| 1 | nov基因的表达与脑的种系发育分化平行相关显示文摘用地高辛标记的cRNA探针原位杂交组织化学方法研究了鲢、鸡、牛、犬和猫脑肾母细胞瘤过度表达基因阳性神经元的比较发育。结果显示 ,低等脊椎动物鲢脑仅有少量novmRNA阳性神经元 ,分布于延髓的迷走神经背核、三叉神经背核、丘脑前核、丘脑后核、下丘脑背侧核、下丘脑外侧核。鸡脑阳性神经元除广泛分布于脑干外 ,小脑、丘脑的多数核团、下丘脑的背侧核、腹侧核和后核以及大脑纹状体等多数脑区也有一定分布。哺乳动物牛、犬和猫脑中novmRNA阳性神经元广泛分布 ,在脑干、小脑、间脑和大脑都检测到强烈阳性信号。以上结果表明 ,在从低等脊椎动物到高等脊椎动物的进化过程中nov基因的表达也从低级中枢向高级中枢扩展 ,提示该基因的表达与脑的种系发育、脑功能的分化平行相关。 | 苏炳银 蔡文琴 张成岗 Bernard Perbal | 2001 | 动物学报2001，47，6： | 3 |
| 2 | 反义nov基因对培养神经元神经递质分泌的影响显示文摘目的 研究 nov基因在神经元功能分化和神经递质分泌中的作用。 方法 利用反义核酸技术构建成反义 nov基因真核表达载体 p BK- nov。采用脂质体转染技术 ,将反义表达载体导入培养的大鼠大脑皮质神经元和纹状体神经元 ,经 G418筛选出被转染的神经元继续培养。RT- PCR检测转染后 nov m RNA的表达量。采用高效氨基酸分析仪和高效液相色谱仪 ,检测抑制 nov基因表达后氨基酸类递质和多巴胺类递质的变化。 结果 谷氨酸和肾上腺素的分泌量明显下降 ,γ-氨基丁酸上升 ,而对多巴胺的分泌没有显著的影响。 结论 以上结果提示 ,nov基因在中枢神经系统神经递质的生成释放和代谢中起作用 ,可能籍此调节神经系统的功能。 | 苏炳银 蔡文琴 黄其林 张成岗 Perbal B | 2001 | 解剖学报2001，32，2： | 2 |
