1Molecular screening of patients with nonsyndromic hearing loss from Nanjing city of China显示文摘Hearing loss is the most frequent sensory disorder involving a multitude of factors,and at least 50% of cases are due to genetic etiology.To further characterize the molecular etiology of hearing loss in the Chinese population,we recruited a total of 135 unrelated patients with nonsyndromic sensorineural hearing loss (NSHL) for mutational screening of GJB2,GJB3,GJB6,SLC26A4,SLC26A5 IVS2-2A>G and mitochondrial 12SrRNA,tRNA Ser(UCN) by PCR amplification and direct DNA sequencing.The carrier frequencies of deafness-causing mutations in these patients were 35.55% in GJB2,3.70% in GJB6,15.56% in SLC26A4 and 8.14% in mitochondrial 12SrRNA,respectively.The results indicate the necessity of genetic screening for mutations of these causative genes in Chinese population with nonsyndromic hearing loss.Yajie Lu Dachun Dai Zhibin Chen Xin Cao Xingkuan Bu Qinjun Wei Guangqian Xing 2011The Journal of Biomedical Research2011,25,5:30
2Substance P-modified human serum albumin nanoparticles loaded with paclitaxel for targeted therapy of glioma显示文摘The blood–brain barrier(BBB) and the poor ability of many drugs to cross that barrier greatly limits the efficacy of chemotherapies for glioblastoma multiforme(GBM). The present study exploits albumin as drug delivery vehicle to promote the chemotherapeutic efficacy of paclitaxel(PTX) by improving the stability and targeting efficiency of PTX/albumin nanoparticles(NPs). Here we characterize PTX-loaded human serum albumin(HSA) NPs stabilized with intramolecular disulfide bonds and modified with substance P(SP) peptide as the targeting ligand. The fabricated SP-HSA-PTX NPs exhibited satisfactory drug-loading content(7.89%) and entrapment efficiency(85.7%) with a spherical structure(about 150 nm) and zeta potential of -12.0 mV. The in vitro drug release from SP-HSA-PTX NPs occurred in a redox-responsive manner. 3Pore scale simulation of liquid and gas two-phase flow based on digital core technology显示文摘Two-phase flow in two digital cores is simulated by the color-gradient lattice Boltzmann method.This model can be applied totwo-phase flow with high-density ratio(on order of 1000).The first digital core is an artificial sandstone core,and itsthree-dimensional gray model is obtained by Micro-CT scanning.The gray scale images are segmented into discrete phases(solid particles and pore space) by the Otsu algorithm.The second one is a digital core of shale,which is reconstructed usingMarkov Chain Monte Carlo method with segmented SEM scanning image as input.The wettability of solid wall and relativepermeability of a cylindrical tube are simulated to verify the model.In the simulations of liquid and gas two phase flow in digital cores,density ratios of 100,200,500 and 1000 between liquid and gas are chosen.Based on the gas distribution in the digital core at different times,it is found that the fingering phenomenon is more salient at high density ratio.With the density ratioincreasing,the displacement efficiency decreases.Besides,due to numerous small pores in the shale,the displacement efficiency is over 20% less than that in the artificial sandstone and the difference is even about 30% when density ratio is greaterthan 500.As the density ratio increases,the gas saturation decreases in big pores,and even reaches zero in some small pores orbig pores with small throats.Residual liquid mainly distributes in the small pores and the edge of big pores due to the wettability of liquid.Liquid recovery can be enhanced effectively by decreasing its viscosity.ZHANG Lei KANG QinJun YAO Jun GAO Ying SUN ZhiXue LIU HaiHu VALOCCHI Albert J. 2015Science China(Technological Sciences)2015,58,8:11
4OSBPL2-disrupted pigs recapitulate dual features of human hearing loss and hypercholesterolaemia显示文摘Oxysterol binding protein like 2(OSBPL2), an important regulator in cellular lipid metabolism and transport, was identified as a novel deafness-causal gene in our previous work. To resemble the phenotypic features of OSBPL2 mutation in animal models and elucidate the potential genotypephenotype associations, the OSBPL2-disrupted Bama miniature(BM) pig model was constructed using CRISPR/Cas9-mediated gene editing, somatic cell nuclear transfer(SCNT) and embryo transplantation approaches, and then subjected to phenotypic characterization of auditory function and serum lipid profiles. The OSBPL2-disrupted pigs displayed progressive hearing loss(HL) with degeneration/apoptosis of cochlea hair cells(HCs) and morphological abnormalities in HC stereocilia, as well as hypercholesterolaemia. High-fat diet(HFD) feeding aggravated the development of HL and led to more severe hypercholesterolaemia. 