| 2 | Hierarchical activation of compartmentalized pools of AMPK depends on severity of nutrient or energy stress显示文摘AMPK, a master regulator of metabolic homeostasis, is activated by both AMP-dependent and AMP-independent mechanisms. The conditions under which these different mechanisms operate, and their biological implications are unclear. Here, we show that, depending on the degree of elevation of cellular AMP, distinct compartmentalized pools of AMPK are activated, phosphorylating different sets of targets. Low glucose activates AMPK exclusively through the AMP-independent, AXIN-based pathway in lysosomes to phosphorylate targets such as ACC1 and SREBP1c, exerting early anti-anabolic and pro-catabolic roles. Moderate increases in AMP expand this to activate cytosolic AMPK also in an AXIN-dependent manner. In contrast, high concentrations of AMP, arising from severe nutrient stress, activate all pools of AMPK independently of AXIN. Surprisingly, mitochondrion-localized AMPK is activated to phosphorylate ACC2 and mitochondrial fission factor (MFF) only during severe nutrient stress. | 3 | An Abrupt Rainfall Decrease over the Asian Inland Plateau Region around 1999 and the Possible Underlying Mechanism显示文摘A decadal change in summer rainfall in the Asian inland plateau(AIP) region is identified around 1999. This decadal change is characterized by an abrupt decrease in summer rainfall of about 15.7% of the climatological average amount,leading to prolonged drought in the Asian inland plateau region. Both the surface air temperature and potential evapotranspiration in the AIP show a significant increase, while the soil moisture exhibits a decrease, after the late 1990s. Furthermore,the normalized difference vegetation index shows an apparent decreasing trend during 1999–2007. Three different drought indices—the standardized precipitation index, the standardized precipitation evapotranspiration index, and the self-calibrating Palmer drought severity index—present pronounced climate anomalies during 1999–2007, indicating dramatic drought exacerbation in the region after the late 1990s. This decadal change in the summer rainfall may be attributable to a wave-like teleconnection pattern from Western Europe to Asia. | 4 | Mapping of quantitative trait loci for fiber and lignin contents from an interspecific cross Oryza sativa×Oryza rufipogon显示文摘Rice straw is always regarded as a by-product of rice production,but it could be a significant energy source for ruminant animals.Knowledge of the genetic variation and genetic architecture of cell wall traits will facilitate rice breeders by improving relevant traits through selective breeding and genetic engineering.The common wild rice,Oryza rufipogon Griff.,which is considered to be the progenitor of Oryza sativa,has been widely utilized for the identification of genes of agronomic importance for rice genetic improvement.In the present study,the mapping of quantitative trait loci (QTLs) for acid detergent fiber (ADF),neutral detergent fiber (NDF),acid detergent lignin (ADL),and ADL/NDF ratio was carried out in two environments using a backcrossed inbred line (BIL) population derived from a cross between the recurrent parent Xieqingzao B (XB) and an accession of Dongxiang wild rice (DWR).The results indicated that all four traits tested were continuously distributed among the BILs,but many BILs showed transgressive segregation.A total of 16 QTLs were identified for the four traits,but no QTLs were in common in two environments,suggesting that environment has dramatic effects on fiber and lignin syntheses.Compared to the QTL positions for grain yield-related traits,there were no unfavorable correlations between grain yield components and cell wall traits in this population.The QTLs identified in this study are useful for the development of dual-purpose rice varieties that are high in grain yield and are also high in straw quality. | Jian-kun XIE Xiang-li KONG Jie CHEN Biao-lin HU Piao WEN Jie-yun ZHUANG Jin-song BAO | 2011 | Journal of Zhejiang University-Science B(Biomedicine & Biotechnology)2011,12,7: | 2 |
