Strategies for reducing speckle noise in digital holography显示文摘Digital holography(DH)has emerged as one of the most effective coherent imaging technologies. The technological developments of digital sensors and optical elements have made DH the primary approach in several research fields, from quantitative phase imaging to optical metrology and 3D display technologies,to name a few.Like many other digital imaging techniques,DH must cope with the issue of speckle artifacts, due to the coherent nature of the required light sources.Despite the complexity of the recently proposed de-speckling methods, many have not yet attained the required level of effectiveness. That is,a universal denoising strategy for completely suppressing holographic noise has not yet been established. Thus the removal of speckle noise from holographic images represents a bottleneck for the entire optics and photonics scientific community. This review article provides a broad discussion about the noise issue in DH,with the aim of covering the best-performing noise reduction approaches that have been proposed so far.Quantitative comparisons among these approaches will be presented.Vittorio Bianco Pasquale Memmolo Marco Leo Silvio Montresor Cosimo Distante Melania Paturzo Pascal Picart Bahram Javidi Pietro Ferraro 2018Light(Science & Applications)2018,7,1:8
Endowing a plain fluidic chip with micro-optics: a holographic microscope slide显示文摘Lab-on-a-Chip(LoC)devices are extremely promising in that they enable diagnostic functions at the point-of-care.Within this scope,an important goal is to design imaging schemes that can be used out of the laboratory. In this paper,we introduce and test a pocket holographic slide that allows digital holography microscopy to be performed without an interferometer setup.Instead,a commercial off-the-shelf plastic chip is engineered and functionalized with this aim.The microfluidic chip is endowed with micro-optics,that is,a diffraction grating and polymeric lenses,to build an interferometer directly on the chip, avoiding the need for a reference arm and external bulky optical components. Thanks to the single-beam scheme, the system is completely integrated and robust against vibrations, sharing the useful features of any common path interferometer.Hence,it becomes possible to bring holographic functionalities out of the lab, moving complexity from the external optical apparatus to the chip itself.Label-free imaging and quantitative phase contrast mapping of live samples are demonstrated, along with flexible refocusing capabilities. Thus,a liquid volume can be analyzed in one single shot with no need for mechanical scanning systems.Vittorio Bianco Biagio Mandracchia Valentina Marchesano Vito Pagliarulo Federico Olivieri Sara Coppola Melania Paturzo Pietro Ferraro 2017Light(Science & Applications)2017,6,1:6

Dehydration of plant cells shoves nuclei rotation allowing for 3D phase-contrast tomography显示文摘Single-cell phase-contrast tomography promises to become decisive for studying 3D intracellular structures in biology. It involves probing cells with light at wide angles, which unfortunately requires complex systems. Here we show an intriguing concept based on an inherent natural process for plants biology,i.e.,dehydration,allowing us to easily obtain 3D-tomography of onion-epidermal cells’nuclei. In fact, the loss of water reduces the turgor pressure and we recognize it induces significant rotation of cells’nuclei. Thanks to the holographic focusing flexibility and an ad-hoc angles’tracking algorithm,we combine different phase-contrast views of the nuclei to retrieve their 3D refractive index distribution.Nucleolus identification capability and a strategy for measuring morphology, dry mass, biovolume, and refractive index statistics are reported and discussed. This new concept could revolutionize the investigation in plant biology by enabling dynamic 3D quantitative and label-free analysis at sub-nuclear level using a conventional holographic setup.Zhe Wang Vittorio Bianco Daniele Pirone Pasquale Memmolo Massimiliano Maria Villone Pier Luca Maffettone Pietro Ferraro 2021Light(Science & Applications)2021,10,10:1
Tomographic flow cytometry by digital holography显示文摘High-throughput single-cell analysis is a challenging task.Label-free tomographic phase microscopy is an excellent candidate to perform this task.However,in-line tomography is very difficult to implement in practice because it requires a complex set-up for rotating the sample and examining the cell along several directions. We demonstrate that by exploiting the random rolling of cells while they are flowing along a microfluidic channel,it is possible to obtain in-line phase-contrast tomography,if smart strategies for wavefront analysis are adopted. In fact, surprisingly,a priori knowledge of the three-dimensional position and orientation of rotating cells is no longer needed because this information can be completely retrieved through digital holography wavefront numerical analysis. This approach makes continuous-flow cytotomography suitable for practical operation in real-world,single-cell analysis and with a substantial simplification of the optical system;that is,no mechanical scanning or multi-direction probing is required.A demonstration is given for two completely different classes of biosamples:red blood cells and diatom algae. An accurate characterization of both types of cells is reported, despite their very different nature and material content, thus showing that the proposed method can be extended by adopting two alternate strategies of wavefront analysis to many classes of cells.Francesco Merola Pasquale Memmolo Lisa Miccio Roberto Savoia Martina Mugnano Angelo Fontana Giuliana D’Ippolito Angela Sardo Achille Iolascon Antonella Gambale Pietro Ferraro 2016Light(Science & Applications)2016,5,1:1

Defining metabolic activity of nephrolithiasis--Appropriate evaluation and follow-up of stone formers显示文摘Considering the variation in metabolic evaluation and medical management of kidney stone disease, this consensus review was created to discuss the metabolic activity of nephrolithiasis, define the difference between single and recurrent stone formers, and develop a schema for metabolic and radiologic follow-up.A systematic review of the literature was performed to identify studies of metabolic evaluation and follow-up of patients with nephrolithiasis. Both single and recurrent stone formers share many similarities in metabolic profiles. The study group determined that based on an assessment of risk for stone recurrence and metabolic activity, single and recurrent stone formers should be evaluated comprehensively, including two 24 h urine studies on a random diet.Targeted medication and dietary recommendations are effective for many patients in reducing the risk of stone recurrence.Follow-up of those with stone disease should be obtained depending on the level of metabolic activity of the patient, the risk of chronic kidney disease and the risk of osteoporosis/osteopenia.A standard scheme includes a baseline metabolic profile,a repeat study 3e6 months after initiation of treatment, and then yearly when stable, with abdominal imaging obtained every 1-2 years.Daniel A.Wollin Adam G.Kaplan Glenn M.Preminger Pietro Manuel Ferraro Antonio Nouvenne Andrea Tasca Emanuele Croppi Giovanni Gambaro Ita P.Heilberg 2018Asian Journal of Urology2018,5,4:1

Quasi noise-free digital holography显示文摘One of the main drawbacks of Digital Holography(DH)is the coherent nature of the light source, which severely corrupts the quality of holographic reconstructions.Although numerous techniques to reduce noise in DH have provided good results, holographic noise suppression remains a challenging task.We propose a novel framework that combines the concepts of encoding multiple uncorrelated digital holograms, block grouping and collaborative filtering to achieve quasi noise-free DH reconstructions. The optimized joint action of these different image-denoising methods permits the removal of up to 98%of the noise while preserving the image contrast. The resulting quality of the hologram reconstructions is comparable to the quality achievable with non-coherent techniques and far beyond the current state of art in DH.Experimental validation is provided for both singlewavelength and multi-wavelength DH,and a comparison with the most used holographic denoising methods is performed.Vittorio Bianco Pasquale Memmolo Melania Paturzo Andrea Finizio Bahram Javidi Pietro Ferraro 2016Light(Science & Applications)2016,5,1:1