5Rational design of MoS_(2) nanosheets decorated on mesoporous hollow carbon spheres as a dual-functional accelerator in sulfur cathode for advanced pouch-type Li–S batteries显示文摘Developing sulfur cathodes with high catalytic activity on accelerating the sluggish redox kinetics of lithium polysulfides(Li PSs) and unveiling their mechanisms are pivotal for advanced lithium–sulfur(Li–S)batteries. Herein, MoS2 is verified to reduce the Gibbs free energy for rate-limiting step of sulfur reduction and the dissociation energy of lithium sulfide(Li2 S) for the first time employing theoretical calculations. The Mo S2 nanosheets coated on mesoporous hollow carbon spheres(MHCS) are then reasonably designed as a sulfur host for high-capacity and long-life Li–S battery, in which MHCS can guarantee the high sulfur loading and fast electron/ion transfer. It is revealed that the shuttle effect is efficiently inhibited because of the boosted conversion of Li PSs. As a result, the coin cell based on the MHCS@Mo S2-S cathode exhibits stable cycling performance maintaining 735.7 mAh g^(-1) after 500 cycles at 1.0 C. More importantly, the pouch cell employing the MHCS@Mo S2-S cathodes achieves high specific capacity of1353.2 m Ah g^(-1) and prominent cycle stability that remaining 960.0 m Ah g^(-1) with extraordinary capacity retention of 79.8% at 0.1 C after 170 cycles. 6GLUT1-mediated effective anti-miRNA21 pompon for cancer therapy显示文摘Oncogenic microRNAs are essential components in regulating the gene expression of cancer cells. Especially miR21, which is a major player involved of tumor initiation, progression, invasion and metastasis in several cancers. The delivery of anti-miR21 sequences has significant potential for cancer treatment. Nevertheless, since anti-miR21 sequences are extremely unstable and they need to obtain certain concentration to function, it is intensely difficult to build an effective delivery system for them.The purpose of this work is to construct a self-assembled glutathione(GSH)-responsive system with tumor accumulation capacity for effective anti-miR21 delivery and cancer therapy. A novel drug delivery nanosphere carrying millions of anti-miR21 sequences was developed through the rolling circle transcription(RCT) method. GSH-responsive cationic polymer polyethyleneimine(pOEI) was synthesized to protect the nanosphere from degradation by Dicer or other RNase in normal cells and optimize the pompon-like nanoparticle to suitable size. Dehydroascorbic acid(DHA), a targeting molecule, which is a substrate of glucose transporter 1(GLUT 1) and highly expressed on malignant tumor cells, was connected to pOEI through PEG, and then the polymer was used for contracting a RNA nanospheres into nanopompons. 7Inhibition of cell proliferation by siRNA targeting hPRLR in breast cancer MCF-7 cell line显示文摘Objective:To study the inhibition of proliferation of breast cancer by small interfering RNA(siRNA) targeting human prolactin(hPRLR) and the underlying mechanisms.Methods:The siRNA targeting hPRLR was chemically synthesized and transfected into MCF-7 cells,the expression of hPRLR was analyzed by real-time quantitive PCR,cell growth inhibition was measured with MTT assay,cell cycle of the transfected cells was examined by flow cytometry,meanwhile,expression of cyclin D1 was tested by semi-quantitative RT-PCR.Results:24 h after transfection with 100 nmol/L siRNA-PRLR,the expression of hPRLR mRNA was suppressed by 65%,cells in G1 phase increased,but cells in S phase decreased.Down regulated hPRLR expression exhibited significant inhibition in cell proliferation.And the expression of cyclin D1 was down regulated.Conclusion:The results indicate that siRNA-hPRLR is a useful tool for silencing hPRLR expression and inhibiting cell proliferation in breast cancer MCF-7 cell line,and it may be a possible new approach for breast cancer gene therapy.Mei Pan Qinjun Wei Fang Cao Yajie Lu Yibao Zhu Yongqian Shu Xin Cao 2007Journal of Nanjing Medical University2007,21,6:5

9IMPACTS OF ANTARCTIC OSCILLATION ON SUMMER MOISTURE TRANSPORT AND PRECIPITATION IN EASTERN CHINA显示文摘Using NCEP/NCAR reanalysis data and monthly precipitation over 160 conventional stations in China, analyses of moisture transport characteristics and corresponding precipitation variation in the east part of China in summer are made, and studies are carried out on possible influence on moisture transport and precipitation in summer by the variation of Antarctic Oscillation (AAO). The results show that the abnormal variation of the AAO affected the summer precipitation in China significantly. 