| 5 | Different copper sources and levels affect growth performance,copper content,carcass characteristics,intestinal microorganism and metabolism of finishing pigs显示文摘Copper(Cu)is an essential trace element in the production of swine.This study was conducted to investigate the effect of 3 different sources of Cu on growth performance,Cu metabolism,and intestinal microorganisms of finishing pigs,so as to estimate the bioavailability of the 3 sources for pigs.A total of 42 male finishing pigs(88.74±5.74 kg)were randomly allocated to 7 treatments.The factors were 3 sources(CuSO4,Cu-glycine,Cu-proteinate)and 2 levels(5 and 20 mg/kg)of Cu,plus one negative control treatment(0 mg/kg added Cu level)for the entire 28-d experiment.The average daily gain(ADG)and feed to gain ratio(F:G)both increased when Cu was added.The Cu level in liver,bile,kidney,serum,lung,urine and feces rose(P<0.001)with increasing dietary Cu level regardless of the source.Meanwhile,pigs receiving organic Cu(glycinate or proteinate)retained more Cu and excreted less Cu than those receiving inorganic Cu(CuSO4),which showed that organic forms were more bioavailable.At the transcriptional level,changes in the level and source of dietary Cu resulted in modulation of transporters.In the jejunal mucosa,import transporter high affinity copper uptake protein 1(CTR1)and export transporter ATPase copper transporting alpha(ATP7A)in supplemental Cu treatments were down-regulated compared to the control.Also,peptide transporter 1(PepT1)and lanine-serine-cysteine transporter,type-2(ASCT2)were significantly(P<0.01)up-regulated in 20 mg/kg Cu-proteinate and Cu-glycinate treatments,respectively.Microbial diversity was lowest in the 20 mg/kg CuSO4 treatment,and the ratio of Firmicutes to Bacteroidetes was higher in added Cu treatments,especially Cu-glycinate treatment.These results indicate that uptake of different Cu forms is facilitated by different transporters and transport mechanisms,and compared with inorganic Cu,organic Cu provides benefits to intestinal microflora and reduces Cu excretion. | Yang Wen Runxian Li Xiangshu Piao Gang Lin Pingli He | 2022 | Animal Nutrition2022,,1: | 2 |
| 6 | Anti-tumor Immune Response Mediated by Newcastle Disease Virus HN Gene显示文摘Hemagglutinin-neuramidinase(HN) is one of the most important surface structure proteins of the Newcastle disease virus(NDV). HN not only mediates receptor recognition but also possesses neuraminidase(NA) activity, which gives it the ability to cleave a component of those receptors, NAcneu. Previous studies have demonstrated that HN has interesting anti-neoplastic and immune-stimulating properties in mammalian species, including humans. To explore the application of the HN gene in cancer gene therapy, we constructed a Lewis lung carcinoma(LLC) solid tumor model using C57BL/6 mice. Mice were injected intratumorally with the recombinant adenovirus expressing HN gene(Ad-HN), and the effect of HN was explored by natural killer cell activity assay, cytotoxic lymphocyte activity assay, T cell subtype evaluation, and Th1/Th2 cytokines analysis. The results demonstrate that HN not only can elicit clonal expansion of both CD4+ and CD8+ T cell populations and cytotoxic T lymphocyte(CTL) and killer cell response, but also skews the immune response toward Th1. | 9 | YB1 regulates miR-205/200b-ZEB1 axis by inhibiting microRNA maturation in hepatocellular carcinoma显示文摘Background:Y-box binding protein 1(YB1 or YBX1)plays a critical role in tumorigenesis and cancer progression.However,whether YB1 affects malignant transformation by modulating non-codingRNAs remains largely unknown.This study aimed to investigate the relationship between YB1 and microRNAs and reveal the underlying mechanism by which YB1 impacts on tumor malignancy via miRNAs-mediated regulatory network.Methods:The biological functions of YB1 in hepatocellular carcinoma(HCC)cells were investigated by cell proliferation,wound healing,and transwell invasion assays.The miRNAs dysregulated by YB1 were screened by microarray analysis in HCC cell lines.The regulation of YB1 on miR-205 and miR-200b was determined by quantitative real-time PCR,dual-luciferase reporter assay,RNA immunoprecipitation,and pull-down assay.The relationships of YB1,DGCR8,Dicer,TUT4,and TUT1 were identified by pull-down and coimmunoprecipitation experiments.The cellular co-localization of YB1,DGCR8,and Dicer were detected by immunofluorescent staining.The in vivo effect of YB1 on tumor metastasis was determined by injecting MHCC97H cells transduced with YB1 shRNA or shControl via the tail vein in nude BALB/c mice.The expression levels of epithelial tomesenchymal transition markerswere detected by immunoblotting and immunohistochemistry assays.Results:YB1 promoted HCC cell migration and tumor metastasis by regulating miR-205/200b‒ZEB1 axis partially in a Snail-independent manner.YB1 suppressedmiR-205 and miR-200b maturation by interacting with the microprocessors DGCR8 and Dicer as well as TUT4 and TUT1 via the conserved cold shock domain.Subsequently,the downregulation of miR-205 and miR-200b enhanced