1海藻糖快速检测方法的建立与初步应用显示文摘海藻糖是生物体合成的、具有抗逆保护作用的天然二糖,在食品加工与贮藏、微生物发酵过程具有重要作用。该文采用前期获得的专一性海藻糖酶,建立了一种定量测定海藻糖的快速方法。该方法对海藻糖的检测限为0.019 mg/m L,定量限为0.1 mg/m L;海藻糖的线性检测范围为0.1 mg/m L^150.0 mg/m L,回收率为98.80%~101.33%。该方法操作简便快捷、特异性高、重复性好,在菌种改良与食品原料营养筛查等海藻糖快速定量检测中具有良好的应用前景。苗佳 王彩喆 牛丹丹 Nokuthula Peace Mchunu 田康明 Suren Singh Kugenthiren Permaul 王正祥 2019食品与发酵工业2019,45,23:5
2疏绵状嗜热丝孢菌外切β-葡聚糖酶的基因克隆与酶学特征显示文摘【目的】疏绵状嗜热丝孢菌是一种嗜热丝状真菌,具有合成与分泌多种耐热酶的能力,从中找寻酶活高、耐热性能优良的β-葡聚糖酶。【方法】对疏绵状嗜热丝孢菌其中一个外切β-葡聚糖酶的编码基因gln B进行克隆,并在毕赤酵母GS115中表达。【结果】在摇瓶水平上重组菌的产酶活为11.5 U/m L,重组酶在SDS-PAGE中的大小约为48 k D。重组Gln B最适作用温度为65°C,最适作用p H为5.0;在低于50°C或p H 3.0-10.0之间具有良好稳定性。重组酶水解海带三糖,首先生成单糖和二糖,延长酶作用时间,可以进一步将其中的二糖部分水解为单糖。【结论】疏绵状嗜热丝孢菌来源的重组酶Gln B为外切β-葡聚糖酶,具有较好的耐热性能和p H稳定性。王瑞杰 Nokuthula Peace Mchunu 牛丹丹 Kugenthiren Permaul Suren Singh 刘晓光 路福平 2016微生物学通报2016,43,2:3
