1Gut-liver axis and probiotics: Their role in non-alcoholic fatty liver disease显示文摘The incidence of obesity and its related conditions, including non-alcoholic fatty liver disease(NAFLD), has dramatically increased in all age groups worldwide. Given the health consequences of these conditions, and the subsequent economic burden on healthcare systems, their prevention and treatment have become major priorities. Because standard dietary and lifestyle changes and pathogenically-oriented therapies(e.g., antioxidants, oral hypoglycemic agents, and lipid-lowering agents) often fail due to poor compliance and/or lack of efficacy, novel approaches directed toward other pathomechanisms are needed. Here we present several lines of evidence indicating that, by increasing energy extraction in some dysbiosis conditions or small intestinal bacterial overgrowth,specific gut microbiota and/or a'low bacterial richness'may play a role in obesity,metabolic syndrome,and fatty liver.Under conditions involving a damaged intestinal barrier('leaky gut'),the gut-liver axis may enhance the natural interactions between intestinal bacteria/bacterial products and hepatic receptors(e.g.,toll-like receptors),thus promoting the following cascade of events:oxidative stress,insulinresistance,hepatic inflammation,and fibrosis.We also discuss the possible modulation of gut microbiota by probiotics,as attempted in NAFLD animal model studies and in several pilot pediatric and adult human studies.Globally,this approach appears to be a promising and innovative add-on therapeutic tool for NAFLD in the context of multi-target therapy.Giulia Paolella Claudia Mandato Luca Pierri Marco Poeta Martina Di Stasi Pietro Vajro 2014World Journal of Gastroenterology2014,20,42:55
2Allelic Analyses of the Arabidopsis YUC1 Locus Reveal Residues and Domains Essential for the Functions of YUC Family of Flavin Monooxygenases显示文摘Flavin monooxygenases(FMOs) play critical roles in plant growth and development by synthesizing auxin and other signaling molecules.However,the structure and function relationship within plant FMOs is not understood.Here we defined the important residues and domains of the Arabidopsis YUC1 FMO,a key enzyme in auxin biosynthesis.We previously showed that simultaneous inactivation of YUC1 and its homologue YUC4 caused severe defects in vascular and floral development.We mutagenized the yuc4 mutant and screened for mutants with phenotypes similar to those of yuc1 yuc4 double mutants.Among the isolated mutants,five of them contained mutations in the YUC1 gene.Interestingly,the mutations identified in the new yuc1 alleles were concentrated in the two GXGXXG motifs that are highly conserved among the plant FMOs.One such motif presumably binds to flavin adenine dinucleotide(FAD) cofactor and the other binds to nicotinamide adenine dinucleotide phosphate(NADPH).We also identified the Ser139 to Phe conversion in yuc1,a mutation that is located between the two nucleotide-binding sites.By analyzing a series of yuc1 mutants,we identified key residues and motifs essential for the functions of YUC1 FMO.Xianhui Hou Sainan Liu Florencia Pierri Xinhua Dai Li-Jia Qu Yunde Zhao 2011Journal of Integrative Plant Biology2011,53,1:4
3Functional conservation of the grapevine candidate gene INNER NO OUTER for ovule development and seed formation显示文摘Seedlessness represents a highly appreciated trait in table grapes.Based on an interesting case of seedless fruit production described in the crop species Annona squamosa,we focused on the Vitis vinifera INNER NO OUTER(INO)gene as a candidate.This gene encodes a transcription factor belonging to the YABBY family involved in the determination of abaxial identity in several organs.In Arabidopsis thaliana,this gene was shown to be essential for the formation and asymmetric growth of the ovule outer integument and its mutation leads to a phenotypic defect of ovules and failure in seed formation.In this study,we identified in silico the V.vinifera orthologue and investigated its phylogenetic relationship to INO genes from other species and its expression in different organs in seeded and seedless varieties.Applying cross-species complementation,we have tested its functionality in the Arabidopsis ino-1 mutant.We show that the V.vinifera INO successfully rescues the ovule outer integument growth and seeds set and also partially complements the outer integument asymmetric growth in the Arabidopsis mutant,differently from orthologues from other species.These data demonstrate that VviINO retains similar activity and protein targets in grapevine as in Arabidopsis.Potential implications for grapevine breeding are discussed.Valentina di Rienzo Zahra Imanifard Isabella Mascio Charles SGasser Debra JSkinner Ciro Leonardo Pierri Martina Marini Valentina Fanelli Wilma Sabetta Cinzia Montemurro Diana Bellin 2021Horticulture Research2021,8,1:2

18The transfer and resulting negative effects of nano-and micro-plastics through the aquatic trophic web—A discreet threat to human health显示文摘The ubiquitous occurrence of plastic nano-and micro-particle contamination has raised concerns about its potential risks and impacts on the global environment,especially in aquatic ecosystems.Hundreds of aquatic species,from different trophic levels,have been affected by this“new”contaminant,which has been reported mainly in their digestive tracts.Consequently,current knowledge about plastic nano-and micro-particle spread and the potential impact on aquatic biota is growing rapidly.However,there is a significant lack of understanding of the trophic spread of microplastic contamination and integration of knowledge derived from laboratory assays with that from field research is difficult.Field experiments are unable to deal with differentiating between directly and indirectly ingested plastic microparticles.On the other hand,laboratory assays evaluating the influence of plastic microparticles and of their adhered or constitutive toxins on representative species cannot satisfactorily simulate the real environment.As a result,little is known about the effective transfer of plastic particles through the trophic net and the resulting human health risks.The present review seeks to gather information that can give a more accurate idea of the current situation and future challenges to be faced in mitigating the environmental and human effects of plastic particle pollution in aquatic,particularly estuarine,ecosystems.Charles V.Neves Christine C.Gaylarde Jose Antonio Baptista Neto Khaue S.Vieira Bruno Pierri Carolina C.C.Waite Daniela C.Scott Estefan M.da Fonseca 2022Water Biology and Security2022,1,4:1