10Lithiated Nafion-garnet ceramic composite electrolyte membrane for solid-state lithium metal battery显示文摘Single-ion conducting solid polymer electrolytes are expected to play a vital role in the realization of solid-state Li metal batteries.In this work,a lithiated Nafion(Li-Nafion)-garnet ceramic Li6.25La3 Zr2 Al0.25O12(LLZAO)composite solid electrolyte(CSE)membrane with 30μm thickness was prepared for the first time.By employing X-ray photoelectron spectroscopy and transmission electron microscope,the interaction between LLZAO and Li-Nafion was investigated.It is found that the LLZAO interacts with the Li-Nafion to form a space charge layer at the interface between LLZAO and Li-Nafion.The space charge layer reduces the migration barrier of Li-ions and improves the ionic conductivity of the CSE membrane.The CSE membrane containing 10 wt%LLZAO exhibits the highest ionic conductivity of2.26×10-4 S cm-1 at 30℃among the pristine Li-Nafion membrane,the membrane containing 5 wt%,20 wt%,and 30 wt%LLZAO,respectively.It also exhibits a high Li-ion transference number of 0.92,and a broader electrochemical window of 0-+4.8 V vs.Li+/Li than that of 0-+4.0 V vs.Li+/Li for the pristine Li-Nafion membrane.It is observed that the CSE membrane not only inhibits the growth of Li dendrites but also keeps excellent electrochemical stability with the Li electrode.Benefitting from the above merits,the solid-state LiFePO4/Li cell fabricated with the CSE membrane was practically charged and discharged at 30℃.The cell exhibits an initial reversible discharge specific capacity of 160 mAh g-1 with 97%capacity retention after 100 cycles at 0.2 C,and maintains discharge specific capacity of 126 mAh g-1 after500 cycles at 1 C.The CSE membrane prepared with Li-Nafion and LLZAO is proved to be a promising solid electrolyte for advanced solid-state Li metal batteries.Jing Gao Qinjun Shao Jian Chen 2020Journal of Energy Chemistry2020,29,7:3
11TREND OF PRECIPITATION VARIATION IN HUBEI PROVINCE SINCE THE 1960S显示文摘Through linear regression analysis to the trend of annual,seasonal and monthly precipitation of 72 meteorological stations in Hubei Province from 1961 to 1995,it is revealed that: 1) annual precipitation was increasing by 61.0mm/10a in the eastern part of Hubei (112°E as a dividing line) and decreasing by 34.9mm/10a in the western part; 2) precipitation in winter and summer (January,February,March,June and July) was increasing in almost whole province which usually with non-uniformity of precipitation distribution from the south to the north. The precipitation in spring,autumn and winter (April,September,November and December) was decreasing in most of the areas which usually with non-uniformity of precipitation distribution from the east to the west. March and December were transition periods between two spatial distribution patterns mentioned above; 3) the eastern part of Hubei has beome one of precipitation increasing centers in China. 14Assessing bacterial communities in the rhizosphere of 8-year-old genetically modified poplar(Populus spp.)显示文摘Microbe communities in rhizosphere ecosystems are important for plant health but there is limited knowledge of them in the rhizospheres of genetically modified(GM) plants, especial for tree species. We used the amplitude sequencing method to analyze the V4 regions of the 16 S r RNA gene to identify changes in bacterial diversity and community structure in two GM lines(D520 and D521), one non-genetically modified(nonGM) line and in uncultivated soil. After chimera filtering,468.133 sequences in the domain Bacteria remained. There were ten dominant taxonomic groups(with [1 % of all sequences) across the samples. 241 of 551 genera(representing a ratio of 97.33 %) were common to all samples.A Venn diagram showed that 1.926 operational taxonomic units(OTUs) were shared by all samples. We found a specific change, a reduction in Chloroflexi, in the microorganisms in the rhizosphere soil planted with poplars. 15Incorporation of layered tin(Ⅳ) phosphate in graphene framework for high performance lithium-sulfur batteries显示文摘To anchor the polysulfide and enhance the conversion kinetics of polysulfide to disulfide/sulfide is critical for improving the performance of lithium-sulfur battery.For this purpose,the graphene-supported tin(Ⅳ) phosphate(Sn(HPO_4)_2·H_2 O,SnP) composites(SnP-G) are employed as the novel sulfur hosts in this work.When compared to the graphene-sulfur and carbon-sulfur composites,the SnP-G-sulfur composites exhibit much better cycling performance at 1.0 C over 800 cycles.Meanwhile,the pouch cell fabricated with the SnP-G-sulfur cathodes also exhibits excellent performance with an initial capacity of1266.6 mAh g^(-1)(S) and capacity retention of 76.9% after 100 cycles at 0.1 C.The adsorption tests,density functional theory(DFT) calculations in