β-MnO_(2) with abundant oxygen vacancies(VO)and graphene oxide(GO)wrapping is synthesized,in which VO in the bulk accelerate the charge/discharge kinetics while GO on the surfaces inhibits the Mn dissolution.This electrode shows a sustained reversible capacity of~129.6 mAh g^(−1) even after 2000 cycles at a current rate of 4C,outperforming the state-of-the-art MnO_(2)-based cathodes.The superior performance can be rationalized by the direct interaction between surface VO and the GO coating layer,as well as the regulation of structural evolution ofβ-MnO_(2) during cycling.The combinatorial design scheme in this work offers a practical pathway for obtaining high-rate and long-life cathodes for AZIBs. | Shouxiang Ding Mingzheng Zhang Runzhi Qin Jianjun Fang Hengyu Ren Haocong Yi Lele Liu Wenguang Zhao Yang Li Lu Yao Shunning Li Qinghe Zhao Feng Pan | 2021 | Nano-Micro Letters2021,13,11: | 3 |
| 19 | Nano CuO/ZSM-5 zeolite as a green and efficient catalyst for dehydration of 1,4-butanediol to tetrahydrofuran显示文摘Nano CuO/ZSM-5 zeolite was prepared and used as a catalyst for dehydration of 1,4-butanediol(BDO) to tetrahydrofuran(THF) in liquid-phase. It was found that the 4.6 wt% CuO/ZSM-5 displayed good catalytic performance, and nearly 100%of BDO conversion and more than 99% of THF selectivity could be achieved by a rotary evaporator reactor at 170 °C under the atmospheric pressure. With such mild reaction conditions, 2400 g BDO could be converted to THF over 1 g catalyst under semi-continuous operation. Characterizations with X-ray diffraction(XRD), temperature-programmed reduction(TPR),NH3-temperature programmed desorption(TPD), X-ray photoelectron spectroscopy(XPS), transmission electron microscope(TEM) and Brunauer-Emmett-Teller(BET) over fresh and used 4.6 wt% CuO/ZSM-5 were conducted. | 20 | China's flourishing synthetic organofluorine chemistry:innovations in the new millennium显示文摘The new millennium has witnessed the rapid development of synthetic organofluorine chemistry all over the world, and chemists in China have made significant contributions in this field. This review aims to provide a brief introduction to China's primary innovations from 2000 to early 2017, covering fluorination,fluoroalkylation, fluoromethylthiolation, fluoroolefination and polyfluoroarylation, as well as synthesis with fluorinated building blocks. Recent advances in the chemistry of difluorocarbene and the chemistry of carbon–fluorine bond activation are also discussed.
