2Detection of soluble TRAIL in HBV infected patients and its clinical implications显示文摘AIM: To detect the expression of soluble TRAIL (TNF-related apoptosis inducing ligand, TRAIL) in the peripheral blood of HBV infected patients and try to elucidate whether the expression level of sTRAIL have any correlativity with the clinical staging, the expression level of HBV markers and the degree of liver damage.METHODS: 52 cases of HBV infected patients were investigated, induding 8 HBV carriers, 30 chronic hepatitis B, 11 drrhotics and 3 HBV infection related hepatocellular carcinoma. Expression of soluble TRAIL and markers of the hepatitis B were mearsured by enzyme-linked immunosorbent assay.RESULTS: The expression level of sTRAIL in the peripheral blood of the HBV infected patients was significantly higher than that of healthy controls (1378.35±540.23 pg/ml vs 613.75±175.80 pg/ml, P<0.001). In the group of chronic hepatitis, the expression level of sTRAIL was coincident with the status of the disease and was significantly correlated with the level of ALT. In the group of cirrhosis and liver cancer, its expression level was significantly higher than that of the healthy persons and HBV carriers, but lower than that of the hepatitis B patients; meanwhile, the expression of siRAIL did not have any correlativity with the functional indexes of the liver. CONCLUSION: The soluble TRAIL in the HBV infected people may participate in the liver damage. 3Hepatitis B virus X protein modulates the apoptosis of hepatoma cell line induced by TRAIL显示文摘The purpose of this study is to observe the effects of HBx on the apoptosis of hepatoma cells induced by TNF-related apoptosis-inducing ligand (TRAIL) and to study prelimi- nary molecular mechanisms for its effects. In order to set up a model in vitro, BEL7402-HBx cell line, stably expressing HBx mRNA, was established by stable transfection of pcDNA-HBx, which contains HBx gene, into hepatoma cell line BEL7402. Control cell line BEL7402-cDNA3, stably transfected with pcDNA3, was set up simultaneously as a control. Trypan blue exclusion test, caspase 3 activity detection and TUNEL assay were performed to detect the apoptosis of BEL7402, BEL7402-cDNA3, BEL7402-HBx induced by TRAIL. The expression of TRAIL recep- tors in three groups was analyzed by Flow cytometry. In addition, phosphorothioated antisense oligonucleotide against the translation initial region of HBx gene (PS-asODNs/HBx) was used to block the expression of HBx in HepG2.2.15 cells and to further confirm the effects of HBx on TRAIL-induced apoptosis. Trypan blue exclusion test indicated that TRAIL had a dose-dependent cytotoxicity on BEL7402, BEL7402-cDNA3 and BEL7402-HBx cells. Under treatment of the same concentration of TRAIL, BEL7402-HBx had a higher apoptosis rate and a higher level of Caspase 3 activation than BEL7402 and BEL7402-cDNA3. TUENL assay showed that the apoptosis rate of BEL7402-HBx induced by 10 μg/L TRAIL was 41.4%±7.2%, signifi- cantly higher than that of BEL7402 and BEL7402-cDNA3 cells. Blockade of HBx expression in Hep G2.2.15 cells partly inhibited the apoptosis induced by TRAIL. The introduction or blockade of HBx did not change the expression pattern of TRAIL receptors. The present study firstly con- firms the effects of HBx on TRAIL- induced apoptosis from two different points and it is not re- lated with the expression level of TRAIL receptors. 5SYNTHESIS OF MESOPOROUS POLY(STYRENE-co-MALEIC ANHYDRIDE)/SILICA HYBRID MATERIALS VIA A NONSURFACTANT-TEMPLATED SOL-GEL PROCESS显示文摘Mesoporous poly (styrene-co-maleic anhydride)/silica hybrid materials have been prepared. The synthesis wasachieved by the HCl-catalyzed sol-gel reactions of tetraethyl orthosilicate (TEOS) and styrene-maleic anhydride copolymerin the presence of 3-aminopropyl triethoxysilane (APTES) as a coupling agent and citric acid as a nonsurfactant template orpore-forming agent, followed by ethanol extraction. Characterization results from nitrogen sorption isotherms and powder X-ray diffraction indicate that polymer-modified mesoporous materials with large specific surface areas (e.g. 900 m^2/g) andpore volumes (e.g. 0.6 cm^3/g) could be prepared. 7Twinning in Intermetallic Compounds Are Long Shear Vectors and/or Shuffles Really Necessary?