combination with physical cha racterizations and electrochemical measurements provide insights into the mechanism of capture-accelerated conversion mechanism of polysulfide at the surface of SnP.DFT calculations indicate that the Li-O bond formed between Li atom(from Li_2 S_n,n=1,2,4,6,8) and O atom(from PO_3-OH in SnP) is the main reason for the strong interactions between Li_2 S_n and SnP.As a result,SnP can effectively restrain the shuttle effect and improving the cycling performance of Li-S cell.In addition,by employing the climbing-image nudged elastic band(ciNEB) methods,the energy barrier for lithium sulfide decomposition(charging reaction) on SnP is proved to decrease significantly compared to that on graphene.It can be concluded that SnP is an effective sulfur hosts acting as dual-functional accelerators for the conversion reactions of polysulfude to sulfide(discharging reaction) as well as polysulfide to sulfur(charging reaction).Haifeng Yuan Na Zhang Leiwu Tian Lei Xu Qinjun Shao Syed Danish Ali Zaidi Jianping Xiao Jian Chen 2021Journal of Energy Chemistry2021,30,2:2
16Application of remote sensing for investigating mining geological hazards显示文摘To investigate geological mining hazards using digital techniques such as highresolution remote sensing,a semi-automatically geological mining hazards extraction method is proposed based on the case of the Shijiaying coal mine,located in Fangshan District,Beijing,China.In the method,the vegetation is first removed using the normalized difference vegetation index(NDVI)on the GeoEye-1 data.Then,geological mining hazards interpretation features are determined after color enhancement using principal component analysis(PCA)transformation.Bitmaps mainly covered by geological mining hazards are isolated by masking operation in the environment for visualizing images software.Next,each bitmap is classified into a two-valued imagery using support vector machine algorithm.In the two-valued imagery,1 denotes the geological mining hazards,while 0 denotes none.Afterwards,the two-valued imagery is converted into a vector graph by corresponding functions in the ArcGIS software and no geological mining hazards regions in the vector graph are deleted manually.Finally,the correlation between factors(such as mining activity,lithology,geological structure,and slope)and geological mining hazards is analyzed using a logistic regression and a hazardous-area forecasting model is built.The results of field verification show that the accuracy of the geological mining hazards extraction method is 98.1%and the results of the hazardous-area forecasting indicate that the logistic regression is an effective model in assessing geological hazard risks and that mining activity is the main contributing factor to the hazards,while geological structure,slope,lithology,roughness of the surface,and aspect are the secondary.Qinjun Wang Huadong Guo Yu Chen Qizhong Lin Hui Li 2013International Journal of Digital Earth2013,6,5:1

18Emergence of superconductivity from fully incoherent normal state in an iron-based superconductor (Ba_(0.6)K_(0.4))Fe_2As_2显示文摘In unconventional superconductors, it is generally believed that understanding the physical properties of the normal state is a pre-requisite for understanding the superconductivity mechanism. In conventional superconductors like niobium or lead, the normal state is a Fermi liquid with a well-defined Fermi surface and well-defined quasipartcles along the Fermi surface. Superconductivity is realized in this case by the Fermi surface instability in the superconducting state and the formation and condensation of the electron pairs(Cooper pairing). The high temperature cuprate superconductors, on the other hand, represent another extreme case that superconductivity can be realized in the underdoped region where there is neither well-defined Fermi surface due to the pseudogap formation nor quasiparticles near the antinodal regions in the normal state. Here we report a novel scenario that superconductivity is realized in a system with well-defined Fermi surface but without quasiparticles along the Fermi surface in the normal state.High resolution laser-based angle-resolved photoemission measurements have been performed on an optimally-doped iron-based superconductor(Ba_(0.6)K_(0.4))Fe_2As_2. We find that, while sharp superconducting coherence peaks emerge in the superconducting state on the hole-like Fermi surface sheets, no quasiparticle peak is present in the normal state. Its electronic behaviours deviate strongly from a Fermi liquid system. The superconducting gap of such a system exhibits an unusual temperature dependence that it is nearly a constant in the superconducting state and abruptly closes at Tc.