ZEB1 expression,thus leading to increased cell migration and invasion.Furthermore,statistical analyses on gene expression data from HCC and normal liver tissues showed that YB1 expression was positively associated with ZEB1 expression and remarkably correlated with clinical prognosis.Conclusion:This study reveals a previously undescribed mechanism by which YB1 promotes cancer progression by regulating the miR-205/200b‒ZEB1 axis in HCC cells.Furthermore,these results highlight that YB1 may play biological functions via miRNAs-mediated gene regulation,and it can serve as a potential therapeutic target in human cancers. | Xiumei Liu Di Chen Huan Chen Wen Wang Yu Liu Yawei Wang Chao Duan Zhen Ning Xin Guo Wuxiyar Otkur Jing Liu Huan Qi Xiaolong Liu Aifu Lin Tian Xia Hong-xu Liu Hai-long Piao | 2021 | Cancer Communications2021,41,7: | 1 |

[Methods] CiteSpace knowledge graph analysis tool, VOSviewer visual analysis software and HistCite citation graph analysis tool were used to analyze the number of publications and subject distribution of duck-related research fields in the Web of Science core collection database. The main countries(regions) and institutions, the main academic journals, the main research scholars, the main research hotspots and their changing trends were used for quantitative analysis. [Results](1) Duck-related research is receiving more and more attention, which further supports ducks’ important significance in the livestock and poultry industry.(2) Countries such as China, the United States, Canada, and France had published more articles in this field, and there was close cooperation between countries.(3) The top two journals with local citations in the duck-related research fields were PLOS ONE and JOURNAL OF VIROLOGY, and the journal with the highest impact factor in 2019 was JOURNAL OF VIROLOGY.(4) High-yield scholars in duck related research fields included Cheng Anchun, Wang Mingshu, Jia Renyong, etc.(5) Infection was a research hotspot in duck-related research. | 17 | Evaluating correlation between the ocular biometry and genetic variants of MYOC and ABCA1 with primary angleclosure glaucoma in a cohort from northern China显示文摘AIM: To investigate whether the gene variants in MYOC and ABCA1 are associated with primary angle-closure glaucoma(PACG) and anterior chamber depth(ACD) and axial length(AL) in samples from northern China. METHODS: The present case-control association study consisted of 500 PACG patients and 720 unrelated controls. Each participant was genotyped for eleven single nucleotide polymorphisms(SNPs) in MYOC and ABCA1 genes(rs12076134, rs183532, rs235875 and rs235913 in MYOC, rs2422493, rs2487042, rs2472496, rs2472493, rs2487032, rs2472459 and rs2472519 near ABCA1) using an improved multiplex ligation detection reaction(iMLDR) technique. The genetic association analyses were performed by PLINK using a logistic regression model. The association between genotypes and ocular biometric parameters was performed by SPSS using generalized estimation equation. Bonferroni corrections wereimplemented and the statistical power was calculated by the Power and Sample Size Calculation. RESULTS: Two SNPs rs183532 and rs235875 as well as a haplotype TTC in MYOC were nominally associated with PACG despite the significance was lost after Bonferroni correction. No association was observed between ABCA1 and PACG, neither did the association between these variants and ACD as well as AL. CONCLUSION: The present study suggests MYOC and ABCA1 do not play a part in the pathogenesis of PACG as well as the regulation of ocular biometric parameters in a northern Chinese population. | 20 | Comprehensive understanding of DC pole-topole fault and its protection for modular multilevel converters显示文摘A modular multilevel converter(MMC)is one of the promising voltage source converter topologies in the field of high-voltage direct current(HVDC)transmission.MMCs are vulnerable to the direct current(DC)short-circuit faults due to the high discharge current stress.This study first introduced the simplified equivalent circuits of MMCs under both normal and DC fault operation conditions.Then the MMC fault spread loops and fault energy distribution principles are presented.Moreover,a detailed comparative study of the existing DC fault handling schemes,which are classified as the rigid blocking and flexible blocking methods are carried out.It provides a technical reference for further study on the MMC DC fault protection schemes for the HVDC transmission system. | Xiaofeng Yang Yao Xue Piao Wen Zejie Li | 2018 | High Voltage2018,3,4: | 0 |