显示文摘In this paper the geometric description and general theory of mechanical twinning are reviewed, the twins in general lattices and superlattices are summarized, and the kinetic process by which mechanical twins form is revisited. A case study of mechanical twinning of HfV2+Nb, (cubic) Laves phase, is presented and the synchroshear of selected atomic layers is proposed to explain the physical process of twin formation. 9SYNTHESIS AND BIOTECHNOLOGICAL APPLICATIONS OF VINYL POLYMERINORGANIC HYBRID AND MESOPOROUS MATERIALS显示文摘We describe the sol-gel synthesis of a new family of organic-inorganic hybrid materials, in which various vinylpolymers are covalently bonded to and uniformly distributed in inorganic oxide matrices. The materials can be tailored tohave both good toughness and hardness while maintaining excellent optical transparency. Doping the sol-gel metal oxideswith optically active compounds such as D-glucose results in new optical rotatory composite materials. Removal of thedopant compounds from the composites affords mesoporous oxide materials, which represents a new, nonsurfactant-templated route to mesoporous molecular sieves. We have successfully immobilized a series of enzymes and other bioactiveagents in mesoporous materials. 10Protective Effects of Dimethyl-4,4'-Dimethoxy-5,6,5',6'-Dimethylene Dioxybiphenyl-2,2'-Dicarboxylate on Damages of Isolated Rat Hepatocytes Induced by Carbon Tetrachloride and D-galactosamine显示文摘The rotective effect of biphenyl dimethyl dicarboxylate(DDB)on chemically induced damages was studied in isolated suspended rat hepatocytes.The experimental results showed that DDB(200μg/10^6cells) efficiently protected the hepatocytes against carbon tetrachloride(CCl4 10mmol·L^-1)and D-galactosamine(1mmol·L^1)induce damages.Membranal lipid peroxidation(malondialdehyde,MDA formation(and glutamic pyruvic transaminase(GPT) release from the hepatocytes were markedly decreased.The damage of the cell surfaces of the hepatocytes were also reduced as seen under a scanning electrom microscope(SEM). 19Snail Hosts of Paragonimus in Asia and the Americas显示文摘We have undertaken a comprehensive review of snail hosts of Paragonimus world-wide exclusive of Africa based on modern malacological data, where available, and with consideration of the phylogeny of the snail groups involved. This is the first comprehensive review since those made by Chen (1979) and Chen et al (1983), and there have been considerable taxonomic changes over the past decade. A number of names and concepts found in the medical malacological and parasitological literature up to the present time require revision or correction. There are vast radiations of snails of the superfamilies Cerithiacea and Rissoacea involved in the transmission of Paragonimus in China. We list 54 species world-wide of which 35(65%) occur in China. Revisions and corrections pertaining to China include: (i) The family Hydrobiidae does not occur in China or S.E. Asia,and the Pomatiopsidae should be used. (ii) The genus Bythinella is entirely European (Hydrobiidae: Amnicolinae). The so-called Bythinella of China belongs to the genus Erhaia (Pomatiopsidae). (iii) The generic name Pseudobythinella described from China is preoccupied, a junior synonym of Pseudobythinella Melville 1956, a fossil from England. All Chinese Pseudobythinella are now classified as Erhaia. (iv) Akiyoshia has been used as a generic name for some snails in Hunan transmitting Paragonimus. Akiyoshia is from Japan and biological/ecological data indicate that the Chinese taxon is not Akiyoshia. (v)The genus Tricula in recent Chinese literature is comprised of four genera determined by detailed comparative anatomical data: Tricula, Neotricula, Gammatricula, and Jinhongia.Shells cannot be used to discriminate among them. (vi) Tricula cristella has been consistently misidentified in collections in China and literature. However, genuine T cristella does transmit Paragonimus skrjabini. (vii) Tricula minutoides in the Chinese literature has been misidentified, and specimens are T. crisIella. (viii) The genus Melamia and the family Melaniidae are used in the Chinese literature. The so-called melaniid snails belong to the families Thiaridae, Pleuroceridae, and Melanopsidae. Asian Semisulcospira is pleurocerid while Melanoides and Tarebia are thiarid. (ix) Semisulcospira libertina is correctly identified for populations ranging throughout Japan to Taiwan. This species is most probably not distributed throughout all south China, and intense systematic research on Semisulcospira is needed. A modern classification is given for snails transmittingG. M.
