2Prediction of coronavirus 3C-like protease cleavage sites using machine-learning algorithms显示文摘The coronavirus 3C-like(3CL)protease,a cysteine protease,plays an important role in viral infection and immune escape.However,there is still a lack of effective tools for determining the cleavage sites of the 3CL protease.This study systematically investigated the diversity of the cleavage sites of the coronavirus 3CL protease on the viral polyprotein,and found that the cleavage motif were highly conserved for viruses in the genera of Alphacoronavirus,Betacoronavirus and Gammacoronavirus.Strong residue preferences were observed at the neighboring positions of the cleavage sites.A random forest(RF)model was built to predict the cleavage sites of the coronavirus 3CL protease based on the representation of residues in cleavage motifs by amino acid indexes,and the model achieved an AUC of 0.96 in cross-validations.The RF model was further tested on an independent test dataset which were composed of cleavage sites on 99 proteins from multiple coronavirus hosts.It achieved an AUC of 0.95 and predicted correctly 80%of the cleavage sites.Then,1,352 human proteins were predicted to be cleaved by the 3CL protease by the RF model.These proteins were enriched in several GO terms related to the cytoskeleton,such as the microtubule,actin and tubulin.Finally,a webserver named 3CLP was built to predict the cleavage sites of the coronavirus 3CL protease based on the RF model.Overall,the study provides an effective tool for identifying cleavage sites of the 3CL protease and provides insights into the molecular mechanism underlying the pathogenicity of coronaviruses.Huiting Chen Zhaozhong Zhu Ye Qiu Xingyi Ge Heping Zheng Yousong Peng 2022Virologica Sinica2022,37,3:1
3Risk factors and clinical outcomes of incomplete endoscopic resection of small rectal neuroendocrine tumors in southern China:a 9-year data analysis显示文摘Background:The histologically complete resection(CR)rate of small rectal neuroendocrine tumors(RNETs)is unsatisfactory at the first endoscopy.Risk factors and clinical outcomes associated with incomplete resection(IR)have not been explicitly elucidated.This study aims to explore the relevant factors of IR.Methods:This retrospective study reviewed patients with small RNETs(10mm)in eight centers from January 2013 to December 2021.Clinicopathological characteristics and clinical outcomes were compared between the CR and IR groups,and the polypectomy and advanced treatment groups.Results:Of the 326 patients included,83(25.5%)were diagnosed with IR.Polypectomy(odds ratio[OR]=16.86),a central depression(OR=7.50),and treatment in the early period(OR=2.60)were closely associated with IR.Further analysis revealed that an atypical hyperemic appearance(OR=7.49)and treatment in the early period(OR=2.54)were significantly associated with the inappropriate use of polypectomy(both P<0.05).In addition,a total of 265(81.3%)were followed up with a median follow-up period of 30.9 months.No death,metastasis,or recurrence was found during the follow-up period.Conclusions:Polypectomy,a central depression,and treatment in the early period were risk factors for IR.Further,an atypical hyperemic appearance and treatment in the early period were significant predisposing factors for inappropriate choice of polypectomy.For histologically incompletely resected small RNETs,follow-up may be a safe and feasible alternative to rigorous salvage therapy.Xiaoduan Zhuang Shaoheng Zhang Guiquan Chen Zongqi Luo Huiqin Hu Wenfeng Huang Yu Guo Yongwen Ouyang Liang Peng Qing Qing Huiting Chen Bingsheng Li Jie Chen Xinying Wang 2023Gastroenterology Report2023,11,1:0

5Effectiveness and safety of red yeast rice predominated by monacolin Kβ-hydroxy acid form for hyperlipidemia treatment and management显示文摘OBJECTIVE:To assess the lipid-lowering activity and safety of a dietary supplement containing monacolin Kβ-hydroxy acid form(MKA),Heye(Folium Nelumbinis),and Cangzhu(Rhizoma Atractylodis Lanceae),compared to lifestyle modifications.METHODS:Totally 117 subjects with moderate to severe dyslipidemia(according to Chinese guidelines)and low CV risk were randomly assigned into three treatment groups:lifestyle modification(LM),LM plus a low dosage of MKA,LM plus a high dosage of MKA,and treated for 60 d.The primary endpoint was the reduction of low density lipoprotein cholesterol(LDL-C)and total cholesterol(TC).Safeties along with Traditional Chinese Medicine Syndromes were assessed through the study.RESULTS:A low dosage of MKA along with lifestyle modifications caused a significant decrease in LDL-C by 15.6%on average(95%CI,9.6%to 21%)with,a decrease in TC by 15.3%on average(95%CI,9.26%to 21.4%),and a decrease in non-HDL-C by 35.4%(95%CI,25.76%to 41.34%).Weak evidence of a reduction of triglycerides but an increment of HDL-C was observed in patients with severe hyperlipidemia.No severe adverse events occurred during the study.CONCLUSION:Our results confirm the LDL-C and TC lowering properties of MKA is clinically meaningful.It also produces a significant reduction of non-HDL-C,and slightly effects on TG and HDL-C as well.CHEN Benjian HUANG Xiaodan PENG Huiting LI Yishi CAO Yongtao WU Huanlin XU Danping 2022Journal of Traditional Chinese Medicine2022,42,2:0
6Role of melatonin receptor 1B gene polymorphism and its effect on the regulation of glucose transport in gestational diabetes mellitus显示文摘Melatonin receptor 1B(MT2,encoded by the MTNR1B gene),a high-affinity receptor for melatonin,is associated with glucose homeostasis including glucose uptake and transport.The rs10830963 variant in the MTNR1B gene is linked to glucose metabolism disorders including gestational diabetes mellitus(GDM);however,the relationship between MT2-mediated melatonin signaling and a high birth weight of GDM infants from maternal glucose abnormality remains poorly understood.This article aims to investigate the relationship between rs10830963 variants and GDM development,as well as the effects of MT2 receptor on glucose uptake and transport in trophoblasts.TaqMan-MGB(minor groove binder)probe quantitative realtime polymerase chain reaction(qPCR)assays were used for rs10930963 genotyping.MT2 expression in the placenta of GDM and normal pregnant women was detected by immunofluorescence,western blot,and qPCR.The relationship between MT2 and glucose transporters(GLUTs)or peroxisome proliferator-activated receptorγ(PPARγ)was established by western blot,and glucose consumption of trophoblasts was measured by a glucose assay kit.The results showed that the genotype and allele frequencies of rs10830963 were significantly different between GDM and normal pregnant women(P<0.05).The fasting,1-h and 2-h plasma glucose levels of G-allele carriers were significantly higher than those of C-allele carriers(P<0.05).Besides,the protein and messenger RNA(mRNA)expression of MT2 in the placenta of GDM was significantly higher than that of normal pregnant women(P<0.05).Melatonin could stimulate glucose uptake and GLUT4 and PPARγprotein expression in trophoblasts,which could be attenuated by MT2 receptor knockdown.In conclusion,the rs10830963 variant was associated with an increased risk of GDM.The MT2 receptor is essential for melatonin to raise glucose uptake and transport,which may be mediated by PPARγ.Lijie WEI Yi JIANG Peng GAO Jingyi ZHANG Xuan ZHOU Shenglan ZHU Yuting CHEN Huiting ZHANG Yuanyuan DU Chenyun FANG Jiaqi LI Xuan GAO Mengzhou HE Shaoshuai WANG Ling FENG Jun YU 2023Journal of Zhejiang University-Science B(Biomedicine & Biotechnology)2023,24,1:0
